Abstract:Surface-enhanced Raman scattering (SERS) offers a route for trace detection of mercury ions (Hg2+) owing to its great enhancement near the surface of the substrate. However, most of the methods are inconvenient due to the long reaction time and the strict measurement conditions. Based on a reaction between silver nanoparticles (AgNPs) and mercury ions, a simple but effective method is proposed in this paper. In the experimental research, a tapered optical fiber (TOF) modified with AgNPs is employed as a sensing probe and Rhodamine 6G (R6G) is taken as the Raman reporter. The SERS substrate is consumed by the Hg2+, causing a spectrum decrease. In the Hg2+ concentration range of 10-12 M to 10-4 M, the spectra evolution is detected using a mobile Raman spectrometer. By analyzing the decrease of the Raman peak after a 5-minutes interval, the concentration of Hg2+ can be quantified. A limit of detection of 5.15 x 10-13 M is achieved, which is much below the maximum Hg2+ content of drinkable water specified by USEPA (10-8 M). Verification tests were performed with the inductively coupled plasma-mass spectrometry (ICP-MS), the results showed that the proposed SERS method is effective for real samples detection. In addition, the reproductivity and the selectivity are experimentally confirmed. With this reliable, rapid, ultrasensitive and accurate method, trace Hg2+ could be detected in many applications.

Abstract:A surface-enhanced Raman scattering (SERS) sensor of mercury ions based on the coordinated catalytic reaction of Hg2+-Ag nanoparticles (NPs) has been designed and constructed for water quality monitoring. We combined Ag NPs (average particle size is 49 nm) with mercury ions to form Hg2+-Ag NPs in aqueous phase by electrostatic interactions. The formed Hg2+-Ag particles can catalyze a redox reaction between o-phenylendiamine (OPD) and dissolved oxygen to form 2,3-diaminephenazine (DAP), which is Raman-active and possesses a strong SERS signal due to the adherence to Ag NPs. Therefore we can trace the SERS intensity of DAP to determine mercury ions and the lowest detectable concentration of mercury is 1.0 nM using this method. This sensor displays higher sensitivity and selectivity and it possesses a certain value in terms of water quality detection.

Abstract:Voltage enrichment-assisted surface-enhanced resonance Raman scattering (SERRS) was employed for monitoring mercury(II) ions based on a nanoporous gold (NPG)/aptamer hybrid sensor. The hybrid sensor based on the coordination chemistry of thymine-Hg2+-thymine interaction and an applied voltage that pushed mercury ions toward the NPG film improved both the speed and sensitivity of Hg2+ ion detection. The detection limit can reach 0.1 pM, even in the aqueous solution containing 13 metal ions, and the sensor also possesses a fine reproducibility and stability, promising great potential in real-time sensing applications.

Abstract:A sensitive and selective surface-enhanced Raman scattering (SERS) sensor for mercury(II) was fabricated based on the target-mediated displacement of a T-rich oligonucleotide strand. A DNA/aptamer duplex was prepared by the hybridization between a tetramethylrhodamine(TMR)-labeled thymine(T)-rich Hg2+-specific aptamer (denoted as TMR-aptamer) and a thiolated adenine-rich capturing DNA. The duplex can be immobilized onto the SERS substrate of the Ag-moiety modified glycidyl methacrylate-ethylene dimethacrylate (denoted as Ag-GMA-EDMA) via self-assembly by the thiol anchor, in which the TMR-aptamer exists in a double-stranded chain. In this case, the label of the TMR moiety approaches the substrate surface and produces a strong SERS signal. Upon the addition of the target, a pair of TMR-aptamers could cooperatively coordinate with Hg2+ to form a stable duplex-like structure mediated by the T–Hg2+–T complex between two adjacent strands, which triggers the release of the TMR-aptamer from the SERS substrate surface, thus drawing the TMR tags away from the substrate with a significant decrease in the SERS signal. This optical sensor shows a sensitive response to Hg2+ in a concentration from 5 nM to 2.0 μM with a detection limit of 2.5 nM. The prepared sensor is negligibly responsive to other metal ions, can be easily regenerated, and shows good performance in real sample analysis.

Abstract:Mercury ions can be enriched along the food chain and even low concentrations of mercury ions can seriously affect human health and the environment. Therefore, rapid, sensitive, and highly selective detection of mercury ions is of great significance. In this work, we synthesized Fe3O4@SiO2@Au three-layer core/shell nanoparticles, and then modified 4-MPy (4-mercaptopyridine) to form a SERS sensor. Mercury ions in water can be easily captured by 4-MPy which were used as the reporter molecules, and the concentration of mercury ions can be evaluated based on the spectral changes (intensification and reduction of peaks) from 4-MPy. After the mercury ion was combined with the pyridine ring, the peak intensity at 1093 cm−1 increased with the concentration of mercury ion in the range of 10 ppm–1 ppb, while the Raman intensity ratio I (416 cm−1)/I (436 cm−1) decreased with the increase of mercury ion concentration. This magnetically separatable and recyclable SERS sensor demonstrates good stability, accuracy, and anti-interference ability and shows the potential to detect actual samples. Furthermore, we demonstrate that the probe is applicable for Hg2+ imaging in macrophage cells.
Au/Ag2S Dimeric Nanostructures for Highly Specific Plasmonic Sensing of Mercury(ii)

Abstract:Although many plasmonic nanosenosrs have been established for the detection of mercury（Ⅱ）(Hg 2+ ),few of them is feasible for analyzing natural samples with very complex matrices because of insufficient method selectivity.To address this challenge,we propose an epitaxial and lattice-mismatch approach to the synthesis of a unique Au/Ag 2 S dimeric nanostructure,which consists of an Au segment with excellent plasmonic characteristics,and a highly stable Ag 2 S portion with minimum solubility product (K sp （Ag 2 S）=6.3×10 -50 ).The detection relies on the chemical conversion of Ag 2 S to HgS when reacting with Hg 2+ ,resulting in a red shift in the absorption band of the connecting Au NPs.The concurrent color changes of the solution from gray purple to dark green and finally to navy correlate well with Hg 2+ concentration,thus enables UV-vis quantitation and a naked-eye readout of the Hg 2+ concentration.This method exhibits superior selectivity towards Hg 2+ over other interfering ions tested because Hg 2+ is the only ion that can react with Ag 2 S to form HgS with even smaller solubility product (K sp （HgS）=4×10 -53 ).The detection limit of this method is 1.21μmol/L,calculated by the signal-to-noise of 3.The practicability of the method was verified by analyzing the Hg 2+ in sewage water samples without sample pretreatment with satisfactory recoveries （93.1%-102.8%） and relative standard deviations （1.38%-2.89%）.We believe this method holds great potential for on-the-spot detection of Hg 2+ in environmental water samples with complex matrices.

Abstract:Developing highly sensitive and specific method is essential for trace Hg2+ quantifying. Herein, we established a background-free surface-enhanced Raman scattering (SERS) platform which allows not only the detection of Hg2+ in homogeneous samples, but also high-precision profiling of Hg2+ distribution in zebrafish. Its superior anti-interference capability is due to the introduction of a high signal-to-noise-ratio (SNR) Raman reporter with an acetylenyl group, which present single nonoverlapping peak at 2125 cm(-1), avoiding interference from other substances. Additionally, the specific recognition of Hg2+-bis-thymine complexes and the signal amplified hybridization chain reaction (HCR) were effectively utilized in this design. Benefited from the above strategies, a detection range for Hg2+ from 1.0 fM to 100 pM was achieved with a detection limit of 0.2 fM. Additionally, Raman imaging was first performed in zebrafish sections to study Hg2+ distribution which provided a reference for the detection of Hg(2+ )pollution in fish. This approach could be extended to target other interested analytes both in simple and biological samples for various purposes.

Abstract:The development of a facile, rapid, precise, and ultrasensitive assay for the detection of mercury ions (Hg2+) is an urgent problem to be solved because excessive Hg2+ poses a great threat to human health and the ecological environment. Herein, a novel surface-enhanced Raman scattering (SERS) sensor which employed phenylacetylene (PA) as a label-free probe was established. The PA functionalized silver nanoparticles (PA-AgNPs) produced an intense SERS signal in the Raman-silent region, which effectively avoided the interference of solvent and overcame the signal overlap problem. In the presence of Hg2+, the PA molecules separated from the Ag surface due to the strong coordination between the alkynyl group and Hg2+, which resulted in a significant SERS signal attenuation. Basing on the proposed SERS platform, a linear relationship with a wide range from 10-10 to 10-8 M and a low detection limit of 87.6 pM were obtained. Besides, the selectivity and reproducibility of the SERS system were also investigated. What's more, we applied the SERS sensor to analyze Hg2+ in lake water with high accuracy and excellent recovery. The obtained results demonstrated that the established SERS platform is expected to be a promising alternative tool for the determination of Hg2+ in practical application.

Abstract:Heavy metal ions, such as Hg(2+) and Ag(+), pose severe risks in human health and the environment. For sensitive detection and selective removal of Hg(2+) and Ag(+) ions, here, we demonstrate a surface-enhanced Raman scattering (SERS)-active platform by employing the oligonucleotide-functionalized magnetic silica sphere (MSS)@Au nanoparticles (NPs). This system exploits mismatched T-Hg-T and C-Ag-C bridges to capture Hg(2+) and Ag(+) ions, exhibiting excellent responses for Hg(2+) ions in the range of 0.1-1000 nM and for Ag(+) in the range of 10-1000 nM. The assay is highly selective for the target ions and does not respond to other metal ions. Additionally, the Hg(2+) and Ag(+) ions in this system can be effectively removed from surrounding solutions by an external magnetic field or through spontaneous precipitation. Moreover, more than 80% of the MSS@Au NPs can be easily recycled with the help of cysteine. We anticipate that the designed strategy could be extended to other analytes that can bind to DNA molecules with a high affinity, and can be used in many potential applications such as environmental renovation, toxin detection, and groundwater analysis.

Abstract:Precisely probing mercury ions (Hg2+) is of essential importance to human health and environmental protection. Although numerous methods have been developed to detect Hg2+ in water, challenges still remain for rapid, accurate, and reliable detection of Hg2+ at the ppt level. In this study, a novel label-free Fe3O4@Ag based surface-enhanced Raman resonance scattering (SERRS) nanosensor has been developed for ultrasensitive and highly selective detection of Hg2+. The detection mechanism is on the basis of competitive binding interaction of Hg2+ and malachite green (MG) with nano-sliver on Fe3O4@Ag magnetic beads (MBs). In the absence of Hg2+, the Raman signal intensity of MG is significantly enhanced by the nano-silver when MG adsorbs on the nano-silver surface via one nitrogen atom. In the presence of Hg2+, the redox reaction occurring between zero-valent nano-silver and Hg2+ leads to the formation of a Ag/Hg amalgam at the Fe3O4@Ag surface, which prevents the adsorption of MG on the nano-silver surface. Thereby, the SERRS signal intensity of MG proportionally decreased with increasing Hg2+ concentration. Under optimized conditions, the proposed label-free nanosensor showed unprecedented Hg2+ detection sensitivity of 10 pM (2 ppt) with excellent selectivity. This simple sensor system can be directly applied for an inexpensive and easy-to-use monitoring method of Hg2+ in drinking water and wastewater treatment effluents that contains complicated organic and inorganic interferents.
Specific Detection of Mercury Ions Based on Surface Plasmon Resonance Sensor Modified with 1, 6-hexanedithiol

Abstract:We report an angle-modulation surface plasmon resonance (SPR) sensor, designed with a five-layer Kretschmann prism-coupled configuration, for the specific detection of mercury ions. To simplify the calculation and obtain accurate results efficiently, we employ the transition matrix method instead of traditional Fresnel reflection law system. Due to the specific linkage among sulfhydryl group, Hg 2+ and Au, we modify 1, 6-hexanedithiol on sensor chips by chemical self-assembly method. A silver/gold (Ag/Au) composite film is employed and optimized to excite SPR effect for higher sensitively detection. Experimental results show that the modified chip can increase the sensor's sensitivity by two orders of magnitude compared with using unmodified chips. A sensitivity of 1.25×10 -2 °/ppm -1 and a detection limit of 80 ppb for Hg 2+ detection has been achieved. In mixed heavy metal ions aqueous medium, solutions containing Hg 2+ causes a red shift of 0.79° in the resonance angle, while that is only 0.235° in solutions without Hg 2+ . The results prove that the 1, 6-hexanedithiol has a good selectivity in detection of Hg 2+ .

Abstract:Precisely probing heavy metal ions in water is important for molecular biology, environmental protection, and healthy monitoring. Although many methods have been reported in the past decade, developing a quantitative approach capable of detecting sub-ppt level heavy metal ions with high selectivity Is still challenging. Here we report an extremely sensitive and highly selective nanoporous gold/aptamer based surface enhanced resonance Raman scattering (SERRS) sensor. The optical sensor has an unprecedented detection sensitivity of 1 pM (0.2 ppt) for Hg2+ ions, the most sensitive Hg2+ optical sensor known so far. The sensor also exhibits excellent selectivity. Dilute Hg2+ ions can be identified in an aqueous solution containing 12 metal ions as well as in river water and underground water. Moreover, the SERRS sensor can be reused without an obvious loss of the sensitivity and selectivity even after 10 cycles.

Abstract:It is of essential importance to precisely probe mercury(II) (Hg2+) ions for environment-protection analysis and detection. To date, there still remain major challenges for accurate, specific, and reliable detection of Hg2+ ions at subppt level. We herein employ gold nanoparticles (AuNPs) decorated silicon nanowire array (SiNWAr) as active surface-enhanced Raman scattering (SERS) substrates to construct a high-performance sensing platform assisted by DNA technology, enabling ultrasensitive detection of trace Hg2+ in similar to 64 min and with low sample consumption (similar to 30 mu L). Typically, strong SERS signals could be detected when the single-stranded DNA structure converts to the hairpin structure in the presence of Hg2+ ions, due to the formation of thymine (T)-Hg2+-T. As a result, Hg2+ ions with a low concentration of 1 pM (0.2 ppt) can be readily discriminated, much lower than those (nM) reported for conventional analytical strategies. Water samples spiked with various Hg2+ concentrations are further tested, exhibiting a good linear relationship between the normalized Raman intensities and the logarithmic concentrations of Hg2+ ranging from 1 pM to 100 nM, with a correlation coefficient of R-2 = 0.998. In addition, such SERS sensor features excellent selectivity, facilely distinguishing Hg2+ ions from various interfering substances. Moreover, this presented SERS sensor possesses good recyclability, preserving adaptable reproducibility during 5-time cyclic detection of Hg2+. Furthermore, unknown Hg2+ concentration in river water can be readily determined through our sensing strategy in accurate and reliable manners, with the RSD value of similar to 9%.

Abstract:It is important to precisely monitor mercury (II) ions (Hg2+) for environment protection and human health monitoring. Although many strategies have been developed in the past decades, there still remains a challenge for developing an ultrasensitive, simple and reliable approach to detect Hg2+. Herein, we report a ratiometric surface-enhanced Raman scattering (SERS) aptasensor by employing aptamer-modified Au@Ag core-shell nanoparticles (Au@Ag NPs) as highly functional sensing probes, allowing for ultrasensitive detection of Hg2+. In principle, the thiolated 5'-Cy3 labeled aptamer probe (Cy3-aptamer) is firstly immobilized on the SERS substrate surface and then hybridizes with the 5'-Rox labeled complementary DNA (cDNA) to form a rigid double-stranded DNA (dsDNA), in which the Cy3 and Rox Raman labels are used to produce the ratiometric Raman signals. In the presence of Hg2+, the aptamer DNA turns into the thymine (T)-Hg2+-T mediated hairpin structure, leading to the dissociation of dsDNA. As a result, the Rox labels are away from the Au@Ag NP SERS substrate while Cy3 labels are close to it. Therefore, the intensity of SERS signal from Cy3 labels increases while that from Rox labels decreases. The ratio between the Raman intensities of Cy3 labels and Rox labels is linear with Hg2+ concentrations in the range from 0.001 to 1.0nM, and the limit of detection is estimated to be 0.4pM. The proposed strategy provides a new rapid, simple and reliable approach for sensitive detection of Hg2+ and may create a universal methodology for developing analogous aptasensors for a wide range of other analytes determination.

Abstract:Due to the nonbiodegradability and accumulation of mercury ion, even in extremely small amount, it will cause varying degrees of harm to environment and human health. Although researchers have developed many strategies to detect and monitor trace Hg2+, only a few provide sensitivities of less than 1.0 pM. Surface Enhanced Raman Spectroscopy (SERS) is a common method to detect mercury ion due to its high sensitivity, rapid detection and easy operation. In this work, we report a new SERS aptasensor based on dual recycling amplification for the detection of trace mercury ion, which combines SERS with nucleic acid signal amplification through functional aptamer and elaborately designed hairpin DNA. Under the optimal experimental conditions, this SERS aptasensor exhibits excellent selectivity and high sensitivity. A linear range (0.2-125 fM) and a low detection limit (0.11 fM) are obtained. By using specific aptamers, the strategy will provide a new idea for the trace detection of toxic contaminants in water environment.
A SERS-based Capillary Sensor for the Detection of Mercury Ions in Environmental Water.

Abstract:Mercury ion ( Hg2+) is one of the most toxic heavy metal ions which will cause permanent damage to the brain and kidneys. So, it is important to develop a sensitive, simple and reliable approach to detect Hg2+. In this work, we report a surface-enhanced Raman scatting (SERS) sensor by decorating the inner wall of capillary with 4,4'-dipyridyl (Dpy) functionalized silver nanoparticles (AgNPs). The main advantage of this sensor is that it can collect samples directly by capillary force and carry out on-site analysis by combining portable Raman spectrometer. In the presence of Hg2+, the Dpy molecules would be separated from the surface of AgNPs and coordinated with Hg2+ resulting in a decrease in the SERS signal. A linear correlation of Raman intensity with Hg2+ concentrations from 1 to 100 part-per-billion (ppb) was obtained for quantitative analysis and the limit of detection (LOD) was determined to be 0.1 ppb. The good reproducibility and selectivity of the sensor were also demonstrated. In addition, the sensors were successfully applied to detect Hg2+ in real environmental water samples, and the sampling process provided operation convenience compared to conventional methods. These results indicated that these capillary sensors had great potential for Hg2+ detection in practical use. (C) 2020 Published by Elsevier B.V.
Highly Sensitive Electrochemical Sensor for Mercury(ii) Ions by Using a Mercury-Specific Oligonucleotide Probe and Gold Nanoparticle-Based Amplification.

Abstract:We report a highly sensitive electrochemical sensor for the detection of Hg(2+) ions in aqueous solution by using a thymine (T)-rich, mercury-specific oligonucleotide (MSO) probe and gold nanoparticles (Au NPs)-based signal amplification. The MSO probe contains seven thymine bases at both ends and a "mute" spacer in the middle, which, in the presence of Hg(2+), forms a hairpin structure via the Hg(2+)-mediated coordination of T-Hg(2+)-T base pairs. The thiolated MSO probe is immobilized on Au electrodes to capture free Hg(2+) in aqueous media, and the MSO-bound Hg(2+) can be electrochemically reduced to Hg(+), which provides a readout signal for quantitative detection of Hg(2+). This direct immobilization strategy leads to a detection limit of 1 microM. In order to improve the sensitivity, MSO probe-modified Au NPs are employed to amplify the electrochemical signals. Au NPs are comodified with the MSO probe and a linking probe that is complementary to a capture DNA probe immobilized on gold electrodes. We demonstrated that this Au NPs-based sensing strategy brings about an amplification factor of more than 3 orders of magnitude, leading to a limit of detection of 0.5 nM (100 ppt), which satisfactorily meets the sensitivity requirement of U.S. Environmental Protection Agency (EPA). This Au NPs-based Hg(2+) sensor also exhibits excellent selectivity over a spectrum of interference metal ions. Considering the high sensitivity and selectivity of this sensor, as well as the cost-effective and portable features of electrochemical techniques, we expect this Au NPs amplified electrochemical sensor will be a promising candidate for field detection of environmentally toxic mercury.

Abstract:In this paper, an electrochemical aptamer sensor with high sensitivity and selectivity was proposed for the detection of mercury ion (Hg 2+ ). The orderly assembled gold nanorods-modified screen-printed electrode (AuNRs/SPE) was used to improve the sensor with higher sensitivity and data reproducibility. The sensor was designed based on the specific recognition of Hg 2+ with aptamer to form T-Hg 2+ -T binding structure. Two aptamer chains were applied in the designed sensor, aptamer S1 with terminal thiolate group was self-assembled on the AuNRs/SPE via Au-S bond, and complementary S2 labeled with ferrocene (Fc) would generate a strong redox electrical signal. The addition of Hg 2+ would mediate the folding of the T-base of S1 to form a hairpin structure, resulting in the dissociation of the Fc-labelled S2 from the sensing system and a significant reduction in the redox current. The electrochemical properties of the sensor were further investigated using cyclic voltammetry (CV), differential pulse voltammetry (DPV) and electrochemical impedance spectroscopy (EIS). The effects of aptamer reaction time, concentration, and target incubation time on the sensor response were also investigated. The results showed that the sensor had a sensitive response to Hg 2+ at concentrations ranging from 1 pM to 1 μM, with a detection limit of 0.3 pM. Furthermore, the proposed sensor was applied in the determination of Hg 2+ in real samples, demonstrating its potential practical value.

Abstract:A localized surface plasmon resonance (LSPR) light-scattering sensor for Hg2+ was developed with unmodified gold nanoparticles (AuNPs) based upon the specific recognition property of Hg2+ with T-T mismatched base pair. Oligonucleotide 5'-GTTCTTTGTCTTCA-3'(oligo-1) and 5'-TGTAGTCTATGTAC-3'(oligo-2) can adsorb on the surface of AuNPs, which can prevent them from aggregation because of electrostatic repulsion. However, the DNA hybridization occurred between oligo-1 and oligo-2 upon addition of Hg2+, can induce the desorption of oligonucleotide from the surface of AuNPs, and trigger the aggregation of AuNPs accompany with the increase of LSPR light-scattering intensity. Under the optimum conditions, the intensity was proportional to the concentration of Hg2+ over the range 53.1-530 nM, and the detection limit was 29.4 nM.

Abstract:We report the rapid and highly sensitive trace analysis of mercury(ii) ions in water using a surface-enhanced Raman scattering (SERS)-based microdroplet sensor. Aptamer-modified Au/Ag core-shell nanoparticles have been fabricated and utilized as highly functional sensing probes. All detection processes for the reaction between mercury(II) ions and aptamer-modified nanoparticles were performed in a specially designed microdroplet channel. Small water droplets that included sample reagents were separated from each other by an oil phase that continuously flowed along the channel. This two-phase liquid-liquid segmented flow system prevented the adsorption of aggregated colloids to the channel walls due to localized reagents within encapsulated droplets. The result was reduced residence time distributions. The limit of detection (LOD) of mercury(II) ions in water was determined by the SERS-based microdroplet sensor to be below 10 pM, which is three orders below the EPA-defined maximum contaminant level. This combination of a SERS-based microfluidic sensor with aptamer-based functional nanoprobes can be used for in-the-field sensing platforms, due to its size and simplicity.
