Abstract:Photoacoustic imaging (PAI) is a noninvasive, nonionizing modality based on the differences in light absorption of various biological tissues. PAI utilizes the endogenous contrast characteristics of traditional optical imaging, while benefiting from high spatial resolution of the ultrasound imaging. A PAI system was developed to reconstruct the two-dimensional cross section image and to visualize the cerebrovascular activities of mouse in vivo. The spatial resolution of the PAI system was determined to be 0.110 mm by a two-point-source phantom with the Rayleigh criterion. The potential applications of the system were clearly demonstrated by successfully mapping a traumatic lesion in the mouse brain cerebral cortex, by its ability to monitor physiological changes in the brain due to carotid ligation and drug stimulation, and two-dimensional sliced images of a traumatic mouse brain at different depths were also provided. Our experimental results indicate that PAI has the potential for studying of traumatic brain injury and physiological functions of the brain.

Abstract:Non-invasive laser-induced photoacoustic tomography ( PAT) is an emerging imaging modality that has the potential to image the dynamic function of the brain due to its unique ability of imaging biological tissues with high optical contrast and ultrasound resolution. Here we report the first application of our finite-element-based PAT for imaging of epileptic seizures in an animal model. In vivo photoacoustic images were obtained in rats with focal seizures induced by microinjection of bicuculline, a GABA(A) antagonist, into the neocortex. The seizure focus was accurately localized by PAT as confirmed with gold-standard electroencephalogram ( EEG). Compared to the existing neuroimaging modalities, PAT not only has the unprecedented advantage of high spatial and temporal resolution in a single imaging modality, but also is portable and low in cost, making it possible to bring brain imaging to the bedside.

Abstract:Epilepsy is a chronic neurologic disorder caused by abnormal discharge of neurons in the brain. Imaging examination is of great significance in the diagnosis and treatment of epilepsy, but some patients failed to find any apparent abnormality in conventional magnetic resonance imaging. Recently, a variety of imaging post-processing techniques are applied in clinical practice, which have significantly improved the detection rate of epileptic lesions. In this paper, the research progress of various imaging post-processing techniques in magnetic resonance imaging negative focal epilepsy is reviewed.

Abstract:Multi-frequency electrical impedance tomography (MFEIT) reconstructs the image of conductivity inside the human body based on the dependence of tissue conductivity on frequency. As there exist differences in the conductivity over frequency between blood, ischemic cortical tissue and normal cortical tissue, MFEIT has potential application in the detection of acute stroke. However, because the conductivity distribution of the human head is highly inhomogeneous and the conductivities of normal head tissue and stroke lesion tissue both change with frequency, the anomaly and normal head tissues are often mixed together in the reconstructed image, which makes it difficult to discern the anomaly. Here we present a spectral decomposition frequency-difference (SD-FD) imaging algorithm in an attempt to address this issue: firstly, we reconstruct so-called EIT spectral images according to the conductivity spectra of tissues; secondly, we obtain the EIT image of the anomaly from the spectral images by using independent component analysis. The results show that the proposed algorithm is capable of detecting the anomaly in a numerical head phantom, as well as in a realistic human head tank with frequency-dependent and heterogeneous conductivities distribution. The proposed SD-FD algorithm may support MFEIT use for human stroke imaging in the future.

Abstract:A real-time three-dimensional (3D) photoacoustic imaging system was developed for epilepsy imaging in small animals. The system is based on a spherical array containing 192 transducers with a 5 MHz central frequency. The signals from the 192 transducers are amplified by 16 homemade preamplifier boards with 26 dB and multiplexed into a 64 channel data acquisition system. It can record a complete set of 3D data at a frame rate of 3.3 f/s, and the spatial resolution is about 0.2 mm. Phantom experiments were conducted to demonstrate the high imaging quality and real time imaging ability of the system. Finally, we tested the system on an acute epilepsy rat model, and the induced seizure focus was successfully detected using this system.

Abstract:While brain imaging and electrophysiology play a central role in neuroscience research and in the evaluation of neurological disorders, a single noninvasive modality that offers both high spatial and temporal resolution is currently not available. Here we show in an acute epilepsy rat model that photoacoustic tomography (PAT) can noninvasively track seizure brain dynamics with both high spatial and temporal resolution, and at a depth that is clinically relevant. The noninvasive yet whole surface and depth capabilities of the PAT system allowed us to actually see what is happening during ictogenesis in terms of seizure onset and spread. Both seizure onset and propagation were tomographically detected at a spatial resolution of 150μm and a temporal resolution of 300ms, respectively. The current study lends support to the theory that seizure onset and spread involves a rich interplay between multiple cortical and subcortical brain areas during the onset and spread of epileptic seizures. Dynamical changes of vasculature during epileptiform events were also detected with high spatiotemporal resolution. Together, these findings suggest that PAT represents a powerful tool for noninvasively mapping seizure onset and propagation patterns, and the ‘functional’ connectivity within epileptic brain networks.

Abstract:For drug-resistant epilepsy patients, epilepsy surgery is the only option. In clinic, this is done with subdural electrode array with a disappointing low spatial resolution of ~ 1cm. In order to improve clinical cure rate, ultrasound imaging technique can be cooperated with Electrocorticography to monitor the maps of seizure. We propose to develop the implanted single ultrasound transducers 3mm×1.5mm×0.8mm to satisfy clinical requirement. For the single transducer recording, the laws of epilepsy corresponding with ultrasonic signals need to be rediscovered. In order to verify our proposals, the phantom experiment is designed to simulate the epileptogenic zone. The experiment result shows the power spectral density is in direct proportion to simulation Cerebral Blood Volume increased. This device makes high resolution 3D seizure maps possibly to guide the epilepsy surgery.

Abstract:Diffuse optical tomography (DOT) is a promising functional imaging modality due to its ability to provide quantitative and dynamic tomographic imaging of brain functions. This pilot study was conducted to demonstrate that DOT can be used to visualize the changes in local hemodynamics during seizures. The focal seizure was induced by microinjection of 10 mu l of 1.9 mM GABA(A) antagonist bicuculline methiodide (BMI) into the left parietal neocortex of male Harlen Sprague-Dawley rats, which was imaged by a multispectral continuous-wave DOT system. Functional images were obtained by our finite element based image reconstruction algorithm. A series of dynamic 2D images were obtained to delineate the time course of concentration changes of oxyhaemoglobin, deoxyhaemoglobin, and total hemoglobin in the rat brain during seizure onset. The BMI induced epileptic foci were localized and observed over time from the images obtained. Our results suggest that diffuse optical tomography may be a promising modality for epilepsy imaging due to its ability to localize epileptic foci as well as its potential to map the functional activity in the area of human cerebral cortex in planning of epilepsy surgery. Abstract:A full-view photoacoustic tomography system with dual-scanning using a linear transducer array for fast imaging of complicated blood network was developed. In this system, a 128-element linear transducer array was used to detect photoacoustic signals by combined scanning of electronic scan and mechanical scan. An improved limited-field filtered back projection algorithm with directivity factors was applied to reconstruct the distribution of the absorbed optical energy deposit. An in vivo experiment on a mouse brain was performed to evaluate the ability of this composite system. A clear view of the cerebrovascular network on the brain cortex was acquired successfully. Furthermore, the reconstruct images with different number of scanning positions were also investigated and analyzed to induce a compromised proposal between scanning time and scanning number. The experimental results demonstrate the multi-element photoacoustic imaging system has the potential to acquire the time-resolved functional information for fundamental research of small animal brain imaging. © 2009 SPEI.

Abstract:We describe a photoacoustic image reconstruction algorithm that is based on the finite-element solution to the photoacoustic wave equation in the frequency domain. Our reconstruction approach is an iterative Newton method coupled with combined Marquardt and Tikhonov regularizations that can extract the spatial distribution of optical-absorption property in heterogeneous media. We demonstrate this algorithm by using phantom and chicken bone measurements from a circular scanning photoacoustic tomography system. The results obtained show that millimeter-sized phantom objects and chicken bones and/or joints can be clearly detected using our finite-element-based photoacoustic tomography method. Abstract:We describe a finite element reconstruction algorithm for time-domain photoacoustic tomography. The algorithm is based on an iterative Newton method coupled with the time-domain Helmholtz-like photoacoustic wave equation subject to the first-order Bayliss-Turkel radiation boundary conditions. The algorithm is tested and evaluated using a series of simulated and tissue-like phantom data where objects are embedded in a circular background. The results show that quantitatively accurate photoacoustic images can be recovered using our time-domain reconstruction algorithm.

Abstract:Photoacoustic imaging combines the contrast advantage of pure optical imaging and the resolution advantage of pure ultrasonic imaging. It has become a popular research subject at present. A fast photoacoustic imaging system based on multi-element linear transducer array and phase-controlled focus method was developed and tested on phantoms and tissues. A Q switched Nd:YAG laser operating at 532nm was used in our experiment as thermal source. The multi-element linear transducer array consists of 320 elements. By phase-controlled focus method, 64 signals, one of which gathered by 11-group element, make up of an image. Experiment results can map the distribution of the optical absorption correctly. The same transducer array also can operate as a conventional phase array and produced ultrasound imaging. Compared to other existing technology and algorithm, the PA imaging based on transducer array was characterize by speediness and convenience. It can provide a new approach for tissue functional imaging in vivo, and may have potentials in developing into an appliance for clinic diagnosis.

Abstract:Photoacoustic imaging is a potential candidate for in vivo brain imaging, whereas, its imaging performance could be degraded by inhomogeneous multi-layered media, consisted of scalp and skull. In this work, we propose a low-artifact photoacoustic microscopy (LAPAM) scheme, which combines conventional acoustic-resolution photoacoustic microscopy with scanning acoustic microscopy to suppress the reflection artifacts induced by multi-layers. Based on similar propagation characteristics of photoacoustic signals and ultrasonic echoes, the ultrasonic echoes can be employed as the filters to suppress the reflection artifacts to obtain low-artifact photoacoustic images. Phantom experiment is used to validate the effectiveness of this method. Furthermore, LAPAM is applied for in-vivo imaging mouse brain without removing the scalp and the skull. Experimental results show that the proposed method successfully achieves the low-artifact brain image, which demonstrates the practical applicability of LAPAM. This work might improve the photoacoustic imaging quality in many biomedical applications which involve tissues with complex acoustic properties, such as brain imaging through scalp and skull.

Abstract:Abstract Novel advances in the field of brain imaging have enabled the unprecedented clinical application of various imaging modalities to facilitate disease diagnosis and treatment. Electrical impedance tomography (EIT) is a functional imaging technique that measures the transfer impedances between electrodes on the body surface to estimate the spatial distribution of electrical properties of tissues. EIT offers many advantages over other neuroimaging technologies, which has led to its potential clinical use. This qualitative review provides an overview of the basic principles, algorithms, and system composition of EIT. Recent advances in the field of EIT are discussed in the context of epilepsy, stroke, brain injuries and edema, and other brain diseases. Further, we summarize factors limiting the development of brain EIT and highlight prospects for the field. In epilepsy imaging, there have been advances in EIT imaging depth, from cortical to subcortical regions. In stroke research, a bedside EIT stroke monitoring system has been developed for clinical practice, and data support the role of EIT in multi-modal imaging for diagnosing stroke. Additionally, EIT has been applied to monitor the changes in brain water content associated with cerebral edema, enabling the early identification of brain edema and the evaluation of mannitol dehydration. However, anatomically realistic geometry, inhomogeneity, cranium completeness, anisotropy and skull type, etc., must be considered to improve the accuracy of EIT modeling. Thus, the further establishment of EIT as a mature and routine diagnostic technique will necessitate the accumulation of more supporting evidence.

Abstract:Epilepsy surgery often depends on the accurate positioning of lesions; however, current positioning methods have some limitations. The cicctrocorticogram ( ECoG) in clinical inspection has a high temporal resolution, but its spatial resolution is below the accepted standard. Based on the neurovascular coupling mechanism in the human brain, we propose a new epileptic foci detection method by detecting cerebral microvascular blood flow with microphotodiodes and ultrasound transducers. The results of experiments conducted on animals show that the light signal on the surface of the cortex has good correspondence with the ECoG signal. The spectra of the ultrasound signal also matches that of the electrical signal within a depth of 1 mm below the cortex. Therefore, we believe that this method can improve the spatial resolution of epileptic lesion detection, and it can potentially be used for depth inspection.

The images are obtained by a finite-element reconstruction algorithm that is based on the Helmholtz-like photoacoustic wave equation in the frequency domain. Our reconstruction approach is an iterative Newton method coupled with combined Marquardt and Tikhonov regularizations that can extract the spatial distribution of relative optical absorption property in heterogeneous media. We demonstrate experimental images in single- and multiple-object configurations with a circular scanning photoacoustic tomographic system. The results obtained show that millimeter-size nanoparticle-containing objects can be clearly detected in terms of position, size, and relative optical properties. Abstract:Minimally-invasive monitoring of electrophysiological neural activities in real-time—that enables quantification of neural functions without a need for invasive craniotomy and the longer time constants of fMRI and PET—presents a very challenging yet significant task for neuroimaging. In this paper, we present in vivo functional PA (fPA) imaging of chemoconvulsant rat seizure model with intact scalp using a fluorescence quenching-based cyanine voltage-sensitive dye (VSD) characterized by a lipid vesicle model mimicking different levels of membrane potential variation. The framework also involves use of a near-infrared VSD delivered through the blood-brain barrier (BBB), opened by pharmacological modulation of adenosine receptor signaling. Our normalized time-frequency analysis presented in vivo VSD response in the seizure group significantly distinguishable from those of the control groups at sub-mm spatial resolution. Electroencephalogram (EEG) recording confirmed the changes of severity and frequency of brain activities, induced by chemoconvulsant seizures of the rat brain. The findings demonstrate that the near-infrared fPA VSD imaging is a promising tool for in vivo recording of brain activities through intact scalp, which would pave a way to its future translation in real time human brain imaging.

Abstract:We present a fast full-view photoacoustic imaging system for visualizing tissue structures using a linear transducer array with combined scan. In this system, a 128-element linear transducer array was used to detect photoacoustic signals by combined scanning of electronic scan and mechanical scan. The experiments of phantoms and in vivo blood vessels in a rat brain were performed with this system. And a clear view of the curve boundaries of objects and the network of blood vessels of rat's brain were acquired. The experimental results demonstrate the multi-element photoacoustic imaging system has the ability of imaging complicated structures of objects.
