Abstract:A facile and practical palladium-catalyzed nucleophilic substitution/CH activation/aromatization cascade reaction has been developed. A range of 6-unsubstituted phenanthridines could be obtained in moderate to good yields (3185%) with readily prepared N-Ms arylamines and commercially available 2-bromobenzyl bromide derivatives as starting materials. The potential application of the protocol was also demonstrated by the expeditious synthesis of the natural alkaloid trisphaeridine.

Abstract:A silver-catalyzed 2-isocyanobiaryls insertion/cyclization with phosphine oxides was described for the construction of the 6-phosphorylated phenanthridines through radical addition of in situ formed P-centered radical to 2-isocyanobiphenyls and homolytic aromatic substitution process. Abstract:A novel and efficient protocol for the creation of 6-amino phenanthridine derivatives by Co(acac)(2)-catalyzed isocyanide insertion with 2-aryl anilines under an O-2 HAS atmosphere via homolytic aromatic substitution (HAS) type C-H ffinctionalization has been developed. This reaction not only proceeds smoothly utilizing O-2 as the oxidant but also provides a new approach to construct phenanthridine derivatives utilizing readily available 2-aryl anilines with isoc-yanides instead of 2-isocyanobiaryls with different radical precursors.

Abstract:An efficient and straightforward procedure toward the synthesis of amides-substituted 2H-benzopyrans H-benzopyrans has been explored through palladium-catalyzed one-pot aminocarbonylation reaction. By using nitro compounds as attractive nitrogen sources, with Mo(CO)6 6 as both CO source and reductant, a variety of amides-containing 2H- H- benzopyrans were obtained in moderate to high yields with good functional group tolerance. This protocol has advantages including atom-economy, no manipulation of toxic CO gas, and broad substrate scope.

Abstract:5,6-Dihydrophenanthridines are common aza heterocycle frameworks of natural products and pharmaceuticals. Herein, we reported the first palladium-catalyzed intramolecular C-H/C-H dehydrogenative coupling reaction of two simple arenes to generate 5,6-dihydrophenanthridines. The approach features a broad substrate scope and good tolerance of functional groups, offering an efficient alternative synthesis route for important 5,6-dihydrophenanthridine compounds.

Abstract:An efficient palladium-catalyzed nucleophilic substitution/C-H activation/aromatization cascade reaction between readily available 2-halo-N-Ms-arylamines (Ms = methanesulfonyl) and benzyl halides/sulfonates has been described. A wide variety of phenanthridines were synthesized in a one-pot fashion in moderate to high yields (37-86 %). Notably, this method provides a straightforward, facile approach for the synthesis of phenanthridines. The practicality was further substantiated by successfully carrying out a gram-scale preparation.

Abstract:A mechanistically new strategy has been described for the simple, practical, and environmentally friendly preparation of 6-(trifluoromethyl)phenanthridine derivatives using ionic isocyanide insertion from biphenyl isocyanide derivatives and Umemoto's reagent. These reactions were promoted only by inorganic base in good-to-excellent chemical yields without any external stoichiometric oxidants and radical initiators.

Abstract:A one-pot process involving cascade C-H activation and insertion between amidines and alkynes in the presence of palladium salt is described. With this methodology, various novel eight-membered cyclopalladated amidines were efficiently constructed. Notably, a series of functionalities with potential application were readily accommodated under the reaction conditions. Isotope effects and H/D exchange suggested that this cascade is initiated by the cleavage of the ortho C-H bond in the N-phenyl ring of amidines. (C) 2020 Elsevier B.V. All rights reserved.

Various quinoxaline‐2,3‐diamines could be obtained in 25%‐95% yields at 100 °C for 3 h. The key to the success of this protocol involves the formation of two C‐N and one C‐C bonds via the double insertion of isocyanide.

Abstract:An isocyanide insertion reaction promoted by the combination of an amide and a photoredox is now presented. By using visible light, 6-amidophenanthridine derivatives can be prepared in good chemical yields under mild and eco-friendly reaction conditions. The reaction was shown to proceed through an electron transfer sequence by mechanistic experiments, and thus enabled the efficient production of 24 compounds, each with one new intramolecular aromatic ring obtained. The synthesized compounds were tested to embody good antitumor, antimicrobial and neuraminidase inhibitory activities.
