Abstract:A concise and efficient synthetic approach was developed for the synthesis of functionalized chromeno[4,3-b]pyrrol-4(1H)-ones via a one-pot, three-component reaction of 4-aminocoumarin, arylglyoxal monohydrates, and aromatic amines promoted by KHSO4 in toluene under reflux condition. The attractive features of this protocol are operational simplicity, short reaction time, easy purification of products and good to excellent yields.

Abstract:A one-pot, three-component reaction involving SNAr-decarboxylative redox isomerization starting from an electron-withdrawing group bearing fluorobenzaldehyde or fluorothiophenecarbaldehyde (electron-pair diacceptor), secondary amine, and trons-4-hydroxy-L-proline or indoline-2-carboxylic acid (electron-pair donor) has been developed to give N-[(pyrrolylmethyl)-substituted (het)aryl]- and N-[(indolylmethyl)-substituted (het)aryllpyrrolidines, -piperidines, -morpholines, and -azocanes. The highlights of this protocol are its great convenience in the simultaneous construction of a C(sp(2))-N bond and a C(sp(3))-N bond with simple operation; the reaction can be performed on a gram scale. This efficient one-pot multicomponent reaction has potential as a novel method for drug synthesis, especially involving polycyclic compounds.

Abstract:The exploration of novel chemical reactivities of readily available feedstocks has always been a hot topic in the synthetic community. We herein report a three-component one-pot reaction with TMSCF 2 Br, p -QMs and 1,3-dicarbonyl compounds and analogues, providing a modular protocol for the construction of densely functionalized tetrasubstituted furans and pyrroles. While TMSCF 2 Br has been widely used as a difluorocarbene or trimethylsilyldifluoromethyl radical precursor, it herein acts as a surrogate of "naked" carbon. Control experiments indicate that repeated substitution/elimination processes are involved. This reaction features high step-economy, mild conditions and excellent chemoselectivity. The phenolic hydroxyl groups of products offer a platform for the late-stage modification of furans and pyrroles, which could enrich the structural diversity of the products rapidly.

Abstract:A chemselective catalyst-free three-component 1,3-dipolar cycloaddition has been described. The unique polycyclic THPI and THIQs were creatively employed as dipolarophiles, which led to the formation of functionalized β-tetrahydrocarboline- and tetrahydroisoquinoline-fused spirooxindoles in 60–94% of yields with excellent diastereoselectivities (10: 1−>99: 1 dr). This reaction not only realizes a concise THPI- or THIQs-based 1,3-dipolar cycloaddition, but also provides a practical strategy for the construction of two distinctive spirooxindole skeletons.

Abstract:A novel method has been developed to synthesize a unique class of highly functionalized isochromeno[4,3-c]pyridazines. This reaction features an intermolecular functionalization of terminal nitrogen atom of diazo group of 4-diazoisochoman-3-imine with two dimethylsulfonium ylide components, followed by a base promoted 6-exo-trig cyclization step. Readily available starting materials, a broad substrate scope, and operationally simple, mild, and catalyst-free reaction conditions are the prominent features of this method.

Abstract:The additive‐free 1,3‐dipolar cycloaddition reaction of isatin‐derived azomethine ylides with α‐cyano‐α,β‐unsaturated compounds was developed, which enabled diversity‐oriented synthesis of a series of novel and structurally complex 3,3′‐pyrrolidinyl‐spirooxindoles derivatives containing four contiguous and two quaternary stereogenic centers in high yields (up to 92%) and excellent diastereoselectivities (up to >25:1 dr). The reaction displays switchable regioselectivity depending on the steric effect of the substrates.

This process represents a novel spiroisocchroman synthesis protocol and is featured by good functional group compatibility, mild conditions and readily available substrates.

Abstract:A novel one-pot pseudo-three-component reaction of 2-amino-4H-chromen-4-ones with 4-benzylidene-5-alkyl-2-aryl-2,4-dihydro-3H-pyrazol-3-ones was investigated for the synthesis of the spiro[chromeno[2,3-b]pyridine-3,4'-pyrazole] derivatives. This procedure involved Michael addition, elimination, and hetero-Diels-Alder sequences, affording a series of spiropyrazolone-chromeno[2,3-b]pyridines in good yields that possessed cis relationships between two aryl groups and the carbonyl of the pyrazolone unit.

Abstract:We reported an efficient three-component reaction to access new spiro heterocycles through the annulation reactions of isatins, substituted ureas, and cyclic ketones under normal laboratory conditions, which is another example of isatins being used to build spiro compounds by the ring-opening and recyclization processes. The wide range of substrates, simple operation, normal experimental conditions, and high yields make the approach of high practical value.

Abstract:Under bifunctional squaramide and thiourea organocatalysis, a series of vicinally bis(trifluoromethyl)-substituted 3,3′-pyrrolidinyl spirooxindoles were successfully synthesized via 1,3-dipolar cycloaddition reactions of 3-(trifluoroethylidene)oxindoles with diethyl 2-((2,2,2-trifluoroethyl)imino)malonate and N-(2,2,2-trifluoroethyl)isatin ketimines. The highlight of this protocol is the efficient construction of the vicinally bis(trifluoromethyl)-substituted pyrrolidine unit of spirooxindoles, including multiple contiguous stereogenic centres, in excellent yields and stereoselectivities.

This strategy can provide direct and rapid access to spiropyrrolizidine oxindoles in high yields (up to 99%) with excellent diastereoselectivities (up to 99: 1 dr). Abstract:An efficient one-pot synthesis of isoquinolines and heterocycle-fused pyridines by three-component reaction of aryl ketones, hydroxylamine, and alkynes is developed. The reaction involves condensation of aryl ketones and hydroxylamine, rhodium(III)-catalyzed C-H bond activation of the in situ generated aryl ketone oximes, and cyclization with internal alkynes. This protocol enables rapid assembly of multisubstituted isoquinolines as well as γ-carbolines, furo[2,3-c]pyridines, thieno[2,3-c]pyridines, and benzofuro[2,3-c]pyridines from readily available substrates.

Abstract:A novel one-pot three-component regioselective synthesis of highly substituted pyridin-2-one derivatives starting from simple and readily available starting materials is described. The reactive zwitterionic intermediate generated by the addition of an isocyanide to dimethyl acetylenedicarboxylate (DMAD) was trapped with N-arylidene-2-cyanoacetohydrazides to afford the title compounds in moderate to good yields at room temperature without using any catalyst and other additives.

Abstract:A facile synthetic protocol for the preparation of spiro diheterocycles from easily available 2'-aminoacetophenones, isocyanates and 1,4-benzoquinones or quinone monoimines under mild conditions has been developed. This multi-component transformation is characterized by efficient construction of two heterocycles and one valuable quaternary carbon in one pot via an in situ cyclization-respiroannulation strategy. Moreover, this reaction showed a broad substrate scope, and generated diverse five-membered oxaspiros.
