Abstract:Amounts of cost-effective biological observing and detecting instruments are needed in biology and medical applications. In these demands, a low-cost, portable microscope with light-weight and tele-communication, is very attractive in the resource-limit area. In this manuscript, a portable singlet phase contrast microscope based on smart-phones are proposed. In the optics hardware, an external singlet lens is designed and closely attached before the rear cameras of a smart-phone, which constructs the imaging part of a simplified microscope. And a circular oblique illumination is adopted. In the computational imaging part, the in-focus images recorded by the smart-phone are virtually enhanced and style-transferred to phase contrast images by deep learning methods. Through pairs of experimental image data, we train the deep learning network and demonstrate the effectiveness of this virtual phase contrast imaging approach using multiple samples, e.g., unstained/transparent tumor tissue pathological slide, and H&E (Hematoxylin and Eosin) stained tumor tissue pathological slide. Our results provide a powerful example of low-cost portable phase contrast microscope with high-speed tele-communication abilities in the 5G + mobile internet era.
Cell-Phone-Based Platform for Biomedical Device Development and Education Applications

Abstract:In this paper we report the development of two attachments to a commercial cell phone that transform the phone's integrated lens and image sensor into a 350x microscope and visible-light spectrometer. The microscope is capable of transmission and polarized microscopy modes and is shown to have 1.5 micron resolution and a usable field-of-view of 150 x 50 with no image processing, and approximately 350 x 350 when post-processing is applied. The spectrometer has a 300 nm bandwidth with a limiting spectral resolution of close to 5 nm. We show applications of the devices to medically relevant problems. In the case of the microscope, we image both stained and unstained blood-smears showing the ability to acquire images of similar quality to commercial microscope platforms, thus allowing diagnosis of clinical pathologies. With the spectrometer we demonstrate acquisition of a white-light transmission spectrum through diffuse tissue as well as the acquisition of a fluorescence spectrum. We also envision the devices to have immediate relevance in the educational field.

Abstract:A smartphone-based analysis system is favored for point-of-care testing applications. The present work proposes a novel micro-spectroscopy/imaging system comprising a portable spectrometer as an optical sensor and a compact homemade microscope to acquire the image and spectra of micron-scale regions. Protein concentration quantification based on the bicinchoninic acid method was demonstrated with the proposed micro-spectroscopy/imaging system to analyse the spectrometer signals. Morphologies of onion endothelial and human breast cancer cells, used as biological sample models, were characterized to demonstrate the microscopic imaging capacity of the device. The ability to simultaneously obtain morphological and spectral information using the proposed portable device was demonstrated by examining the 10 mu m sub-pixels of a smartphone screen. These results highlight the potential for adopting a smartphone-based micro-spectroscopy/imaging system for point-of-care testing.

Abstract:In this paper we present for the first time a system comprised of two mobile phones, one for illumination and the other for microscopy, as a portable, user-friendly, and cost-effective microscopy platform for a wide range of applications. Versatile and adaptive illumination is made with a Retina display of an Apple mobile phone device. The phone screen is used to project various illumination patterns onto the specimen being imaged, each corresponding to a different illumination mode, such as bright-field, dark-field, point illumination, Rheinberg illumination, and fluorescence microscopy. The second phone (a Nokia phone) is modified to record microscopic images about the sample. This imaging platform provides a high spatial resolution of at least 2 μm, a large field-of-view of 3.6 × 2.7 mm, and a working distance of 0.6 mm. We demonstrate the performance of this platform for the visualization of microorganisms within microfluidic devices to gather qualitative and quantitative information regarding microorganism morphology, dimension, count, and velocity/trajectories in the x-y plane.

Abstract:High-throughput single-cell heterogeneity imaging and analysis is essential for understanding complex biological systems and for advancing personalized precision disease diagnosis and treatment. Here, we present a miniaturized lensless chemiluminescence chip for high-throughput single-cell functional imaging with subcellular resolution. With the sensitive chemiluminescence sensing and wide field of view of contact lensless imaging, we demonstrated the chemiluminescent imaging of over 1000 single cells, and their membrane glycoprotein and the high-throughput single-cell heterogeneity of membrane protein imaging were examined for precision analysis. Furthermore, the functional adhesion and heterogeneity of single live cells were imaged and explored. This miniaturized lensless on-chip CL-CMOS imaging platform enables high-throughput single-cell imaging and analysis with high sensitivity and subcellular resolution, providing new techniques for the cellular study of biological heterogeneity and has potential application in precision disease diagnosis and treatment at the point-of-care settings.

Abstract:We present a novel, low-cost, portable smartphone-based fluorescence microscope capable of directly detecting single molecules without signal amplification. The setup leverages the image sensors and data handling capacity of mass-produced smartphones, making it adaptable to any smartphone and capable of detecting single molecules across the visible spectral range. We showcase this capability through single-molecule measurements on DNA origami models and super-resolution microscopy of biological cells by single-molecule localization microscopy. This development paves the way for biotechnology innovations making use of massively distributed or personalized assays with single-molecule sensitivity with the potential to revolutionize digital bioassays, point-of-care diagnostics, field expeditions, STEM outreach, and life science education.

Abstract:We demonstrate wide-field fluorescent imaging on a cell-phone, using compact and cost-effective optical components that are mechanically attached to the existing camera unit of the cell-phone. Battery powered light-emitting diodes (LEDs) are used to side-pump the sample of interest using butt-coupling. The pump light is guided within the sample cuvette to excite the specimen uniformly. The fluorescent emission from the sample is then imaged with an additional lens that is put in front of the existing lens of the cell-phone camera. Because the excitation occurs through guided waves that propagate perpendicular to the detection path, an inexpensive plastic color filter is sufficient to create the dark-field background needed for fluorescent imaging. The imaging performance of this light-weight platform (~28 grams) is characterized with red and green fluorescent microbeads, achieving an imaging field-of-view of ~81 mm(2) and a spatial resolution of ~10 μm, which is enhanced through digital processing of the captured cell-phone images using compressive sampling based sparse signal recovery. We demonstrate the performance of this cell-phone fluorescent microscope by imaging labeled white-blood cells separated from whole blood samples as well as water-borne pathogenic protozoan parasites such as Giardia Lamblia cysts.

Abstract:The increasing capabilities and ubiquity of mobile phones and their associated digital cameras offer the possibility of extending low-cost, portable diagnostic microscopy to underserved and low-resource areas. However, mobile phone microscopes created by adding magnifying optics to the phone's camera module have been unable to make use of the full image sensor due to the specialized design of the embedded camera lens, exacerbating the tradeoff between resolution and field of view inherent to optical systems. This tradeoff is acutely felt for diagnostic applications, where the speed and cost of image-based diagnosis is related to the area of the sample that can be viewed at sufficient resolution. Here we present a simple and low-cost approach to mobile phone microscopy that uses a reversed mobile phone camera lens added to an intact mobile phone to enable high quality imaging over a significantly larger field of view than standard microscopy. We demonstrate use of the reversed lens mobile phone microscope to identify red and white blood cells in blood smears and soil-transmitted helminth eggs in stool samples.

Abstract:This article proposes a new device named Wireless Cellscope that combining mobile phone and optical microscope together. The established wireless microscope platform consists of mobile phone, network monitor, miniaturized microscope or high resolution microscope etc. A series of conceptual experiments were performed on microscopic observation of ordinary objects and mice tumor tissue slices. It was demonstrated that, the new method could acquire microscopy images via a wireless way, which is spatially independent. With small size and low cost, the device thus developed has rather wide applicability in non-disturbing investigation of cell/tissue culture and long distance observation of dangerous biological sample etc.

Abstract:We report on the construction of a Fourier plane imaging system attached to a cell phone. By illuminating particle suspensions with a collimated beam from an inexpensive diode laser, angularly resolved scattering patterns are imaged by the phone's camera. Analyzing these patterns with Mie theory results in predictions of size distributions of the particles in suspension. Despite using consumer grade electronics, we extracted size distributions of sphere suspensions with better than 20 nm accuracy in determining the mean size. We also show results from milk, yeast, and blood cells. Performing these measurements on a portable device presents opportunities for field-testing of food quality, process monitoring, and medical diagnosis.

Abstract:In this article, we demonstrated a handheld smartphone fluorescence microscope (HSFM) that integrates dual-functional polymer lenses with a smartphone. The HSFM consists of a smartphone, a field-portable illumination source, and a dual-functional polymer lens that performs both optical imaging and filtering. Therefore, compared with the existing smartphone fluorescence microscope, the HSFM does not need any additional optical filters. Although fluorescence imaging has traditionally played an indispensable role in biomedical and clinical applications due to its high specificity and sensitivity for detecting cells, proteins, DNAs/RNAs, etc., the bulky elements of conventional fluorescence microscopes make them inconvenient for use in point-of-care diagnosis. The HSFM demonstrated in this article solves this problem by providing a multifunctional, miniature, small-form-factor fluorescence module. This multifunctional fluorescence module can be seamlessly attached to any smartphone camera for both bright-field and fluorescence imaging at cellular-scale resolutions without the use of additional bulky lenses/filters; in fact, the HSFM achieves magnification and light filtration using a single lens. Cell and tissue observation, cell counting, plasmid transfection evaluation, and superoxide production analysis were performed using this device. Notably, this lens system has the unique capability of functioning with numerous smartphones, irrespective of the smartphone model and the camera technology housed within each device. As such, this HSFM has the potential to pave the way for real-time point-of-care diagnosis and opens up countless possibilities for personalized medicine.
Improving Cellphone Microscopy Imaging with Contact Ball Lenses

Abstract:It is shown that the resolution of cellphone-based microscopy can be improved with the use of millimeter-scale ball lenses with index n=1.85 placed in contact with fluorescent (FL) objects represented by dye-doped microspheres. Using the virtual imaging through such ball lenses, the resolution improvement from ~35 μm to ~2 μm is achieved over a field-of-view of about 0.1 × 0.1 mm 2 . Further improvement of resolution and field-of-view can be expected with optimization of parameters of the contact ball lenses that can lead to wide applications of the proposed technology in mobile biomedical imaging.

Abstract:By taking advantage of the inherent flexibility of low-cost 3D printing materials, we achieved an optical focus tuning accuracy of ~5 micron with a novel structure design. It shrinks the mechanical displacement by a factor of ~11 through a seesaw-like component. Combing with the built-in flashlight illumination and an off-the-shelf smartphone lens, the total manufacturing cost of our smartphone-based microscope is less than 4 USD. We demonstrated the capability of this design in imaging thick biological specimens. We further applied this device in the cell culture monitoring of VX2 tumor cells because of its portability and flexibility.

Abstract:Photomicrographs of thin sections provide a swift and efficient means of sharing information for consultation, education, documentation and publication within the Geosciences and related areas. In general terms, the main way to capture digital microscopic images involves the use of a mounted camera unit on a high-end costly benchtop microscope. However, freehand methods to capture microscope-scale images using a smartphone, as well as smartphone adapters that can be attached to a microscope have emerged during recent years. This paper presents the design features of a costless system able to obtain photomicrographs without requiring a conventional microscope. The imaging device is comprised of a mini-objective lens attached to a smartphone and a structure that allows it to focus as well as to rotate the stage and to insert/remove a polarized sheet. The quality and magnification of the images attainable from the new design is comparable to the images normally obtained by a conventional petrographic microscope using a ×4 objective and a ×10 ocular (total magnification ×40).

Abstract:Multi-spectral imaging technology is helpful to obtain better contrast and more details of images at different wavelengths compared with monochromatic imaging or conventional RGB imaging technologies. And in resource limited areas, microscopes with features of low-cost and portability are extremely attractive. In this manuscript, we demonstrate that cost-effective lens-less microscopy can be combined with the multi-spectral imaging technology, which can be alternative to conventional approaches and applied in biology, chemistry, environmental science. As a proof of this concept, we present a novel portable multi-spectral lens-less microscope (MSLLM). An outstanding highlight of our setup is its wavelength-self-calibrating imaging sensor (WSCIS), which can provide computationally optimized wavelength value (COWV) for LED illumination in real time. For each illumination, 5 holograms of the sample at 5 different heights are captured. Through the COWVs and phase retrieval algorithms, 7 multi-spectral images (FOV: 4.8 mm*4.6 mm, resolution: similar to 1 mu m) can be reconstructed. We believe that our MSLLM can be widely applied to multi-spectral microscopy in low-cost required scenes.

Abstract:Spectroscopy plays a pivotal role in various applications for industrial and commercial analysis. Conventional spectroscopy instruments include various discrete optical components like light sources, monochromators and detectors. These components are mechanically intricate and require space for the mechanisms that adjust and calibrate them, which increases the instrument’s footprint and make them less adjustable for portable applications. This drawback initiated the increase in the demand for miniatured spectral detection systems. Here, we present a low-cost ultra-compact miniaturized spectral detection system, which integrates both linear variable light sources, sample chambers and detectors within a centimetre-scale chip. The linear variable light generation across wavelengths ranging from 480 nm to 600 nm is achieved by stimulating different regions of the ZnCdSeS bandgap-graded semiconductor nanofilm with ultraviolet light. We demonstrate the capability of the system with precise identification of liquids. Our device overcomes the typical miniaturization limits imposed by discrete optical components, and long light paths in conventional spectroscopy instruments and opens up numerous potential applications across various fields due to its portability, ease of use, and lower cost.

Abstract:Microscopy is an important tool in biology and medicine, but it is often limited to high numerical aperture (NA) with a short working distance(w(d)), such as NA=0.6 then W-d <1mm. For imaging living cells in culture, a high numerical aperture and long working distance of optical imaging structure is required, and the common microscopy objective is no good here. In order to meet the single living cell imaging, a novel optical design has been performed in this paper, which is character of ultra-long working distance W-d = 15mm and high numerical aperture NA=0.7. An optical imaging system with dual modes information of fluorescence and spectrum can be developed for the subcellular imaging of cells attached on surface of vessel or free-floating in culture.

Abstract:Mobile-phones have facilitated the creation of field-portable, cost-effective imaging and sensing technologies that approach laboratory-grade instrument performance. However, the optical imaging interfaces of mobile-phones are not designed for microscopy and produce spatial and spectral distortions in imaging microscopic specimens. Here, we report on the use of deep learning to correct such distortions introduced by mobile-phone-based microscopes, facilitating the production of high-resolution, denoised and colour-corrected images, matching the performance of benchtop microscopes with high-end objective lenses, also extending their limited depth-of-field. After training a convolutional neural network, we successfully imaged various samples, including blood smears, histopathology tissue sections, and parasites, where the recorded images were highly compressed to ease storage and transmission for telemedicine applications. This method is applicable to other low-cost, aberrated imaging systems, and could offer alternatives for costly and bulky microscopes, while also providing a framework for standardization of optical images for clinical and biomedical applications.

Towards ultra-low-cost smartphone microscopy

Abstract:The outbreak of COVID-19 exposed the inadequacy of our technical tools for home health surveillance, and recent studies have shown the potential of smartphones as a universal optical microscopic imaging platform for such applications. However, most of them use laboratory-grade optomechanical components and transmitted illuminations to ensure focus tuning capability and imaging quality, which keeps the cost of the equipment high. Here we propose an ultra-low-cost solution for smartphone microscopy. To realize focus tunability, we designed a seesaw-like structure capable of converting large displacements on one side into small displacements on the other (reduced to ~9.1%), which leverages the intrinsic flexibility of 3D printing materials. We achieved a focus-tuning accuracy of ~5 micron, which is 40 times higher than the machining accuracy of the 3D-printed lens holder itself. For microscopic imaging, we use an off-the-shelf smartphone camera lens as the objective and the built-in flashlight as the illumination. To compensate for the resulting image quality degradation, we developed a learning-based image enhancement method. We use the CycleGAN architecture to establish the mapping from smartphone microscope images to benchtop microscope images without pairing. We verified the imaging performance on different biomedical samples. Except for the smartphone, we kept the full costs of the device under 4 USD. We think these efforts to lower the costs of smartphone microscopes will benefit their applications in various scenarios, such as point-of-care testing, on-site diagnosis, and home health surveillance.

Abstract:Phase contrast imaging is widely employed in the physical, biological, and medical sciences. However, typical implementations involve complex imaging systems that amount to in-line interferometers. We adapt differential phase contrast (DPC) to a dual-phone illumination-imaging system to obtain phase contrast images on a portable mobile phone platform. In this dual phone differential phase contrast (dpDPC) microscope, semicircles are projected sequentially on the display of one phone, and images are captured using a low-cost, short focal length lens attached to the second phone. By numerically combining images obtained using these semicircle patterns, high quality DPC images with ≈ 2 micrometer resolution can be easily acquired with no specialized hardware, circuitry, or instrument control programs.
