Abstract:In EtOH/benzene, 2-butyn-1, 4-diol and its derivatives can be halogenated by cupric halide stereospecifically to obtain (E)-2,3-dihalo-2-buten-1,4-diol derivatives. The isolated yields were 100%, and the purity of crude products attained 98%(GC).

Abstract:3-Hydroxybenzylhydrazine dihydrochloride was synthesized from 3-hydroxybenzaldehyde and methoxycarbonylhydrazine via condensation, hydrogenation, decarboxylation and salt formation in a total yield of 77.1%. The yield of condensation was 94.6% and of hydrogenation was 99.8% by using Pd/C (5%) which can be reused 3 times at the optimal quantity of 3% (W/W). The yield of salt formation was 81.7%. This route is easy to industrialize for its simple operation, mild reacting condition and low cost.

Abstract:2, 4-Dimethoxybenzaldehyde was prepared using 1, 3-dimethoxybenzene as raw material through Vilsmeier formylation with DMF directly. The compound was characterized by IR and NMR as a qualified product. Under the optimal conditions the product obtained from the orthogonal experiments could be given in 94.2% yield with purity of 98.64%, after purification the purity of final compound could be reached 99.6%.

Abstract:A parallel process for the new synthesis of 2,7-dimethyl-2,4,6-octatriene-1,8-dial was investigated.3-(5,5-Dimethyl-1,3-dioxan-2-yl)-2-butenal was obtained by a process involving aldehyde protection,ester exchange and catalytic oxidation from 4-acetoxy-2-methyl-2-butenal.A new catalytic oxidation process was proposed to get 3-(5,5-dimethyl-1,3-dioxan-2-yl)-2-butenal by oxidating 3-(5,5-dimethyl-1,3-dioxan-2-yl)-2-buten-1-ol with copper(Ⅰ) chloride、2,2,6,6-tetramethyl-1-piperidinyloxy and oxygen.3-(5,5-Dimethyl-1,3-dioxan-2-yl)-2-buten-1-ol was chlorizated with SOCl_2 in DMF to get 2-(3-chloro-1-methyl-1-propenyl)-5,5-dimethyl-1,3-dioxane.2,7-Dimethyl-2,4,6-octatriene-1,8-dial was acquired in an overall yield of 48% by one-pot synthesis method from 3-(5,5-dimethyl-1,3-dioxan-2-yl)-2-butenal,2-(3-chloro-1-methyl-1-propenyl)-5,5-dimethyl-1,3-dioxane and triethylphosphite.

Abstract:1-Chloro-2,5-dimethoxybenzene was prepared from p-benzoquinone through chlorination and methylation with total yield of 78%. p-Benzoquinone was firstly converted catalytically to 1-chloro-2,5-dihydroxybenzene in the binary system of dichloroethane and hydrochloric acid at 35℃ with yield of 89%, then the final product was synthesized by methylation of obtained 1-chloro-2,5-dihydroxybenzene with dimethyl sulfate in the presence of sodium hydroxide. The intermediate 1-chloro-2,5-dihydroxybenzene and the obtained title compound 1-chloro-2,5-dimethoxybenzene were characterized by IR and GC-MS respectively. The results showed that their chemical structures were correct and the purity of title compound was over 98%.

Abstract:2-Bromo-5-hydroxy-4-methoxybenzaldehyde(I)and 5-hydroxy-4-methoxy-2-nitrobenzaldehyde(II)were synthesized.The intermediates 2-bromo-4,5-dimethoxybenzaldehyde and 4,5-dimethoxy-2-nitrobenzaldehyde were synthesized firstly from veratraldehyde through bromination and nitration respectively.In the presence of molar quantity methionine,the 5th-position methoxys of the intermediates were in turn hydrolysed to hydoxy by concentrated sulfuric acid to give I and II.The chemical structures of the titled compounds were confirmed by 1H-NMR,IR and MS spectrum.The yields of compounds I and II are 85.0% and 65.0% respectively.The orientations of the bromination and nitration steps were suggested and discussed.The mechanism of methionine in the hydrolysis step may involve in the formation of the multiple hydrogen bonded association complex,and the calculated single point energy(E=-1532.5 hartree)of the multiple hydrogen bonded association complex seems to show it is possibly existed at the RHF/6-31G level.

Abstract:Improved synthesis of 4-benzylidene-2,6-di-tert-butylcyclohexa-2,5-dienone has been developed. The dropwise addition of secondary amines was shown to reduce the dosages of amines drastically. In optimized conditions, the dosages of the catalysts were only 0.3–0.5 M of 2,6-di-tert-butylphenol. Removing the water generated in the step of Mannich base formation was found to hugely accelerate the whole reaction and improve the product yield. The catalytic activity of several annular secondary amines including morpholine and piperidine in this reaction has been researched. Most of the amines led to good yields. Using this method, 2,6-di-tert-butylphenol reacted with a wide range of aromatic aldehydes to produce nine diverse 4-benzylidene-2,6-di-tert-butylcyclohexa-2,5-dienone derivatives of up to 75 % yield.

Abstract:Novel cyclic esters (1R, 2R, 3R, 5S, 7S, 9R, 12R)-3-(t-butyldimethylsilyl) oxy-7-methoxymethyloxy-2, 10-dimethyl-12-oxatricyclo [7.2.1.05,12] dodecane were prepared when their precursor 1 was treated with SOCl 2/pyridine. A plausible mechanism was hypothesized.

Abstract:Bicyclohexane-4,4′-dione was synthesized and the carbonyl groups were partially protected.Bicyclohexane-4,4′-diol was oxidized to bicyclohexane-4,4′-dione in 75.4% yield by using chromic acid at 30 ℃ for 3 h.Under the catalysis of potassium bisulfate,bicyclohexane-4,4′-dione was subjected to dehydrating condensation with ethylene glycol ［n(bicyclohexane-4,4′-dione)∶n(ethylene glycol)=1.2∶1.0］.The unprotected material was partially removed by aqueous sodium bisulfite solution from the toluene solution.By using high concentration aqueous sodium bisulfite solution,bisulfite addition product of bicyclohexane-4,4′-dione mono-ethyleneketal formed in ethyl acetate.The addition product was treated with 100 g/L aqueous ammonia to obtain bicyclohexane-4,4′-dione mono-ethyleneketal with overall yield of 31％ and purity of 99.7% by GC.
Enantioselective Aldol Reaction of Acetone and Benzofuran-2,3-Diones

Abstract:The 3-substituted 3-hydroxybenzofuran-2-one unit is encountered in a large variety of natural products with a wide spectrum of biological activities. Therefore, development of new methodologies to the construction of chiral quaternary center at the 3-position of benzofuran-2-one compounds were highly desired. A highly enantioselective Aldol reaction of benzofuran-2,3-ones with ketone catalyzed by N-(2-thiophenesulfonyl)prolinamide through enamine activation strategy was developed. As a result, a number of chiral 3-hydroxyl substituted benzofuran-2-ones derivatives were synthesized with very good yields(up to 96%) and moderate to good enantioselectivities(up to 91% e.e.). This methodology substantially broadens the benzofuranone chemistry and offer functionalized chiral products.

Abstract:The combination of zirconocene-mediated coupling of silylated alkynes with a protonation-desilylation or bromination-desilylation process afforded otherwise unavailable butadiene derivatives. When (E,E)-2,3-dialkyl-1,4-bis(trimethylsilyl)-1,3-butadienes were treated with 3 equiv of Br-2 in CH2Cl2, (E)-2,3-dialkyl-1,1,4,4-tetrabromo-2-butenes were obtained in excellent yields with perfect stereoselectivity.

Abstract:Z and E configuration 4-aryl-but-2-en-1-ols were isolated from several terrestrial plants, and (Z)-4-aryl-but-2en-1-ols were found to have choleretic activity. Strategies have been reported to synthesize (E)-4-aryl-but-2-en-1ols with high selectivity. However, there is no method to obtain (Z)-4-aryl-but-2-en-1-ols with high selectivity now. We developed a Suzuki-Miyaura cross-coupling reaction of 1,2-oxaborol-2(5H)-ols with benzyl bromides to synthesize (Z)-4-aryl-but-2-en-1-ols, the products were obtained in up to 94% isolated yield.

Abstract:Z(E)-ethyl 3-oxo-2-(4-phenyl-1,3-dithiolan-2-ylidene)butanoate (1) was prepared in moderate (40.3%) yield by the reaction of ethyl 3-oxobutanoate with carbon disulfide and 1-(1,2-dibromoethyl)benzene with potassium carbonate as the base in DMF.Subsequently,the condensation reaction of 1 with arylalde- hydes was performed in the presence of C_2H_5ONa in C_2H_5OH,in which the product of condensation and ester hydrolysis:(2Z(E),4E)-3-oxo-5-phenyl-2-(4-phenyl-1,3-dithiolan-2-ylidene)pent-4-enoic acid (2) were isolated in good yield.

Abstract:We report herein the synthesis of highly substituted and stereochemically well-defined vinylbenziodoxole (VBX) derivatives through hydrochlorination and iodochlorination of ethynylbenziodoxoles. The hydrochlorination is achieved using pyridine hydrochloride as an HCl source in an anti-fashion under mild, open-air conditions to afford a 2-chlorinated VBX product, which serves as a useful building block for the stereoselective synthesis of trisubstituted alkenes. Meanwhile, iodochlorination with iodine monochloride proceeds in an unusual syn-pathway, stereoselectively affording a tetrasubstituted VBX derivative.

Abstract:A facile method for the preparation of tert-butyl (3R,5S)-6-hydroxy-3,5-O-isopropylidene-3,5-dihydroxyhexanoate is described by a chemoenzymatic approach. In this method, one hydroxyl stereocenter at C5 is obtained with a high ee value (up to 98.0%) via an enzymatic transesterification resolution of l-chloro-3-(4-methylbenzyloxy)-2-propanol. The other hydroxyl stereocenter at C3 was built with 98.0% de, by acid-hydrolysis of a 1,3-diol-acetonide syn/anti-10. It is noteworthy that the reduction of β-hydroxy ketone 8 with sodium borohydride can be carried out smoothly in aqueous isopropyl alcohol with a high diastereomeric ratio syn/anti (drs:a) of 4.0:1.

Abstract:(Z)‐alkenes are useful synthons but thermodynamically less stable than their (E)‐isomers and typically more difficult to prepare. The synthesis of 1,4‐hetero‐bifunctionalized (Z)‐alkenes is particularly challenging due to the inherent regio‐ and stereoselectivity issues. Herein we demonstrate a general, chemoselective and direct synthesis of (Z)‐2‐butene‐1,4‐diol monoesters. The protocol operates within a Pd‐catalyzed decarboxylative acyloxylation regime involving vinyl ethylene carbonates (VECs) and various carboxylic acids as the reaction partners under mild and operationally attractive conditions. The newly developed process allows access to a structurally diverse pool of (Z)‐2‐butene‐1,4‐diol monoesters in good yields and with excellent regio‐ and stereoselectivity. Various synthetic transformations of the obtained (Z)‐2‐butene‐1,4‐diol monoesters demonstrate how these synthons are of great use to rapidly diversify the portfolio of these formal desymmetrized (Z)‐alkenes.

Abstract:(E)-4-bromo-3-methyl-2-butenoic acid was conveniently synthesized from 3-methyl-2-butenoic acid in 29% overall yield.In this procedure,the title compound was easily obtained by hydrolysis of (E)-trimethylsilyl 4-bromo-3-methyl-2-butenoate,which could be isolated from its mixture of Z-/E-isomer by distillation in vacuo.The isomeric mixture was prepared via the esterification of 3-methyl-2-butenoic acid with chlorotrimethylsilane,followed by bromination with NBS.

Abstract:Stereo-defined 1,1,4,4-tetrahalo- and 1,1,4,4-mixed-tetrahalo-1,3-butadienes were obtained in excellent yields via desilylation–halogenation of their corresponding 1,4-bis(trimethylsilyl)-1,4-dihalo-1,3-dienes, which could be readily prepared from the zirconocene-mediated high-yield and high-regioselective cyclo-dimerization of 1-trimethylsilyl-1-alkynes followed by halogenation. These poly-functionalized gem-dihalodienes are potentially useful building blocks.

Abstract:A stereoselective synthesis of (4E,7S)-(-)-7-methoxy dodec-4-enoic acid has been accomplished in eight steps from hex: anal in 24% overall yield. Abstract:The enantiopure 1,1'-spirobiindane-7,7'-diol (SPINOL) and its 4,4'-derivatives 4,4'-diiodo-1,1'-spirobiindane-7,7'-dioI (DISPINOL) and 4,4'-dimethyl-1, 1'-spirobiindane-7,7'-diol (DMSPINOL) have been synthesized effectively via (1S)-(-)-4,4'-dibromo-1,1'-spirobiindane-7,7'-diyl bismenthyl carbonate with BuLi following the cleavage of the bismenthyl carbonate. Their absolute configurations were assigned by chemical correlation. They are also applied in the form of their Ti-alkoxides as catalysts for the addition of diethylzinc to aldehydes. The addition reactions proceeded with high conversions and enantioselectivities up to 88%. The effect of substitution of the substrate was studied.
