Abstract:Novel cADPR mimics, which integrate nucleobase, northern and southern ribose modifications were synthesized. The key steps of the synthesis were a Cu(I)-catalyzed Huisgen [3+2] cycloaddition and a microwave-assisted intramolecular pyrophosphorylation. Preliminary biological investigations showed that these cADPR mimics are membrane-permeating agonists of the calcium signaling pathway. The introduction of chlorine or fluorine at the 2'-position of the southern riboses led to a decrease of activity. The existence of a hydrophobic group on the 3'-OH of the southern riboses does not obviously alter the agonistic activity.

Abstract:New cADPR mimic 5 that integrates the simplified nucleobase and ribose was designed and synthesized by using Cu(Ⅰ)-catalyzed Huisgen [3+2] cycloaddition to build the simplified triazole nucleobase and microwave-assisted reaction to carry out intramolecular pyrophosphorylation.Primary biological investigation showed that the introduction of an ether strand to replace northern and southern riboses could retain calcium inducing activity,but the introduction of a carbon strand to replace the northern ribose could lead to decreased activity.

Abstract:Cyclic ADP-ribose (cADPR) is an endogenous Ca2+-mobilizing second messenger in many cell types and organisms. Although the biological activity of several modified analogues of cADPR has been analyzed, most of these structures were still very similar to the original molecule. Recently, we have introduced simplified analogues in which the northern ribose (N-1-linked ribose) was replaced by an ether strand (Gu, X., Yang, Z., Zhang, L., Kunerth, S., Fliegert, R., Weber, K., Guse, A. H., and Zhang, L. ( 2004) J. Med. Chem. 47, 5674 - 5682). Here we also demonstrate that the southern ribose ( N-9-linked ribose) can be replaced by an ether strand resulting in N-1-[(phosphoryl-O-ethoxy)methyl]- N-9-[(phosphoryl-O-ethoxy)- methyl]-hypoxanthine-cyclic pyrophosphate (cIDP-DE). This minimal structural analogue of cyclic ADP-ribose released Ca2+ from intracellular stores of permeabilized Jurkat T lymphocytes. In intact T lymphocytes initial subcellular Ca2+ release events, global Ca2+ release, and subsequent global Ca2+ entry were observed. Cardiac myocytes freshly prepared from mice responded to cIDP-DE by increased recruitment of localized Ca2+ signals and by global Ca2+ waves.

Abstract:Ca2+ signaling is one of the most important intracellular signal transduction pathways in diverse cells. Local Ca2+ signals are mainly due to release from internal stores and these calcium release events are controlled by various second messengers and regulated by Ca2+ channel-associated proteins. Cyclic ADP-ribose (cADPR), a metabolite of NAD+, was discovered in 1987 and is a signaling molecule to regulate calcium mobilization via ryanodine receptors (RyR) from intracellular stores in a wide variety of biological systems. A main approach to explore the molecular mechanism of calcium release is to investigate the relationship of the structure of cADPR and cADPR analogues with their respective biological activities. In addition to the structure activity relationship, such studies result in novel antagonists and agonists which serve as the tools for mechanistic studies in the complex cellular system. This review outlines the design and discovery of structural mimics of cADPR (cIDPRE and cIDPDE) with replacement of one or both of the ribose moieties by ether bridges. Compounds of the type cIDPDE or cIDPRE may thus both be used as membrane-permeant, agonists of the cADPR/Ca2+ signaling system in intact cells.

Abstract:A series of cyclic ADP-ribose (cADPR) analogs, in which modifications mainly focused on riboses, was synthesized in order to explore the molecular mechanism Of calcium release regulated by cADPR. Biological activities investigated in intact T-lymphocytes showed that the structurally simplified analogs, NI-ethoxyrnethyl-substituted cyclic inosine diphosphoribose (cIDPRE), N1,N9-diethoxymethyl-substituted cyclic inosine diphosphoribose (cIDPDE), and N1-ethoxymethil-substituted cyclic adenosine diphosphoribose (cADPRE) in which the northern ribose or both northern and southern riboses were replaced by ether linkages are membrane-permeant and induce calcium release from intracellular stores. This research has provided novel molecules to probe cADPR-mediated calcium signaling and enlarges Our knowledge of the structure-activity relationships of cADPR analogs.

Abstract:Cyclic adenosine diphosphate ribose is an endogenous Ca(2+) mobilizer involved in diverse cellular processes. A cell membrane-permeable cyclic adenosine diphosphate ribose analogue, cyclic inosine diphosphoribose ether (cIDPRE), can induce Ca(2+) increase in intact human Jurkat T-lymphocytes. Here we synthesized a coumarin-caged analogue of cIDPRE (Co-i-cIDPRE), aiming to have a precisely temporal and spatial control of bioactive cIDPRE release inside the cell using UV uncaging. We showed that Co-i-cIDPRE accumulated inside Jurkat cells quickly and efficiently. Uncaging of Co-i-cIDPRE evoked Ca(2+) release from endoplasmic reticulum, with concomitant Ca(2+) influx in Jurkat cells. Ca(2+) release evoked by uncaged Co-i-cIDPRE was blocked by knockdown of ryanodine receptors (RyRs) 2 and 3 in Jurkat cells. The associated Ca(2+) influx, on the other hand, was abolished by double knockdown of Stim1 and TRPM2 in Jurkat cells. Furthermore, Ca(2+) release or influx evoked by uncaged Co-i-cIDPRE was recapitulated in HEK293 cells that overexpress RyRs or TRPM2, respectively, but not in wild-type cells lacking these channels. In summary, our results indicate that uncaging of Co-i-cIDPRE incites Ca(2+) release from endoplasmic reticulum via RyRs and triggers Ca(2+) influx via TRPM2.

Abstract:For the investigation of the structure-activity relationship of the analogues of cyclic ADP-ribose (cADPR), a novel cyclic IDP-ribose analogue, cIDPRN, in which the northern ribose was replaced by N-carbobenzyloxy-alkylimine bridge was designed and synthesized. The enzymatic stability of cIDPRN was evaluated by the incubation with Jurkat T-lymphocytes for 0, 2 and 18 h. The result analyzed by capillary electrophoresis indicated that cIDPRN antagonized the hydrolysis, whereas cADPR was easily hydrolyzed. The Ca2+ signal release behavior was investigated in intact Jurkat T-lymphocytes by spectrofluorometer. It showed that cIDPRN induced calcium release either in extracellular Ca2+-containing or Ca2+-free condition. The result indicated that cIDPRN was a mild membrane-permeant agonist of cADPR/RyR signaling system. This study provided further information for understanding the effect of structure of northern ribose moiety of cIDPR on the calcium motivation activity.

Abstract:A purine nucleobase-simplified cyclic ADP ribose (cADPR) analogue was synthesized, in which a 1,2,3-triazole-4-amide was constructed, instead of a purine moiety, and the northern ribose was replaced by an ether strand. Compound exhibits calcium release activity in intact T-lymphocytes and indicates that it is a membrane-permeable cADPR mimic. Thus, the cADPR analogue containing 1,2,3-triazole-4-amide provides a novel template for further designing cADPR analogues and elucidating their structure-activity relationships.

Abstract:Cyclic ADP-ribose (cADPR) is not only a potent endogenous calcium modulator but also a second messenger. However, studies on the mechanism of cADPR action were limited due to its instability and lack of available structural modifications in the N1-glyosyl unit of cADPR. In the present work, a series of N1-glycosyl mimics with different configurational glycosyls or an ether strand were designed and synthesized mimicking the furanose ring. S(N)2 substitutions were carried out between the protected inosine and glycosyl triflates to form the N1-glycosylinosine derivatives, accompanied with some O6-glycosyl-substituted as side products. The intramolecular cyclization was followed the strategy described by Matsuda et al. It was found that the 8-unsubstituted substrate could also be used to construct the intramolecular cyclic pyrophosphate. The activities of N1-glycosyl-substituted cADPR mimics were evaluated by induced Ca2+ release in rat brain microsomes and HeLa cells. It was found that the configuration of the N1-glycosyl moiety in cADPR is not a critical structural factor for retaining the activity of mobilizing Ca2+ release. More interestingly, the N1-acyclic analogue 6 exhibited strong activity by inducing Ca2+ release in both rat brain microsomes and HeLa cells. It constitutes a useful tool for further studies.

Abstract:Cyclic ADP-ribose (cADPR) is a naturally occurring metabolite of NAD+ that is capable of mobilizing calcium from intracellular stores in a variety of biological systems. This chapter presents the procedures for the synthesis of three 8-substituted (8-amino-, 8-azido-, and 8-bromo-) analogs and a caged analog of cyclic ADP-ribose. The use of these analogs demonstrates that the occupation of the cADPR receptor site does not necessarily lead to Ca2+ release; appropriate interactions between the 8-position of the ligand and receptor are also required. All three compounds are competitive antagonists that bind to the same site as cADPR. The 8-azido-cADPR, with its photoactive azido group, is particularly useful as a photoaffinity probe for the identification and characterization of cADPR-binding proteins. The synthesis of high specific activity 8-azido-[32P]cADPR for use in photoaffinity labeling experiments is also described in the chapter. The use of photoactivable “caged” compounds represents a powerful tool for controlling the release of biologically active molecules in intact cells. Generally, the caging group incorporated into an active molecule is designed to render the molecule inactive. Removal of the caging group by photolysis results in liberation of the active substance, which can be controlled both spatially and temporally.

Abstract:Cyclic adenosine diphosphate ribose (cADPR) is an endogenous Ca2+ mobilizer involved in diverse cellular processes. Mimics of cADPR play a crucial role in investigating the molecular mechanism(s) of cADPR-mediated signaling. Here, compound 3, a mimic of cADPR in which a neutral triazole moiety and an ether linkage were introduced to substitute the pyrophosphate and northern ribose components, respectively, was synthesized for the first time. The pharmacological activities in Jurkat cells indicated that this mimic is capable of penetrating plasma membrane and inciting Ca2+ release from the endoplasmic reticulum (ER) through the action of ryanodine receptors (RyRs) and triggering Ca2+ influx. Furthermore, a uridine moiety was introduced in place of adenine and the new cADPR mimics 4 and 5 were synthesized. The results of biological investigation showed that these mimics also targeted RyRs and retained moderate Ca2+ agonistic activities. The results indicated that the neutral cADPR mimics had the same targets for inducing Ca2+ signaling.

Abstract:Cyclic nucleotides are second messengers exerting their cellular effects mainly through protein phosphorylation. A new member of this family, cyclic ADP-ribose, is involved, instead, in mediating mobilization of Ca+2 from intermal stores. The structure of this nucleotide has now been determined by X-ray crystallography and accumulating evidence indicates it may be an endogenous modulator of the Ca+2 release mechanism. This article summarizes the current knowledge of the structure, the mechanism of action and the metabolic enzymes of this novel nucleotide. With this new addition, the signalling functions of the cyclic nucleotide family are now extended from protein phosphorylation to Ca+2 signalling.

Abstract:Novel analogues of cADPR with adenine as base and ether (10a) or different alkane chain (10b-d) substitutions of the northern ribose were synthesized from protected imidazole nucleoside 1 in good yields. The pharmacological activities of cyclic inosine diphosphoribose ether (cIDPRE) and the compounds (10a-d) were analyzed in intact human Jurkat T-lymphocytes. The results indicate that the analogues 10a-d permeate the plasma membrane and are weak agonists of the cADPR/ryanodine receptor signaling system in intact human Jurkat T cells. They are the first membrane-permeant and biologically active cADPR analogues that contain ether or alkane bridges instead of the northern ribose and retain adenine as its base.

Abstract:ADP-ribosyl cyclase and CD38 are multi-functional enzymes involved in calcium signaling. Both can cyclize NAD and its guanine analog, NGD, at two different sites of the purine ring, N1 and N7, respectively, to produce cyclic ADP-ribose (cADPR) and cyclic GDP-ribose, a fluorescent but inactive analog. Both enzymes can also catalyze the exchange of the nicotinamide group of NADP with nicotinic acid, producing yet another potent activator of Ca2+ mobilization, nicotinic acid adenine dinucleotide phosphate (NAADP). The Ca2+ release mechanism activated by NAADP is totally independent of cADPR and inositol trisphosphate indicating it is a novel and hitherto unknown Ca2+ signaling pathway. This article summarizes the current results on the structures and activities of cADPR, NAADP and the enzymes that catalyze their syntheses. A comprehensive model accounting for the novel multi-functionality of ADP-ribosyl cyclase and CD38 is presented.

Abstract:Cyclic ADP-ribose (cADPR) is a Ca2+-mobilizing cyclic nucleotide derived from NAD(+) Accumulating evidence indicates that it is an endogenous modulator of the Ca2+-induced Ca2+ release mechanism in cells. In this study, we show that ADP-ribosyl cyclase catalyzes the cyclization of not only NAD(+) but also several of its analogs with various purine bases (guanine, hypoxanthine, or xanthine) substituting for adenine. Unlike cADPR, the resulting cyclic products are fluorescent. Comparisons with various model compounds indicate that only 7-methyl substituted purine nucleosides and nucleotides are fluorescent, and the pH-dependence of their UV spectra is most similar to that of the fluorescent cADPR analogs, indicating that the site of cyclization of these analogs is at the N7-position of the purine ring. This finding is novel since the site of cyclization is at the N1-position for cADPR as determined by X-ray crystallography. That a single enzyme can cyclize a variety of substrates at two different sites has important implications mechanistically, and a model is proposed to account for these novel catalytic properties. Among the analogs synthesized, cyclic GDP-ribose is highly resistant to hydrolysis, while cyclic IDP-ribose can be readily hydrolyzed by CD38, a bifunctional enzyme involved in the metabolism of cADPR. These unique properties of the analogs can be used to develop fluorimetric assays for monitoring separately the cyclization and hydrolytic reactions catalyzed by the metabolic enzymes of cADPR. The convenience of the method in measuring kinetic parameters, pH-dependence, and modulator activity of the metabolic enzymes of cADPR is illustrated.

Abstract:Cyclic ADP-ribose (cADPR) is a recently discovered cyclic nucleotide with Ca2+ signaling functions. There is a growing recognition that it is an endogenous modulator of the Ca2+-induced Ca2+ release mechanism in cells. The cyclic structure of cADPR has now been confirmed by X-ray crystallography. A series of analogs of cADPR has been synthesized, including antagonists and a novel analog, cyclic GDP-ribose. Considerable prepress has been made in characterizing ADP-ribosyl cyclase, the synthetic enzyme, and cADPR hydrolase, the hydrolytic enzyme. A new class of bifunctional enzymes has been identified which catalyses both the synthesis and hydrolysis of cADPR. CD38, a lymphocyte differentiation antigen, is a member of this class. The understanding of the mechanisms of regulation of the metabolic enzymes and signaling by cADPR is likely to have important implications and several possibilities are discussed in this article.

Abstract:Cyclic ADP-ribose, (cADPR) is a novel endogenous metabolite of nicotinamide adenine dinucleotide (NAD(+)), and is a recently discovered second messenger. cADPR is active in mobilizing intracellular Ca2+ in invertebrate as well as mammalian cells. The mechanism of releasing Ca2+ from internal pool(s) has been studied. cADPR may bind its receptor, which in turn induces the release of Ca2+ from cADPR-sensitive Ca2+ pool(s) through ryanodine receptor or ryanodine receptor-like mediated Ca2+ channel. In addition, a possible intracellular signal transduction pathway involving nitric oxide(NO), cyclic guanylic acid (cGMP) and cADPR has been suggested to exist in various cells.
