Automated Synthesis of Novel F-18-Labeled Diarylideneacetone Derivatives for in Vivo Imaging of Beta-Amyloid Plaque

Abstract:1105 Objectives Alzheimer’s disease is pathologically characterized by deposition of misfolded β-amyloid (Aβ) as senile plaques and neurofibrillary tangles in the brain. We have shown novel 18F-diarylideneacetone derivatives with 18F atom directly attached to the aromatic benzene or pyridine ring have high affinities to Aβ (Ki = 3.0 - 135 nM) and improved properties (ineligible defluorination, high brain uptake, and better brain pharmacokinetics). Herein we discuss fully automated two-step radiosyntheses of novel 18F-diarylideneacetones using base-catalyzed Claisen condensation. Methods The precursors for radiolabeling were synthesized by converting the N,N’-dialkyl arylaldehydes (benzene or pyridine ring) to their corresponding quaternary ammonium salts. The radiosyntheses were performed on an automated GE FXFN Tracerlab. The SNAr reaction of precursors with 18F anion resulted in the corresponding 18F-fluoroarylaldehydes, which upon condensation with substituted benzylideneacetones resulted in labeled diarylideneacetones. The crude products were purified by semi-prep HPLC after neutralization. Isolated products were confirmed by co-injection with the standards on an analytical HPLC. A variety of substituted benzylideneacetones were tested for condensation. Results 18F-labeled novel diarylideneacetones were successfully synthesized with chemical and radiochemical purity of > 99 % and specific activity of 1.8 ± 0.4 mCi/nmol (EOS). The overall radiochemical yields were 7-8 % (EOB). Conclusions Novel diarylideneacetones were successfully prepared by two-step automated radiosyntheses. Further research to increase the radiochemical yields by varying conditions is in progress. Preclinical imaging studies with microPET are underway.
