Abstract:We report on the synthesis and application of a new hydrogel based on a methacrylate substituted polyphosphazene. Through ring-opening polymerization and nucleophilic substitution, poly[bis(methacrylate)phosphazene] (PBMAP) was successfully synthesized from hexachlorocyclotriphosphazene. By adding PBMAP to methacrylic acid solution and then treating with UV light, we could obtain a cross-linked polyphosphazene network, which showed an ultra-high absorbency for distilled water. Lipase from Candida rugosa was used as the model lipase for entrapment immobilization in the hydrogel. The influence of methacrylic acid concentration on immobilization efficiency was studied. Results showed that enzyme loading reached a maximum of 24.02 mg/g with an activity retention of 67.25% when the methacrylic acid concentration was 20% (w/w).
Surface Modification of Cyclomatrix Polyphosphazene Microsphere by Thiol-Ene Chemistry and Lectin Recognition

Abstract:A new synthetic route leading to functional polyphosphazene cyclomatrix microsphere has been developed via thiol-ene click modification. Hexacholorocyclophosphazene (HCCP) was crosslinked with both bisphenol-S and 4,4 '-dially1 bisphenol-S to obtain vinyl polyphosphazene microspheres (VPZM) in order to ensure high crosslinking degree and introduce vinyl moieties. Compared to the microspheres obtained by HCCP and bisphenol-S, the size of VPZM was broadly dispersed from 400 nm to 1.40 mu m. Thiol-ene click reactions were carried out to attach functional groups, such as glucosyl, carboxyl, ester and dodecyl groups onto polyphosphazene microspheres, which demonstrated no change in morphology and size after modification. Solid state NMR (SSNMR) and Fourier transform infrared spectoscopy (FT-IR) results showed that the vinyl moieties were introduced in the period of crosslinking and functionalization was also successful via click reactions. Moreover, the microspheres presented a little difference in thermal properties after modification. Concanavalin A (Con-A) fluorescent adsorption was also observed for glucosyl microspheres. Thus, the thiol-ene modified polyphosphazene microspheres displayed chemical flexibility in post-functionalization. Abstract:Water-soluble hydrogel nanoparticles have been synthesized by the incorporation of the stimuli-sensitive monomer of N-isopropylacrylamide (NIPAM) to a catalytic, molecularly imprinted system. The unique properties of pNIPAM copolymerized with cationic and anionic polymers offers a homogenously biocompatible catalytic system with switchable response to pH change. The maximum catalytic efficiency can thus be significantly increased.

Precursors containing thiol or epoxy groups are facilely synthesized by polycondensation and radical polymerization respectively, which are used to in situ form hydrogels under physiological conditions within tens of seconds via thiol-epoxy reaction. Abstract:Injectable hydrogels with pH and temperature triggered drug release capability were synthesized based on biocompatible glycol chitosan and benzaldehyde-capped poly(ethylene glycol)-block-poly(propylene glycol)-block-poly(ethylene glycol) (PEO-PPO-PEO). Aqueous solutions of the above polymers formed hydrogel under physiological conditions, allowing a desirable injectability, through the formation covalent benzoic-imine bond with pH and temperature changes. Rheological characterization demonstrated that the gelation rate and the moduli of the hydrogels were able to be tuned with chemical composition as well as pH and temperature of the polymer solution. Both hydrophobic and hydrophilic drugs could be incorporated inside the hydrogel through the in situ gel forming process and undergo a controlled release by altering pH or temperature. In vivo tests proved the formation and biocompatibility of the hydrogel in rat model.
"One-step" preparation of thiol-ene clickable PEG-based thermoresponsive hyperbranched copolymer for in situ crosslinking hybrid hydrogel

Abstract:A well-defined poly(ethylene glycol) based hyperbranched thermoresponsive copolymer with high content of acrylate vinyl groups was synthesized via a "one-pot and one-step" deactivation enhanced atom transfer radical polymerization approach, which provided an injectable and in situ crosslinkable system via Michael-type thiol-ene reaction with a thiol-modified hyaluronan biopolymer. The hyperbranched structure, molecular weight, and percentage of vinyl content of the copolymer were characterized by gel permeation chromatography and (1)H NMR. The lower critical solution temperature of this copolymer is close to body temperature, which can result in a rapid thermal gelation at 37 °C. The scanning electron microscopy analysis of crosslinked hydrogel showed the network formation with porous structure, and 3D cell culture study demonstrated the good cell viability after the cells were embedded inside the hydrogel. This injectable and in situ crosslinking hybrid hydrogel system offers great promise as a new class of hybrid biomaterials for tissue engineering.
Strong and Reversible Covalent Double Network Hydrogel Based on Force‐Coupled Enzymatic Reactions

Abstract:Biological load-bearing tissues are strong, tough, and recoverable under periodic mechanical loads. However, such features have rarely been achieved simultaneously in the same synthetic hydrogels. Here, we use a force-coupled enzymatic reaction to tune a strong covalent peptide linkage to a reversible bond. Based on this concept we engineered double network hydrogels that combine high mechanical strength and reversible mechanical recovery in the same hydrogels. Specifically, we found that a peptide ligase, sortase A, can promote the proteolysis of peptides under force. The peptide bond can be re-ligated by the same enzyme in the absence of force. This allows the sacrificial network in the double-network hydrogels to be ruptured and rebuilt reversibly. Our results demonstrate a general approach for precisely controlling the mechanical and dynamic properties of hydrogels at the molecular level.

Abstract:We describe a versatile approach to synthesize gly- cosylated polyphosphazenes with controllable density of glyco- syl groups. These glycopolymers have been synthesized by the nucleophilic substitution of poly(dichlorophosphazene) with propargylamine and subsequent ''thiol-yne'' click reaction between poly(di(propargylamine)phosphazene) and 2,3,4,6- tetra-O-acetyl-1-thio-b-D-glucopyranose (SH-GlcAc4). The poly- mers were characterized with FTIR and 1 H NMR. We found that the high steric hindrance of SH-GlcAc4 plays a key role in the overall reaction process, and � 55% of the alkyne groups partic- ipate in the ''thiol-yne'' click reaction. About 8% of the alkyne groups convert to alkene groups at the end of click reaction. The substitution of alkyne/alkane mixture was conducted to reduce the alkyne density in the side groups of polyphospha- zenes and minimize the influences of this steric effect. Mixed- substituent polyphosphazene was synthesized with 2:3 ratio of alkyne and alkane. In this case, almost no alkyne group remains after the ''thiol-yne'' click reaction, and thus the glyco- sylated polyphosphazene is able to form into micelles through self-assembly process. Abstract:A new strategy is introduced to prepare an adaptive polymer gel that has a unique adaptability in response to environmental stimuli. This gel is prepared by the thiol-ene click reaction between a bisvinyl [2]catenane and a poly(ethylene glycol) derivative containing multiple thiol groups. The catenane crosslinker is responsive to external stimuli due to the existence of intercomponent hydrogen bonding (IHB). The strong IHB restricts the rotation and movement of the crosslinker, giving it a rigid feature; however, the crosslinker becomes flexible when the IHB is destroyed. In consequence, the resulting gel can be reversibly switched between tough and soft states under stimulations.

Abstract:Sequential thiol-ene click reaction is reported for amphiphilic glycosylated polyphosphazene. Poly [bis(allylamino)phosphazene] was used as precursor to go through UV irradiation with 2,3,4,6-tetra-O-acetyl-1-thiol-beta-D-glucopyranose (SH-GlcAc(4)) and 1-dodecanethiol in sequence. Variation of the reaction conditions, including click reaction time and the dose of photoinitiator, led to different hydrophilic/hydrophobic ratios. As a result, glycosylated polyphosphazenes were synthesized with 533%, 77.7% and 85.0% of glucose moieties. The different residual composition could give rise to different self-assembly behaviors. Micelles of amphiphilic polyphosphazenes were formed in aqueous solution and the CMC value (0.79 x 10(-3)-4.00 x 10(-3) mg/mL) as well as mean diameter (170-220 nm) varied along with the hydrophilic glucose moiety/hydrophobic dodecyl moiety ratio. Abstract:Stimuli‐responsive hydrogels with decent electrical properties are a promising class of polymeric materials for a range of technological applications, such as electrical, electrochemical, and biomedical devices. In this paper, thermally responsive and conductive hybrid hydrogels are synthesized by in situ formation of continuous network of conductive polymer hydrogels crosslinked by phytic acid in poly(N‐isopropylacrylamide) matrix. The interpenetrating binary network structure provides the hybrid hydrogels with continuous transporting path for electrons, highly porous microstructure, strong interactions between two hydrogel networks, thus endowing the hybrid hydrogels with a unique combination of high electrical conductivity (up to 0.8 S m−1), high thermoresponsive sensitivity (significant volume change within several seconds), and greatly enhanced mechanical properties. This work demonstrates that the architecture of the filling phase in the hydrogel matrix and design of hybrid hydrogel structure play an important role in determining the performance of the resulting hybrid material. The attractive performance of these hybrid hydrogels is further demonstrated by the developed switcher device which suggests potential applications in stimuli‐responsive electronic devices.

Abstract:Polyelectrolyte hydrogels, combining the conductivity of polyelectrolytes with the flexibility of hydrogels, have become a popular candidate for flexible sensors, soft robotics, and electronic skins due to their remarkable stability in electrical conductivity. However, their mechanical properties and adhesive strength are limited because of their single-type bonding interaction. This work introduces a double-networked (DN) polyelectrolyte hydrogel formed through polymer chain entanglements, chemical crosslinking, and the incorporation of multiple strong and/or weak bonding interactions. The first network is a chemically crosslinked polyacrylamide (PAAm). The second network consists of polyelectrolytes (poly(diallyldimethylammonium chloride) (PDDA) and poly(methacrylic acid sodium salt) (PMAANa)), which form numerous weak and/or strong ionic bonds. Weak and/or strong hydrogen bonds are present within and between two networks. The mechanical properties and adhesive strength of the polyelectrolyte DN hydrogel can be tailored through modulating the content of PAAm, polyelectrolytes, and co-solvents. The optimal compositions have a tensile modulus of 10.8 kPa, tensile fracture strain of 1000%, and adhesive strength of 37.8 kPa. The hydrogel sensors are successfully applied to flexible electrodes for various devices for detecting human motion, handwriting recognition, and continuous monitoring of electrophysiological signals. The distinctive solvent-adjustable transparency of the gel also allows for its utilization in information encryption and decryption.

Abstract:A new visible light induced graft polymerization method was utilized to prepare pH-sensitive hydrogel layers covalently attached to polymer substrates for drug delivery. In our strategy, isopropyl thioxanthone semi-pinacol (ITXSP) dormant groups were firstly introduced on the surface of a polycaprolactone (PCL) film by a UV-induced abstracting hydrogen-coupling reaction. Then visible light induced graft cross-linking polymerization was performed to initiate polymerization of poly(ethylene glycol) diacrylate (PEGDA) and acrylic acid (AA), resulting in the formation of a hydrogel layer. The thickness of the hydrogel film can be controlled by varying the exposure time and monomer composition. The grafted hydrogel layers showed a flat morphology and dense structure, which is different from the traditional reported porous structure. The water contact angle of the hydrogel layer exhibited a reversible change from 381 to 181 when the film was alternatively treated in buffers of pH 2.0 and 7.4, respectively. Patterned hydrogel layers were prepared as a model to determine the change in the height of the grafted hydrogel layer as a function of pH. As the pH changed from 2.0 to 7.4, the hydrogel pattern showed an increase in height due to the swelling of the hydrogel network, and the hydrogel layer formed by 0.2 wt% PEGDA and 25 wt% AA showed the most increase (30%) in height. Bovine serum albumin (BSA) and lysozyme as models of protein drugs were incorporated in the hydrogel network, and their release also showed obvious pH-sensitivity. At pH 2.0, hydrogels present a faster initial burst release due to the squeezing mechanism. Tertiary structure analysis showed that encapsulation and release did not affect the protein conformation. These findings have improved our understanding of hydrogel thin films, which may be useful as potential vehicles of therapeutic proteins in drug delivery applications.
Synthesis of Multiresponsive and Dynamic Chitosan-Based Hydrogels for Controlled Release of Bioactive Molecules

Abstract:An inexpensive, facile, and environmentally benign method has been developed for the preparation of multiresponsive, dynamic, and self-healing chitosan-based hydrogels. A dibenzaldehyde-terminated telechelic poly(ethylene glycol) (PEG) was synthesized and was allowed to form Schiff base linkages between the aldehyde groups and the amino groups in chitosan. Upon mixing the telechelic PEG with chitosan at 20 °C, hydrogels with solid content of 4-8% by mass were generated rapidly in <60 s. Because of the dynamic equilibrium between the Schiff base linkage and the aldehyde and amine reactants, the hydrogels were found to be self-healable and sensitive to many biochemical-stimuli, such as pH, amino acids, and vitamin B6 derivatives. In addition, chitosan could be digested by enzymes such as papain, leading to the decomposition of the hydrogels. Encapsulation and controlled release of small molecules such as rhodamine B and proteins such as lysozyme have been successfully carried out, demonstrating the potential biomedical applications of these chitosan-based dynamic hydrogels.

The glucose-substituted polyphosphazene was synthesized by nucleophilic substitution of poly(dichlorophosphazene) with propargylamine and the subsequent azide/alkyne "click" reaction of poly[di(propargyl-amine)phosphazene] (PDPAP) with azidosugar. The relative proportion of the glucose substituent can be easily controlled over a broad range by varying the amount of azidoglucose used in the azide/alkyne "click" reaction.

Abstract:A green route is reported for functionalizing polyphosphazene via the thiol‐ene click reaction in an aqueous medium. Poly[bis(allylamino)phosphazene] is used as the precursor polymer, which is easily dissolved in water containing 5% (v/v) phosphoric acid, but barely dissolved in other acidic aqueous solutions with the same pH value. Three thiol reagents, namely, 3‐mercapto‐1,2‐propanediol, 2‐mercaptoethoxy ethanol, and l‐cysteine, are then reacted with the precursor in the phosphoric acid aqueous solutions. 1H and 31P NMR analyses confirm that the allyl polyphosphazene can be quantitatively modified by the mercaptans without hydrolysis degradation during the synthesis and purification processes. Moreover, these polyphosphazenes exhibit higher regioselectivity than those functionalized in organic solvents. This method provides a facile route for the green synthesis of functional polyphosphazenes without organic solvents. image

Abstract:Stimuli-responsive polymeric hydrogels based on host-guest interactions have demonstrated potential as smart drug delivery platforms. However, host-guest interactions alone have been repeatedly highlighted to provide insufficient mechanical strength for the formed hydrogels likely due to the generally low grafting degrees of functional groups and steric hindrance of polymer backbone and grafted functions. Herein, a combination strategy of reversible addition-fragmentation chain-transfer polymerization (RAFT) and dual crosslinking was employed to fabricate thermo-and pH-dual responsive hydrogels with enhanced mechanical properties. Spe-cifically, two kinds of poly (N-isopropyl acrylamide) (PNIPAM)-based well-defined thermo-sensitive polymers, denoted as PH and PG, consisting of beta-cyclodextrin (beta-CD) and adamantane (Ad) groups were synthesized via RAFT copolymerization. A facile mixing of PH and PG with commercially available adipic dihydrazide led to the construction of hydrogels via host-guest interactions and dynamic acylhydrazone bonding. The resulting hydrogels not only exhibited pH-switchable sol-gel transitions and temperature-dependent swelling behaviors, but also possessed good stretchability (fracture stress up to 39.97 kPa) and self-healing property (self-healing efficiency above 90%). Notably, the resulting hydrogels showed pH-responsive controlled spatiotemporal release administration of rhodamine B due to the presence of dynamic acylhydrazone links. Together with good cyto-compatibility, the hydrogels developed herein represent promising candidates for potential transdermal drug delivery.
Reversible Shape-Shifting of an Ionic Strength Responsive Hydrogel Enabled by Programmable Network Anisotropy.

Abstract:Reversible shape-shifting hydrogels exhibit great potential in diverse fields. Repeatable programmability for the shape transformation has newly been enabled in thermally responsive hydrogels via engineering of the chain orientation of the polymer network, which substantially promotes the transformation capability. However, diversified responsive behavior and the enabling mechanism require further investigation. Herein, we develop an ionic strength (IS) responsive hydrogel enabling the programmable reversible shape transformation based on a semi-interpenetrating network of poly(acrylic acid) (PAA) and poly(vinyl alcohol) (PVA). Deformation of the hydrogel upon external force can be fixed due to crystallization of PVA that underwent cyclic freezing-thawing. Therefore, the chain orientation can be retained in the deformed area, enabling the programmable IS responsive actuation. In contrast to the thermally responsive actuation originated from the lower critical solution temperature phase transition, the IS responsive actuation does not accompany any phase change and the corresponding mechanism is proposed. Reversible bending providing an actuation angle as large as 80° can be achieved after optimization of the PVA content. The PVA crystals can be melted upon heating, and the responsive actuation can thus be reprogrammed. In addition, utilizing a digital light 3D printer, the hydrogels are further fabricated into arbitrary geometries, thus realizing more complex actuations. Overall, our work provides a general strategy to develop reversible shape-shifting hydrogels and paves the way for soft actuators.

A thiol–ene click-based strategy to customize injectable polymer–nanoparticle hydrogel properties for therapeutic delivery

Abstract:Polymer–nanoparticle (PNP) hydrogels are a promising injectable biomaterial platform that has been used for a wide range of biomedical applications including adhesion prevention, adoptive cell delivery, and controlled drug release. By tuning the chemical, mechanical, and erosion properties of injected hydrogel depots, additional control over cell compatibility and pharmaceutical release kinetics may be realized. Here, we employ thiol–ene click chemistry to prepare a library of modified hydroxypropylmethylcellulose (HPMC) derivatives for subsequent use in PNP hydrogel applications. When combined with poly(ethylene glycol)-b-poly(lactic acid) nanoparticles, we demonstrate that systematically altering the hydrophobic, steric, or pi stacking character of HPMC modifications can readily tailor the mechanical properties of PNP hydrogels. Additionally, we highlight the compatibility of the synthetic platform for the incorporation of cysteine-bearing peptides to access PNP hydrogels with improved bioactivity. Finally, through leveraging the tunable physical properties afforded by this method, we show hydrogel retention time can be dramatically altered without sacrificing mesh size or cargo diffusion rates. This work offers a route to optimize PNP hydrogels for a variety of translational applications and holds promise in the highly tunable delivery of pharmaceuticals and adoptive cells.
