Abstract:Different voltages were applied to anaerobic treatment to investigate the enhancement effects and the changes of microbial community structure. The results indicated that the best appropriate voltage was 0.10 V, COD removal increased by 16.72% at first 6 h and cumulative CH4 production increased by 23.39%. Average methane yield was 15.69% higher than that of control. The sludge measurements indicated that voltage addition could promote the interspecies electron transfer to produce more methane. The strengthening effect of voltage could be sustained for a short period of time when the voltage was removed. Microbial community analysis revealed that the changes of Methanothrix and Methanolinea resulted in higher biogas production. The increases of Smithella and Geobacter improved the possibility of "electronic syntrophism" between microorganisms and promoted the performance of DIET process. The results would provide the theoretical supports for enhancing the anaerobic treatment efficiency by voltages.

Abstract:Electrode biofilm and planktonic microorganisms are two important parts in bioelectrochemical system, but the mechanism of planktonic microorganisms affect biofilms is not clear. Waste sludge lysate produced from sludge pyrolysis contains melanoidins as complex carbon source that are refractory. Anaerobic digestion companied with weak electrostimulation is adopted to treat the wastewater and explore the mechanism in electron transfer, biofilm activity and microbial community structure. In the presence of planktonic microorganisms, electrode biofilm still plays a major role that about 70% of organics was removed by biofilm, the energy recovery efficiency increased by 10.81%, and the main electron transfer mechanism of biofilm is unchanged, but extra electron transfer pathway (Ef = 0.404 V vs Ag/AgCl) is generated. More cytochrome is produced in biofilm and charge transfer resistance decreases to facilitate electron transfer. The planktonic microorganisms cause more electrochemically active microorganisms and methanogens enrich in the electrode biofilm to enhance the direct interspecies electron transfer.
Electrostimulation Triggers an Increase in Cross-Niche Microbial Associations Toward Enhancing Organic Nitrogen Wastewater Treatment.

Abstract:As an efficient wastewater pretreatment biotechnology, electrostimulated hydrolysis acidification (eHA) has been used to accelerate the removal of refractory pollutants, which is closely related to the effects of electrostimulation on microbial interspecies associations. However, the ecological processes underpinning such linkages remain unresolved, especially for the microbial communities derived from different niches, such as the electrode surface and plankton. Herein, the principles of cross-niche microbial associations and community assembly were investigated using molecular ecological network and phylogenetic bin-based null model analysis (iCAMP) based on 16S rRNA gene sequences. The electrostimulated planktonic sludge and electrode biofilm displayed significantly (P < 0.05) 1.67 and 1.53 times higher organic nitrogen pollutant (azo dye Alizarin Yellow R) degradation efficiency than non-electrostimulation group, and the corresponding microbial community composition and structure were significantly (P < 0.05) changed. Electroactive bacteria and functional degraders were enriched in the electrode biofilm and planktonic sludge, respectively. Notably, electrostimulation strengthened the synergistic microbial associations (1.8 times more links) between sludge and biofilm members. Additionally, both electrostimulation and cross-niche microbial associations induced greater importance of deterministic assembly. Overall, this study highlights the specificity of cross-electrode surface microbial associations and ecological processes with electrostimulation and advances our understanding of the manipulation of sludge microbiomes in engineered wastewater treatment systems.

Abstract:A dielectric barrier discharge (DBD) pretreatment of waste activated sludge (WAS) was developed for boosting the releasing of organic matter from floc structures and microbial cells. The changes of protein and polysaccharide in different extracellular polymeric substances layers of SCOD releasing from sludge were determined by using three-dimensional fluorescence spectra. Significantly improved cumulative methane production was achieved via enhancing anaerobic digestion, which was 83.5% higher than that of the control. The formed center dot OH radicals play a key role in sludge hydrolysis, thus improving the biodegradability of WAS. Furthermore, highthroughput sequencing analysis showed that DBD pretreatment improved the relative abundances of the main functional microbes with respect to sludge hydrolysis and anaerobic digestion. Therefore, this study verifies the feasibility of DBD pretreatment in sludge hydrolysis and is expected to provide a practical new technology for anaerobic digestion of WAS.
Electrical Stimulation Enhanced Denitrification of Nitrite-Dependent Anaerobic Methane-Oxidizing Bacteria

Abstract:As a process utilizing methane as the sole electron source to reduce nitrite, nitrite-dependent anaerobic methane-oxidizing (n-damo) shows high potential to be applied in energy-efficient wastewater treatment. However, the metabolism of n-damo bacteria is far too low for practical application. In this study, bioelectrical reactor (BER) was used to enhance the metabolism. The nitrite removal rate was accelerated under applied electric field. The best stimulating effect was obtained at 1 V applied with the denitrification rate increased to 1.8 fold compared to conventional n-damo process. The enhancement of nitrite consumption was consistent well with the acceleration of methane depletion, further confirmed the electrical stimulation of n-damo process. However, the enhancement could only be realized under continuous electrical exposure mode, rather than intermittent mode. The above results demonstrated that electrical stimulation could lead to an enhanced denitrification of n-damo bacteria. (c) 2015 Elsevier B.V. All rights reserved.
Intermittent Electrostimulation-Modified Direct Interspecies Electron Transfer for Enhanced Methanogenesis in Anaerobic Digestion of Sulfate-Rich Wastewater

Abstract:Methane recovery and organics removal in sulfate (SO42-)-rich 4 2- )-rich wastewater anaerobic digestion are hindered by electron competition between methanogenesis and sulfidogenesis. Here, intermittently electrostimulated bioelectrodes were developed to facilitate direct interspecies electron transfer (DIET)-driven syntrophic methanogenesis, increasing substrate competition among methanogenic archaea (MA). By optimising the electrochemical environment, MA was able to employ electron transfer more efficiently than sulfate-reducing bacteria (SRB), resulting in significant methane accumulation (58.1 f 1.0 mL-CH4/m3 4 /m 3 reactor) ) and COD removal (90.5 f 0.5 %) at lower COD/SO42- 4 2- ratio. Intermittent electrostimulation improved the metabolic pathway for electroactive bacteria to utilize acetate and direct electrons to electrotrophic MA, decreasing SRB abundance and affecting the sulfate reduction pathway. Intermittently electrostimulated biofilms significantly increased gene levels of key enzymes in electron transport for cytochrome and e-pili biosynthesis, crucial for DIET, demonstrating enhanced DIET-driven syntrophic methanogenesis. This study provides a strategic approach to optimize methanogenesis in sulfate-rich wastewater anaerobic digestion.

Abstract:Introduction of voltage or conductive materials have been proven to be effective strategies for enhancing the stability and efficiency of sludge digestion for methane recovery. This experiment simulated a two-phase anaerobic system, consisting of an acid-producing phase with alkali pretreatment and a methane-producing phase with immobilized electrodes and voltage enhancement. Various effective and comparative experiments were conducted to investigate the crucial roles of immobilized electrodes like carbon brush (CB) and nickel foam (NF), electroactive biofilm and voltage during the sludge methane-producing phase. The experimental results indicate that directly application of immobilized CB and NF electrode to promote sludge anaerobic digestion achieves better performance than additional voltage supply. The electrodes can increase methane production by 38% to 94%, whereas under additional voltage supply, methane production only increases by 22%–53% compared to the control. Through the analysis of key transformations in organic substrates and changes in microbial community structure, the inherent advantages of electrode materials in promoting the syntrophic methanogenesis processes are confirmed. Compared to suspended sludge, the types and relative abundance of typical syntrophic methane-producing microorganisms, Methanosarcina , and acetogenic bacteria, Syntrophomonas on the electrode surface significantly increase by 8.5% and 1.7%. This process is notably superior to the methane generation process dominated by hydrogenotrophic methanogensis after applying voltage. The results indicate that CB and NF are suitable conductive materials to promote anaerobic methane recovery, and their effectiveness surpasses that of applying voltage. These research outcomes provide new insights for the development of novel anaerobic digestion enhancement systems.

Abstract:Bioelectrochemical is a potential measure for treating waste activated sludge(WAS) and recovering energy by improving methane production efficiency of anaerobic digestion(AD). A lab-scale digester, which equipped with a MEC bioanode and cathode(MEC-AD), was operated under three voltages (0V, 0.6V, 0.8V) at 35 ± 1℃ in order to examine the effects of the microbial electrolysis cell (MEC) reactions on anaerobic digestion of WAS . The concentration of acetic acid, propionic acid, n-butyric acid, iso-butyric acid, n-valeric acid and iso-valeric acid increased with the increasing of voltage due to the direct oxidation of the short-chain fatty acids at the bioanode as well as the indirect contribution of low acetic acid concentration on beta-oxidation. The time required for COD and protein degradation of the MEC-AD at 0.6V and 0.8V were shortened more than 7 days compared to 0V. At the end of the reaction, the concentration of sugar and protein in the MEC-AD at both 0.6V and 0.8V were 300 mg/L and 200 mg/L lower than that at 0V, respectively. Compared to 0V, The methane production cycle at 0.6V of MEC-AD was shortened about 7 days as well, indicating that the applied voltage was a very effective way to achieve high-efficiency anaerobic digestion of WAS.

Abstract:It has been more than one century since the activated sludge process was invented. Despite its proven stability and reliability, the energy (especially the electrical energy) use in wastewater treatment should evolve to meet the increasingly urgent demand of energy efficiency. This paper discusses how the energy utilized in conventional biological wastewater treatment can be altered by switching the indirect energy input to a direct electricity injection, which is achieved by the electrode integration providing extra thermodynamic driving force to biodegradation. By using electrodes instead of oxygen as terminal electron acceptors, the electrical energy can be utilized more efficiently, and the key of direct use of electrical energy in biodegradation is the development of highly active electroactive biofilm and the increase of electron transfer between microbes and the electrode. Furthermore, the synergy of different microbial electrochemical units has additional benefit in energy and resource recovery, making wastewater treatment more sustainable.

Abstract:Batch tests were operated to investigate effects of initial pH (3,5,7,9,11) on anaerobic digestion of waste activated sludge under 0.6V direct electrical stimulation. Initial pH had a great influence on biogas production and VS removal efficiency. At the optimal initial pH of 9, maximum methane yield of 224mLCH4/g VS and VS removal efficiency of 38.1% were achieved after 32days digestion. Initial pH could accelerate hydrolysis and acidification process of waste activated sludge. Alkaline environment (pH 9, 11) enhanced SCOD and VFAs concentrations in suspensions, especially at pH 11. Acetic acid and butyric acid were dominant at pH 3and 11while acetic acid and propionic acid were main products at pH 5, 7 and 9. Initial pH facilitates release of ammonium from waste activated sludge and in acidic environment ammonium was released in excess.

Abstract:The operation of biological processes may face the challenges of system instability and performance deterioration under different stresses, including transient shock of toxic matters/substrates in wastewater and cold temperatures. Bioaugmentation (BA) has been considered as a useful strategy playing an excellent role for performance enhancement and recovery in biosystems under various stresses due to the improvement of detrimental conditions, retention and adaptation of BA consortium, reactivation, and quorum-sensing effect. However, BA faces the potential risk of functional failure, even negative effects on biosystems. Measures should be used to resolve this risk, such as balancing the relationship between BA functional consortium and indigenous organisms, substrate, physical and chemical conditions, and so on. The application and mechanisms of BA, as an effective tool widely used in different stress scenarios, are detailed and discussed. Especially, the key point of successful application of BA is quantifying when and how BA is effectively implemented. Therefore, in-depth research on the BA time, BA dosage, BA interval with their relationship in different biosystems was suggested in the future.

Abstract:An increasing interest is devoted to combined microbial electrolysis cell-anaerobic digestion (MEC-AD) system which could convert waste activated sludge into biogas. In this study series tests were initially conducted to study the effect of alkaline pretreatment on AD system and the results showed that alkaline pretreatment could promote the dissolution of organic matters in the sludge and thus improve the methane production. Then, the methane production in combined MEC-AD system fed with alkaline-pretreated sludge was investigated. The results indicated that the methane productions increased by 37% and 42% when applied voltage was 0.5 V and 0.8 V. The microbial electrochemical system strongly promoted the growth of Euryarchaeota (Methanosaeta and Methanobacterium ). Meanwhile, the abundance of Paraclostridium increased from 17.9% to 38.5% when applied voltage was 0.8 V, suggesting an enhanced fermentation and acetogenesis process. The results of energy balance estimation indicated that MEC-AD system at 0.5 V could achieve higher net energy output.

Abstract:The invention discloses a method for promoting anaerobic digestion of sludge through microbial electrical mediation, and relates to a method for fast generating methane from organic waste biomass and at the method is used for solving the technical problems that the existing sludge anaerobic fermentation technology is difficult in sludge floc wall breakage, organic matters in sludge microbial cells is difficult to fully release to a liquid phase, and the utilization rate of organic matters of the sludge and the recovery rate of methane are low. The method comprises the following steps: I, pretreating sludge; II, performing the function microbial acclimation of an anode electrolysis flora; III, promoting the anaerobic digestion of sludge through microbial electrical mediation. According to the method, protons in a system can be more quickly converted by means of the electric mediation action of a microbial catalytic electrolysis system, complicated organic acid ingredients can be effectively converted in an electronic manner so as to eliminate the inhibition role of acid accumulation in the system, and the anaerobic digestion of sludge and the speed and yield of the generated methane can be effectively promoted. The method belongs to the fields of promoting sludge to generate methane through microbial electrolysis.
Integrated microbial electrolysis with high-alkali pretreated sludge digestion: Insight into the effect of voltage on methanogenesis and substrate metabolism

Abstract:Integrated microbial electrolysis with anaerobic digestion is proved to be an effective way to improve methanogenesis efficiency of waste activated sludge (WAS). WAS requires pretreatment for efficient improvement of acidification or methanogenesis efficiency, but excessive acidification may inhibit the methanogenesis. In order to balance these two stages, a method for efficient WAS hydrolysis and methanogenesis has been proposed in this study by high-alkaline pretreatment integrated with microbial electrolysis system. The effects of pretreatment methods and voltage on the normal temperature digestion of WAS have also been further investigated with emphasis on the effects of voltage and substrate metabolism. The results show that compared to low-alkaline pretreatment (pH = 10), high-alkaline pretreatment (pH > 14) can double the SCOD release and promote the VFAs accumulation to 5657 ± 392 mg COD/L, but inhibit the methanogenesis process. Microbial electrolysis can alleviate this inhibition effectively through the rapid consumption of VFAs and speeding up of the methanogenesis process. The optimal methane yield of the integrated system is 120.4 ± 8.4 mL/g VSS at the voltage of 0.5 V. Enzyme activities, high-throughput and gene function prediction analysis reveal that the cathode and anode maintain the activity of methanogens under high substrate concentrations. Voltage positively responded to improved methane yield from 0.3 to 0.8 V, but higher than 1.1 V is found to be unfavorable for cathodic methanogenesis and results in additional power loss. These findings provide a perspective idea for rapid and maximum biogas recovery from WAS.

Abstract:Electrostimulated hydrolysis acidification (eHA) has been used as an efficient biological pre-treatment of refractory industrial wastewater. However, the effects of electrostimulation on the function and assembly of planktonic anaerobic sludge microbial communities are poorly understood. Using 16S rRNA gene and metagenomic sequencing, we investigated planktonic sludge microbial community structure, composition, function, assembly, and microbial interactions in response to electrostimulation. Compared with a conventional hydrolysis acidification (HA) reactor, the planktonic sludge microbial communities selected by electrostimulation promoted biotransformation of the azo dye Alizarin Yellow R. The taxonomic and functional structure and composition were significantly shifted upon electrostimulation with azo dyes degraders (e.g. Acinetobacter and Dechloromonas) and electroactive bacteria (e.g. Pseudomonas) being enriched. More microbial interactions between fermenters and decolorizing and electroactive bacteria, as well as fewer interactions between different fermenters evolved in the eHA microbial communities. Moreover, the decolorizing bacteria were linked to the higher abundance of genes encoding for azo- and nitro-reductases and redox mediator (e.g. ubiquinone) biosynthesis involved in the transformation of azo dye. Microbial community assembly was more driven by deterministic processes upon electrostimulation. This study offers new insights into the effects of electrostimulation on planktonic sludge microbial community function and assembly, and provides a promising strategy for the manipulation of anaerobic sludge microbiomes in HA engineering systems.
Facilitating Solid-State Anaerobic Digestion of Food Waste Via Bio-Electrochemical Treatment

Abstract:Food waste has become a global environmental concern over emissions of greenhouse gas and odorous pollutants. Anaerobic digestion is gaining increasing attention as an effective food waste disposal method combining waste minimization and bioenergy recovery. Solid-state anaerobic digestion can handle food waste containing high contents of solids and has the merits of higher disposal volumes and less parasitic energy input, however, the low energy/mass transfer impairs the treatment efficiency. This study incorporated bio-electrochemical treatment into solid-state anaerobic digestion of food waste to overcome these shortcomings for enhanced performances. Food waste liquid leachate was used as feedstock to validate the bio-electrochemical treatment effects under different conditions. At a low applied voltage of 0.7 V and initial pH of 8.13, the methane yield increased by 77.5% while the carbon dioxide yield decreased by 16.0% compared with the control without electrodes. Although higher voltages (1.1-2.0 V) contributed to higher methane and hydrogen yields, the fast consumption and breakages of anodes significantly decreased the treatment consistency and material lifespan. Thereby, 0.7 V was selected as the applied voltage when incorporating the bio-electrochemical unit into the solid-state digester treating food waste. Cathodic hydroxide generation increased the buffering capacity, thus contributing to a more stable start-up process. With an immersed electrode surface area of 25.2 cm2/L, the highest methane yield of 526.7 mL/gVS was recorded with lower carbon dioxide and hydrogen contents initially, and the peak gaseous hydrogen sulfide emission was significantly reduced by 71.5% mainly due to the ferrous ions release from low carbon steel anode for sulfide precipitation.

Both biochar and electric field stimulation accelerated AD start-up, and the coupling strategy promoted volatile fatty acids (VFA) oxidation and methanogenesis, leading to a 34.1 % increase in accumulated methane yield during a 50-day operation period. Microbial characterization analysis revealed that biochar primarily upregulated the secretion of protein-like compounds in tightly-bound extracellular polymeric substances, and the electroactive matrix involving bulk sludge and biochar was likely established to mediate syntrophic methanogenesis through extracellular electron transfer (EET). Comparatively, electric field mainly governed the electrochemical behaviors and microbial community evolution of the electrode biofilm, which boosting the electrochemical activity of both anode and cathode, and increasing the cell viability of cathodic biofilm. It also selectively enriched Syntrophomonas in the anode and filamentous Methanospirillum in the cathode, which contributed to syntrophic methanogenesis in the electric stimulation groups. Overall, the outcome of this research underlined the different roles of biochar and electric field in boosting methanogenesis, and confirmed the synergistic benefits of the coupling stimulation strategy.

Abstract:This study explores the treatment of low-organic strength wastewater by integrating bio-electrochemical technology with low-temperature anaerobic digestion . Composite electrode plates acted as carriers, maintaining an operating temperature of 17 °C through gradual acclimation. Results indicate that the bio-electrochemical reactor (R1) with an applied voltage of 0.6 V and electrode carriers, and the reactor (R2) with electrode carriers but without applied voltage, achieved soluble chemical oxygen demand (SCOD) removal rates of 92.2 % and 91.2 %, respectively. These rates were approximately 1.3 times higher than those of the conventional reactor (R3). Furthermore, the average SCOD concentration in R1 effluent was 15.95 % lower than that in R2. The application of voltage notably enhanced wastewater treatment efficacy at 17 °C in R1. Abundant electroactive microorganisms capable of direct interspecies electron transfer (DIET) were observed in both R1 and R2, promoting an accelerated electron transfer rate. Additionally, the electrodes facilitated a coexisting system wherein methane production increased due to a combination of a methane-producing system driven by H 2 mediation and an electroactive methanogenic system.

Abstract:Anaerobic digestion is widely used for the treatment of wastewater for its low costs and bioenergy production, but the performances of anaerobic digestion often need improving in practical applications. The addition of conductive materials could lead to direct interspecies electron transfer (DIET) among the anaerobic microorganisms, and consequently enhance the efficiencies of anaerobic digestion. In this paper, the effects of DIET via conductive materials on chemical organic demand (COD) removal, volatile fatty acid (VFA) consumption and methane production were reviewed. The reports on the increase of conductive microorganisms due to the addition of conductive materials were discussed. Results regarding activities of microorganisms and morphology and properties of sludge were described and commented, and future research needs were also proposed which included better understanding of the roles of DIET in each step of anaerobic digestion, mechanisms of metabolism of pollutants in DIET-established systems and inhibition of excessive dosage of conductive materials.

Abstract:Due to highly recalcitrant and toxicological nature of pyridine, the conventional anaerobic bioprocess is often limited by low removal rate and poor process stability. In this study, an electricity-assisted anaerobic system was developed in order to enhance biodegradation of pyridine from wastewater. The results showed that the performance and stability of the anaerobic reactor was remarkably improved for pyridine biodegradation with the applied direct current of 0.3 mA, where the efficiencies of pyridine and total organic carbon removal as well as NH4+-N formation were as high as 100.0%, 96.1 ± 1.2% and 60.1 ± 2.1% respectively. The compact biofilm due to electrical stimulation as well as the microaerobic environment in the bioanode might promote pyridine bio-mineralization in the anaerobic reactor. Moreover, the species related to pyridine biodegradation (Desulfovibrio, Dokdonella, Hydrogenophaga, and Paracoccus) were enriched in the anodic biofilm, which would be another reason for better reactor performance. This study demonstrated that electrical stimulation would be a potential alternative for the enhancement of pyridine removal from wastewater in anaerobic systems.
