Abstract:Platelet activation plays an important role in thrombosis. Platelet glycoprotein Ⅱb/Ⅲa (GP Ⅱb/Ⅲa) is the receptor of fibrinogen. Platelet cross-linking with fibrinogen through GPⅡb/Ⅲa is the process of thrombosis. Ca 2+ is an important intracellular second messenger in platelet activation. It has been reported that GPⅡb/Ⅲa receptors were involved in the calcium influx of activated platelet, and GPⅡb/Ⅲa receptor had characteristics of calcium channel or an adjacent calcium channel. There also are chloride channels in platelet membrane. The activity of the channels depends on the platelet free calcium concentration ([Ca 2+]i). 4,4,-disothiocyano-2,2,disulfonic acid stilbene (DIDS), the chloride channel blocker, could inhibit the platelet aggregation and thrombin induced increase in platelet [Ca 2+]i. 1,2 Department of Hematology, 2nd Affiliated Hospital of Sun Yat-Sen University, Guangzhou 510120, China (Yin SM, Xie SF, Nie DN, Li YQ, Li HM, Ma LP, Wang XJ, Wu YD and Feng JH)
