Abstract:Aims: The effects of exogenously applied salicylic acid (SA) on gas exchange characteristics, photosynthetic pigments and chloroplast ultrastructure were investigated in gerbera at their reproductive stage under salt-stressed conditions. Methodology: A pot experiment was conducted under glasshouse conditions at the Zhejiang University, Hangzhou, China, (30° N/120° E) between February 2008 and March 2009.Plants, pretreated with foliar applications of 0, 0.5, and 1.0 mmoldm-3 SA at the onset of flower initiation were irrigated with 100 mmoldm-3NaCl(aq) for two weeks, starting after three days from the SA pretreatment. Control did not receive either NaCl or SA.Photosynthetic rate, gas exchange, photosynthetic pigments content and chloroplast ultrastructure were investigated against treatments. All data were subjected to analysis of variance (ANOVA) and Generalized Linear Model (GLM) using SAS statistical software. Pearson’s correlation test was carried out to study the relationships among the parameters. The means were compared using Duncan’s multiple range test (DMRT). For all the tests, P< .05 was considered statistically significant. Results: Salt stress adversely affected the gas exchange characteristics, photosynthetic pigment contents and chloroplast ultrastructure. SA application significantly increased the net photosynthesis, stomatal conductivity, intra-cellular CO2 content and transpiration rate but decreased the stomatal limitation, compared to those of untreated salt-stressed plants. Further, the enhanced photosynthetic pigment contents and notably undamaged chloroplast ultrastructure were evident of the ameliorative effects of SA on photosynthetic system under salt stress. Of the two concentrations tested, 0.5 mmoldm-3 SA concentration seemed to have greater effect throughout the experiment showing no significant variation from control in some attributes (chlorophyll contents and chloroplast ultrastructure). Conclusion: Responses of plants pretreated with SA spraying and significant correlation among them plausibly suggest SA-induced enhancement of photosynthetic system as another target for conferring salt tolerance in crop plants.

Abstract:5-Aminolevulinic acid (ALA) is a common precursor of tetrapyrroles as well as a crucial growth regulator in higher plants. ALA has been proven to be effective in improving photosynthesis and alleviating the adverse effects of various abiotic stresses in higher plants. However, little is known about the mechanism of ALA in ameliorating the photosynthesis of plant under abiotic stress. In this paper, we studied the effects of exogenous ALA on salinity-induced damages of photosynthesis in cucumber (Cucumis sativus L.) seedlings. We found that the morphology (plant height, leave area), light utilization capacity of PS II [qL, Y(II)] and gas exchange capacity (Pn, gs, Ci, and Tr) were significantly retarded under NaCl stress, but these parameters were all recovered by the foliar application of 25 mg L-1 ALA. Besides, salinity caused heme accumulation and up-regulation of gene expression of ferrochelatase (HEMH) with suppression of other genes involved in chlorophyll synthesis pathway. Exogenously application of ALA under salinity down-regulated the heme content and HEMH expression, but increased the gene expression levels of glutamyl-tRNA reductase (HEMA1), Mg-chelatase (CHLH), and protochlorophyllide oxidoreductase (POR). Moreover, the contents of intermediates involved in chlorophyll branch were increased by ALA, including protoporphyrin IX (Proto IX), Mg-protoporphyrin IX (Mg-Proto IX, protochlorophyllide (Pchlide), and chlorophyll (Chl a and Chl b) under salt stress. Ultrastructural observation of mesophyll cell showed that the damages of photosynthetic apparatus under salinity were fixed by ALA. Collectively, the chlorophyll biosynthesis pathway was enhanced by exogenous ALA to improve the tolerance of cucumber under salinity.

Abstract:5-aminolevulinic acid (ALA) has been used as a plant growth regulator and can affect physiochemical processes, including carbon fixation and nutrient assimilation. Here, we investigated the effect of ALA on sulfur metabolism and glucosinolate (GSL) biosynthesis in rape (Brassica napus L.) seedlings, which were treated with 0, 0.5, 1.0, 5.0, and 10.0 mg/L ALA supplemented in a Hoagland solution. After 28 d of treatment, the effect of ALA on thiol and GSL contents and transcriptional profile of associated genes was analyzed. Results showed that low ALA concentrations increased cysteine, GSL, and total soluble thiol contents, and upregulated expression of genes such as BnSULTR1.1, BnSULTR2.2, and BnAPK1 that regulate sulfur uptake and metabolism. ALA also increased the GSL content, particularly the aliphatic GSLs, due to the improvement of sulfur metabolism and assimilation to methionine. Other GSLs biosynthesis stages, such as desulfo-GSL glycosylation, were also significantly improved by the ALA applications as indicated by the increased expression of BnUGT74B1 and BnUGT74C1. High ALA concentrations negatively affected sulfur metabolism and GSL synthesis by inflicting photo-oxidative stress that damaged cellular components. Moderate ALA concentrations promoted sulfur acquisition, assimilation, and GSL biosynthesis.

Abstract:Application of 5-Aminolevulinic acid (ALA) to plants not only affects their growth, but also influences the nutritional values of plant organs. Oilseed rape (Brassica napus L.) is an important source of vegetable oil and various compounds benefiting human health. However, limited information is available regarding the effect of ALA on the growth of this important crop. Here, we investigated the consequence of ALA application on oilseed rape seedlings. We found that moderate ALA concentrations (0.5 or 1 mg/L) caused an increase in the dry weight of the seedling biomass and the contents of various bioactive compounds, i.e. soluble sugar, phenolic compounds, ascorbate, and glucosinolates etc, and the upregulated expressional levels of the genes encoding RuBisCO, namely, BnRBCL and BnRBCS, and other genes such as BnUGT79B1, BnMYB12 and MYB28 that regulate the synthesis of phenolic compounds and glucosinolates respectively. On the other hand, high ALA concentrations (5 and 10 mg/L) gave rise to oxidative stress that negatively affected oilseed rape growth, and caused deleterious effects by reducing various bioactive compounds, but increasing tocopherols that may serve as a stress mitigatory agent. With the high ALA concentrations, the ultrastructure of chloroplasts was notably damaged, featured with large starch grains and fewer plastoglobules.

Abstract:5-Aminolevulinic acid (ALA) is an important plant growth regulator which is derived from 5-carbon aliphatic amino acid. The present study investigates the interaction of increasing NaCl-salinity and ALA on plant growth, leaf pigment composition, leaf and root Na+/K+ ratio and chloroplast ultrastructure in mesophyll cells of oilseed rape (Brassica napus) leaves. The plants were treated hydroponically with three different salinity levels (0, 100, 200 mM) and foliar application of ALA (30 mg l(-1)) simultaneously. Ten days after treatment, higher NaCl-salinity significantly reduced the plant biomass and height. However, ALA application restored the plant biomass and plant height under saline conditions. A concentration-dependent increase in Na+ uptake was observed in the aerial parts of B. napus plants. On the other hand, ALA reduced Na+ uptake, leading to a significant decrease in Na+/K+ ratio. Accumulation of Na+ augmented the oxidative stress, which was evident by electron microscopic images, highlighting several changes in cell shape and size, chloroplast swelling, increased number of plastogloubli, reduced starch granules and dilations of the thylakoids. Foliar application of ALA improved the energy supply and investment in mechanisms (higher chlorophyll and carotenoid contents, enhanced photosynthetic efficiency), reduced the oxidative stress as evident by the regular shaped chloroplasts with more intact thylakoids. Abstract:This study examined the effects of 5-aminolevulinic acid (ALA) application on photosynthesis, activity and gene expression of key antioxidant enzymes, and on proline accumulation in tomato (Lycopersicon esculentum Mill. ‘Hezuo 903’) seedlings under NaCl stress. NaCl stress significantly decreased the net photosynthetic rates and inhibited the activity of photosystem II, whereas exogenous ALA application significantly restored the net photosynthetic rates, quantum yield of electron transport, and energy conversion efficiency of photosystem II of tomato under NaCl stress. Production of superoxide, hydrogen peroxide, and malondialdehyde strongly increased in response to NaCl stress, and these increases were significantly counteracted by ALA. ALA increased the activity of reactive oxygen species (ROS) scavenging antioxidant enzymes, including superoxide dismutase, catalase, ascorbate peroxidase, and peroxidase, and upregulated the expression of SOD, APX, and POD, genes that encode these enzymes in NaCl-treated plants. ALA simultaneously increased proline accumulation in tomato seedlings under NaCl stress by regulating the expression of genes that encode ALA biosynthetic enzymes and that control proline biosynthesis and metabolism, for example, expression of GluRS and GluTR was downregulated, accompanied by a significant increase in the expression of P5CS and decline in the expression of ProDH. ALA provided protection against NaCl stress by increasing photosynthetic capacity, regulating antioxidant enzyme gene expression and proline accumulation, and decreasing ROS accumulation and lipid peroxidation in tomato.

Abstract:To study the effects of exogenous salicylic acid pretreatment on thermotolerance and photosynthesis,two pakchoi varieties,'Xia-di' and 'Suzhou-qing',were sprayed with salicylic acid(SA) and then subjected to high temperature stress.The results showed that SA pretreatment prevented lipid peroxidation on pakchoi leaves under high temperature stress by reducing the electrolyte leakage,MDA content,proline content and net photosynthetic rate(Pn).Meanwhile,the soluble sugar content,soluble protein content,SOD activity and POD activity increased to make the pakchoi adapt to high temperature.However,two pakchoi varieties showed the opposite trend beween stomatal conductance(Gs) and transpiration rate(Tr) after spraying SA,with no significant alteration on intercellular CO2 concentration(Ci).As compared with heat tolerant cultivar 'Xia-di',the effects of SA pretreatment on heat susceptible cultivar 'Suzhou-qing' were more significant under high temperature stress.

Abstract:In the present study, the roles of foliarly applied 5-aminolevulinic acid (ALA) in photosynthetic parameters, ion contents and expression of several photosynthetic genes were investigated in rapeseed (Brassica napus L.) under well watered and drought conditions. Drought stress reduced relative plant growth rate and leaf water content (RWC), and contents of photosynthetic pigments, starch, soluble proteins and ions in roots. This type of stress, however, stimulated the accumulation of soluble sugars and free amino acids, as well as leaf proline contents. The transcript levels of photosynthetic genes were also down-regulated under drought stress. Exogenously applied ALA partially reduced growth inhibition and increased RWC and chlorophyll contents under drought conditions. The expression of photosynthetic genes was induced by ALA under both normal and drought conditions, which may contribute to an increased photosynthetic capacity. The ALA application further improved the accumulation of hexose and sucrose, accompanied by the maintenance of starch contents in the leaves and possibly relatively higher RWC under drought stress. Based on these results, we suggest that ALA promotes plant growth under drought stress via improving photosynthetic assimilation, increasing chlorophyll content and inducing the expression of several photosynthetic genes.
5-Aminolevulinic Acid Activates Antioxidative Defence System and Seedling Growth in Brassica Napus L. under Water-Deficit Stress

Abstract:The present study assesses the effects of 5-aminolevulinic acid (ALA, 0, 0.1, 1 and 10 mg l(-1)) on the growth of oilseed rape (Brassica napus L. cv. ZS758) seedlings under water-deficit stress induced by polyethylene glycol (PEG 6000, 0 and -0.3 MPa). Water-deficit stress imposed negative effects on seedling growth by reducing shoot biomass, cotyledon water potential, chlorophyll content and non-enzymatic antioxidants (glutathione and ascorbic acid) levels. On the other hand, water-deficit stress enhanced the malondialdehyde (MDA) content, reactive oxygen species (ROS) production, enzymatic antioxidants activities, reduced/oxidized glutathione ratio (GSH/GSSG) and reduced/oxidized ascorbic acid (ASA/DHA) ratio in seedlings. Application of ALA at lower dosages (0.1 and 1 mg l(-1)) improved shoot weight and chlorophyll contents, and decreased MDA in rape seedlings, whereas moderately higher dosage of ALA (10 mg l(-1)) hampered the growth. The study also indicated that 1 mg l(-1) ALA improved chlorophyll content, but reduced MDA content and ROS production significantly under water-deficit stress. Lower dosages of ALA (0.1 and 1 mg l(-1)) also enhanced GSH/GSSG and ASA/DHA as compared to the seedlings under water-deficit stress. The antioxidant enzymes (ascorbate peroxidase, peroxidase, catalase, glutathione reductase and superoxide dismutase) enhanced their activities remarkably with 1 mg l(-1) ALA treatment under water-deficit stress. It was also revealed that 1 mg l(-1) ALA treatment alone induced the expression of APX, CAT and GR substantially and under water-deficit stress conditions ALA treatment could induce the expression of POD, CAT and GR to a certain degree. These results indicated that 0.1-1 mg l(-1) ALA could enhance the water-deficit stress tolerance of oilseed seedlings through improving the biomass accumulation, maintaining a relative high ratio of GSH/GSSG and ASA/DHA, enhancing the activities of the specific antioxidant enzymes and inducing the expression of the specific antioxidant enzyme genes.
Promotive Effect of 5-Aminolevulinic Acid on the Antioxidant System in Ginkgo Biloba Leaves

Abstract:The effect of 5-aminolevulinic acid (ALA), a key precursor in the biosynthesis of porphyrins, at low levels (10 and 100 mg l-1) on the antioxidant system in Ginkgo biloba leaves were investigated. The results showed that ALA-treatment accelerated hydrogen peroxide accumulation and temporarily increased the activities of superoxide dismutase (SOD), ascorbate peroxidase (APX), catalase (CAT) at day 4 and 8. The activities of glutathione reductase (GR) and dehydroascorbate reductase (DHAR) and the contents of ascorbic acid (AsA) and glutathione (GSH) were all significantly increased by ALA, while dehydroascorbate (DHA) content was decreased and no variety was observed in oxidized glutathione (GSSG) content after ALA treatment. The ALA-treated leaves had higher values of AsA/DHA and GSH/GSSG than those in the control leaves. These results suggested that foliar treatment with a low concentration of ALA might provide a useful means of improving beneficial antioxidant activity in G. biloba leaves.

Abstract:Salinity is one of the major constraints in oilseed rape ( Brassica napus L.) production. One of the means to overcome this constraint is the use of plant growth regulators to induce plant tolerance. To study the plant response to salinity in combination with a growth regulator, 5-aminolevulinic acid (ALA), oilseed rape plants were grown hydroponically in greenhouse conditions under three levels of salinity (0, 100, and 200 mM NaCl) and foliar application of ALA (30 mg/l). Salinity depressed the growth of shoots and roots, and decreased leaf water potential and chlorophyll concentration. Addition of ALA partially improved the growth of shoots and roots, and increased the leaf chlorophyll concentrations of stressed plants. Foliar application of ALA also maintained leaf water potential of plants growing in 100 mM salinity at the same level as that of the control plants, and there was also an improvement in the water relations of ALA-treated plants growing in 200 mM. Net photosynthetic rate and gas exchange parameters were also reduced significantly with increasing salinity; these effects were partially reversed upon foliar application with ALA. Sodium accumulation increased with increasing NaCl concentration which induced a complex response in the macro-and micronutrients uptake and accumulation in both roots and leaves. Generally, analyses of macro- (N, P, K, S, Ca, and Mg) and micronutrients (Mn, Zn, Fe, and Cu) showed no increased accumulation of these ions in the leaves and roots (on dry weight basis) under increasing salinity except for zinc (Zn). Foliar application of ALA enhanced the concentrations of all nutrients other than Mn and Cu. These results suggest that under short-term salinity-induced stress (10 days), exogenous application of ALA helped the plants improve growth, photosynthetic gas exchange capacity, water potential, chlorophyll content, and mineral nutrition by manipulating the uptake of Na + .

Abstract:A number of studies have established that plant growth and development in oilseed rape (Brassica napus L.) are hampered by salinity stress. Nowadays, researchers have focused on the use of plant growth regulators to increase plant tolerance against salinity. An experiment was performed to evaluate the effects of 5-aminolevulinic acid (ALA, 30 mg l−1) on Brassica napus L. (cv. ‘ZS 758’) plants under NaCl (100, 200 mM) salinity. Data presented here were recorded on two different leaf positions (first and third) to have a better understanding of the ameliorative role of ALA on NaCl-stressed oilseed rape plants. Results have shown that increasing salinity imposed negative impact on relative growth rate (root and shoot) and leaf water relations (osmotic potential and relative water content), whereas enhanced the level of relative conductivity, malondialdehyde (MDA) content, osmolytes (soluble sugar, soluble protein, free amino acid and proline) concentration, reactive oxygen species (ROS), and enzymatic (ascorbate peroxidase, guaiacol peroxidase, catalase and superoxide dismutase) and non-enzymatic (reduced glutathione and ascorbate) antioxidants activity in two different leaf position samples. Foliar application of ALA improved relative growth rate (root and shoot) and leaf water relations (osmotic potential and relative water content), and also triggered the further accumulation of osmolytes (soluble sugar, soluble protein, free amino acid and proline) as well as enzymatic (ascorbate peroxidase, guaiacol peroxidase, catalase and superoxide dismutase) and non-enzymatic (reduced glutathione and ascorbate) antioxidants activity in both leaf samples, whereas decreased the membrane permeability, MDA content and ROS production. Our results also indicate that osmolytes are preferentially accumulated in younger tissues.
Substrate Application of 5-Aminolevulinic Acid Enhanced Low-temperature and Weak-light Stress Tolerance in Cucumber (Cucumis sativus L.)

Abstract:5-Aminolevulinic acid (ALA) is a type of nonprotein amino acid that promotes plant stress tolerance. However, the underlying physiological and biochemical mechanisms are not fully understood. We investigated the role of ALA in low-temperature and weak-light stress tolerance in cucumber seedlings. Seedlings grown in different ALA treatments (0, 10, 20, or 30 mg ALA·kg−1 added to substrate) were exposed to low temperature (16/8 ̊C light/dark) and weak light (180 μmol·m−2·s−1 photosynthetically active radiation) for two weeks. Treatment with ALA significantly alleviated the inhibition of plant growth, and enhanced leaf area, and fresh and dry weight of the seedlings under low-temperature and weak-light stress. Moreover, ALA increased chlorophyll (Chl) a, Chl b, and Chl a+b contents. Net photosynthesis rate, stomatal conductance, transpiration rate, photochemical quenching, non-photochemical quenching, actual photochemical efficiency of photosystem II, and electron transport rate were significantly increased in ALA-treated seedlings. In addition, ALA increased root activity and antioxidant enzyme (superoxide dismutase, peroxidase, and catalase) activities, and reduced reactive oxygen species (hydrogen peroxide and superoxide radical) and malondialdehyde accumulation in the root and leaf of cucumber seedlings. These findings suggested that ALA incorporation in the substrate alleviated the adverse effects of low-temperature and weak-light stress, and improved Chl contents, photosynthetic capacity, and antioxidant enzyme activities, and thus enhanced cucumber seedling growth.

Abstract:The vital signaling molecule 5-Aminolevulinic acid (ALA) plays critical roles in signal transduction and biological modulation under abiotic stresses. In this study, we explored the effects of exogenous ALA on low-light (LL) stress-induced photosynthesis and antioxidant system damage in tall fescue (Festuca arundinacea Schreb.) seedlings. LL stress decreased morphological index values and chlorophyll contents, while also reduced net photosynthetic rate (Pn) and the maximum quantum yield of photosystem II photochemistry (Fv/Fm). Notably, these restrictions were substantially alleviated by exogenous ALA. Moreover, the contents of chlorophyll and its synthetic precursors were significantly increased after ALA treatment. Meanwhile, ALA observably enhanced expression level of FaCHLG, FaHEMA, FaPOR, and FaCAO, which encode the chlorophyll precursors biosynthesis enzymes. Exogenous ALA repaired the damage to the chloroplast ultrastructure caused by LL stress and promoted the formation of ordered thylakoids and grana lamella. ALA also improved Rubisco activity and expression level of the photosynthetic enzyme genes FaRuBP, FaPRK, and FaGADPH. Additionally, application of exogenous ALA decreased relative electrolytic leakage and the accumulation of malondialdehyde (MDA), hydrogen peroxide (H2O2), and superoxide radicals (O2∙-), and increased the gene expression levels and activity of antioxidant enzymes. The ratios of ascorbic acid (AsA) to dehydroascorbic acid (DHA) and reduced glutathione (GSH) to oxidized glutathione (GSSG) were also increased significantly by application of ALA. Furthermore, all responses could be reversed by treatment with levulinic acid (LA). Thus, these results indicated that ALA protects tall fescue from LL stress through scavenging ROS, improving photosynthetic enzyme activity levels, increasing photosynthetic pigments contents, repairing chloroplast damage, and enhancing the photosynthesis rate.

Abstract:Salinity is among the main abiotic stresses affecting crop growth and productivity in many regions across the globe. This study aimed to assess the potential of foliar application of 5-aminolevulinic acid (ALA) in improving the salt tolerance in sunflower under an arid climate. The experiment comprised of two sunflower hybrids (Parsun and NK-Senji), foliage sprayed with five levels of ALA (control, 25, 50, 75, and 100 mg L −1 ), under two soil salinity levels (control, 40, and 80 mM NaCl). The results revealed improved plant height, number of leaves, stem diameter, and dry biomass traits in sunflower hybrid 'NK-Senji', while foliar applied ALA at 75 mg L −1 was more useful for improvement in these traits under both growth environments, but the effect was more prominent under saline conditions. Improvement in yield and morphological traits in sunflower due to foliage application of ALA was attributed to an improvement in chlorophyll contents and increased actions of catalase, peroxidase, and superoxidase dismutase. It is concluded that foliar applied ALA at 75 mg L −1 in sunflower hybrid 'NK-Senji' is advantageous for improving the sunflower yield in saline soils.

Abstract:In the present study, the ameliorating effects of 5-aminolevulinic acid (ALA) under cadmium (Cd) stress conditions were studied with special emphasis on root morphology and ultra-structure in oilseed rape. For this purpose, plants were treated hydroponically at three different Cd levels (0, 100, 500μM) and foliar spray of ALA with three concentrations (0, 12.5, 25mg/l) simultaneously. The results showed that foliar application of ALA improved the plant growth, root morphology and reduced the reactive oxygen species and malondialdehyde contents in roots under Cd stress conditions. The higher concentration of Cd (500μM) decreased the activities of antioxidants enzymes like catalase (CAT), superoxide dismutase (SOD), peroxidase (POD) and glutathione reductase (GR) and also reduced the oxidized glutathione and total glutathione contents in roots. Application of ALA at 25mg/l dosage significantly enhanced the antioxidant activities e.g. APX, SOD, POD, and GSH contents under Cd stress. The microscopic micrographs showed that application of exogenous ALA improved the cell structure under Cd toxicity. A whole cell with developed nucleus, nuclear membrane, smooth cell wall, continuous endoplasmic reticulum, and well shaped mitochondria was observed under the combine application of ALA and Cd. These results suggest that, application of ALA helped the plants to improve root growth, root antioxidant enzymes, and ultra-structural changes in root tip cells under fifteen days Cd-induced stress.
Exogenous Salicylic Acid Alleviates Salt Stress by Improving Leaf Photosynthesis and Root System Architecture in Cucumber Seedlings

Abstract:Exogenous salicylic acid (SA) can alleviate salt stress in plants. Previous researches have been focused on SA's role in leaf morphological and physiological characteristics, however, little information is available on its role in both leaf photosynthesis and root system architecture (RSA). A hydroponic study was conducted to analyze biomass, leaf photosynthesis, RSA parameters and expression of RSA genes in cucumber seedlings grown under four treatments, including control (CK), 50 mM NaCl (NaCl), 0.3 mM SA (SA) and 50 mM NaCl plus 0.3 mM SA (NaCl + SA). The results showed that compared with CK, both NaCl and SA treatments severely reduced relative growth rate (RGR) and leaf photosynthetic parameters (F-v/F-m, Phi(II) and Pn). However, compared with NaCl treatment, NaCl + SA treatment significantly increased RGR, leaf photosynthetic (F-v/F-m, Phi(II) and Pn) and RSA (total root length, root surface area, number and total length of primary and secondary LR) parameters. In addition, compared with CK, although both NaCl and SA treatments down-regulated expression of GL2 gene and up-regulated expression of RHD2 gene, NaCl + SA treatment notably up-regulated expression of NAC1, NAC2, GL2, EXP and down-regulated expression of RHD2. It is concluded that exogenous SA alleviated salt stress via effectively promoting leaf photosynthesis and RSA in cucumber seedlings. SA-mediated increment in expression of RSA genes, total length and number of primary and secondary LR could contribute to the improved RSA.
Effect of ALA on Fast Chlorophyll Fluorescence Induction Dynamics of Watermelon Leaves under Chilling Stress

Abstract:Protection of 5-aminolevulinic acid(ALA)on fast chlorophyll fluorescence induction in watermelon(Citrullus lanatus Thunb.Mansfeld)leaves under 4 ℃ low temperature was studied by using a plant efficiency analyzer(PEA)and JIP-test.The results showed that JIP phases of chlorophyll a fluorescence curves in the watermelon leaves decreased during 4 ℃ low temperature stress,whereas much higher fluorescence remained in the leaves treated by ALA.The performance index on an absorption basis(PIABS),performance due to the trapping probability(PTR)and performance due to the conversion of excitation energy to electron transport(PET)of watermelon leaves under 4 ℃ low temperature stress were decreased significantly.However,ALA treatment improved PIABS,PTR and PET of watermelon leaves.After low temperature stress for 96 h,ALA treatment increased the maximum quantum yield of PSⅡ(φPo),the quantum yield of electron transport(φEo),and the efficiency that a trapped exciton moves an electron into the electron transport chain beyond QA-(Ψo)of the watermelon leaves under low temperature stress,while decreased the approximate initial slope of the fluorescence transient(Mo)and relative variable fluorescence intensity at the J-step(Vj)greatly,the latter were assumed to be related with closure of reaction centers.ALA treatment increased the density of reaction centers(RC/CS).And simultaneously,Fk-j,Fj-i and Fi-p,three parameters which were possibly related with the electron acceptability in the acceptor side of electron transport chain of PSⅡ reaction centers were increased greatly in the leaves of ALA treatment.Combined with previous reports,this work proves that ALA improvement on photosynthesis may be correlated with the higher ability of the acceptor side of PSⅡ reaction center to transfer electron to downstream under chilling stress condition.

Abstract:This study evaluates the role of exogenous foliar application of 5-aminolevulinic acid (ALA) on water relations, gas exchange, chlorophyll fluorescence, and the activities and gene expression patterns of antioxidant enzymes in leaves of oilseed rape under drought stress and recovery conditions. Seedlings at four-leaf stage were imposed to well-watered condition (80 % of water-holding capacity) or drought stress (40 % of water-holding capacity) and subsequently foliar sprayed with water or ALA (30 mg l−1). Drought suppressed the accumulation of plant biomass and decreased chlorophyll content and leaf water status (relative water content and water potential). The actual quantum yield of photosystem II and electron transport rates were hampered in parallel to net photosynthetic rate. However, drought stress induced the accumulation of malondialdehyde (MDA) and hydrogen peroxide, enhanced the activities of catalase (CAT), peroxidase (POD), ascorbate peroxidase (APX), glutathione reductase (GR) and superoxide dismutase and up-regulated the expression of APX and GR. After rehydration for 4 days, the growth of drought-treated seedlings was restored to normal level for most of the physiological parameters. Foliar application of ALA maintained relatively higher leaf water status and enhanced chlorophyll content, net photosynthetic rate, actual quantum yield of photosystem II, photochemical quenching, non-photochemical quenching and electron transport rates in stressed leaves. Exogenous ALA also alleviated the accumulation of MDA and hydrogen peroxide, increased the activities of antioxidant enzymes and enhanced the expression of CAT and POD in drought-treated plants. These results indicate that ALA may effectively protect rapeseed seedlings from damage induced by drought stress.
