Abstract:Polymeric microneedles have attracted increasing interest for transdermal drug delivery owing to the unique advantages of minimal invasiveness, biocompatibility, biodegradability, and efficient drug-loading capacity. However, drug release from polymeric microneedles lacks controllability during transdermal administration. Herein, a poly(ethylene glycol) diacrylate/polyvinylpyrrolidone (PEGDA/PVP) microneedle patch with tailorable matrix constitutions is designed and fabricated using a two-step ultraviolet (UV) light-induced polymerization method. Scanning electron microscopic characterization reveals a non-uniform interior matrix of microneedles, resulting from the aggregation of PVP in the polymerized PEGDA matrix induced by UV irradiation. The fabricated microneedles are found to show sufficient mechanical strength for skin penetration and they can be removed intact after application. The controllable release property of rhodamine B (RhB)-loaded PEGDA/PVP microneedles is demonstrated in vitro by tuning the fraction of PVP content. The typical characteristics including initial burst and subsequent sustained release are attributed to the rapid dissolution of PVP and prolonged swelling of PEGDA, respectively. In addition, the microneedle insertion site can be completely recovered within 1 h on a porcine skin, indicating the minimally invasive feature of microneedle-mediated therapeutic delivery. This scalable microneedle platform may serve as a convenient and effective drug carrier for transdermal delivery of various pharmaceuticals under precise control.

Abstract:Microneedles are primarily designed for enhancing transdermal drug delivery in a minimally invasive manner. In particular, coated microneedles have attracted increasing attention due to the resulting mode of rapid and highly efficient drug administration. In this study, we developed a novel gelatin/sucrose film-coated poly(ethylene glycol)diacrylate (PEGDA) microneedle patch, which can effectively improve the skin permeability of therapeutic drugs and realize convenient and efficient drug administration. A simple and reliable technique was proposed to produce uniform film coatings on microneedles at room temperature. After introducing a prepolymer solution into the mold cavity, a film-coated microneedle patch was fabricated via a photo-induced polymerization process. Four categories of molecular models, including rhodamine B (RhB), bovine serum albumin (BSA), doxorubicin (DOX) and indocyanine green (ICG), were encapsulated into the gelatin/sucrose film to evaluate their transdermal delivery and therapeutic effects both in vitro and in vivo. The presented methodology represents a facile technique for rapid formation of film-coated microneedles for efficient transdermal delivery of small molecular drugs and large proteins.

Abstract:Dissolving microneedles have been employed as a safe and convenient transdermal delivery system for drugs and vaccines. To improve effective drug delivery, a multilayered pyramidal dissolving microneedle patch, composed of silk fibroin tips with the ability of robust mechanical strength, rapid dissolution and drug release supported on a flexible polyvinyl alcohol (PVA) pedestal is reported. To show the utility of this approach the ability of the fabricated microneedles to deliver insulin is demonstrated. The dissolving microneedles have sufficient mechanical strength to be inserted into abdomen skin of mice to a depth of approximately 150μm, and release their encapsulated insulin into the skin to cause a hypoglycemic effect. The fabrication of microneedles avoids high temperature which benefits storage stability at room temperature for 20d. This result indicates >99.4% of insulin remained in the microneedles. In comparison to traditional needle-based administration, the proposed multilayered pyramidal dissolving microneedle patches enable self-administration, miniaturization, pain-free administration, drug delivery and drug stability, all being important features in needle free drug delivery.
A universal powder-laden crosslinked chitosan microneedle patch for high-dose controllable drug delivery

Abstract:In this study, we constructed a novel powder-laden core-shell crosslinked chitosan microneedle patch for high-dose and controllable delivery of various drugs, including both macromolecular biological drugs and small-molecule chemical drugs. Direct loading of drug powders greatly improved drug loading capacity and minimized degradation. The results of the in vitro drug release study suggested that the release behaviors of the most tested drugs (both macromolecular drugs and small-molecule drugs) can be tuned by adjusting the crosslink density of the microneedle shell to achieve either rapid or sustained release of the loaded drug. The in vivo hypoglycemic efficacy test in streptozotocin-induced diabetic mice further proved that the onset and duration of the insulin-laden patch can be customized by adjusting the crosslink density. Furthermore, a combination of microneedle patches with different crosslink densities not only rapidly reduced blood glucose levels to normoglycemic levels (within 1 h) but also maintained normoglycemia for up to 36 h. The insulin loaded in the patch also showed good stability during storage at 40 °C for 6 months. Our results suggest that this powder-laden patch represents a strong candidate for addressing the multiple challenges in the preparation and application of polymeric microneedles and shows promise in clinical applications.

Abstract:The use of microneedles has facilitated the painless localized delivery of drugs across the skin. However, their efficacy has been limited by slow diffusion of molecules and often requires external triggers. Herein, an autonomous and degradable, active microneedle delivery platform is introduced, employing magnesium microparticles loaded within the microneedle patch, as the built-in engine for deeper and faster intradermal payload delivery. The magnesium particles react with the interstitial fluid, leading to an explosive-like rapid production of H-2 bubbles, providing the necessary force to breach dermal barriers and enhance payload delivery. The release kinetics of active microneedles is evaluated in vitro by measuring the amount of IgG antibody (as a model drug) that passed through phantom tissue and a pigskin barrier. In vivo experiments using a B16F10 mouse melanoma model demonstrate that the active delivery of anti-CTLA-4 (a checkpoint inhibitor drug) results in greatly enhanced immune response and significantly longer survival. Moreover, spatially resolved zones of active and passive microneedles allow a combinatorial rapid burst response along with slow, sustained release, respectively. Such versatile and effective autonomous dynamic microneedle delivery technology offers considerable promise for a wide range of therapeutic applications, toward a greatly enhanced outcome, convenience, and cost.

Abstract:Microneedle (MN) patches are promising stratagems for transdermal drug delivery owing to its minimal invasiveness and portability. Particularly, separable MN patches attract more attention for durable and long-term drug delivery. Here, a new strategy for designing controllably separable MN patches by integrating photothermal responsive phase-changing microspheres (MPs) into conical hydrogel MNs, is presented. Such MPs consist of near-infrared (NIR)-responsive black-phosphorus (BP) and phase-changing gelatin (GT), and are fabricated using a simple capillary microfluidic device. When the composite MN patch is exposed to NIR, the contained BP/GT MPs can transform light into heat, increase local temperature, turn into a liquid state, and form cavities in MNs, resulting in the convenient detachment of the backing plate and the needles by shear force. Subsequently, the embedded needles can sustainably release the drug. In addition to intelligently controlling the detachment of MNs, the BP/GT MPs can load macromolecular drugs and release them in response to NIR stimulation. Results further demonstrated that such controllably separable MN patch can effectively load and release two drugs to ameliorate disease progression for systemic lupus erythematosus treatment. These features indicate the practical values of controllably separable MN patches for versatile drug delivery and clinical applications.

Abstract:In this study, we propose a novel method for the preparation of dissolvable microneedle (DMN) patch directly attached on a medical adhesive tape for transdermal drug delivery. The fabrication process is simple and time-saving without second curing procedures and the fabricated DMN patch exhibits high flexibility and dosage accuracy owing to the property of the medical adhesive tape. Sodium carboxymethyl cellulose (SCMC) was adopted as matrix material due to its biodegradability and mechanical hardness. The MN patch shows great ability to penetrate skin ex vivo and in vivo by utilizing porcine skin and the living mouse. Most importantly, it possesses good capacity to carry drugs and locally release drug molecules into mouse skin by adopting Rhodamine B as the drug analog.
Dual Cross-Linking Mechanism of Sodium Alginate Composite Hydrogel for Adhesive Microneedle Patch Preparation

Abstract:The introduction of microneedles has revolutionized transdermal drug delivery. However, solid microneedle patches often fail to conform to the epidermis and adapt to changes in skin curvature, limiting their effectiveness. Using sodium alginate hydrogel to create flexible microneedle patches can address this issue, but conventional sodium alginate hydrogels suffer from inhomogeneous structure, dehydration, and poor mechanical properties. These drawbacks result in reduced drug delivery efficacy and insufficient mechanical robustness. To enhance the performance of sodium alginate hydrogels, this study presents a novel sodium alginate composite hydrogel with a dual cross-linking mechanism. The hydrogel, with a gelatin to sodium alginate ratio of 1:1, exhibited optimal surface morphology and mechanical properties suitable for microneedle patch preparation. The resulting microneedle patch demonstrated excellent water adhesion, securely attaching to skin and oral mucosa surfaces. In vitro experiments confirmed the microneedle's ability to puncture and release the drug, dissolving and releasing the drug within 15minutes. The patch also showed superior piercing and dissolution properties on the mouth palate, highlighting it may serve as a potential delivery location for more efficient microneedle patch drug administration. In the future, this technology will show great potential in expanding the scope of drug administration for transdermal drug delivery and in reducing the work pressure of healthcare professionals.

Abstract:Microneedle (MN) patches consisting of miniature needles have emerged as a promising tool to perforate the stratum corneum and translocate biomolecules into the dermis in a minimally invasive manner. Stimuli-responsive MN patches represent emerging drug delivery systems that release cargos on-demand as a response to internal or external triggers. In this review, a variety of stimuli-responsive MN patches for controlled drug release are introduced, covering the mechanisms of action toward different indications. Future opportunities and challenges with respect to clinical translation are also discussed.

Abstract:Abstract Microneedles offer a convenient transdermal delivery route with potential for long term sustained release of drugs. However current microneedle technologies may not have the mechanical properties for reliable and stable penetration (e.g. hydrogel microneedles). Moreover, it is also challenging to realize microneedle arrays with large size and high flexibility. There is also an inherent upper limit to the amount and kind of drugs that can be loaded in the microneedles. In this paper, we present a new class of polymeric porous microneedles made from biocompatible and photo-curable resin that address these challenges. The microneedles are unique in their ability to load solid drug formulation in concentrated form. We demonstrate the loading and release of solid formulation of anesthetic and non-steroidal anti-inflammatory drugs, namely Lidocaine and Ibuprofen. Paper also demonstrates realization of large area (6 × 20 cm 2 ) flexible and stretchable microneedle patches capable of drug delivery on any body part. Penetration studies were performed in an ex vivo porcine model supplemented through rigorous compression tests to ensure the robustness and rigidity of the microneedles. Detailed release profiles of the microneedle patches were shown in an in vitro skin model. Results show promise for large area transdermal delivery of solid drug formulations using these porous microneedles.

Abstract:For successful treatment of diseases, sufficient therapeutics must be provided to the body. Microneedle applications in therapeutic delivery and analytics sampling are restricted because of various issues, including smaller area for drug loading and analytics sampling. To achieve sufficient drug loading and analytics sampling and improve drug penetration while maintaining painless administration, patch-type microneedle arrays were designed and fabricated using polymer casting from a conical cavity mold. Microcavities were formed on a carbon plate via micromechanical machining. A porous polymer layer was coated on a microneedle patch (MNP). The pores of the porous polymer layer provided space and channels for drug delivery. A pH-sensitive polymer layer was employed to cap the porous polymer layer, which prevented drug leakage during storage and provided a stimulus drug release in response to body pH conditions. The drug can be delivered through holes connected to both sides of the patch. The drug release of the MNP was investigated in vitro and in vivo and showed conceptual proof that these MNs have the potential to enhance treatment protocols for various diseases with the flexibility of coating and therapeutic materials and offer significant scope for further variations and advancement.

Polymer homologue-mediated formation of hydrogel microneedles for controllable transdermal drug delivery

Abstract:Poly(ethylene glycol) diacrylate (PEGDA) microneedles (MNs) are hydrogel-based devices that achieve controlled drug delivery kinetics by adjusting the crosslinking density. However, the biosafety of many crosslinking agents used to regulate crosslinking density is not ideal. To avoid crosslinking agents and simplify the preparation process, using two types of polymer homologues with different number-average molecular weights, we have successfully developed a series of PEGDA MNs with controllable crosslinking density (abbreviated as TP-X MNs). The research showed that the mechanical properties and drug release behavior of TP-X MNs could be tuned by simply controlling the weight proportion of two different PEGDA components in MNs. Ex vivo drug delivery experiments indicated that all TP-X MNs exhibited a sustained release profile, and their control range of 336-hour accumulative release rates was from 6.24% to 40.93%. Moreover, we prepared a novel dual-layer PEDGA MN, which can customize the drug loading and release rate in each layer of MN. This work demonstrates a new way to develop hydrogel MNs with adjustable crosslink density and broadens the applications of PEGDA MN in the biomedical field.
Near‐infrared Triggered Biodegradable Microneedle Patch for Controlled Macromolecule Drug Release

Abstract:Transdermal drug delivery of macromolecule drugs has attracted significant attention due to the advantage of convenience and biocompatibility. However, the practical usage of it is limited by the low delivery efficiency and poor drug absorption. To develop an efficient, safe, and controllable transdermal delivery method, the near‐infrared (NIR) triggered calcium sulfate and gelatin biodegradable composite microneedle (MN) patches have been developed. The MN patches were fabricated by polydimethylsiloxane (PDMS) molds, and the structure data could be adjusted by changing the molds. Such an MN patch could release both macro and micro molecule drugs. After loaded with photothermal converter IR780, which could transfer energy of light to heat, the release of macromolecule drugs in MNs could be controlled by applying NIR irradiation. The control effect could be enhanced by spraying 1‐tetradecanol (TD) coating and optimizing the ratio (weight) of gelatin and calcium sulfate to 2:6. Besides, the MN patch could deliver drugs through the skin barrier, and the process could be controlled by NIR. Moreover, the insulin‐loaded MN patch exhibits some therapeutic effects on healthy mice. This work suggests that biodegradable MNs could achieve controllable drug delivery and potentially be applied in individual treatment via transdermal ingestion. Abstract:Patch-type drug delivery has garnered increased attention as an attractive alternative to the existing drug delivery techniques. Thus far, needle phobia and efficient drug delivery remain huge challenges. To address the issue of needle phobia and enhance drug delivery, we developed a needle-free and self-adhesive microcup patch that can be loaded with an ultrathin salmon DNA (SDNA) drug carrier film. This physically integrated system can facilitate efficient skin penetration of drugs loaded into the microcup patch. The system consists of three main components, namely, a cup that acts as a drug reservoir, an adhesive system that attaches the patch to the skin, and physical stimulants that can be used to increase the efficiency of drug delivery. In addition, an ultrathin SDNA/drug film allows the retention of the drug in the cup and its efficient release by dissolution in the presence of moisture. This latter feature has been validated using gelatin as a skin mimic. The cup design itself has been validated by comparing its deformation and displacement with those of a cylindrical structure. Integration of the self-adhesive microcup patch with both ultrasonic waves and an electric current allows the model drug to penetrate the stratum corneum of the skin barrier and the whole epidermis, thereby enhancing transdermal drug delivery and reducing skin irritation. This system can be used as a wearable biomedical device for efficient transdermal and needle-free drug delivery.

Abstract:The birth of microneedles, an array of needles sufficiently long to penetrate epidermis but small enough to do not cause skin injury and pain feeling, has offered a highly promising solution for non-invasive delivery of protein and peptide drugs, a long-cherished desire over eighty years. However, the attempts to develop clinically feasible microneedle transdermal delivery methods encountered series of difficulties, for which a decade research efforts have yet to result in a single product. Microneedles may be incorporated into devices as skin pre-treatment tools, skin microinjectors as well as transdermal patches by their functions in drug delivery. They may also be categorized to insoluble solid microneedles, hollow microneedles, soluble/degradable solid microneedles and phase-transition microneedles by their structure and forming materials. This review article is aimed to update the progress and discuss the technical challenges raised in developing protein/peptide loaded microneedle patches.

Abstract:Wound healing remains a serious challenge in the biomedical field due to its complex physiological processes. To address this issue, microneedle patches are proposed as an effective strategy for wound management. Herein, we propose novel breathable microneedle patches containing therapeutic drugs and functional materials for wound healing, which are fabricated using extrusion 3D printing technology. The microneedle patch is composed of needle tips and a breathable scaffold backing layer made of spidroin hydrogel, polyurethane, and aloe vera gel, providing excellent biocompatibility and self-healing ability. Furthermore, eutectic Galium-Indium (EGaIn) was incorporated into the dressing, enabling the near-infrared light response function. The highly efficient photothermal conversion allowed for superfast self-healing in seconds. The microneedle dressing was sensitive enough to monitor large-scale movements at different joints and small-scale motions of the throat during swallowing. Combining biocompatible microneedles with a temperature-responsive hydrogel (pNIPAM), stable controllable drug delivery is achieved using near-infrared (NIR) light. The stable photothermal performance enabled the device to release a controlled dose of the drug over multiple light triggers. With NIR-triggered pulse drug release, VEGF-mediated treatment on wound healing was significantly improved in mouse models by accelerating tissue repair and reducing the inflammatory response.

Abstract:Microneedles offer minimally invasive, user-friendly, and subcutaneously accessible transdermal drug delivery and have been widely investigated as an effective transdermal delivery system. Ibuprofen is a common anti-inflammatory drug to treat chronic inflammation. It is crucial to develop microneedle patches capable of efficiently delivering ibuprofen through the skin for the effective treatment of arthritis patients requiring repeated medication. In this study, the mechanical properties of a new type of polymer microneedle were studied by finite element analysis, and the experimental results showed that the microneedle could effectively deliver drugs through the skin. In addition, a high ibuprofen-loaded microneedle patch was successfully prepared by micromolding and subjected to evaluation of its infrared spectrum morphology and dissolve degree. The morphology of microneedles was characterized by scanning electron microscopy, and the mechanical properties were assessed using a built linear stretching system. In the in-vitro diffusion cell drug release test, the microneedle released 85.2 ± 1.52% (210 ± 3.7 μg) ibuprofen in the modified Franz diffusion within 4 h, exhibiting a higher drug release compared to other drug delivery methods. This study provides a portable, safe and efficient treatment approach for arthritis patients requiring daily repeated medication.

Abstract:Dissolving polymer microneedles have attracted much attention for their biocompatibility, fast dissolution, and high drug loading. Among them, polyvinylpyrrolidone (PVP) is widely used, but its high water absorption and poor mechanical properties constrain its broad applications. Herein we show that adding cyclodextrin (CD) to form PVP-CD inclusion complexes can alleviate these problems. The water absorption of PVP was reduced by 36-40% at different RHs as the PVP-CD inclusion complexes formed. Attractively, the water absorption at 10 and 20 days remained almost the same for the complexes while it could dramatically increase for the pure PVP samples, particularly in high humidity environments, indicating a possibly longer storage time for the complexes. It was also found that the Young's modulus and hardness of the PVP-CD could be greatly improved, especially for low molecular weight PVP. Furthermore, the glass transition temperature (Tg) of the PVP-CD increased by up to 39 °C. With the improved properties, the fabricated PVP-CD microneedles possessed much sharper needle tips and the patch had less cracks than those made from pure PVP. Pig skin application results suggested that the PVP-CD microneedle arrays were able to reliably pierce the stratum corneum of the skin while it was not achievable for the PVP microneedles with the same geometry. We anticipate that these PVP-CD complex microneedles are more suitable for vaccine and drug delivery because of their superior properties.
