Abstract:To investigate the possible mechanisms of the abnormal expression patterns of many genes in the embryos in vitro, the expression of histone acetyltransferase GCN5 and histone deacetylase 1 (HDAC1) were detected in mouse preimplantation embryos. For the in vitro group, the pronucleus embryos were obtained from superovulated mice, and cultured in vitro to get the two-cell, four-cell, eight-cell, morula and blastocyst stages embryos. For the in vivo group, embryos at different stages were obtained from pregnant mice directly. Reverse transcription polymerase chain reaction (RT-PCR) and immunocytochemistry were used to detect the mRNA and protein expression levels of GCN5 and HDAC1. Compared with the embryos in vivo, the expression levels of Gcn5 in the embryos in vitro significantly increased except those in four-cell embryos. The expression of Gcn5 in eight-cell embryos in vivo and in four-cell and eight-cell embryos in vitro was higher than those at other stages within the same group. Compared with the expressions of Hdac1 in embryo in vivo, only those at two-cell embryo in vitro showed decreased level. The expression of Hdac1 enhanced after two-cell embryo stage in vitro, but no difference showed in vivo. The protein expression of GCN5and HDAC1 in the embryo in vitro at every stage showed a lower level with the control of those in the embryos in vivo. Our studies indicated that in vitro culture could induce the expressed alteration of GCN5 and HDAC1, which might be related to the expression patterns of many other epigenetically regulated genes.
Expression of HLA-G Protein in Human Preimplantation Embryos

Abstract:Objective: To investigate the function and significance of human leukocyte antigen-G(HLA-G) protein in human preimplantation embryos and immunological tolerance. Methods: Human preimplantation embryos, which were residual and contributed, were obtained during assisted conception process by IVF-ET. Immunof- luorescence staining was used to detect HLA-G protein in varies stages of human preimplantation embryos. Results: Twenty 2-cell to 8-cell human embryos were stained with MEM-G/9, a monoclonal antibody specific for HLA-G. HLA-G labeling was found in 15/20 2-cell to 8-cell human embryos. HLA-G positive labeling was observed in all 3 morulas and 5 blastocysts. Conclusions: Human preimplantation embryos express HLA-G protein.
Sexing of Mouse Preimplantation Embryos Using Polymerase Chain Reaction

Abstract:In order to determine the sex of mouse embryo, 1 or 2 blastomeres were biopsied from Kun-ming-white mouse preimplantation embryo at 4-8 cells stage. The gDNA of the single-blastomere was abstracted. According to the base sequence of 145C5, a repititive sequence of Y chromosome of mouse C57BL6, a pair of primer were asigned and synthesized. The gDNA was amplified using these primers. 108 mouse preimplantation embryos were sexed via this technique. 46 male embryos and 62 female embryos were transfered into five pseudopreganant mothers respectively. 4 male litters and 9 female litters were obtained. The diagnosis positive　rate was 100%(4/4) and 70% (9/13)respectively. The result of PCR indecated that there was no difference between the repititive sequence of Y chromosome in mouse C57BL6 and Kun-ming-white mouse. The technique developed in this study might be further used for preimplantation genetic diagnosis of single-gene defects.

Abstract:Objective To investigate the expression of HLA-G (human leukocyte antigen-G) and its isoforms in human preimplantation embryos in order to discuss the significance of HLA-G in the early embryo development. Methods The human embryos were donered by the volunteers from the residual fertilized ovum after accepting IVF from January of 2003 to December of 2005 in the Reproductive Center of Tangdu Hospital,Fourth Military Medical University. Human embryo mRNA was isolated using Dynabeads mRNA DIRECTTM Kit. Nested RT-PCR was performed using SuperScriptTM One-Step RT-PCR Platinum Taq Kit and Platinum PCR SuperMix High Fidelity Kit. Results HLA-G and its isoforms in human cleavage stage embryos:a total of twenty individual cleavage stage human embryos were examined (five 2-cell, five 4-cell, five 6-cell and five 8-cell). Only 7/20 embryos expressed HLA-G and its partial isoforms mRNA. HLA-G mRNA isoforms in human morulas:five individual four-day human embryos were examined. All of them expressed HLA-G and its partial isoforms mRNA. HLA-G mRNA isoforms in human blastocysts:twenty-five six-day expanded blastocysts were studied. All of them were found to express HLA-G but different isoforms mRNA. G1 was detected in 20/25 (80%), G2 in 4/35 (16%), G3 in 25/25 (100%), G4 in 24/25 (96%), G5 in 5/25 (20%) and G6 in 8/25 (32%). Conclusions The HLA-G expression increases along with the stages of embryo development.Human preimplantation embryos diferentially express HLA-G mRNA isoforms in different development stages.
Differential Expression of Alternatively Spliced Transcripts of HLA-G in Human Preimplantation Embryos and Inner Cell Masses

Abstract:It has been reported that preimplantation human embryos secrete HLA-G, and the levels may be predictive of their ability to implant. However, it is not known which of the membrane-bound (HLA-G 1-4) and soluble (HLA-G 5-6) alternatively spliced forms are present, nor the developmental stage at which they appear. Therefore, we have investigated HLA-G mRNA isoform expression on single embryos at the two-, four-, six-, and eight-cell, morula, and blastocyst stages. The percentage of embryos expressing each HLA-G isoform mRNA increased with developmental stage, but contrary to expectation, HLA-G5 mRNA was not detected in single two- to eight-cell embryos and was only expressed by 20% of morulae and blastocysts. Similarly, soluble HLA-G6 mRNA was not detected until the blastocyst stage and then in only one-third of embryos. In contrast, labeling with MEM G/9 Ab (specific for HLA-G1 and -G5) was observed in 15 of 20 two- to eight-cell embryos and 5 of 5 blastocysts. This disparity between mRNA and protein may be due to HLA-G protein remaining from maternal oocyte stores produced before embryonic genome activation and brings into question the measurement of soluble HLA-G for clinical evaluation of embryo quality. Although RLA-G is expressed in the preimplantation embryo, later it is primarily expressed in the invasive trophoblast of the placenta rather than the fetus. Therefore, we have investigated whether down-regulation of HLA-G first occurs in the inner cell mass (precursor fetal cells) of the blastocyst and, in support of this concept, have shown the absence HLA-G1 and -G5 protein and mRNA.

Abstract:INTRODUCTION:HLA-G is a major histocompatibility complex (MHC), class Ib molecule that is selectively expressed at the fetal-maternal interface. It is thought to play a role in protecting the fetus from the maternal immune response. Interestingly, the preimplantation embryo development (Ped) gene product Qa-2 is also a mouse MHC class Ib protein that affects cleavage and division of preimplantation mouse embryos and subsequent embryonic survival. Data from many human in vitro fertilization (IVF) clinics suggest that the mouse Ped phenomenon also exists in human because embryos fertilized at the same time have different cleavage rates and consequently different IVF outcomes. As HLA-G is expressed in human early embryos, it is highly regarded as the functional homologue of Qa-2. Whether HLA-G expression is correlated with the cleavage rate of human embryos has great potential clinical value.METHODS:In this study, 45 human early abnormal fertilized embryos (3 PN) from patients undergoing in vitro fertilization were used to test the effects of HLA-G knock-down via infection with adenovirus carrying its specific siRNA on the cleavage rate in a 2-day culture period. One-way ANOVA, Post hoc and Chi-square were used to compare groups. A p-value smaller than 0.05 was considered statistically significant.RESULTS:Knocking-down HLA-G in human pre-implantation stage embryos resulted in a higher cell arrest rate and a slower cleavage rate.CONCLUSION:The results from the present study suggested that HLA-G might play an important role in early human embryo development.

Abstract:The effects of reproductive hormone on mouse embryo sexes were studied with in vitro fertilization (IVF), in vitro culture (IVC) and sex identification. Results showed that it had no adverse effects on embryos cleavage and development after the embryos were incubated with the fertilization media with 17beta-E2, FSH and progesterone respectively at the concentration of this study. Mice embryos sex ratios significantly deviated from 1:1(P < 0.01) and skewed in favor of males at the concentration of 17beta-E2 attained 70 pg/mL and progesterone attained 99 pg/mL in IVF culture media respectively. 0.01-0.04 IU/mL FSH had no effects on the mouse embryo sexes.
Effect of the FBS and hLIF of Different Concentrations on the Viability of Mouse Pronuclear Zygote in Vitro

Abstract:The objective of this study was to probe into some culture conditions of mouse pronuclear zygote and establish a kind of steady culture system of mouse pronuclear zygote in vitro through the appending of the FBS and hLIF of different concentration in mM16.The ultama result indicated in mM16,the addition of 10% or 15% FBS could decrease the viability of zygote in vitro.The ratio of blastocyst formation to experimental and control groups were different(73.3% vs 32.6% and 73.3% vs 35.8%),respectively;the different concentrations of hLIF(500,1?000 and 1?500?IU/ml) can increase the development ratio of mouse zygote in vitro,which there were no obvious difference among of three groups.But this group added with 1?000?IU/ml hLIF,the ratio of morula,blastocyst and prolapsing blastocyst from the zona pellucida were the highest compared with other groups.The conclusion was the addition of FBS did not effectively accelerate the viability of mouse zygote in vitro,the appending of 1?000?IU/ml hLIF can remarkably increase development ratio of mouse zygote in vitro.
The correlation between the quantity of soluble forms of human leukocyte antigen-G in human embryo cultures during cleavage period and pregnancy & implantation rates.

Abstract:Objective:To investigate the relationship between the quantity of soluble Human Leukocyte Antigen-G(sHLA-G) in human embryo culture and pregnancy rate(PR) & implantation rate(IR) during the cleavage period.Methods:By an enzyme-linked immunosorbent assay(ELISA),we tested 80 media of single embryo culture from 40 patients in IVF cycles.Results:Thirty-five media of single embryo culture had sHLA-G expression in 80 cultures,the positive rate was 43.8,and the average quantity was(4.68±1.29)U/ml.All the patients were transferred two embryos.27 patients had got at least one sHLA-G positive embryos transferred and 19 patients were pregnant.The PR was 70.4%(19/27) and IR was 46.2%(25/54).While 13 patients were transferred sHLA-G negative embryos,only 3 patients got pregnant.The PR and IR were 23.1%(3/13) and 15.4%(4/26) respectively.The two groups had a significant difference(P<0.01).Conclusion:The level of sHLA-G expresses in embryo culture.There is a significant difference between sHLA-G positive group and sHLA-G negative group in PR and IR.We consider that there is a definite correlation between sHLA-G level and PR & IR.The level of sHLA-G can forecast the potential of embryo development.So the sHLA-G ELISA can be a useful assay in addition to embryo morphology in embryo selection for transfer in IVF treatment.
Rabbit Antiserum to Mouse Embryonic Stem Cells Delays Compaction of Mouse Preimplantation Embryos.

Abstract:BACKGROUND:Mouse embryonic stem (ES) cells are derived from the inner cell mass (ICM) of the preimplantation blastocysts. So it is suggested that ES and ICM cells should have similar cellular surface molecules and antiserum to ES cells can inhibit ICM development.OBJECTIVE:The objective of this study was to evaluate the effect of rabbit antiserum to ES cells on mouse preimplantation embryo development and chimera production.MATERIALS AND METHODS:Mouse 4-cell embryos were matured in vitro at 37.5(o)C, in humidified 5% CO2 atmosphere for 12-36 h. The embryos were cultured in KSOM medium with or without antiserum for 12-36 h. The ratios of in vitro embryo development of the blastocysts, cell division, attachment potential, alkaline phosphatase activity, post-implantation development, and chimera production were assessed and compared with the control group. P<0.05 was considered as significant.RESULTS:The rabbit antiserum to mouse ES cells showed delay in embryo compaction and induced decompaction at 8-cell stage. The development of 4-cell embryos in the presence of the antiserum for 36h did not lead to a reduced or absent ICM. These embryos still displayed positive alkaline phosphatase activity, normal cell division, embryo attachment, outgrowth formation, implantation and post-implantation development. In addition, decompaction induced by antiserum did not increase production and germline transmission of chimeric mice.CONCLUSION:The results showed that antiserum to ES cells delayed embryo compaction and did not affect post-implantation development and chimera production.

Abstract:Expressions of GCN5 and HDAC1 in in vivo-and in vitro-cultured mouse preimplantational embryos were studied by immunocytochemistry.Results:(1) In in-vivo group: GCN5 fluorescence signals were evident in the cytoplasm of preblastocyst embryos,but in blastocysts,it disappeared.In two-cell embryos,HDAC1 staining was observed in the cytoplasm and nuclei,but in the following cleavage-stage embryos,it was mainly localized in the nuclei.(2) In in-vitro group: there were no GCN5 signals at all stages of embryos.The expression pattern of HDAC1 was similar to that of in-vivo group,but its level decreased.Those results show the early cleavage environment influence GCN5 and HDAC1 expressions in mouse preimplantational embryos.

Abstract:AIM: [JP3]To investigate the expression of c-myc gene in mouse preimplantation embryos. METHODS: In situ hybridization and Immunohistochemical staining (ABC method) were used to detect c-myc mRNA and c-myc protein, espectively. RESULTS: c-myc mRNA and c-myc protein were found in 2-cell, 4-cell, 6-cell, 8-cell and blastsyst stages of mouse preimplantation embryos. The positive staining substance was distributed in cytoplasm and nucleus. CONCLUSION: These results suggest that c-myc gene is expressed in mouse preimplantation embryos and may have an essential role in mouse early embryonic development.

Abstract:The goal of preimplantation development is to establish the fates of the embryonic and extra-embryonic cells. However, when and how cell fates are determined during early mammalian embryonic development remains unclear. We report that the high mobility group (HMG) protein family member HMGA1 was distributed differentially in mouse two-cell blastomeres. Knockdown of Hmga1 expression in one of the two cells reduced the number of cells contributing to the inner cell mass (ICM), suggesting that differential distribution of HMGA1 in the blastomeres in two-cell mouse embryos affected the selection of embryonic cell lineages. Mechanistically, HMGA1 promotes the expression of the ICM-specific gene Sox2. The results of this study show that mouse embryos demonstrate heterogeneity as early as the two-cell stage, and that these differences are related to cell-fate differentiation in early mouse embryos.

Abstract:Objective: This research investigated the effects of human chorionic gonadotropin (HCG)-producing peripheral blood mononuclear cells (PBMCs) on the implantation rate and embryo attachment in mice. Methods: In this experimental study, a DNA fragment of the HCG gene was cloned into an expression vector, which was transfected into PBMCs. The concentration of the produced HCG was measured using enzyme-linked immunosorbent assay. Embryo attachment was investigated on the co-cultured endometrial cells and PBMCs in vitro. As an in vivo experiment, intrauterine administration of PBMCs was done in plaque-positive female mice. Studied mice were distributed into five groups: control, embryo implantation dysfunction (EID), EID with produced HCG, EID with PBMCs, and EID with HCG-producing PBMCs. Uterine horns were excised to characterize the number of implantation sites and pregnancy rate on day 7.5 post-coitum. During an implantation window, the mRNA expression of genes was evaluated using real-time polymerase chain reaction. Results: DNA fragments were cloned between the BamHI and EcoRI sites in the vector. About 465 pg/mL of HCG was produced in the transfected PBMCs. The attachment rate, pregnancy rate, and the number of implantation sites were substantially higher in the HCG-producing PBMCs group than in the other groups. Significantly elevated expression of the target genes was observed in the EID with HCG-producing PBMCs group. Conclusion: Alterations in gene expression following the intrauterine injection of HCG-producing PBMCs, could be considered a possible cause of increased embryo attachment rate, pregnancy rate, and the number of implantation sites.

Abstract:Purpose By comparing the chromosomal constitution among the arrested cleavage-stage embryos, blastocysts and human embryonic stem cells (hESCs) which are all derived from monopronuclear (1PN) zygotes, it is aimed to determine whether chromosomally normal embryos can be reliably selected by blastocyst culture. Methods After 1PN zygotes are sequentially cultured for 5 days, the blastocysts and arrested cleavage-stage embryos were analyzed by fluorescence in situ hybridization (FISH) with probes for chromosomes 18, X and Y; G-banding analysis was adopted to analyze the karyotype of 1PN hESCs. Results The diploid rate of blastocysts was 74.6%, which was significantly ( P < 0.001) higher than that of arrested cleavage-stage embryos (31.6%), and the diploid rate of hESCs was 97.0%, which was significantly ( P < 0.01) higher than that of blastocysts; the haploid embryos were excluded by blastocyst culture; nevertheless, there still existed such chromosomal abnormalities as mosaic and monosomic in blastocysts and trisomy in hESCs. Conclusions Blastocyst culture is an effective method to select against chromosomal abnormalities, especially the haploids in 1PN embryos; however, development to the blastocyst stage is not a reliable marker for mosaicism or aneuploidy.

Abstract:We report on the birth of 39 mice following single cell embryo biopsy and precise sex determination following in-vitro fertilization. Polymerase chain reaction was used to amplify fragments of the mouse testis-specific gene sequence (pYMT2/B) on the Y chromosome and the ovary-specific gene (ZP3) sequence on chromosome 5 from the single biopsied cell. Embryo biopsy was not associated with any deleterious effects.
Effect of Monoclonal Antibody to Bull Sperm Sp18 on Murine Fertilization and Embryos Development

Abstract:Western blot analysis revealed that one IgG 1 monoclonal antibody (mAb) to sp18 family membrane proteins (Mr. 14, 16 and 18 ku) of bovine sperm reacted faintly with protein bands of 14, 18, 22, 30 and 60 ku (reducing) in samples of mouse sperm. The mAb also reacted to protein of egg lysozyme. Using a laser confocal microscope, indirect immunofluorescence (IIF) showed that the sp18 antigens were present in the posterior head of murine sperm. In murine in vitro fertilization (IVF) and embryo development trails, a total of 426 oocytes from C 57 ,BL/6 and F 1 hybrid strain (CD 1 × C 57 BL/6 cross) of 12 female mice were used in 3 independent trails. After preincubating capacitated sperm with 182 μ/mL of sp18 mAb in the modified TYH IVF medium for 15–20 min, cumulus-oocyte-complexes were introduced. The fertilization rate in sp18 rnAb groups was 77.1 %, which was not significantly (P > 0.05) different from the nonspecific mouse IgG (79.2 %) and non-lgG (80.3 %) control groups. Fertilized oocytes had been continuously cultured in modifed CZB medium. 100%, 100% and 97.9% of 1cell embryos developed to 2-cell stage in sp18 mAb, nonspecific mouse IgG and non-lgG group 30 h after the start of fertilization, respectively. In the nonspecific mouse IgG and nonlgG groups, 64.1% and 64.3% of embryos developed to the 4-cell stage, respectively, but all developing eggs in sp18 mAb groups arrested development in vitro at 2-cell stage. After zonae of 2-cell blocked embryos were enzymatically removed with 0.5% pronase, detection of sp18 antigens by IIF indicated that the fluorescence scattered on two embryonic cells. For embryos fertilized in vivo and co-cultured with 182 μg/mL sp18 mAb, the numbers of 1-cell embryos reaching the 2-cell and 4-cell stage were 95.2% and 70.5%, which were not significantly ( P > 0.05) different from the control group (92.9% and 77.9%). These results indicate that the sp18 antigens on posterior head of mouse sperm were incorporated into the egg plasma membrane during fertilization, and played an active role in development of murine preimplantation embryo.

Abstract:The objective was to determine the effects of adding progesterone, 17β-estradiol (17β-E2), and FSH during in vitro fertilization on development and sex ratio of murine embryos. Progesterone (33–330pg/mL), 17β-estradiol (17β-E2; 10–70pg/mL), and FSH (0.01–0.05IU/mL), were added to human tubal fluid (HTF); this medium (with or without hormones) was used to pre-incubate sperm (2h) and to co-incubate sperm and oocytes (6h). Thereafter, the ova were washed and incubated in mM16 medium and embryo sex was determined (by PCR) on Day 4 (insemination=Day 0). There was no effect (P>0.05) of hormone treatments on rates of cleavage (6h after cessation of co-incubation with sperm). The only significant effects of added hormones on development were a decrease in the rate of development to at least the morula stage in 165pg/mL progesterone (0.46±0.03 vs. 0.54±0.05 in the control, mean±S.D.; P<0.05) and a decrease in the blastocyst rate in 0.03IU/mL FSH (0.34±0.00 vs. 0.42±0.04 in the control, P<0.05). However, the ratio of male to female embryos was 1.61 and 2.90 following the addition of 99pg/mL progesterone and 70pg/mL 17β-E2, respectively; both of these ratios were different (P<0.01) than in the control group (1.20). In contrast, the addition of FSH to the medium had no significant effect on this ratio (range, 0.78–1.02). We concluded that the addition of progesterone and estradiol to the media during in vitro fertilization did not enhance embryonic development, but significantly increased the proportion of male murine embryos.

Abstract:Abstract Background Morphological features are the most common criteria used to select human embryos for transfer to a receptive uterine cavity. However, such characteristics are not valid for embryos in cellular arrest. Even aneuploid embryos can have normal morphology, and some euploid embryos have aberrant morphology. The aim of this study was to quantify the expression profile of hsa-miR-21-3 p , -24-1-5 p , -191-5 p , and -372-5 p in culture media on day 5 of in vitro embryo development, and compare the profiles of two groups of media classified by outcome: successful (n = 25) or unsuccessful (n = 25) implantation pregnancy. Methods Fifty patients were accepted in the Department of Reproductive Biology of a Hospital in México City, based on the Institutional inclusion criteria for in vitro fertilization. Embryos were transferred to the women on day 5 of cultivation, and the culture media were collected. RNA was isolated from each culture medium with TRIzol reagent, and microRNA (miRNA) expression was detected through RT-PCR with specific primers. Expression bands were quantified by reading optical density. Results There was a 5.2-fold greater expression of hsa-miR-191-5 p in the pregnancy-related culture media ( p ≤ 0.001) and a 1.6-fold greater level of hsa-miR-24-1-5 p ( p = 0.043) in the media corresponding to non-pregnant women. No significant difference existed between the two groups hsa-miR-21-3 p ( p = 0.38) or hsa-miR-372-5 p ( p = 0.41). Conclusions Regarding adequate in vitro embryo development, hsa-miR-191-5 p could possibly represent a positive biomarker, while has-miR-24-1-5 p may indicate poor prognosis. This former miRNA modulates IGF2BP-1 and IGF2R, associated with the implantation window. On the other hand, hsa-miR-24-1-5 p may be related to a poor prognosis of human embryo development.

Abstract:Although the biochemical purified gonadotropins have been utilized mainly in the studies on the mammals reproductive processes(including the early embryonic development),the problem of those so called contaminants in gonadotropins purified by such a way has still existed.It means that these contaminants might affect the real function of those biochemically purified hormones on the mammals propagation.On the other hand,by the recombinant DNA technology,their recombinant gonadotropinic products could escape most of the problems in the regularly biochemical purified ones.The biological stability of such recombinant hormones is necessary for the effective research work on mammals reproductive processes.In this experiment,both the recombinant human follicular stimulating hormone(r hFSH)and recombinant human luteinizing hormone(r hLH)were applied for superovulating the immature prepubertal Kunming mice,then,those flushed preembryos were in vitro cultured for observation of their blastocyst formation capacity.In groups superovulating with r hFSH(20 IU)and r hLH(0～20 IU),the number of preembryos for mice was in dose dependent with r hLH(3 1±0 8 to 0 3±2 1),their blastocystal rates were also in such a relation with it(0%～7 96%).In groups superovulating with r hFSH(20 IU)combining a ratio of r hLH(0～20 IU),the number of preembryos for mice was similar among those different groups,but their blastocyst rates were somewhat dose dependent with r hLH(0～8 11%).The results showed that r hLH probably affect the early embryonic development in mice negatively, especially for their blastocystal formation.
