Abstract:A mapping population of 81 F11 lines (recombinant inbred lines: RILs), derived from a cross between a japonica variety Kinmaze and an indica variety DV85 by the single-seed descent methods, was used to detect quantitative trait loci (QTLs) for leaf bronzing index (LBI) and plant height (PH) under Fe2+ stress condition. Two parents and 81 RILs were phenotyped for the traits by growing them in Fe2+ toxicity nutrient solution. A total of three QTLs were detected on chromosome 3, with score LOD ranging from 3.79 to 5.89. Two QTLs controlling LBI was located at the region of X279-C25 and X144-X362 , and their contributions to total variation were 17.38 % and 22.07%, respectively. One QTL for PH was located at the region of R1468A-R1468B, with contribution rate 23.18%. Comparing with the other mapping results, the QTL for LBI located at the region of X279-C25 on chromosome 3 was identical with the QTL for chlorophyll content on a rice function map. Another QTL for LBI located at the region of X144-X362 linked with QTL for chlorophyll content which located at the region of C136-C944 on a rice function map, the genetic distance between the two QTLs was 25.6 cM. The results indicated that ferrous iron toxicity of rice was characterized by bronzing spots on the lower leaves, which spread over the whole leaves, causing the lower leaves to turn dark gray and producing chlorophyll catabolites or derivatives which reduced cytotoxicity of some heavy metals, such as ferrous iron.
Mapping QTL for Traits Associated with Resistance to Ferrous Iron Toxicity in Rice (oryza Sativa L.),Using Japonica Chromosome Segment Substitution Lines

Abstract:A mapping population of 66 japonica chromosome segment substitution lines (CSSLs) in indica genetic background, derived from a cross between a japonica variety Asominori and an indica variety IR24 by the single-seed descent, backcrossing and marker-assisted selection, was used to detect quantitative trait loci (QTLs) for leaf bronzing index (LBI), stem dry weight (SDW), plant height (PH), root length (RL) and root dry weight (RDW) under Fe2+ stress condition in rice. Two parents and 66 japonica CSSLs were phenotyped for the traits by growing them in Fe2+ toxicity nutrient solution. A total of fourteen QTLs were detected on chromosome 3, 6, 7, 9, 11 and 12, respectively, with LOD of QTLs ranging from 2.72 to 6.63. Three QTLs controlling LBI were located at the region of C515-XNpb279, R2638-C1263 and G1465-C950 on chromosome 3, 9 and 11, their contributions to whole variation were 16.45%, 11.16% and 28.02%, respectively. Comparing with the other mapping results, the QTL for LBI located at the region of C515-XNpb279 on chromosome 3 was identical with the QTL for chlorophyll content on a rice function map. The results indicated that ferrous iron toxicity of rice is characterized by bronzing spots on the lower leaves, which spread over the whole leaves, causing the lower leaves to turn dark gray and to product chlorophyll catabolites or derivatives which reduce cytotoxicity of some heavy metals, such as ferrous iron. Furthermore, the QTL for LBI, SDW and RDW located at the region of G1465-C950 on chromosome 11 is a major QTL. Whether the QTL for SDW, PH, RL and RDW at the region of XNpb386-XNpb342 on chromosome 6 is associated with resistance to ferrous iron toxicity need further studies. Our goal is to identify breeding materials for resistance to Fe2+ toxicity through marker-assisted selection based on the detected markers.
Rice Genotype Differences in Nutrient Status under Excessive Ferric‐iron Conditions

Abstract:To investigate the relationship between rice genotypic variation in tolerance to iron (Fe) toxicity and nutrient element status, 10 rice genotypes with different growing performances under Fe toxicity were grown under normal culture solution and with excessive ferrous (Fe2+)-Fe concentrations of 250 and 500 mg Fe2+ L-1. A close relationship was obtained between the relative ratio of symptomatic leaf numbers to total leaf numbers (SLN/TLN) and a relative decrease in dry matter under Fe2+-toxicity conditions. The genotypic variations in nitrogen (N), phosphorus (P), potassium (K), and magnesium (Mg) uptake were evaluated by the relative decrease in the N, P, K, and Mg content in the plants. Remarkable genotypic variation in tolerance to excessive Fe2+ was observed. The results indicated that excessive Fe2+ reduced N, P, K, and Mg uptake. The nutrient element concentrations, however, were still higher above deficient criteria even in severely affected plants, suggesting that the retardation of growth may not be intirely due to the deficiency of these elements in plants at the seedling stage. Significant correlations were found between the genotypic variation and the decrease in N, P, K, and Mg uptake and in their tolerance to Fe2+ toxicity, which suggests that the ability to maintain higher nutrient element uptake under a Fe2+-toxic condition contributes the tolerance to Fe2+ toxicity.

Abstract:A mapping population of 96 BC 1 F 9 lines (Backcross Inbred Lines: BILs),derived by a single-seed descent method rom a backcross of Nipponbare ( japonica ) / Kasalath ( indica // Nippon are, was used to detect quantitative trait loci (QTLs) for leaf bronzing index (LBI), stem dry weight (SDW), tiller number (TN) and root dry weight (RDW) under Fe 2+ stress condition in rice. Two parents and 96 BILs were phenotyped for the traits by growing them in Fe 2+ toxicity nutrient solution. A total of four QTLs were detected on chromosome 1 and 3, respectively, with LOD of QTLs ranging from 3.17 to 7.03. One QTL controlling LBI, DW, N and RDW was located at the region of C955-C885 on chromosome 1, and their contributions to whole variation were 20.5%, 36.9%, 43.9% and 38.8%,respectively. The QTL located at the region of C955-C885 on chromosome 1 may be important to ferrous iron toxicity tolerance in rice. Another QTL for SDW and RDW was located at the region of C25-C515 on chromosome 3, with respective contributions of 47.9% and 35.0% to whole variation. Further, two QTLs on chromosome 1 were located for RDW at the region of R2329-R210 and for TN at the region of R1928-C178. Comparing with the other mapping results, the QTL located at the region of C955-C885 on chromosome 1 was identical with the results reported previously. There is a linkage between a TL detected under Fe 2+ stress condition for stem and root dry weight and a QTL detected under phosphorus-deficiency condition for dry weight on chromosome 3.
Mapping of QTLS for Ferrous Iron Toxicity Tolerance in Rice (oryza Sativa L.).

Abstract:Ferrous iron toxicity is the main factor limiting the productivity of rice in gleyic paddy soils. In this study, an F2 and an equivalent F3 populations derived from a japonica/indica cross of rice, Longza8503/IR64, were raised under iron-enriched solution cultures, and used to map QTLs controlling ferrous iron toxicity tolerance. A genetic linkage map consisting of 101 SSR markers was constructed to determine the position and nature of quantitative trait loci (QTLs) affecting Fe2+ toxicity tolerance. Three characters, i.e., leaf bronzing index (LBI), plant height (PH) and maximum root length (MRL) were evaluated for the F2 plants and F3 lines and the parents at the seedling stage in nutrient solution. A total of 20 QTLs for LBI, PH and MRL under the Fe2+ stress were detected over 10 of the 12 rice chromosomes, reflecting multigenic control of these traits. QTLs controlling LBI were located at the region of RM315-RM212 on chromosome 1, RM6-RM240 on chromosome 2 and RM252-RM451 on chromosome 4. Compared with other mapping results: (1) the QTL for LBI located at the region of RM252-RM451 on chromosome 4 was identical with the QTL for decreased chlorophyll content on a rice function map. Another QTL for LBI located at the region of RM315-RM212 on chromosome 1 was linked with the QTL for chlorophyll content which located at the region of C178-R2635 on a rice function map. (2) The third QTL for LBI located at the region of RM6-RM240 on choromosome 2 was linked with the QTL for potassium uptake located at the region of RZ58-CDO686 under potassium deficiency stress.
The Iron Contents of the Transgenic Rice Expressing a Pea Ferritin Gene As Affected by the Foliar Application of Ferrous Sulphate

Abstract:Using homozygous transgenic rice line (Fer34)transformed with a pea ferritin gene and its control rice Xiushui 11 as materials, variations in the accumulation of the iron content in the iron-rich transgenic lines (Fer34)and Xiushui 11 were compared. The results showed that the Fer34 line had a stronger capacity to accumulate the iron contents than the control, but the high iron contents were dramatically affected by the growing environment such as soil and climate. Foliar application of ferrous sulphate at filling stage increased the iron contents significantly, but had no negative effect on the grain appearance, eating quality and starch rapid visco analyzer (RVA)parameters. Therefore, the technical practice of using transgenic iron-rich transgenic rice in combination with foliar application of ferrous sulphate at filling stage facilitated the production of the iron- rich rice, which helped to solve the problem of iron nutrition deficiency in human.

Abstract:AbstractAfter seven generations of self-crossing assisted by β-glucuronidase (GUS) detection, 26 homozygous transgenic rice lines were obtained. Transgene effects on iron content, agronomic traits, rice quality and stress tolerance were studied. The results indicated that the average iron content in the milled grain of the transgenic (pea ferritin, Fer) rice (Oryza sativa) was 10.37 μg/g among all the homozygous lines, and only four lines (Fer34, Fer36, Fer39 and Fer65) had significantly higher iron contents than that of the control, Xiushui11 (6.46 μg/g). Significant differences in main agronomic traits, such as days from sowing to heading, plant height, main panicle length, total grains of main panicle, seed setting rate, 1000-grain weight, and yield per plant were observed among some homozygous lines, but similar flag leaf length and number of panicles per plant were found. This showed that insertion of the foreign gene (Fer) had a slight impact on the main agronomic traits, but that the transgene had no negative effects. The quality traits of homozygous rice lines, such as the percentage of brown rice, milled rice and full milled rice, grain length, grain width, ratio of grain length and width, and amylose content were all similar to those of the control. However, variations were noticed in chalkiness, translucency, alkali spreading value and gel consistency among partial homozygous lines. Responses of homozygous lines to low and high temperatures were not significantly correlated with the iron contents, and resistance to rice blast (Pyricularia oryzae), bacterial blight (Xanthomonas oryzae) and brown planthopper (Niaparvata lugens) were enhanced with increased iron contents.

Abstract:Mineral elements in brown rice grain play an important role in human health. In this study, variations in the content of iron (Fe), zinc (Zn), selenium (Se), cadmium (Cd), and lead (Pb) in 378 accessions of brown rice were investigated, and association mapping was used to detect the quantitative trait loci (QTLs) responsible for the variation. Among seven subpopulations, the mean values of Zn and Cd in the japonica group were significantly higher than in the indica groups. The population structure accounted for from 5.7% (Se) to 22.1% (Pb) of the total variation. Correlation analyses showed that Pb was positively correlated with the other minerals (P < 0.001) except for Se. For the five mineral elements investigated, 20 QTLs, including some previously reported and new candidate loci, were identified. Particularly, three cases of QTL colocalization, i.e. Cd and Pb on chromosome 5, Zn and Pb on chromosome 7, and Se and Pb on chromosome 11, were observed. This study suggested that the identified markers could feasibly be used to enhance desired micronutrients while reducing the heavy metal content in whole rice grain by marker-assisted selection (MAS).

Abstract:Iron (Fe) toxicity is a major issue adversely affecting rice production worldwide. Unfortunately, the physiological and genetic mechanisms underlying Fe toxicity tolerance in rice remain relatively unknown. In this study, we conducted a genome–wide association study using a diverse panel consisting of 551 rice accessions to identify genetic mechanisms and candidate genes associated with Fe toxicity tolerance. Of the 29 quantitative trait loci (QTL) for Fe toxicity tolerance detected on chromosomes 1, 2, 5, and 12, five (qSH_Fe5, qSFW_Fe2.3, qRRL5.1, qRSFW1.1, and qRSFW12) were selected to identify candidate genes according to haplotype and bioinformatics analyses. The following five genes were revealed as promising candidates: LOC_Os05g40160, LOC_Os05g40180, LOC_Os12g36890, LOC_Os12g36900, and LOC_Os12g36940. The physiological characteristics of rice accessions with contrasting Fe toxicity tolerance reflected the importance of reactive oxygen species–scavenging antioxidant enzymes and Fe homeostasis for mitigating the negative effects of Fe toxicity on rice. Our findings have clarified the genetic and physiological mechanisms underlying Fe toxicity tolerance in rice. Furthermore, we identified valuable genetic resources for future functional analyses and the development of Fe toxicity–tolerant rice varieties via marker–assisted selection.

Abstract:Fe is an essential mineral element that plants need for their growth. When there is low soil availability of Fe, plants show severe deficiency symptoms. Under Fe-deficiency conditions, plants alter a number of processes to acquire Fe from soil. Genes involved in these mechanisms have been identified from different model plants, including Arabidopsis and rice. Fe transport within plants is also tightly regulated. In this study, we used H9405, a cultivar of rice with high Fe accumulation in seeds, and Yangdao 6, a cultivar with low seed Fe accumulation, to study their responses under different Fe conditions. Our results showed that genes involved in acquisition of Fe from soil in these two cultivars were both up-regulated in roots under Fe-deficiency conditions, and the elevation of the expression was much higher in Yangdao 6 than in H9405. However, remobilization-related genes in shoot vasculature were expressed in an opposite way between the two cultivars. In H9405, the expression of these genes was up-regulated; but in Yangdao 6, their expression was reduced. Our results showed that the differential expression of root-uptake and shoot-remobilization genes in the two cultivars is correlated to the Fe content in roots, shoots, and seeds. Strategies to biofortify rice cultivars with different characteristics were also discussed based on our discovery.

Abstract:Iron (Fe) is an essential nutrient for almost all organisms. However, free Fe within cells can lead to damage to macromolecules and oxidative stress, making Fe concentrations tightly controlled. In plants, Fe deficiency is a common problem, especially in well-aerated, calcareous soils. Rice ( Oryza sativa L.) is commonly cultivated in waterlogged soils, which are hypoxic and can cause Fe reduction from Fe 3+ to Fe 2+ , especially in low pH acidic soils, leading to high Fe availability and accumulation. Therefore, Fe excess decreases rice growth and productivity. Despite the widespread occurrence of Fe excess toxicity, we still know little about the genetic basis of how rice plants respond to Fe overload and what genes are involved in variation when comparing genotypes with different tolerance levels. Here, we review the current knowledge about physiological and molecular data on Fe excess in rice, providing a comprehensive summary of the field.

Ameliorating Effect of Potassium on Iron Toxicity in Hybrid Rice

Abstract:Toxicity of Fe2+ has been a constraint to lowland rice production, potassium has the ameliorating effect on Fe2+ toxicity .Hybrid rice (Oryza sativa L.) were grown in nutrient solution with three K supplies (0.26, 0.51 and 1.02 mmol/L) and five Fe2+ levels (0.09, 0.18, 0.36, 0.54, and 0.90 mmol/L). Excessive Fe2+ greatly decreased root and shoot dry weights, root length, and shoot height of the hybrid rice, and this depression was obvious even at relatively low Fe2+ levels (above 0.18 mmol/L). Excessive Fe2+ levels notably influenced morphological parameters and concentrations of K, P, and N in the plants. High K supplies ameliorated the negative effects of higher Fe2+, as indicated by increases in biomass and nutrient contents. Proper K supply reduced translocation of Fe2+ from roots to shoots, especially to the upper leaves. Phosphorus, which is antagonistic to Fe2+, was significantly increased by K supply. Ameliorating effects of K may be attributed to the antagonistic effect of K on Fe absorption and translocation to shoots, especially to upper leaves, and to the improvement of plant nutritional status.

Abstract:Iron (Fe) toxicity is a major abiotic stress in lowland rice production. Breeding tolerant varieties has proven challenging due to the complex genetic architecture of Fe toxicity tolerance and the strong genotype-by-environment interactions. Additionally, conventional methods for phenotyping visible stress symptoms are often inaccurate, inconsistent, and lack reproducibility. In our previous work, we identified that ascorbate redox regulation, mediated by the activities of dehydroascorbate reductase (DHAR) and ascorbate oxidase (AO), contributed to high tolerance in an indica rice genotype across various environments. To explore whether this mechanism is common among other rice genotypes, we selected ten genotypes with contrasting stress symptoms under Fe-toxic conditions to examine the roles of DHAR and AO in regulating Fe toxicity tolerance. Additionally, we aimed to develop objective and accurate image-based phenotyping methods to replace the traditional leaf bronzing scoring method. Among the ten genotypes we tested, we found significant positive correlations between DHAR activity and stress symptoms in plants grown under both Fe toxicity and control conditions, suggesting a general link between ascorbate redox regulation and Fe toxicity tolerance. Using RGB signals from leaf images of plants exposed to 1000 mg/L Fe 2+ , we evaluated 36 different color indices to quantify stress symptoms. We identified the normalized green‒red difference index as most significant in quantifying stress symptoms under Fe toxicity conditions. Our findings suggest that DHAR activity could be potentially employed as a biomarker in the screening of rice germplasms and breeding tolerant cultivars to Fe toxicity.

Latent Function of Microorganisms on Plant Iron Acquisition

Abstract:Iron deficiency in plants is a worldwide agricultural problem. It has been proved that soil microorganisms have a beneficial effect on the iron acquisition of some plants, but the underlying mechanism is still unclear. Based on last ten years' research results, possible mechanism on how the microorganisms favor plant iron acquisition was discussed in this review. Iron-deficiency-induced root exudates, such as phenolic and flavin compounds, acting as inhibitors and carbon sources, affected the architecture of microorganisms in the rhizosphere and induced a formation of a specified microorganism community. Those specified microorganisms in turn benefited the plant iron acquisition through the secretion of siderophore which could improve the bioavailability of iron in soil. In addition, the symbiosis between some microorganisms and plants could also improve plant iron nutrient status. The underlying processes may involve: the formation of rhizobium nodulation enhanced plant iron-deficient response, and mycorrhizal fungi infected root which improved the iron nutrient status through enlarging absorption area and excreting the materials which chelated iron of plant.
Shoot-Root Communication Plays a Key Role in Physiological Alterations of Rice ( Oryza sativa ) Under Iron Deficiency.

Abstract:Iron (Fe) is an essential mineral element required for plant growth, and when soil availability of Fe is low, plants show symptoms of severe deficiency. Under conditions of Fe deficiency, plants alter several processes to acquire Fe from soil. In this study, we used rice cultivars H 9405 with high Fe accumulation in seeds and Yang 6 with low Fe accumulation in seeds to study their physiological responses to different conditions of Fe availability. In both shoots and roots, the responses of ROS enzymes, leaf and root ultrastructure and photosynthetic system to iron deficiency in Yang 6 were much sensitive than those in H 9405. For the distribution of iron, the iron content was much higher in roots of Yang 6, in contrast to higher shoot content in H 9405. Differential responses were shown with the Fe content in roots and shoots, which were the opposite in the two varieties; thus, we proposed the existence of long-distance signals. Then split root and shoot removal experiments were used to demonstrate that a long-distance signal was involved in the iron-deficient rice plant, and the signal strength was highly correlated with the functional leaves.

Abstract:Iron (Fe) distribution in plant of a high Fe concentration genotype IR68144 and a low one IR64 was compared. In experiment I, Fe supply was stopped at the fourth leaf stage for 30 d, and the results showed that 6.37 µg Fe/plant in older leaves and 5.5 µg Fe/plant in leaf sheath were retranslocated into newly growing leaves and tillers in IR68144, which was 1.9 and 3.4 times of those in IR64. In experiment II, plants were treated with three different Fe levels to anthesis and then stopping Fe supply until full mature; the results showed that total Fe uptake of both genotypes increased with Fe treated levels, but Fe concentration in brown rice did not. After stopping Fe supply, more Fe was moved out from flag leaf into newly growing organs in IR68144. Those results indicated IR68144 has a greater ability to reutilize Fe from source tissues than IR64.

Abstract:The chemical form and physiological activity of iron (Fe) in soil are dependent on soil pH and redox potential (Eh), and Fe levels in soils are frequently elevated to the point of causing Fe toxicity in plants, with inhibition of normal physiological activities and of growth and development. In this review, we describe how iron toxicity triggers important physiological changes, including nitric-oxide (NO)-mediated potassium (K + ) efflux at the tips of roots and accumulation of reactive oxygen species (ROS) and reactive nitrogen (RNS) in roots, resulting in physiological stress. We focus on the root system, as the first point of contact with Fe in soil, and describe the key processes engaged in Fe transport, distribution, binding, and other mechanisms that are drawn upon to defend against high-Fe stress. We describe the root-system regulation of key physiological processes and of morphological development through signaling substances such as ethylene, auxin, reactive oxygen species, and nitric oxide, and discuss gene-expression responses under high Fe. We especially focus on studies on the physiological and molecular mechanisms in rice and Arabidopsis under high Fe, hoping to provide a valuable theoretical basis for improving the ability of crop roots to adapt to soil Fe toxicity.

Abstract:In pot experiments, uptake of zinc (Zn), copper (Cu), iron (Fe), and manganese (Mn) by hybrid rice from different soil types was compared with a traditional rice (Oryza sativa L.) cultivar. The concentration and total uptake of Fe in the shoots of hybrid rice grown in Oxisol and Ultisol were lower than those of the traditional cultivar. The concentration and total uptake of Zn in the shoots of hybrid rice grown in the Inceptisol (calcareous) were significantly higher than those of the traditional cultivar. Higher ratios of Zn and Fe in upper leaves (UL) to the lower leaves (LL) were found in hybrid rice grown in the calcareous Zn-deficiency soil. The results indicated that hybrid rice root avoided absorbing excess Fe from Fe-toxic soils due to its higher oxidizing power, and was more efficient in absorbing Zn from calcareous Zn-deficient soils than the traditional cultivar.
