Abstract:Aromatic aldehydes are very important organic intermediates,which are widely used in pharmaceuticals,pesticides and other areas.In this paper,an improved synthetic method of aromatic aldehydes containing electrowithdrawing groups in ortho or para(or both) position of the aromatic ring was introduced.4-choloro-2-nitrotoluene,4-nitrotoluene and 4-cyanotoluene were used as raw materials,respectively,via enamination with N,N-dimethylformamide dimethyl acetal(DMFDMA) in the present of N,N-dimethylformamide(DMF) as solvent and under reflux,the corresponding styrylamine compounds were produced;then the intermediates were further oxidized by H2O2 under different reaction conditions.The effects of solvent species,reaction temperature and the used amount of H2O2 on the product yield were studied,and the optimal reaction conditions were determined.Under those optimal conditions,the best yields of different aldehydes are 75.6%(4-chloro-2-nitrobenzaldehyde),58.3%(4-nitrobenzaldehyde) and 68.2%(4-cyanobenzaldehyde),respectively with the advantages of simple process,low cost and being environment-friendly..The main intermediates and objective products were confirmed by HPLC and 1H-NMR.
Synthesis and in vitro Antibacterial Activities of Some Bis-Oxazolidinones

Abstract:Aryl diamines were condensed with (R)-glycidyl butyrate to form (R,R)-l,4-bis-[(2-hydroxyl-3-butyryloxy)-propylamino] benzene derivatives (3a and 3b), which were cyclized with carbonyldiimidazole to get the bis-oxazolidinones(4a and 4b), the latter were hydrolyzed into (R,R)-l,4-bis-(5-hydroxymethyl-2-oxo-oxazolidin-3-yl)benzene derivatives (5a and 5b). Compounds 5a and 5b were converted via 6a, 6b and 7a, 76 to (S,S)-1,4-bis-(5-acetamidomethyl-2-oxo-oxazolidinyl-3)benzene derivatives (8a and 8b). The structures of these new compounds were confirmed by HNMR and MS. The in vitro antibacterial activities on 20 bacteria were tested and the result showed that compd 5b had better activity than linezolid against Staphylococcus epidermidis, Staphylococcus enteridis and Staphylococcus nonhemolyticus.
Synthesis and antibacterial activities of dihydrothiazole derivatives

Abstract:Seventeen novel dihydrothiazole derivatives were synthesized from compounds A and A'in one step. The structures of the compounds were characterized by IR, 1H-NMR, 13C-NMR,and MS analysis.The bioas-say showed that some of the synthesized dihydrothiazole derivatives exhibited excellent antibacterial activities. Especially,compounds 5,5'and 6 exhibited much higher activities against bacillus subtilis and staphylococcus aureus than penicillin.

Abstract:A high pharmacological ability of aromatic benzaldehydes makes them important intermediates for the synthesis of medicinal preparations, such as anticancer, bactericidal, antifungal, and herbicidal drugs. The purpose of this work is the synthesis of biologically active compounds, based on 4-hydroxybenzaldehyde and 4-hydroxy-3-methoxybenzaldehyde and the establishment of the structure of the synthesized compounds. Results and discussion. New carbonodithioates, based on O-aromatic systems have been synthesized by the interaction of 4-hydroxybenzaldehyde and 4-hydroxy-3-methoxybenzaldehyde with carbon disulfide in the presence of sodium hydroxide in ethanol at the room temperature. As a result of the reactions, sodium O-(4-formylphenyl)carbodithioate (86 %) and sodium O-(4-formyl-2-methoxyphenyl)carbodithioate (80%) have been isolated. The interaction of sodium xanthates with acid chlorides (4-methoxy-, 4-nitro-, 2,4-dinitrobenzoic) in chloroform has led to the formation of aromatic thioanhydrides of carbonodithioic acids in 55-80 % yields. The reactivity of hydroxybenzaldehydes and their dithiocarboxylic derivatives has been studied in the propargylation reaction. Propargylation of 4-hydroxybenzaldehyde and 4-hydroxy-3-methoxybenzaldehyde has been carried out with propargyl bromide in the presence of a 3-fold excess of K2CO3 in acetone at the temperature of 60°C. The propargylation reaction of sodium xanthate has been carried out with propargyl bromide in acetone at the room temperature. Conclusion. As a result of the reactions, carbonodithioates, thioanhydrides, acetylenic and thioacetylenic ethers have been synthesized based, on O-aromatic systems (4-hydroxybenzaldehyde and 4-hydroxy-3-methoxybenzaldehyde). The structure of the synthesized compounds has been established on the basis of elemental analysis data, IR spectra, 1Н and 13С NMR spectroscopy.

Abstract:Eighteen novel 2-(1-aryl-5-methyl-1,2,3-triazol-4-yl)-1,3,4-oxadiazole derivatives and two acylhydrazone intermediate compounds were synthesized by various pathways starting from I -aryl-5-methyl-1,2,3-triazol-4-formhydrazide (1). All products were identified by spectroscopic analysis, and 2-(l -aryl-5-methyl- 1,2,3-triazol-4-yl)-5-benzalthio-1,3,4-oxadiazole was further validated by X-ray crystallography. Results from primary antibacterial activity tests indicated that most of the compounds were effective against E coli, P. aeruginosa, B, subtilis and S. aureus.

Abstract:A series of halogenated 2-arylimino-3-ary-thiazolidine-4-ones containing (5-furan-2-yl)-benzoic acid or (5-thiophene-2-yl)-benzoic acid fragments was synthesized, and evaluated in vitro for their antibacterial activity, antibiofilm activity, erythrocyte hemolysis and cytotoxicity. Compounds (7c, 7f, 7i, 7n and 7p) exhibited excellent potency in inhibiting the growth of Staphylococcus epidermidis ATCC35984 (minimal inhibitory concentration (MIC): 0.8 mu g/mL). The five compounds also presented promising antibiofilm activity against S. epidermidis ATCC35984 and none of them showed obvious hemolytic activity and cytotoxicity. Further antibacterial effects of the selected five compounds (7c, 7f, 7i, 7n and 7p) against clinical isolates (methicillin-resistant Staphylococcus epidermidis and methicillin-resistant Staphylococcus aureus) were investigated in comparison with methicillin and ampicillin. 7p showed the most potent antibacterial activities among the synthesized compounds.

Abstract:In this work, twenty hydrazide-hydrazone and 4-thiazolidinone derivatives were synthesized starting from m-cresol. Antimicrobial evaluation was carried out by microdilution method against Enterococcus faecalis and Staphylococcus aureus as Gram-positive bacteria and Escherichia coli and Pseudomonas aeruginosa as Gram-negative bacteria, and three pathogenic fungi Candida albicans , Candida parapsilosis and Candida krusei . Some compounds possessed considerable antimicrobial properties against the tested microorganisms, particularly against E. coli . 4-Thiazolidinones containing 3-methoxyphenyl and 3,5-dichlorophenyl moieties ( 4h and 4i ) were found to be the most active derivatives with MICs of 2 μg/mL against E. coli . N '-[(3,5-dichlorophenyl)methylidene]-2-(3-methylphenoxy)acetohydrazide ( 3i ) also displayed antifungal activity against Candida krusei that was comparable to fluconazole. Calculated drug-likeness and ADMET parameters of the most active compounds confirmed their potential as antimicrobial drug candidates. Molecular docking investigations were carried out in the thiamine diphosphate-binding site of pyruvate dehydrogenase multienzyme complex E1 component (PDHc-E1) to clarify the potential antibacterial mechanism against E. coli. Abstract:A series of 4‐azido‐N'‐(benzylidene)benzohydrazides have been synthesized from amino benzoic acid in excellent yields via a four steps procedure. The products have been fully characterized and their antimicrobial activity evaluated by disc diffusion procedure. Benzohydrazide analogues are interesting compounds in the field of pharmaceutical, medicinal chemistry, and coordination chemistry, since they are diverse in biological activities, and they are contributing in medicines and drugs. Abstract:Antimicrobial potential of 3-hydroxy-2-methylene-3-phenylpropionic acid derivatives Twenty Baylis-Hillman adducts were synthesized from different aromatic aldehydes and activated vinyl derivatives. The adducts, which are differently substituted 3-hydroxy-2-methylene-3-phenylpropionic acid derivatives, were screened for their antimicrobial activity in vitro by the serial dilution method. Many of these molecules displayed potent antibacterial and antifungal activities. The ease of synthesis from low-cost starting materials along with potent antimicrobial activity of these molecules provide the lead for further improvement of activity and reflect the possibility of therapeutic use.

Abstract:Hydrazides are very important precursors for the synthesis of a wide variety of derivatives, including heterocyclic compounds of biological interest in drug design and discovery. Acid hydrazides with a pyrrole, thiophene, furan, quinoline, isoquinoline, isoxazole and benzimidazole core in the structure were found to be used in medicine, pharmacy, agriculture and many other fields. In the search for compounds with antibacterial properties, mono- and dihydrazides were used in this study to prepare 1-(4-substituted phenyl)-5-oxopyr-rolidine-3-carbohydrazide derivatives bearing the corresponding hydrazone and azole substituents. The formation of the target compounds was performed via condensation reactions with the chosen aromatic aldehydes and isatin, which led to the construction of the appropriate hydrazone-type structures as well as with diketones which afforded compounds with pyrazole and pyrrole nuclei. In addition, a preliminary antibacterial evaluation of the synthesised compounds was performed using the gram-positive Bacillus subtilis and the gramnegative Escherichia coli bacterial strains. As it was expected, the evaluation revealed potential antibacterial candidates that can help address the global challenges of antibiotic resistance and infectious disease outbreaks.

Abstract:A series of novel 2-(substituted-2-benzylidenehydrazinyl)-3-chloropyridine derivatives were designed and synthesized from 2,3-dichloropyridine as starting materials, and their structures were confirmed by H-1 NMR, MS and elemental analysis. The antifungal activity of title compounds was determined. The antifungal assay showed that some of title compounds displayed excellent activity, especially against Stemphylium lycopersici (Enjoji) Yamamoto and Fusarium oxysporum. sp. cucumebrium.

Abstract:2,4-Dimethylbenzoylhydrazones 1-30 were synthesized by condensation reactions of 2,4-dimethylbenzoylhydrazide with various aromatic aldehydes and characterized. The assigned structures of compounds 10, 15 and 22 were further supported by single-crystal X-ray diffraction data. The synthesized compounds were evaluated for their in vitro DPPH radical scavenging activity. They exerted varying degree of scavenging activity toward DPPH radical with IC₅₀ values between 25.6-190 µM. Compounds 1, 4, 2, 3, 7, and 6 have IC₅₀ values of 25.6, 28.1, 29.3, 29.8, 30.0 and 30.1 µM respectively, showing better activity than an n-propyl gallate standard (IC₅₀ value = 30.30 µM). For super oxide anion scavenging activity compounds 1, 2 and 3 with IC₅₀ values of 98.3, 102.6, and 105.6, respectively, also showed better activity than the n-propyl gallate standard (IC₅₀ value = 106.34 µM).

Abstract:The synthesis and biological assessment of 2,5-disubstituted-1,3,4-oxadiazole derivatives from benzo hydrazide and amino acids as novel potential antioxidant and antibacterial agents have been reported. The structures of the new compounds were characterized by physicochemical properties and spectral methods. The synthesized compounds were screened for their in vitro antibacterial activity against three Gram-positive bacterial strains, namely Staphylococcus aureus ATCC 25923, Bacillus cereus ATCC 14579, Listeria innocua ATCC 33090, and two Gram-negative bacterial strains, namely Pseudomonas aeruginosa ATCC 27853, Escherichia coli ATCC 25922, and antifungal activity against Candida albicans ATCC 10231 in comparison with Amoxicillin, Tetracycline, Gentamicin and Oxacillin as standards. Most of the compounds have excellent efficacy, and some of them, such as 5i , 5g , 5d , 5c , and 5j can inhibit their activity better or very close to that of Amoxicillin, Tetracycline, Gentamicin, Oxacillin used as standards. Compounds 5b and 5i provided good results against L. innocua with inhibition values of IZ = 14 mm and IZ = 22 mm, respectively, while the rest of the compounds and antibiotics were unable to inhibit it. Compounds 5c , 5d , 5g and 5j showed excellent activity against C. albicans with values between 31 mm and 34 mm. These results were better than all the standards used. The MIC value (25 μg/ml) for derivatives 5(c-e) , 5g and 5(i-j) against B. cereus represent the best activity of the tested compounds. All the target compounds were also screened for radical scavenging antioxidant activities by DPPH, FRAP, and TAC assays and found to be excellent antioxidant agents. According to the results, it was observed that most derivatives synthesized showed excellent activity with a concentration of 250 μg/ml as an antioxidant agent (76 % < RSA < 95.5 %) which gave an inhibition percentage that was better or very close to that of ascorbic acid and better than BHT.

Abstract:Nowadays, searching for novel antimicrobial agents is crucial due to the increasing number of resistant bacterial strains. Moreover, cancer therapy is a major challenge for modern medicine. Currently used cytostatics have a large number of side effects and insufficient therapeutic effects. Due to the above-mentioned facts, we undertook research to synthesize novel compounds from the acylhydrazone group aimed at obtaining potential antimicrobial and anticancer agents. As a starting material, we employed hydrazides of 2-, 3- or 4-iodobenzoic acid, which gave three series of acylhydrazones in the condensation reaction with various aldehydes. The chemical structure of all obtained compounds was confirmed by IR, 1H NMR, and 13C NMR. The structure of selected compounds was determined by single-crystal X-ray diffraction analysis. Additionally, all samples were characterized using powder X-ray diffraction. The other issue in this research was to examine the possibility of the solvent-free synthesis of compounds using mechanochemical methods. The biological screening results revealed that some of the newly synthesized compounds indicated a beneficial antimicrobial effect even against MRSA-the methicillin-resistant Staphylococcus aureus ATCC 43300 strain. In many cases, the antibacterial activity of synthesized acylhydrazones was equal to or better than that of commercially available antibacterial agents that were used as reference substances in this research. Significantly, the tested compounds do not show toxicity to normal cell lines either.

Abstract:Ester 2 was produced by reacting thiourea derivative 1 with ethyl 2-chloro-3-oxobutanoate in MeOH containing piperidine. Hydrazide 3 was produced by reacting the latter ester with hydrazine hydrate in EtOH at reflux. By reacting hydrazide 3 with aromatic/heterocyclic aldehydes, twelve derivatives of hydrazide hydrazone 5a–l were produced. Spectral measurements and elemental analysis verified the molecular structure. Compounds 2, 5a, 5c, 5d, and 5f had strong effects on all the pathogenic bacterial strains according to an evaluation of the antimicrobial qualities of the synthetic compounds. With inhibitory zone diameters ranging from 16 to 20.4 mm, hydrazide hydrazone 5f exhibited the strongest activity. Additionally, the minimum inhibitory concentration (MIC) was assessed. The best outcomes were found with hydrazones 5c and 5f. For B. subtilis, the MIC of 5c was 2.5 mg/mL. For E. coli and K. pneumoniae, the MIC of 5f was 2.5 mg/mL. The molecular mechanics-generalized born surface area (MM/GBSA) was utilized to compute binding free energies via a molecular dynamics simulation analysis of the most active compounds, 5f and 5c. Moreover, computational analyses demonstrated that 5f had a substantial affinity for the active site of DNA gyrase B, suggesting that this compound could be a strong platform for new structure-based design efforts.

Abstract:Novel benzimidazoles, benzothiazoles, and benzoxazoles linked to N -arylacetamide were synthesized in good yields by reacting a series of 2-oxo-2-(arylamino)ethyl 4-formylbenzoates with o -phenylenediamine, 2-aminothiophenol, or 2-aminophenol in ethanol at reflux in the presence of NaHSO 3 . Attempts to use the new aldehydes as adaptable precursors for the synthesis of fused dihydropyran or fused dihydropyridine via Michael or Hantzsch's reactions were unsuccessful; instead, the processes produced the corresponding arylidene derivatives. The structures of the novel compounds were verified with a variety of spectra. All synthesized compounds were evaluated for antibacterial activity against four different bacterial strains. Compound 6a had good activity against Escherichia coli in an agar diffusion assay, with an inhibitory zone width of 30 mm compared to ofloxacin (20 mm). Compounds 18a and 18b were most effective against Bacillus subtilis , with MICs of 156.3 and 312.5 μg/mL, respectively. Molecular docking investigations demonstrated that compounds 6a, 18a , and 18b had high binding affinities for bacterial tyrosyl-tRNA synthetase and DNA gyrase.

Abstract:A series of novel 1-(acetyl,carboxamide,carbothioamide)substituted-5-(substituted-phenyl)-3-methy-4,5-dihydropyrazole derivatives have been synthesized and characterized by 1H NMR, IR, ESI-MS, and elemental analysis. Compounds 6h and 6q were further characterized by single crystal X-ray structural analysis. All of the compounds have been screened for their antibacterial potential in vitro against Bacillus subtilis ATCC 6633, Escherichia coli ATCC 35218, Pseudomonas fluorescens ATCC 13525, and Staphylococcus aureus ATCC 6538. Among the tested compounds, some of them display significant activity against the tested strains, and compounds 5ac and 6h show potent activity with a minimum inhibitory concentration value of 1.562 μg mL–1 against B. subtilis ATCC 6633, which is comparable to the positive control penicillin. Structure–effect relationships are also discussed based on the experimental data.

Abstract:A series of dihydropyrazole derivatives containing morpholine was designed and synthesized as antimicrobial agents. All of the synthesized compounds were characterized by 1H-NMR and MS. Afterwards they were evaluated for in vitro antibacterial activity against four bacteria strains. Along with S. aureus TyrRS inhibition and cytotoxicity examination, some compounds proved to be of low toxicity and potent, especially against Gram-positive bacteria strains. Compound 4s exhibited the potential to be new a antibacterial drug with strong broad-spectrum antimicrobial activity and enzyme inhibitory activity. Docking simulation was performed to position compound 4s into the S. aureus TyrRS structure active site to investigate the probable binding mode. A 3D-QSAR model was also established to explain how structural alterations affect the activity and guide further study.

Abstract:Seven new 5-(2-hydroxy-6-methoxylphenyl)-3-methyl-4,5-dihydropyrazole-1-carbothioamide derivatives were synthesized and were characterized by ~1H NMR,IR and elemental analysis.The biological activity tests showed that some of the derivatives exhibited antibacterial activity.

Abstract:As an archetypal molecule, hydrazides have a crucial vital role in numerous applications, so hydrazide-related inhibitors, especially sulfur-enriched, are favored. In the present work, we designed, synthesized and characterized fifteen novel benzohydrazide derivatives containing 4-methanesulfonyloxy and arylidene building blocks with a four-step synthesis pathway. The inhibitory potential of the compounds was assessed using human carbonic anhydrases I and II (hCA I and II) isozymes and the results were compared to those of the standard inhibitor, acetazolamide (AZA). The antioxidant activity profiles for all compounds were also examined using various bioanalytical methods and the results were compared with the standards. The hCA I and II were the best inhibited by compounds 5f, 5g , and 5i with inhibition constants IC 50 in the range 20.45–51.43 nM (AZA: IC 50 =218.38) for hCA I and 33.54–42.45 nM (AZA: IC 50 =44.39) for hCA II. Structure-activity relationships were also discussed and discovered that hCA I and II inhibition was unaffected by the presence of an electron-withdrawing or releasing group. This effectiveness was the only result of the sort of substituted group(s), which was located at the reagent. Molecular docking and dynamics simulations showed that compounds 5f, 5g , and 5i have strong and stable interactions with key amino acids and zinc ions in the active sites of enzymes, which supports their ability to block enzyme activity. In silico ADME studies predicted favorable drug-like properties and high human oral absorption for all synthesized compounds. These findings highlight the multifunctional potential of the synthesized benzohydrazide derivatives as hCA I and II inhibitors and antioxidants, paving the way for their further development and optimization for therapeutic applications.
