Abstract:In this contribution, we devoted ourselves to fabricating aggregation-induced emission (AIE) activity copolymers via one-pot combination of RAFT polymerization and Biginelli reaction for the first time. When the feeding ratio of TPB was 33.5%, the molar fraction of TPB was, respectively, about 14.2 and 22.5% in PEG-PTE1 copolymers by two-step strategy and PEG-PTE2 copolymers by one-pot strategy with the similar structure. The Mn of PEG-PTE1 increased to 59,300 from 52,800 of PEG-AE presoma with narrow PDI, which was more than Mn of PEG-PTE2 with 52,300. As compared with PEG-PTE2, when the feeding ratio of TPB was 48.6%, the molar fraction of TPB increased to 32.6% in PEG-PTE3. In aqueous solution, the as-obtained PEG-PTE2 copolymers can self-assemble into fluorescent organic nanoparticles (FONs) with 100–180 nm spherical morphology, the maximal emission peak of which presented at 460 nm with the obvious AIE phenomenon. Moreover, due to the low toxicity and excellent cell dyeing behavior, the as-prepared PEG-PTE2 copolymers displayed great potential for biomedical applications.

Abstract:9-fluorenylmethoxycarbonyl-diphenylalanine (Fmoc-FF), has been has been extensively explored due to its ultrafast self-assembly kinetics, inherent biocompatibility, tunable physicochemical properties, and especially, the capability of forming self-sustained gels under physiological conditions. Consequently, various methodologies to develop Fmoc-FF gels and their corresponding applications in biomedical and industrial fields have been extensively studied. Herein, we systemically summarize the mechanisms underlying Fmoc-FF self-assembly, discuss the preparation methodologies of Fmoc-FF hydrogels, and then deliberate the properties as well as the diverse applications of Fmoc-FF self-assemblies. Finally, the contemporary shortcomings which limit the development of Fmoc-FF self-assembly are raised and the alternative solutions are proposed, along with future research perspectives.

Abstract:In this paper, a fluorescein methacryloyl monomer (named Flu-MA) was firstly fabricated by the esterification reaction of fluorescein and methacryloyl chloride; subsequently, a novel amphiphilic fluorescent copolymer (Flu-PEG) with smart pH sensitivity was successfully prepared by RAFT copolymerizing of Flu-MA and poly(ethylene glycol) monomethacrylate (PEGMA). The result of 1H NMR and FTIR implied that both Flu-MA and PEGMA monomers were successfully incorporated into Flu-PEG copolymers, the molecular weight (M n) of which was about 39,800 with polydispersity index (PDI) as about 1.30. In aqueous solution, Flu-PEG copolymers could self-assemble into fluorescent organic nanoparticles (FONs) with about 200 nm diameters due to high water dispersibility and good fluorescence, respectively, endowed by PEGMA and Flu-MA, the fluorescence intensity of which was high sensitive to pH value of solution. More importantly, due to their high water dispersibility, excellent biocompatibility and good fluorescence, the as-prepared Flu-PEG FONs were high attractive for the application in cell imaging.

Abstract:Recently, various AIE-based materials with new characteristics and functions have been developing for different fields. Many chiral polymers are attracting the research interests for the new multifunctional materials in biomedicine, chiral recognition, etc. In this work, novel AIE copolymers with chirality activity were successfully fabricated for the first time. Firstly, a new fluorescent monomer (R)-THM with AIE effect and chirality activity was firstly synthesized by Suzuki reaction and enzymatic transesterification reaction. The quantum yield of (R)-THM increased to 0.643 from 0.100 of its substrate TBP with remarkable red-shift of emission spectra, which was consistent with their quantum chemical calculations of HOMO and LUMO situations, and its specific optical rotation <math>[α]D25°C</math> was about −9.67°. Subsequently, a series of chiral AIE copolymers of PEG-(R)-THM1 and PEG-(R)-THM2 were prepared via RAFT polymerization of (R)-THM and PEGMA with different feeding ratios. Their molecular weights were about 2.0 × 104 g/mol with narrow polydispersity index (PDI), and the kinetics analysis presented a pseudo-first-order linear procedure. For the molar 24.2%–32.3% feeding ratio of (R)-THM, the actual fraction of (R)-THM in the copolymers PEG-(R)-THM1 and PEG-(R)-THM2 was respectively about 22.3% and 30.2%, and the <math>[α]D25°C</math> value of copolymers PEG-(R)-THM1 and PEG-(R)-THM2 was about −2.51° and −3.74°. The amphiphilic structure of the obtained PEG-(R)-THM make them be easily dispersed in water solution and tend to self-assemble into fluorescent organic nanoparticles (FONs) with 100–150 nm diameter by TEM, and their maximum emission wavelength presented at 495 nm with obvious AIE effect. Moreover, the PEG-(R)-THM FONs had good biological compatibility, desirable low cytotoxicity and good cells absorption effect, which showed the great potential in bioimaging application.

Abstract:Nanocapsules made of a cross-linked poly(dodecafluoroheptyl acrylate) shell were successfully synthesized by interfacial RAFT miniemulsion polymerization. The well-defined hollow fluorinated polymer structures were obtained by the subsequent removal of the liquid core materials. The core/shell ratio could be easily tuned up to 1:1 by changing the conditions. In the cases of high core/shell ratio of 1:2 and 1:1, the nanocapsules were severely deformed, which was improved by increasing the cross-linker amount up to 30 wt.-%. It was concluded that the utility of the interfacial RAFT miniemulsion polymerization overcame the thermodynamic and kinetic barriers in a conventional miniemulsion polymerization.

Abstract:Fluorescent organic nanoparticles (FNPs) with intensive luminescence are widely applied in various biomedical fields, but the malicious aggregation caused quenching effect of conventional organic dyes hampers their biomedical application. In this work, we report a facile and highly efficient method for fabrication of FNPs with a stable cross-linked structure and enhanced luminescence via a one-pot mercaptoacetic acid locking imine reaction, in which a hydrophobic aggregation induced emission (AIE) dye (AHAn) and hydrophilic polylysine (Plys) were used as two reaction components, while mercaptoacetic acid acted as the lock to conjugate AHAn and Plys. The final amphiphilic fluorescent materials (Plys&AHAn&Plys) are readily formed FNPs in aqueous solution and exhibit high water dispersibility and strong luminescence. Furthermore, Plys&AHAn&Plys FNPs display low toxicity and good biological imaging performance. Altogether, we have developed a rather facile one-pot multicomponent reaction (MCR) to fabricate AIE active FNPs, which shows many promising properties for biomedical applications. More importantly, the MCR strategy could also be used for fabrication of many other multifunctional AIE active polymeric nanoprobes due to its good substrate adaptability and high efficiency.

Abstract:Facile one-pot preparation of cross-linked amphiphilic fluorescent polymer based on aggregation induced emission (AIE) dyes and 2-isocyanatoethyl methacrylate (IM) has been developed. This was carried out first by free radical polymerization between AIE monomer (PhE) and IM, and then polyethyleneimine (PEI) was introduced to obtain the cross-linked fluorescent polymer. The resulted cross-linked amphiphilic polymer was prone to self-assemble into stable nanoparticles in aqueous solution with surplus amino groups on the surface which made them highly water dispersible and can be further functionalized. The as-prepared fluorescent polymer nanoparticles (PhE-IM-PEI FPNs) were fully characterized by a series of techniques including (1)H NMR spectrum, X-ray photoelectron spectroscopy, Fourier transform infrared spectroscopy, transmission electron microscopy, dynamic light scattering, UV-vis absorption spectrum, and fluorescence spectra. Such FPNs demonstrated intense orange fluorescence with a high quantum yield of about 40%. Biocompatibility evaluation and cell uptake behavior of the nanoparticles were further investigated to explore their potential biomedical applications; the demonstrated excellent biocompatibility made them promising for cell imaging.

Abstract:In this work, we reported a rather facile method for fabrication of ultrabright, well dispersible and biocompatible fluorescent organic nanoparticles (FONs) with aggregation-induced emission (AIE) properties through combination of esterification and ring-opening reaction. The hydroxyl groups of Pluronic F127 was first reacted with the chloride of trimellitic anhydride chloride (TMAC), and its anhydride groups were further reacted with the amino groups of amino-terminated AIE dye (PhNH2) through ring-opening reaction. The optical properties, biocompatibility as well as cell uptake behavior of these obtained AIE-active nanoparticles (F127-TMAC-PhNH2 FONs) were examined by a series of characterization techniques and assays. We demonstrated that uniform organic nanoparticles with high water dispersibility, strong luminescence and desirable biocompatibility can be facilely obtained, which are promising for biological imaging applications. More importantly, a number of carboxyl groups were introduced into these AIE-active nanoparticles, which can be further utilized for further conjugation reaction and carrying anticancer drugs such as cisplatin. Therefore, the strategy of described in this work should be a simple and useful route for fabrication of multifunctional AIE-active luminescent nanotheranostic systems.

Abstract:Oligomer of styrene and maleic anhydride synthesized by bulk RAFT polymerization (SMA-RAFT) was used to construct a novel strategy for robust nanoencapsulation via interfacially confined controlled/living radical miniemulsion polymerization. After ammonolysis, SMA-RAFT becomes amphiphilic and can be used as a surfactant to prepare miniemulsion. The ammonolyzed SMA-RAFT molecules would self-assemble on the interface of water/droplets. This self-assembly property combining with the RAFT living polymerization chemistry demands the polymer chains to grow inwards gradually in particles, leading to the formation of a polymer shell. The hydrophilicity of ammonolyzed SMA-RAFT agent tuned by the ammonolyzed degree or structures of SMA-RAFT agent was found to play a key role in the final morphology. The well-defined nanocapsules with little solid particles can be obtained by using partially ammonolyzed SMA-RAFT with 0.5wt% SDS as a co-surfactant.

Abstract:In recent years, the fluorescent polymeric nanoparticles (FPNs) with aggregation-induced emission (AIE) feature have been extensively exploited in various biomedical fields owing to their advantages, such as low toxicity, biodegradation, excellent biocompatibility, good designability and optical properties. Therefore, development of a facile, efficient and well designable strategy should be of great importance for the biomedical applications of these AIE-active FPNs. In this work, a novel method for the fabrication of AIE-active FPNs has been developed through the self-catalyzed photo-initiated reversible addition fragmentation chain transfer (RAFT) polymerization using an AIE dye containing chain transfer agent (CTA), which could initiate the RAFT polymerization under light irradiation. The results suggested that the final AIE-active FPNs (named as TPE-poly(St-PEGMA)) showed great potential for biomedical applications owing to their optical and biological properties. More importantly, the method described in the work is rather simple and effective and can be further extended to prepare many other different AIE-active FPNs owing to the good monomer adoptability of RAFT polymerization.

Abstract:Fluorescent organic nanoparticles (FONs) have attracted intensive research interest for a variety of biomedical applications. Therefore, preparation of novel FONs with excellent optical properties and good biocompatibility in rather facile manner is of utmost importance. In this work, FONs based on dopamine (DA) containing copolymers were facilely fabricated through the combination of reversible addition–fragmentation chain transfer (RAFT) polymerization and self-polymerization of DA containing copolymers (poly(AC-co-PEGMA)), which were obtained by the one-pot polymerization of poly(ethylene glycol) methacrylate and acryloyl chloride in the present of DA. To generate these FONs, polyethyleneimine (PEI) was mixed with polymer-DA conjugates under rather mild experimental conditions (e.g. room temperature, absent of catalysts, air atmosphere and aqueous solution). Results confirmed polymer-DA-PEI FONs owned excellent water dispersibility and strong green fluorescence and desirable biocompatibility. All of these features make polymer-DA-PEI FONs alternative candidates for biomedical applications. In conclusion, we developed a rather facile strategy to fabricate novel types of FONs through the combination of RAFT polymerization and self-polymerization of DA. Because of the good monomer adaptability, this strategy could also be used for fabrication of many other FONs with different functional groups and properties.

Abstract:Fluorescent probes have long been regarded as tools for imaging living organisms with advantages such as high sensitivity, good designability and multifunctional potential. Many fluorescent probes, especially the probes based on aggregation-induced emission (AIE) dyes, have received increasing attention since the AIE phenomenon was discovered. These AIE dye-based fluorescent probes could elegantly overcome the notorious quenching effect caused by aggregation of conventional organic dyes. However, it is still difficult to directly apply these AIE-active dyes for biomedical applications owing to their hydrophobic nature. Therefore, the development of novel and facile strategies to endow them with water dispersibility is of critical importance. In this work, we exploit an efficient and simple strategy to fabricate an AIE dye-based fluorescent copolymer through the combination of reversible addition-fragmentation chain transfer and the Biginelli reaction. Moreover, the copolymer can self-assemble to fluorescent polymeric nanoparticles (FPNs) in water solution. Hydrophilic poly(PEGMA-co-AEMA) was reacted with the AIE-active dye 4',4‴-(1,2-diphenylethene-1,2-diyl)bis([1,1'-biphenyl]-4-carbaldehyde (CHO-TPE-CHO) to form amphiphilic luminescent polymers using urea as the connection bridge. The successful synthesis of the final products (poly(PEGMA-co-AEMA-TPE) FPNs) was confirmed by various instruments. Furthermore, Transmission electron microscopy (TEM) images manifest that poly(PEGMA-co-AEMA-TPE) copolymers will self-assemble into spherical nanoparticles in aqueous environments with sizes between 100 nm and 200 nm. The cell uptake and bioimaging experiment confirm that poly(PEGMA-co-AEMA-TPE) FPNs have excellent biocompatibility and emit strong green fluorescence in a cellular environment. Thus, poly(PEGMA-co-AEMA-TPE) FPNs are excellent candidates for biomedical applications.

Abstract:Multicomponent reactions (MCRs) have recently received increasing attention for the synthesis of structural complexity in a single step from three or more reactants. They have also been considered as a powerful tool for the construction of sequence-controlled multifunctional polymers owing to their good substrate adaptability, simple operation and high efficiency. In this work, we reported methods that are a combination of the three-component mercaptoacetic acid locking imine (MALI) reaction and reversible addition fragmentation chain transfer (RAFT) polymerization in one pot to form luminescent organic nanoparticles (LONs) with aggregation-induced emission (AIE) features, high-brightness, great water dispersibility, ultra-small nanoscale size and excellent biocompatibility. In the reaction system, the MALI reaction and RAFT polymerization happened simultaneously in a "one-pot'' route. On the one hand, the AIE-active organic dye with one amino group ((Z)-3-(4-aminophenyl)-2-(10-hexadecyl-10H-phenothiazin-3yl) acrylonitrile) (named as Phe-NH2) was conjugated with an aldehyde-containing monomer (10-undecenal) by the MALI reaction, while the aldehyde-containing monomer was copolymerized with the hydrophilic monomer polyethylene glycol methyl methacrylate (PEGMA) through RAFT polymerization at the same time. Compared with other fabrication strategies, "one-pot'' strategies possess some advantages such as high efficiency, simplicity, and atom economy. On the other hand, due to the good applicability of RAFT polymerization and the MALI reaction, many other multifunctional AIE-active LONs could also be fabricated via adjusting the function of the substrates. Therefore, this strategy should be a general and important route for fabrication of AIE-active materials for different applications.
Facile Fabrication of Amphiphilic AIE Active Glucan Via Formation of Dynamic Bonds: Self Assembly, Stimuli Responsiveness and Biological Imaging

Abstract:Fluorescent organic nanoparticles (FONs) with aggregation induced emission (AIE) properties have recently emerged as one of the most promising luminescent nanomaterials for biomedical applications due to their unique AIE feature. In this study, we reported the preparation of AIE active FONs through mixing AIE dye (TPE-CHO), 3-aminobenzeneboronic acids (ABBA) and glucan in one-pot. ABBA acted as a molecular "bridge" to conjugate TPE-CHO with glucan via formation of a Schiff base and phenyl borate. The resultant products (Glu-TPE FONs) showed amphiphilic properties and could self-assemble into nanoparticles in an aqueous solution. Glu-TPE FONs showed strong luminescence intensity and high water dispersibility because of the AIE properties of TPE-CHO and hydrophilic nature of glucan. To examine the biomedical application potential of glucan-AIE FONs, the responsiveness, biocompatibility and cell uptake behavior of Glu-TPE FONs were subsequently examined. We demonstrated that Glu-TPE FONs possess good biocompatibility and can be potentially used for biological imaging applications. More importantly, it is well known that the Schiff base and phenyl borate can respond to pH and glucose. Therefore, Glu-TPE FONs can be used for the fabrication of multifunctional biomaterials with stimuli responsiveness.
