Abstract:This study introduces a novel ice lithography system integrated with a low-vibration micro-machined Joule-Thomson cooler. Ice lithography is an eco-friendly method for high-resolution nanofabrication on delicate substrates and requires to operate below 130 K in the vacuum of a microscope chamber. Previously, liquid nitrogen rather than cryocoolers was used to cool the system due to its low vibration, but it is both bulky and costly. To overcome these challenges, this study employs a low-vibration micro-machined cooler in a scanning electron microscope (SEM) for ice lithography. The design and methodology of the system are described in detail. The results show that the substrate can reach a temperature of 105 K within 30 minutes with a measured mechanical vibration of less than 10 nm, enabling high-resolution nanofabrication. As proof of concept, the system successfully fabricates nanoscale patterns on a silicon wafer. This low-vibration cooling system has great potential for use in cryogenic electron microscopes (cryo-EMs) in the future.

Abstract:In the practices of cryobiology, selection of an optimum freeze/thawing program and an idealistic cryoprotective agent often requires rather tedious, time consuming and repetitive tests. Integrating the functions of sample preparation and viability detection, the concept of biochip technology was introduced to the field of cryopreservation, aiming at quickly finding an optimum freezing and thawing program. Prototype devices were fabricated and corresponding experimental tests were performed. It was shown that microflow-channel chip could not offer a high quality solution distribution. As an alternative, the spot-dropping chip proved to be an excellent way to load the sample quickly and reliably. Infrared thermal mapping on such a chip showed that it had a rather uniform heat transfer boundary. Applying the spot-dropping chip combined with the thermoelectric cooling device, the final output of cryopreservation of multiple samples was tested, and the optimal freeze/thawing program as well as the potentially best concentration of the cryoprotective agent was found by analyzing the results. Further, application of this technique to measure the thermo-physical properties of the cryo-protective agent was also investigated. The study demonstrated that a biochip with integrated automatic loading and inspection units opens the possibility of a massive optimization of the complex cryopreservation program in a quicker and more economical way.
A thermo-responsive PNIPAAm-grafted-PDMS surface used for cell culture in microfluidic channels

Abstract:A thermo-responsive PNIPAAm-grafted-PDMS (PNIPAAm-g-PDMS) surface was used for cell culture. Cos7 cells could adhere to the PNIPAAm-g-PDMS surface and proliferated therein at 37°C. The cultured cells could detach from the surface at room temperature without trypsin digestion. The viability of cells harvested from the PNIPAAm-g-PDMS surface via lowering temperature was higher than that of cells harvested from native PDMS surface via trypsin digestion. A PNIPAAm-g-PDMS microchannel was designed for the in-channel thermo-modulated cell culture/harvest. Cells could be long-term cultivated and be harvested without enzymatic treatment. Successful thermo-modulated cell passage operation was demonstrated in the microfluidic chips with the PNIPAAm-gPDMS channels.

Abstract:Cell culture and harvest are the most upstream operation for a completely integrated cell assay chip. In our previous work, thermoresponsive poly(N-isopropylacrylamide) (PNIPAAm) was successfully grafted onto polydimethylsiloxane (PDMS) surface via benzophenone-initiated photopolymerization. In the present work, the PNIPAAm-grafted-PDMS (PNIPAAm-g-PDMS) surface was explored for thermomodulated cell culture and noninvasive harvest in microfluidic channels. Using COS 7 fibroblast from African green monkey kidney as the model cells, the thermomodulated adhering and detaching behaviors of the cells on the PNIPAAm-g-PDMS surfaces were optimized with respect to PNIPAAm-grafting yields and gelatin modification. The viability of the cells cultured on and harvested from the PNIPAAm-g-PDMS surface with the thermomodulated noninvasive protocol was estimated against the traditional cell culture∕harvest method involving trypsin digestion. The configuration of the microchannel on the PNIPAAm-g-PDMS chip was evaluated for static cell culture. Using a pipette-shaped PNIPAAm-g-PDMS microchannel, long-term cell culture could be achieved at 37 °C with periodic change of the culture medium every 12 h. After moving the microchip from the incubator set at 37 °C to the room temperature, the proliferated cells could be spontaneously detached from the PNIPAAm-g-PDMS surface of the upstream chamber and transferred by a gentle fluid flow to the downstream chamber, wherein the transferred cells could be subcultured. The thermomodulated cell culture, harvest, and passage operations on the PNIPAAm-g-PDMS microfluidic channels were demonstrated.
Investigation of Rapid Rewarming Chips for Cryopreservation by Joule Heating

Abstract:Rapid and uniform rewarming is critical to cryopreservation. Current rapid rewarming methods require complex physical field application devices (such as lasers or radio frequencies) and the addition of nanoparticles as heating media. These complex devices and nanoparticles limit the promotion of the rapid rewarming method and pose potential biosafety concerns. In this work, a joule heating-based rapid electric heating chip (EHC) was designed for cryopreservation. Uniform and rapid rewarming of biological samples in different volumes can be achieved through simple operations. EHC loaded with 0.28 mL of CPA solution can achieve a rewarming rate of 3.2 × 10<sup>5</sup> °C/min (2.8 mL with 2.3 × 10<sup>3</sup> °C/min), approximately 2 orders of magnitude greater than the rewarming rates observed with an equal capacity straw when combined with laser nanowarming or magnetic induction heating. In addition, the degree of supercooling can be significantly reduced without manual nucleation during the cooling of the EHC. Subsequently, the results of cryopreservation validation of cells and spheroids showed that the cell viability and spheroid structural integrity were significantly improved after cryopreservation. The viability of human lung adenocarcinoma (A549) cells postcryopreservation was 97.2%, which was significantly higher than 93% in the cryogenic vials (CV) group. Similar results were seen in human mesenchymal stem cells (MSCs), with 93.18% cell survival in the EHC group, significantly higher than 86.83% in the CV group, and cells in the EHC group were also significantly better than those in the CV group for further apoptosis and necrosis assays. This work provides an efficient rewarming protocol for the cryopreservation of biological samples, significantly improving the quantity and quality of cells and spheroids postcryopreservation.

In this paper a series of directional freezing processes and cell responses in a culture microchip were experimentally investigated. The temperature in the microchip was manipulated by a thermoelectric cooler. The surviving cells, necrotic and apoptotic cells under different cryotreatment (duration of the freezing process, freeze-thaw cycle, post-culture et al) were stained and distinguished by PI and FITC-Annexin V. The locations of the ice front and cell death boundary were observed and recorded through an inverted microscopy. By controlling the cooling process in a microfluidic channel, it is possible to recreate a sketch of biological effect during the process of simulated cryosurgery.
Premelting-directed interfacial design of nanoshell-coated cells in directional freezing and thawing

Abstract:Freezing interface interacting with soft cells is a core issue in cryopreservation. However, most of existing cryopreservation approaches face challenges such as complex processing and poor controllability. Herein we report a new, nanotechnology-based method to modulate interfacial interactions between ice and cells via the premelting theory. Through the interfacial design by controllably modifying biocompatible nanoshells on the surface of cells, the effective Hamaker constant between cells and ice can be modified. The thickness of premelted films between cells and ice is further regulated, and directional migration of coated cells occurs in a thermal gradient to achieve directional thawing of coated cells. The decreased mortality of freezing coated cells suggests the premelting modulation in freezing and thawing can effectively protect cells from mechanical damage of ice formation during freezing and ice recrystallization during thawing.

In clinical applications, the growth of ice ball must reach a suitable size as cells could not be all killed at the ice periphery. The cell death ratio at the ice periphery is important for the control of the freezing destruction. The mechanisms of cryoinjury around the ice periphery need thorough understanding. In this paper, a primary freeze-thaw control was carried out in a cell culture microchip. A series of directional freezing processes and cell responses was tested and discussed. The temperature in the microchip was manipulated by a thermoelectric cooler. The necrotic and apoptotic cells under different cryotreatment (duration of the freezing process, freeze-thaw cycle, postculture, etc.) were stained and distinguished by propidium iodide and fluorescein isothiocyanate (FITC)-Annexin V. The location of the ice front was recorded and a cell death boundary which was different from the ice front was observed. By controlling the cooling process in a microfluidic channel, it is possible to recreate a sketch of biological effect during the process of simulated cryosurgery.

Abstract:On-chip cell culture is a basic technique for an integrated lab on a chip for cell-based assays.However,the process has low cell viability and contamination problems of the on-chip cell culture.This paper presents a protocol including polydimethylsiloxane(PDMS)-glass microfluidic chip fabrication,preparation,and cell treatment methods for on-chip cell culture.Several factors to improve the viability of the cells cultured in the channels are given including cell preparation tips and ways to avoid bubbles,chip surface treatments,and medium serum concentrations.Several mammalian cells were successfully cultured on such chips using these methods.

Abstract:The storage of living cells is the major challenge for cell research and cell treatment. Here, we introduced a novel supramolecular gel cryopreservation system which was prepared in the microchannel, and the supramolecular gel (BDTC) was self-assembled by gelator Boc-O-dodecyl-L-tyrosine (BDT). This cryopreservation system could obviously minimize the cell injury because the BDTC supramolecular gel had a more compact three-dimensional network structure when the BDT gelator self-assembled in the confined space of microchannel. This compact structure could confine the growth of the ice crystal, reduce the change rate of cell volumes and osmotic shock, decrease the freezing point of the cryopreservation system, and possess better protection capability. Furthermore, the results of functionality assessments showed that the thawed cells could grow and proliferate well and remain the same growth trend of the fresh cells after the RSC96 cells flowed out from the microchannel. This novel method has potential to be used for the cryopreservation of cells, cell therapy, and tissue engineering.

Abstract:Preservation of evolved biological structure and function in robust engineering materials is of interest for storage of biological samples before diagnosis and development of vaccines, sensors, and enzymatic reactors and has the potential to avoid cryopreservation and its associated cold-chain issues. Here, we demonstrate that "freezing cells in amorphous silica" is a powerful technique for long-term preservation of whole mammalian cell proteomic structure and function at room temperature. Biomimetic silicification employs the crowded protein microenvironment of mammalian cells as a catalytic framework to proximally transform monomeric silicic acid into silicates forming a nanoscopic silica shell over all biomolecular interfaces. Silicification followed by dehydration preserves and passivates proteomic information within a nanoscale thin silica coating that exhibits size selective permeability (<3.6 nm), preventing protein leaching and protease degradation of cellular contents, while providing access of small molecular constituents for cellular enzymatic reaction. Exposure of dehydrated silicified cells to mild etchant or prolonged hydrolysis removes the silica, completely rerevealing biomolecular components and restoring their accessibility and functionality.

Abstract:Oocyte cryopreservation is essential in the field of assisted reproduction, but due to the large size and poor environmental tolerance of oocytes, cell freezing technology needs further improvement. Here, we ingeniously devised a Y‐shaped microfluidic chip based on three‐dimensional graphene by combining laser‐induced graphene (LIG) technology and fiber etching technology. The prepared LIG/PDMS microfluidic chip can effectively suppress ice crystal size and delay ice crystal freezing time by adjusting surface hydrophobicity. In addition, LIG endows the microfluidic chip with an outstanding photothermal effect, allowing us to sharply increase its surface temperature from 25 °C to 71.8 °C with 10 s of low‐power 808 nm laser irradiation (0.4 W/cm2). Notably, the LIG/PDMS microfluidic chip not only replaces the traditional cryopreservation carriers, but also effectively reduces the dosage of cryoprotectants (CPAs) needed in mouse oocyte cryopreservation. Even when the concentration of CPAs is cut in half (final concentration of 7.5% EG and 7.5% DMSO), the survival rate of oocytes is still as high as 92.4%, significantly higher than the control group's 85.8%. Therefore, our work provides a novel design strategy to construct multifunctional microfluidic chips for high‐performance oocytes cryopreservation. This article is protected by copyright. Abstract:Abstract Background The cryopreservation and thawing processes are known to induce many deleterious effects in cells and might be detrimental to several cell types. There is an inherent variability in cellular responses among cell types and within individual cells of a given population with regard to their ability to endure the freezing and thawing process. The aim of this study was to evaluate the fate of cryopreserved cells within an optical cryo apparatus, the individual-cell-based cryo-chip (i3C), by monitoring several basic cellular functional activities at the resolution of individual cells. Results In the present study, U937 cells underwent the freezing and thawing cycle in the i3C device. Then a panel of vital tests was performed, including the number of dead cells (PI staining), apoptotic rate (Annexin V staining), mitochondrial membrane potential (TMRM staining), cytoplasm membrane integrity and intracellular metabolism (FDA staining), as well as post-thawing cell proliferation assays. Cells that underwent the freezing - thawing cycle in i3C devices exhibited the same functional activity as control cells. Moreover, the combination of the multi-parametric analysis at a single cell resolution and the optical and biological features of the device enable an accurate determination of the functional status of individual cells and subsequent retrieval and utilization of the most valuable cells. Conclusions The means and methodologies described here enable the freezing and thawing of spatially identifiable cells, as well as the efficient detection of viable, specific, highly biologically active cells for future applications.

Abstract:Cryopreservation of biological samples is an important technology for expanding their applications in the biomedical field. However, the quality and functionality of samples after rewarming are limited by the toxicity of commonly used cryoprotectant agents (CPAs). Here, we developed a novel preservation system by combining the natural amino acid l-proline (L-Pro) with gelatin methacryloyl (GelMA) hydrogels. Compared with dimethyl sulfoxide (DMSO), L-Pro and GelMA demonstrated excellent biocompatibility when co-culturing with cells. Cryopreservation procedures were optimized using 3T3 as model cells. The results showed that rapid cooling was the most suitable cooling procedure for L-Pro and GelMA among the three cooling procedures. Co-culturing with cells for 3 h before cryopreservation, 6 % L-Pro +7 % GelMA had the highest survival rate, reaching up to 80 %. Differential Scanning Calorimetry (DSC) analysis showed that 6 % L-Pro + 7 % GelMA lowered the freezing point of the solution to -4.2 °C and increased the unfrozen water content to 20 %. To the best of our knowledge, this is the first report of cell cryopreservation using a combination of L-Pro and GelMA hydrogels, which provides a new strategy for improving cell cryopreservation.
Enhancing cell cryopreservation with acidic polyamino acids integrated liquid marbles

Abstract:Cryopreservation is highly desired for long-term maintenance of the viability of living biosamples, while effective cell cryopreservation still relies heavily on the addition of dimethyl sulfoxide (DMSO) and fetal bovine serum (FBS). However, the intrinsic toxicity of DMSO is still a bottleneck, which could not only cause the clinical side effect but also induce cell genetic variants. In the meantime, the addition of FBS may bring potentially the risk of pathogenic microorganism contamination. The liquid marbles (LMs), a novel biotechnology tool for cell cryopreservation, which not only have a small volume system that facilitated recovery, but the hydrophobic shell also resisted the harm to cells caused by adverse environments. Previous LM-based cell cryopreservation relied heavily on the addition of FBS. In this work, we introduced acidic polyaspartic acid and polyglutamic acid as cryoprotectants to construct LM systems. LMs could burst in an instant to facilitate and achieve ultrarapid recovery process, and the hydrophilic carboxyl groups of the cryoprotectants could form hydrogen bonds with water molecules and further inhibit ice growth/formation to protect cells from cryoinjuries. The L929 cells could be well cryopreserved by acidic polyamino acid-based LMs. This new biotechnology platform is expected to be widely used for cell cryopreservation, which has the potential to propel LMs for the preservation of various functional cells in the future.

Abstract:Full‐Cycle Cryogenic Protection In article number 2402312, Huanzhong Liu, Yunxia Cao, Gang Zhao, and co‐workers propose a multi‐scale ice inhibition platform, which effectively suppresses ice crystal morphology, ice recrystallization, thermal stress, and devitrification. Based on this ice inhibition platform, they achieve high‐efficiency cryopreservation of mouse oocyte using the low concentration of penetrating cryoprotective agents. Meanwhile, recovered oocytes retain normal fertilization, embryonic development, and birth to healthy offspring, offering promising prospects for achieving high‐quality fertility preservation.

Abstract:Abstract Ovarian follicle cryopreservation is a promising strategy for fertility preservation; however, cryopreservation protocols have room for improvement to maximize post-thaw follicle viability and quality. Current slow-freezing protocols use either manual ice-seeding in combination with expensive programmable-rate freezers or other clinically incompatible ice initiators to control the ice-seeding temperature in the extracellular solution, a critical parameter that impacts post-cryopreservation cell/tissue quality. Previously, sand has been shown to be an excellent, biocompatible ice initiator, and its use in cryopreservation of human induced pluripotent stem cells enables high cell viability and quality after cryopreservation. This study applies sand as an ice initiator to cryopreserve multicellular microtissue, preantral ovarian follicles, using a simple slow-freezing protocol in the mouse model. Ovarian follicles cryopreserved using the sand partially embedded in polydimethylsiloxane (PDMS) film to seed ice in the extracellular solution exhibit healthy morphology, high viability, and the ability to grow similarly to fresh follicles in culture post-thaw. This sand-based cryopreservation strategy can facilitate convenient ovarian follicle cryopreservation using simple equipment, and this study further demonstrates the translatability of this strategy to not only single cells but also multicellular tissues.

Abstract:The solution exchange process is crucial in cell cryopreservation, an assistive reproductive technique that enhances reproductive autonomy and helps women overcome infertility challenges. Such a task is time-critical in the sense that the duration of cell exposure to the solutions significantly impacts cell viability. In this paper, a new micromanipulation system has been developed to automate such a task, where the contributions can be summarized as follows. This paper addresses the challenge of tracking cell positions within a microfluidic chip, due to the limitations of the microscope’s field of view (FOV). By utilizing a region control approach with visual feedback, the proposed method ensures that the cell remains centered in the image. Additionally, a real-time tracking method based on correlation filtering is presented, which precisely localizes cells and controls the syringe pump flow rate to mitigate delays and prevent cell loss. Experimental results illustrate the consistent positioning of the micro objects/cells at each step, the satisfactory success rate, and the robustness to the loss of vision feature. Integrated with novel manipulation strategies, our intelligent manipulation system offers a promising solution for in vitro fertilization (IVF), characterized by an embryologist-centered configuration and standardized robotic manipulation. This study aims to standardize clinical cryopreservation by transitioning from manual operation to a more efficient and reliable automated process. Furthermore, this approach can simplify procedures and enhance oocyte viability. Note to Practitioners —The motivation for this study was to propose an automatic method to address the challenges of manual cell cryopreservation, such as high operational complexity, low efficiency, and significant cell damage. Existing automated cell cryopreservation methods have shortcomings as they fail to simultaneously achieve comprehensive cell tracking and avoid damage caused by direct contact between cells and microtools. Additionally, the high complexity of robotic operations makes clinical application difficult. To overcome these limitations, this study introduces a novel robotic micromanipulation method based on microfluidic chip technology. This method enables full-process cell tracking, FOV tracking of the motorized stage, and non-contact manipulation during the exchange process of cells and solutions. Experimental results showed that the proposed method outperformed existing manual methods in terms of oocyte viability. For fertility experts and medical staff, such a robotic micromanipulation system can be combined with other automatic machines to provide a solution to cell surgery, achieving higher accuracy and efficiency.
In Situ Vitrification of Lung Cancer Organoids on a Microwell Array

Abstract:Three-dimensional cultured patient-derived cancer organoids (PDOs) represent a powerful tool for anti-cancer drug development due to their similarity to the in vivo tumor tissues. However, the culture and manipulation of PDOs is more difficult than 2D cultured cell lines due to the presence of the culture matrix and the 3D feature of the organoids. In our other study, we established a method for lung cancer organoid (LCO)-based drug sensitivity tests on the superhydrophobic microwell array chip (SMAR-chip). Here, we describe a novel in situ cryopreservation technology on the SMAR-chip to preserve the viability of the organoids for future drug sensitivity tests. We compared two cryopreservation approaches (slow freezing and vitrification) and demonstrated that vitrification performed better at preserving the viability of LCOs. Next, we developed a simple procedure for in situ cryopreservation and thawing of the LCOs on the SMAR-chip. We proved that the on-chip cryopreserved organoids can be recovered successfully and, more importantly, showing similar responses to anti-cancer drugs as the unfrozen controls. This in situ vitrification technology eliminated the harvesting and centrifugation steps in conventional cryopreservation, making the whole freeze–thaw process easier to perform and the preserved LCOs ready to be used for the subsequent drug sensitivity test.
