Abstract:Two potent anti-MRSA tanshinone glycosides (1 and 2) were discovered by targeted microbial biotransformation, along with rapid identification via MS/MS networking. Serial reactions including dehydrogenation, demethylations, reduction, glycosylation and methylation have been observed after incubation of tanshinone IIA and fungus Mucor rouxianus AS 3.3447. In addition, tanshinosides B (2) showed potent activities against serial clinical isolates of oxacillin-resistant Staphylococcus aureus with MIC values of 0.78 μg/mL. This is the first study that shows a significant increase in the level and activities of tanshinone glycosides relative to the substrate tanshinone IIA.

Abstract:Cryptotanshinone (1) is one of the major bioactive constituents in Salvia miltiorrhiza Bunge. Preparative-scale biotransformation of cryptotanshinone by Cunninghamella elegans (AS 3.2082) produced three new products, which were identified as (3R, 15R)-3-hydroxycryptotanshinone (2), (3S, 15R)-3-hydroxycryptotanshinone (3), and (4S, 15R)-18-hydroxycryptotanshinone (4), respectively. The structural elucidation was based primarily on 1D and 2D NMR and HR-ESI-MS analyses. The absolute configuration of these three products was confirmed by comparison of their circular dichroism spectra with those of the known compounds. These biotransformed metabolites were used as for the comparison of in vivo metabolites in rat bile sample after intravenous administration and they are identical to three of the minor hydroxylated metabolites in vivo, which suggested that microbial biotransformation model was a useful and feasible approach for the preparation of mammalian metabolites in trace.

Abstract:Biotransformation of methyl protodioscin (1) by Cunninghamella elegans (AS 3.1207) was investigated. Nine bioconversion products were isolated and identified. Eight of the bioconversion products were pregnane glycoside or steroidal saponins. It was found that steroidal saponin skeleton could be converted to pregnenolone skeleton only using microbial methods, which must have chemical procedures in the reported literatures. The found enriched the types of bioconversion reaction and provided a new way for the production of androstenedione. Most bioconversion products showed considerable cytotoxic activities against HepG2, NCI-H460, MCF-7 and HeLa cell lines compared to methyl protodioscin.

Abstract:Microbial transformation of the steroidal sapogenin diosgenin (1) by resting cells of the filamentous fungus, Cunninghamella echinulata CGMCC 3.2716 was studied. Four metabolites were isolated and unambiguously characterized as (25R)-spirost-5-ene-3β,7β-diol-11-one (2), (25R)-spirost-5-ene-3β,7β-diol (3), (25R)-spirost-5-ene-3β,7β,11α-triol (4), and (25R)-spirost-5-ene-3β,7β,12β-triol (5), by various spectroscopic methods (1H, 13C NMR, DEPT, 1H–1H COSY, HMBC, HSQC and NOESY). Compound 2 is a new metabolite. The NMR data and full assignment for the known metabolites (25R)-spirost-5-ene-3β,7β-diol (3) and (25R)-spirost-5-ene-3β,7β,11α-triol (4) are described here for the first time. The biotransformation characteristics observed included were C-7β, C-11α and C-12β hydroxylations. Compounds 1–5 exhibited no significant cytotoxic activity to human glioma cell line U87.
Cell-Free Production of Pentacyclic Triterpenoid Compound Betulinic Acid from Betulin by the Engineered Saccharomyces cerevisiae.

Abstract:Betulinic acid is a product of plant secondary metabolism which has shown various bioactivities. Several CYP716A subfamily genes were recently characterized encoding multifunctional oxidases capable of C-28 oxidation. CYP716A12 was identified as betulin C-28 oxidase, capable of modifying betulin. This study aimed to induce the transformation of betulin to betulinic acid by co-expressing enzymes CYP716A12 from Medicago truncatula and ATR1 from Arabidopsis thaliana in Saccharomyces cerevisiae. The microsome protein extracted from the transgenic yeast successfully catalyzed the transformation of betulin to betulinic acid. We also characterized the optimization of cell fragmentation, protein extraction method, and the conversion conditions. Response surface methodology was implemented, and the optimal yield of betulinic acid reached 18.70%. After optimization, the yield and the conversion rate of betulin were increased by 83.97% and 136.39%, respectively. These results may present insights and strategies for the sustainable production of betulinic acid in multifarious transgenic microbes.

Abstract:BackgroundTanshinones are diterpenoid compounds that are used to treat cardiovascular diseases. As current extraction methods for tanshinones are inefficient, there is a pressing need to improve the production of these bioactive compounds to meet increasing demand.ResultsOverexpression of SmMDS (2-c-methyl-d-erythritol 2,4-cyclodiphosphate synthase, a tanshinone biosynthesis gene) in transgenic Salvia miltiorrhiza hairy roots significantly increased the tanshinone yield compared to the control, and total tanshinone content in SmMDS-overexpressing lines increased after elicitor treatment. Total tanshinones increased to 2.5, 2.3, and 3.2mg/g DW (dry weight) following treatment with Ag+, YE (yeast extract), and MJ (methyl jasmonate), respectively, compared with the non-induced transgenic line (1.7mg/g DW). Also, qRT-PCR analysis showed that the expression levels of two pathway genes was positively correlated with increased accumulation of tanshinone.ConclusionsOur study provides an effective strategy for increasing the content of tanshinones and other natural compounds using a combination of genetic engineering and elicitor treatment.

Abstract:Betulinic acid and its derivatives are potential bioactive compounds present in nature. This study investigated the biotransformation of betulin to betulinic acid by Cunninghamella blakesleeana cells. LC–MS analysis demonstrated that betulin could be transformed into at least five products from cultured C. blakesleeana cells, among which betulinic acid was the most important. The presented method provides an attractive alternative approach to chemical synthesis, because is less time-consuming and more environmentally friendly. C. blakesleeana can transform betulin into potent derivatives with high pharmacological activities.

Abstract:Microbial transformation of cardamonin by Mucor spinosus (CGMCC 3.3450) in preparative scale resulted in the isolation of two new products. Their structures were elucidated unambiguously by ESI-MS, 1H NMR, 13C NMR and 2D NMR spectra analyses as 4-O-beta-D-glucopyranosyl-6-hydroxy-2-methoxychalcone (1, 4-GluC) and 6-O-beta-D-glucopyranosyl-4-hydroxy-2-methoxychalcone (2, 6-GluC), respectively. The time-course of biotransformation by M. spinosus showed that both 4-GluC and 6-GluC appeared on the 2nd day. The optimal biotransformation temperature was 28 degrees C, the optimal biotransformation time was 72 h and the optimal concentration for cardamonin was 40 mg x mL(-1). This is the first time for successful microbial glycosylation of cardamonin in present research.

Abstract:Biotransformation of ent-kaur-16-en-19-oic acid using fungus Cunninghamella echinulata resulted in two novel hydroxylated metabolites together with five known compounds. Their structures were elucidated by means of extensive NMR and HR-ESI-MS data analysis. The eight compounds were measured for their cytotoxicity against the human breast carcinoma (MCF-7) and human hepatoblastoma (HepG-2) cell lines. Seven compounds showed no cytotoxicity to the two cell lines. One compound displayed moderate cytotoxicity against HepG-2 and MCF-7 with the IC50 values of 12.6 and 27.1 mu M, respectively.

Abstract:Microbial transformation of ginsenosides to increase its pharmaceutical effect is gaining increasing attention in recent years. In this study, Cellulosimicrobium sp. TH-20, which was isolated from soil samples on which ginseng grown, exhibited effective ginsenoside-transforming activity. After protopanaxadiol (PPD)-type ginsenoside (Rb1) and protopanaxatriol (PPT)-type ginsenosides (Re and Rg1) were fed to C . sp . TH20, a total of 12 metabolites, including 6 new intermediate metabolites, were identified. Stepwise deglycosylation and dehydrogenation on the feeding precursors have been observed. The final products were confirmed to be rare ginsenosides Rd, GypXVII, Rg2 and PPT after 96 h transformation with 38–96% yields. The four products showed improved anti-inflammatory activities by using lipopolysaccharide (LPS)-induced murine RAW 264.7 macrophages and the xylene-induced acute inflammatory model of mouse ear edema. The results indicated that they could dramatically attenuate the production of TNF-α more effectively than the precursors. Our study would provide an example of a unique and powerful microbial cell factory for efficiently converting both PPD-type and PPT-type ginsenosides to rare natural products, which extends the drug candidates as novel anti-inflammatory remedies.

Abstract:Microbial transformations of two tetracyclic beyerane-type diterpenes, ent-16 beta-oxobeyeran-19-oic acid (1) and its chemical reduction product, ent-16 beta-hydroxybeyeran-19-oic acid (2), by the filamentous fungus Cunninghamella echinulata ATCC 8688a yielded eight metabolites (3-10). Incubation of the substrate 2 with C. echinulata afforded three new hydroxylated ones (3-5) along with two known ones (6-7), while incubation of 1 gave three known ones (8-10). The new compounds were characterized by 1D and 2D NMR as well as HRESIMS analysis, and the stereostructures of 3 and 4 were confirmed by X-ray crystallography. The bioreactions were involved not only in stereoselective incorporation of hydroxyl groups at inert positions C-7, -9, -12, and -14 of the two beyerane diterpenes but also in glucosidation at C-19 of 2. This is the first report on the biotransformation of the diterpenes by using C. echinulata. All compounds were assayed for their alpha-glucosidase inhibitory, neurotrophic, anti-inflammatory, and phytotoxic activity, and only in neurotrophic assay compounds, 2 and 9 were found to display nerve growth factor-mediated neurite-outgrowth promoting effects in PC12 cells; the others were inactive.

Abstract:Glycosylation is an effective approach to improve the druggability of natural products by increasing their water solubility. In this work, we report the glycosylation of oleanane-type triterpenoids by a recombinant microbial glycosyltransferase YjiC1. A preliminary screening test indicated YjiC1 exhibited robust capabilities for O-glycosylation of triterpenoids, based on LC/MS analysis. Among the products, two new compounds (2a and 3a), together with a known one (1a), were isolated and characterized. These products exhibited improved water solubility, and 3a showed moderate anti-HIV activities at 100 μM. This reaction provides a facile and efficient approach to synthesize the glucosides of triterpenoids.

Abstract:Background: Tanshinone IIA is effective in the treatment of various cardiovascular diseases. Currently, tanshinone IIA is primarily derived from the roots and rhizomes of Salviamiltiorrhiza Bunge. However, resources of S.miltiorrhiza are already facing scarcity. Objectives: The aim of this study was to obtain an alternative and high-yield mutagenic strain of tanshinone IIA, thus establishing a cost-effective and non-plant-derived substitute for tanshinone IIA. Methods: The mutants with enhanced tanshinone IIA production were selected through protoplast mutation induced by ultraviolet radiation (UV) and sodium nitrite (NaNO2). The yield of tanshinone IIA (YT) was determined using a TU-1810 ultraviolet-visible (UV/Vis) Spectrophotometer and LC-2010A high-performance liquid chromatography (HPLC). The selected mutant strains with the highest yield were subjected to continuous cultivation, and the dry weight of mycelium (DW) and the YT in the first generation were used as controls to assess passage stability. Fungal genomic DNA from the wild-type and mutant strains was extracted from axenic cultures using the modified cetyltrimethyl ammonium bromide (CTAB) method. Results: The colony morphological characteristics exhibited significant differences between the wild-type TR21 and the mutant NU204. The NU204 demonstrated a significantly higher tanshinone IIA yield at 165 ± 2.52 μg/g, which was 4.67-fold greater than that of TR21 (P < 0.01), indicating a substantial enhancement in NU204 production compared to TR21. The NU204 underwent continuous cultivation for five generations, and there were no significant differences in the DW and YT among the different generations (P > 0.05). The observed genetic changes between TR21 and NU204 were induced by the protoplast mutation. Conclusions: The mutant NU204 is emerging as a promising novel alternative source for tanshinone IIA, offering potential practical applications in production.
Transcriptome Sequencing of Endophytic Fungi with High Yield Tanshinone IIAand Exploration of Key Genes in Metabolic Pathways

Abstract:Abstract Background: Medicinal plant endophytic fungi can produce the same or similar biologically active secondary metabolites as the host.In this study, the endophytic fungus T21 in the roots of Salvia miltiorrhiza can produce tanshinone IIA, the active substance of S. miltiorrhiza .We compared the differential expression between strain T21 and its high-yielding mutant strain F3-4 by transcriptome sequencing, so as to determine the key genes and pathways of tanshinone IIA synthesis in fungi. Methods and Results: Research materials were studied by transcriptome sequencing and quantitative real-time polymerase chain reaction (qRT-PCR).A total of 11.31 Gb clean data were obtained. For each sample, there were 2.71 Gb clean data, and the Q30 base percentage of 86.15% and above was achieved. A total of 13,573 Unigene pieces were acquired after de novo assembly. Functional annotation of Unigene was performed, involving the comparison with non-redundant protein sequence (NR), swiss-prot, kyoto encyclopedia of genes and genomes(KEGG), cluster of orthologous groups of proteins(COG)and gene ontology(GO)databases. Moreover, 10,218 annotation results of Unigene were achieved. This study delved into the gene expression differences between the two fungi and acquired 88 differentially expressed genes.Then, the relative expression levels of 7 genes in the MVA pathway and the highly differentially expressed gene NRPS were detected by qRT-PCR technology. Conclusion: As revealed from this finding, tanshinone IIA may be synthesized via MVA pathway in fungi, and NRPS gene also participates in the synthesis of tanshinone IIA.
Increased accumulation of the cardio-cerebrovascular disease treatment drug tanshinone in Salvia miltiorrhiza hairy roots by the enzymes 3-hydroxy-3-methylglutaryl CoA reductase and 1-deoxy-d-xylulose 5-phosphate reductoisomerase

Abstract:Tanshinone is widely used for treatment of cardio-cerebrovascular diseases with increasing demand. Herein, key enzyme genes SmHMGR (3-hydroxy-3-methylglutaryl CoA reductase) and SmDXR (1-deoxy-d-xylulose 5-phosphate reductoisomerase) involved in the tanshinone biosynthetic pathway were introduced into Salvia miltiorrhiza (Sm) hairy roots to enhance tanshinone production. Over-expression of SmHMGR or SmDXR in hairy root lines can significantly enhance the yield of tanshinone. Transgenic hairy root lines co-expressing HMGR and DXR (HD lines) produced evidently higher levels of total tanshinone (TT) compared with the control and single gene transformed lines. The highest tanshinone production was observed in HD42 with the concentration of 3.25 mg g−1 DW. Furthermore, the transgenic hairy roots showed higher antioxidant activity than control. In addition, transgenic hairy root harboring HMGR and DXR (HD42) exhibited higher tanshinone content after elicitation by yeast extract and/or Ag+ than before. Tanshinone can be significantly enhanced to 5.858, 6.716, and 4.426 mg g−1 DW by YE, Ag+, and YE-Ag+ treatment compared with non-induced HD42, respectively. The content of cryptotanshinone and dihydrotanshinone was effectively elevated upon elicitor treatments, whereas there was no obvious promotion effect for the other two compounds tanshinone I and tanshinone IIA. Our results provide a useful strategy to improve tanshinone content as well as other natural active products by combination of genetic engineering with elicitors.

[Microbial Transformation of Glycyrrhetinic Acid by Cunninghamella Blakesleeana].

Abstract:A study on the microbial transformation of glycyrrhetinic acid (GA) was conducted by a fungus, Cunninghamella blakesleeana CGMCC 3.970 systematically. After incubation with the cell cultures of C. blakesleeana CGMCC 3.970 at 25 degrees C for 7 days on a rotary shaker operating at 135 r x min(-1), GA was converted into one major product and five minor products. The products were extracted and purified by solvent extraction, macroporous adsorbent resin, silica gel column chromatography, and semi-preparative RP-HPLC chromatography. Their structures were identified as 3-oxo-15α-hydroxy-18β-glycyrrhetinic acid(1), 3-oxo-15β-hydroxy-18β-glycyrrhetinic acid (2), 7β,15α-dihydroxy-18β-glycyrrhetinic acid (3), 3-oxo-7β, 15α-dihydroxy-18β-glycyrrhetinic acid (4), 7β-hydroxy-18β-glycyrrhetinic acid(5) and 15α-hydroxy-18β-glycyrrhetinic acid(6) by the analyses of MS, 1H-NMR and 13C-NMR spectroscopic data respectively. Among them, 2 was a new compound. These results suggest that C. blakesleeana CGMCC 3.970 has the capability of selective ketonization and hydroxylation for GA. Abstract:Structural modification of natural products by biotransformation with fungi is an attractive tool to obtain novel bioactive derivatives. In the present study, cryptotanshinone (1), a quinoid abietane diterpene from traditional Chinese medicine Salvia miltiorrhiza (Danshen), was transformed by two marine-derived fungi. By using Cochliobolus lunatus TA26-46, one new oxygenated and rearranged product (2), containing a 5,6-dihydropyrano [4,3-b]chromene moiety, together with one known metabolite (10), were obtained from the converted broth of cryptotanshinone (1) with the isolated yields of 1.0% and 2.1%, respectively. While, under the action of Aspergillus terreus RA2905, seven new transformation products (3-9) as well as 10 with the fragments of 2-methylpropan-1-ol and oxygenated p-benzoquinone were produced and obtained with the isolated yields of 0.1%-1.3%. The structures of the new compounds were elucidated by comprehensive spectroscopic analysis including High Resolution Electrospray Ionization Mass Spectroscopy (HRESIMS), Nuclear Magnetic Resonance (NMR) and Electronic Circular Dichroism (ECD). The metabolic pathways of cryptotanshinone by these two fungi were presumed to be the opening and rearrangement of furan ring, and/or oxygenation of cyclohexane ring. Cryptotanshinone (1) and its metabolites displayed anti-inflammatory activities against NO production in LPS-stimulated BV-2 cells and antibacterial activities towards methicillin-resistant Staphylococcus aureus. These findings revealed the potential of marine fungi to transform the structures of natural products by biotransformation.
Preparation of Some Metabolites of Timosaponin BII by Biotransformation in Vitro

Abstract:Timosaponin BII (TBII) is an active constituent of some Chinese medicinal herbs, and is abundant in Anemarrhena asphodeloides Bunge. To better understand the pharmaceutical mechanisms of TBII, we prepared its deglycosylated derivatives through biotransformation with fungi Colletotrichum gloeosporioides, Acremonium alternatum and Aspergillus niger. In the presence of 10 g/L glucose, transformation with C. gloeosporioides yielded 4 products: timosaponin AIII (P3) and AI (P5) and their beta isoforms (P2 and P4 respectively). However, P4 and P5 could not be detected when 0-5 g/L glucose was applied; P2 and P3 disappeared at 20 g/L glucose. P4 is a newly discovered compound. We firstly report that fungi in whole cell model can hydrolyze C3-Omicron-glucopyranosyl residues of steroidal saponins after removing the C26 sugar. Therefore, the transformation efficiency depends on glucose. Transformation of TBII, and maybe other steroidal saponins, with C. gloeosporioides is an ideal method to prepare the metabolites in vitro. (C) 2015 Elsevier Ltd. All rights reserved.

Abstract:Three 20(S)-protopanaxatriol-type saponins, ginsenoside-Rg(1) (1), notoginsenoside-R-1 (2), and ginsenoside-Re (3), were transformed by the fungus Absidia coerulea (AS 3.3389). Compound 1 was converted into five metabolites, ginsenoside-Rh-4 (4), 3 beta, 2 beta, 25-trihydroxydammar-(E)-20(22)-ene-6-O-beta-D-glucopyranoside (5), 20(S)-ginsenoside-Rh-1 (6), 20(R)-ginsenoside- Rh-1(7), and a mixture of 25-hydroxy-20(S)-ginsenoside-Rh-1 and its C-20(R) epimer (8). Compound 2 was converted into 10 metabolites, 20(S)-notoginsenoside-R-2 (9), 20(R)-notoginsenoside-R-2 (10), 3 beta, 12 beta, 25-trihydroxydammar( E)-20(22)-ene-6-O-beta-D-xylopyranosyl-(1 -> 2)-beta-D-glucopyranoside (11), 3 beta, 12 beta-dihydroxydammar-(E)-20(22), 24-diene-6- O-beta-D-xylopyranosyl-(1 -> 2)-beta-D-glucopyranoside (12), 3 beta, 12 beta, 20,25-tetrahydroxydammaran-6-O-beta-D-xylopyranosyl(1 -> 2)-beta- D-glucopyranoside (13), and compounds 4-8. Compound 3 was metabolized to 20(S)-ginsenoside-Rg(2) (14), 20(R)-ginsenoside-Rg(2) (15), 3 beta, 12 beta, 25-trihydroxydammar-(E)-20(22)-ene-6-O-alpha-L-rhamnopyranosyl-(1 -> 2)-beta-D-glucopyranoside (16), 3 beta, 12 beta-dihydroxydammar-(E)-20(22), 24-diene-6-O-alpha-L-rhamnopyranosyl-(1 -> 2)-beta-D-glucopyranoside (17), 3 beta, 12 beta, 20,25-tetrahydroxydammaran-6-O-alpha-L-rhamnopyranosyl-(1 -> 2)-beta-D-glucopyranoside (18), and compounds 4-8. The structures of five new metabolites, 10-13 and 16, were established by spectroscopic methods.

Abstract:Cryptotanshinone (1), a major bioactive constituent in the traditional Chinese medicinal herb Dan-Shen Salvia miltiorrhiza Bunge, has been reported to possess remarkable pharmacological activities. To improve its bioactivities and physicochemical properties, in the present study, cryptotanshinone (1) was biotransformed with the fungus Cunninghamella elegans AS3.2028. Three oxygenated products (2-4) at C-3 of cryptotanshinone (1) were obtained, among them 2 was a new compound. Their structures were elucidated by comprehensive spectroscopic analysis including HRESIMS, NMR and ECD data. All of the biotransformation products (2-4) were found to inhibit significantly lipopolysaccharide-induced nitric oxide production in BV2 microglia cells with the IC50 values of 0.16-1.16 mu M, approximately 2-20 folds stronger than the substrate (1). These biotransformation products also displayed remarkably improved inhibitory effects on the production of inflammatory cytokines (IL-1 beta, IL-6, TNF-alpha, COX-2 and iNOS) in BV-2 cells via targeting TLR4 compared to substrate (1). The underlying mechanism of 2 was elucidated by comparative transcriptome analysis, which suggested that it reduced neuroinflammatory mainly through mitogen-activated protein kinase (MAPK) signaling pathway. Western blotting results revealed that 2 downregulated LPS-induced phosphorylation of JNK, ERK, and p38 in MAPK signaling pathway. These findings provide a basal material for the discovery of candidates in treating Alzheimer's disease.
