Preliminary Studies of Human Sodium Iodide Symporter Gene Imaging and Therapy Through 188re: A Human Glioma Study in Model Mice

Abstract:1409 Objectives With a higher energy than 131I, we investigated the role of 188Re in human sodium iodide symporter (hNIS) mediated gene imaging and therapy in human glioma in model mice. Methods Human glioma cell line U87 transfected with recombinant lentivirus encoding hNIS gene under the control of the cytomegalovirus (CMV) promoter was constructed (U87-hNIS), hNIS was expressed in the cell line. The uptake and efflux of 188Re were determined after incubation of the cells with 188Re, the sodium perchlorate depression test was carried out. 188Re mediated U87-hNIS nude mice imaging was tested. Both in vitro and in vivo therapy were peformed. Results hNIS expressed cell line (U87-hNIS) was established. The uptake of 188Re in U87-hNIS cells increased up to 26-fold compared with control U87 cells, but released rapidly, with a half-life of about 4 minutes. Cellular 188Re accumulation was depressed by sodium perchlorate. Transfected cells were selectively killed by exposure to 188Re, with a survival of 21.31%. In nude mice, U87-hNIS cells accumulated more 188Re than that of the control mice. Unlike in vitro studies, U87-hNIS cells showed an markedly increased 188Re retention, even 48 h after 188Re injection (Fig.1). There is a difference in tumor size between U87-hNIS mice (317 ±67 mm3) and U87 mice (861 ± 153 mm3) treated with 188Re for 4 weeks (P Conclusions The study indicated that the U87-hNIS cell line is sufficient to induce specific 188Re uptake both in vitro and in vivo, outlining the potential of this novel cancer gene imaging and therapy approach for a targeted radiotherapy.
