Abstract:A convenient method to perdeuterate aromatic compounds is reported. Commercially available nonaflic acid (C4F9SO3H) in combination with C6D6 as the deuterium source provided access to a series of highly labeled arenes. Some of them were then used as building blocks for the preparation of functionalized deuterated products. A wide range of synthetic procedures were investigated. They involved traditional methods such as halogenation or nucleophilic substitution, but also metal‐catalyzed cross coupling and C−X bond activation. The transformations comprised redox‐neutral, oxidative as well as reductive reactions, and mechanistically, ionic, radical and pericyclic processes were covered. Under many conditions, the overall degree of deuteration remained high; in C−X activation reactions, a more pronounced re‐protonation was observed.

Abstract:To date, although tremendous advances have been made in N-heteroaryl C-H functionalization, there remain significant unmet challenges in the deconstruction of extensively applied but poorly reactive N-heteroaromatics to useful frameworks. Here, by a strategy merging hydrogen transfer and selective coupling, we present a ruthenium-catalyzed deconstruction of N-heteroaromatics to functionalized arylamines with 2-aminoaryl methanols. The reaction is achieved via sequential functionalization of the beta and alpha sites of the initially formed N-heteroarenium salts followed by a C-N cleavage, which proceeds with the striking features of broad substrate scope, excellent functional group tolerance, high chemoselectivity and atom efficiency, and suitability for a streamlined synthesis of some biomedical molecules. The strategy utilized will pave the way for further development of catalytic transformations of inert organo systems to functional frameworks.

Abstract:Functionalized arenes, such as aniline and phenol derivatives, are important structures in natural products, drug molecules, and functional materials. Thus, the development of methods for the efficient synthesis of functionalized arenes from different precursors under mild conditions have been a long‐term topic in organic synthesis and related areas. To date, great efforts have been devoted the Ar‐X bond‐forming processes from aromatic precursors. In addition, synthesis of functionalized arenes from aliphatic architectures represents an attractive yet challenging alternative to access functionalized arenes, which construct aromatization during the reaction process along with the formation of C‐X bonds. Recently, a new strategy for functionalized arenes from non‐aromatic precursors enabled by radicals has received considerable attention and represents a new direction to access functionalized arenes from broader precursors at room temperature.

Abstract:Aromatization-driven deconstruction and functionalization of spiro dihydroquinazolinones via dual photoredox/nickel catalysis is developed. The aromatization effect was introduced to synergistically drive unstrained cyclic C-C bond cleavage, with the aim of overcoming the ring-size limitation of nitrogen-centered radical induced deconstruction of carbocycles. Herein, we demonstrate the synergistic photoredox/nickel catalyzed deconstructive cross-coupling of spiro dihydroquinazolinones with organic halides. Remarkably, structurally diverse organic halides including aryl, alkenyl, alkynyl, and alkyl bromides were compatible for the coupling. In addition, this protocol is also characterized by its mild and redox-neutral conditions, excellent functional group compatibility, high atom economy, and easy scalability. A telescoped procedure involving condensation and ring-opening/coupling was found to be accessible. This work provides a complementary strategy to the existing radical-mediated C-C bond cleavage of unstrained carbocycles.

Abstract:Diboron-mediated rhodium-catalyzed transfer hydrogenation of functionalized arenes is reported. In addition to good functional group tolerance, the reaction features operational simplicity and controllable chemoselectivity. The general applicability of this procedure is demonstrated by the selective hydrogenation of a range of arenes, including functionalized benzenes, biphenyls, and polyaromatics.

Abstract:m-Disubstituted arenes are important in various fields but difficult to be directly synthesized by the classical methods in the case of two electron-donating substituents due to the o,p-orientation. Dehydrogenative aromatization from non-aromatic cyclohexenone motifs and nucleophiles via 1,2- and 1,4-addition to produce m-disubstituted arenes is promising as the novel process overcoming the intrinsic problems derived from using aromatic compounds as the substrates; however, there are no reports on tandem (one-step) synthesis via simultaneous 1,2- and 1,4-addition or m-phenylenediamine derivative synthesis via dehydrogenative aromatization because of the quite difficult product selectivity control. Here, by utilizing unique dehydrogenation catalysis of CeO2-supported Au nanoparticles, we have developed tandem aerobic oxidation to various m-phenylenediamine derivatives via selective dehydrogenative aromatization from cyclohexenone motifs and secondary amines. In addition, the product selectivity switch to enaminones or N,N-disubstituted anilines was also achieved by tuning reaction conditions.

Abstract:AbstractIn arene chemistry, CH direct functionalization has become one of the most attractive and challenge strategies for the introduction of new fragments bringing molecular complexity. Among a great number of transition metals involved in reaction approaches, Ru‐catalyzed CH functionalization of directed aryl‐containing compounds certainly provided unique reaction modes and new synthetic consideration in molecular design. In this article, we have overviewed the recent advances in the development of new synthetic approaches within the field of Ru‐catalyzed remote CH functionalization of directed arenes. The developments of the methodologies, synthetic applications, and reaction mechanisms on typical reaction modes have been discussed, and examples for accessing molecules in medicinal chemistry were also included.

Abstract:The direct functionalization of C–H bonds has revolutionized the field of synthetic organic chemistry by enabling efficient and atom-economical modification of arenes by avoiding prefunctionalization. However, the inherent challenges of inertness and regioselectivity in different C–H bonds, particularly for distal positions, necessitate innovative approaches. In this aspect, photoredox catalysis by utilizing both transition metal and organic photocatalysts has emerged as a powerful tool for addressing these challenges under mild reaction conditions. This review provides a comprehensive overview of recent progress in regioselective C–H functionalization in arenes via photocatalysis. Emphasizing the strategies for achieving ortho, meta, and para-selectivity, we explore the mechanistic insights, catalyst designs, and the novel methodologies that have expanded the scope of C–H bond functionalization. This discussion aims to offer valuable perspectives for advancing the field and developing more efficient and sustainable synthetic methodologies.

Abstract:Dearomatization of arenes emerges as a reliable strategy for crafting intricate 3D polycyclic frameworks. Nonetheless, the development of an efficient method for dearomatization across diverse arenes remains a formidable challenge, particularly in the case of unactivated benzene. In this study, we present a facile dearomative difunctionalization approach for unactivated arenes through a highly selective radical relay reaction. The reaction operates under mild conditions, devoid of metals, photocatalysts, and additives. This method exhibits a broad substrate scope and exceptional functional group tolerance, successfully transforming various aromatics, including inert naphthalene and benzene rings, indoles, pyrroles, benzofuran, and thiophene. Bioactivity assessments reveal a significant inhibitory effect of the synthesized complex polycyclic frameworks on HCT 116 cancer cells, with subsequent mouse experiments providing compelling evidence of its feasibility.

Abstract:Decarboxylative C-H functionalization reactions are highly attractive methods for forging carbon-carbon bonds considering their inherent step- and atom-economical features and the pervasiveness of carboxylic acids and C-H bonds. An ideal approach to achieve these dehydrogenative transformations is through hydrogen evolution without using any chemical oxidants. However, effective coupling of decarboxylative carbon-carbon bond formation with proton reduction remains an unsolved challenge. Herein, we report an electrophotocatalytic approach that merges organic electrochemistry with photocatalysis to achieve efficient direct decarboxylative C-H alkylation and carbamoylation of heteroaromatic compounds through hydrogen evolution. The electrophotocatalytic method, which combines the high efficiency and selectivity of photocatalysis in promoting decarboxylation and electrochemistry in effecting proton reduction, allows efficient coupling of a wide range of heteroaromatic bases with a variety of carboxylic acids and oxamic acids. Advantageously, this method is scalable to decagram scale and applicable to the late-stage functionalization of drug molecules.

Abstract:Although dearomative functionalizations enable the direct conversion of flat aromatics into precious three-dimensional architectures, the case for simple arenes remains largely underdeveloped due to the high aromatic stabilization energy. We herein report a dearomative sequential addition of two nucleophiles to arene π-bonds via umpolung of chromium-arene complexes. This mode enables divergent dearomative carbonylations of benzene derivatives by tolerating various nucleophiles in combination with alcohols or amines under CO-gas-free conditions, thus providing modular access to functionalized esters or amides. The tunable synthesis of 1,3- or 1,4-cyclohexadienes as well as the construction of all-carbon quaternary centers further highlight the versatility of this dearomatization. Diverse late-stage modifications and derivatizations towards synthetically challenging and bioactive molecules reveal the synthetic utility. A possible mechanism was proposed based on control experiments and intermediate tracking.

Abstract:Nonactivated arenes, such as benzene derivatives, are chemically inert due to their intrinsic aromaticity and low polarity. The catalytic asymmetric dearomatization (CADA, coined by You and co-workers) of the nonactivated arenes represents a formidable challenge. We herein demonstrated an organocatalytic asymmetric dearomatizing hetero-Diels-Alder reaction of benzene derivatives. The tunable regioselectivity of this strategy allowed delivery of a diversity of stereochemically complex polycyclic compounds and oxahelicenes with excellent stereoselectivity. The high complexity and three-dimensionality of the products are crucial for their potential applications in materials science and drug discovery. Mechanistic studies suggested that this reaction proceeds through a chiral tetra-substituted vinylidene ortho-quinone methide (VQM) intermediate, which is extremely active to overcome the loss of aromaticity of benzene derivatives with concomitant chirality transfer.

Abstract:Abstract While a great number of ortho C–H functionalization reactions have been developed and several breakthroughs have been achieved in meta C–H activation, para C–H functionalization is still in its infancy stage. In this article, a versatile strategy for the synthesis of para-substituted arenes has been developed via a tandem process consisting of palladium-catalyzed C–H functionalization and subsequent copper-catalyzed protodecarboxylation of benzoic acids. Both electron-withdrawing and electron-donating functionalities can be introduced into the para positions of arenes bearing a variety of substituents.

Abstract:Terpenes, consisting of isoprene monomer units, represent a family of naturally abundant compounds. The difunctionalization of terpenes is highly appealing yet remains challenging, since the multiple unbiased C=C bonds of terpenes lead to difficulty in controlling the regioselectivity. Herein, a cobalt(iii)-catalyzed C-H activation strategy has been developed to facilitate hydroxymethylarylation of terpenes with formaldehyde and arenes with high chemo- and regio-selectivities. These (chemo- and regio-) selectivities are governed by the coordination abilities of isoprene, directing groups and the steric effect. This terpene difunctionalization also features high atom and step economy through a C-H addition pathway.

Abstract:A general and practical approach for diverse dealkenylative functionalization of olefin-containing substrates has been developed through the one-pot formation and utilization of pro-aromatic 1,4-dihydropyridazines using tetrazine as the key cycloaddition reagent. Triggered by either excitation or oxidation, the targeted C-C bonds in the 1,4-dihydropyridazine intermediates could be readily cleaved to generate alkyl radicals for subsequent transformations. Diverse carbon-carbon and carbon-hetero bond forming protocols, including Giese-type addition, hydrazination, borylation, Minisci-type alkylation, copper-catalyzed NH alkylation, acylation, alkynylation, cyanation, and azidation, are achieved in a highly modular fashion.

Abstract:A novel α-arylation of deoxybenzoins with non-prefunctionalized arenes is developed through an iron-catalyzed oxidative dehydrogenative approach. Abstract:The palladium-catalyzed functionalization of 5-membered ring heteroaromatics, through a C-H bonds activation, for access to arylated heteroaromatics represent a very attractive alternative to more classical cross-coupling reactions, as it saves steps and reduces the amount of wastes. Despite these advantages, these transformations initially suffered from major limitations in terms of sustainable chemistry concerning the reaction media, the nature of the catalyst, the catalyst loading, the aryl source or the regioselectivity control. Tremendous improvements allowing more sustainable reaction conditions using greener solvents, new aryl sources or employing easily removable catalysts and lower catalyst loadings have been described during the last decades. This review summarizes some of the most important contributions made in these directions.
Access to Arynes from Arenes via Formal Dehydrogenation: Increased Efficiency and Scope, Synthetic Applications and Mechanistic Analysis

Abstract:Arynes undergo a wide range of diverse chemical transformations making them highly versatile reactive in-termediates for organic synthesis. Access to arynes has long been dominated by pre-functionalized reagents such as the venerable o-trimethylsilylaryl triflate class of substrates. However, a move toward the development of methods to access arynes that are both mild and efficient has prompted research into aryl “onium” aryne precursors. We have leveraged aryl “onium” species as intermediates in an overall net dehydrogenation of simple arenes as an efficient way to generate and trap arynes. Herein, we describe two strategies to achieve mild and efficient access to arynes. 1) The development of a one-pot method that converts simple arenes into aryl thianthrenium salts and uses them in situ to generate arynes; the net transformation of arene to trapped aryne is achieved within two hours. 2) The development of a two-step process to convert arenes into aryl(Mes)iodonium salts and ultimately trapped arynes to expand the scope of arenes compatible; the net transformation is complete within four hours. Mechanistic analysis through competition experiments, deuterium kinetic isotope effects (DKIE) and Density Functional Theory (DFT) provide key insight into the similarity and differences of the two approaches described in this work and yield a user’s guide for selecting the appropriate strategy based on the arene.
