Abstract:The aim of this study was to develop a bioactive, degradable, and cytocompatible akermanite (Ca2MgSi2O7) scaffold with high porosity and pore interconnectivity. In brief, porous akermanite scaffolds were prepared using polymer sponge method. The porosity and corresponding compressive strength were evaluated. The in vitro degradability was investigated by soaking the scaffolds in Ringer's solution. Hydroxyapatite (HAp)-formation ability of akermantite scaffolds in simulated body fluid (SBF) and the effect of ionic products from the scaffolds dissolution on osteoblasts were investigated. In addition, bone marrow stromal cells (BMSC) adhesion and proliferation on the scaffolds were evaluated. Differentiation of the cells was assessed by measuring alkaline phosphatase (ALP) activity. The results showed that akermanite scaffolds possessed 63.5-90.3% of porosity, with a corresponding compressive strength between 1130 and 530 kPa. The weight loss of the scaffolds and ionic content of the Ringer's solution increased with the increase in soaking time, indicating the degradability of scaffolds. HAp was formed on the scaffolds in SBF and the ionic products from akermanite scaffolds dissolution stimulated osteoblasts proliferation, indicating good in vitro bioactivity. Furthermore, BMSC adhered and spread well on akermanite scaffolds and proliferated with the increase in the culture time, and the differentiation rate of osteoblasts on scaffolds was comparable to that on blank culture plate control. Our results suggested that akermanite scaffolds were bioactive, degradable, and cytocompatible, and might be used as bone tissue engineering materials.
Akermanite scaffolds reinforced with boron nitride nanosheets in bone tissue engineering

Abstract:Akermanite (AKM) is considered to be a promising bioactive material for bone tissue engineering due to the moderate biodegradability and excellent biocompatibility. However, the major disadvantage of AKM is the relatively inadequate fracture toughness, which hinders the further applications. In the study, boron nitride nanosheets (BNNSs) reinforced AKM scaffolds are fabricated by selective laser sintering. The effects of BNNSs on the mechanical properties and microstructure are investigated. The results show that the compressive strength and fracture toughness increase significantly with BNNSs increasing from 0.5 to 1.0 wt%. The remarkable improvement is ascribed to pull out and grain wrapping of BNNSs with AKM matrix. While, overlapping sheets is observed when more BNNSs are added, which results in the decline of mechanical properties. In addition, it is found that the composite scaffolds possess good apatite-formation ability when soaking in simulated body fluids, which have been confirmed by energy dispersed spectroscopy and flourier transform infrared spectroscopy. Moreover, MG63 osteoblast-like cells and human bone marrow stromal cells are seeded on the scaffolds. Scanning electron microscopy analysis confirms that both cells adhere and proliferate well, indicating favorable cytocompatibility. All the facts demonstrate the AKM scaffolds reinforced by BNNSs have potential applications for tissue engineering.

Abstract:The poor mechanical properties of akermanite (AKM), especially fracture toughness, limits its applications in bone tissue engineering, although it possesses favorable biological performance. In this research, silicon carbide whiskers (SiCw) are added in order to reinforce AKM scaffolds with controllable porous structures fabricated by selective laser sintering (SLS). The mechanical properties, microstructure and toughening mechanisms are analyzed. The results indicate that the compressive strength and fracture toughness increases with increasing SiCw from 5 to 20 wt.%, and then decreases when more SiCw are incorporated. The strengthening and toughening mechanisms are attributed to interface debonding, whisker fracture and whisker pull-out, and the main fracture mode becomes transgranular fracture. Moreover, the phase composition of SiCw remains constant, confirmed by X-ray diffraction (XRD) analysis. The bioactivity and degradation behaviour of the scaffolds were evaluated by soaking them in simulated body fluid (SBF). The results show that the composite scaffolds with 20 wt.% SiCw exhibits good apatite-mineralization ability and a moderate degradation rate in SBF medium. Moreover, scanning electron microscopy (SEM) analysis, MTT assay and alizarin red staining of human bone marrow stromal cells (hBMSCs) seeded scaffolds confirm the stem cell attachment, viability, proliferation and differentiation on the scaffolds. Thus, the overall study proves that SiCw reinforced AKM scaffolds have the potential to be used in bone tissue engineering.

Abstract:Akermanite had attracted great attention due to the favourable mechanical properties and excellent biological performance. In this research, the microstructure and mechanical properties of akermanite scaffolds fabricated via laser sintering under different process conditions were studied and characterized. The results showed that the akermanite particles gradually mixed together, grew up and reached complete densification with the scanning speed decreasing from 450 to 150mm/min, while micro defects such as air holes occurred at 50mm/min. simultaneously, the compressive strength of the scaffolds went up and then descended, and the optimum value was 5.92 +/- 0.41 MPa. The Vickers hardness and fracture toughness increased consistently and then tended to stabilize. X-ray diffraction (XRD) results indicated no new phase appeared under all process conditions. MG-63 cell culture revealed that cell adhesion and proliferation occurred, indicating excellent cytocompatibility of the scaffolds. Moreover, in vitro bioactivity tests showed that the apatite layer formed on the scaffolds and became dense and thick with the increase of soaking time in simulated body fluid (SBF), and this fact was further confirmed by energy-dispersive spectroscopy (EDS).

Abstract:Porous akermanite (Ca2MgSi2O7, AKT) ceramic scaffolds are regarded as a potential bioactive material for bone tissue engineering due to their biodegradability and osteostimulatory property. However, the main disadvantages of the porous AKT ceramic scaffolds are their relatively low mechanical strength. The purpose of this study is to prepare poly(d, l-lactic acid) (PDLLA)-reinforced AKT scaffolds (PR-AKT) and further investigate the effect of PDLLA modification on the mechanical strength, biodegradability and bioactive properties. The results showed that the compressive strength of AKT scaffolds was significantly improved by the modification of PDLLA, which was greatly dependent on the PDLLA concentrations. Furthermore, the PR-AKT scaffolds still maintained porous structure with large-pore size of 200–500µm. The porosity of scaffolds could be effectively tailored by controlling the mass ratio of porogen and AKT powders. When PR-AKT prepared scaffolds were soaked in Tris–HCl solution, the pH value of Tris–HCl solution was reduced indicating the neutralizing effect of PDLLA on the released ions (e.g. Ca2+, Mg2+ and SiO44−) from AKT scaffolds. In addition, it was found that PR-AKT scaffolds maintained good apatite-mineralization ability in simulated body fluids (SBF) and improved the attachment and proliferation of MC3T3-E1 cells. Our results indicate that PR-AKT scaffolds have potential for bone tissue engineering application by the virtue of improved mechanical strength, modulated degradability and enhanced cell attachment and proliferation.
Fabrication of β-TCP/ Akermanite composite scaffold via DLP and in-situ modification of micro-nano surface morphology for bone repair

Abstract:Although β-TCP bioceramics possess favorable biocompatibility and bioactivity, their inherent brittleness restricts further applications. In this study, we aimed to enhance the mechanical performance and biocompatibility of β-TCP by incorporating 15% Akermanite (AK) into the structure. The fabrication process involved the use of digital light processing (DLP) technology to fabricate ceramic green bodies, followed by high-temperature debinding and sintering to obtain the ceramic scaffold. Subsequently, we employed an in-situ growth technique to create micro/nano surface topography on the scaffolds, facilitating synergistic regulation of cell behavior in conjunction with the controlled release of bioactive agents. Scanning electron microscopy (SEM) revealed that the addition of AK enlarged the grain size and reduced the number of micropores in the composite scaffold. Furthermore, after the in-situ growth treatment, a significant increase in micro/nano surface features was observed. Mechanical testing demonstrated that the incorporation of AK effectively improved the mechanical properties of the composite scaffold, resulting in an increase in strength of approximately 20%. Cell experiments indicated that AK supplementation further enhanced the material's cell compatibility, and the presence of micro/nano structures effectively promoted cell adhesion, proliferation, and expression of osteogenic genes. Consequently, we propose a strategy for fabricating porous bioceramics that stimulate bone growth, exhibit high strength, and ameliorate the degradation microenvironment. This approach opens up promising avenues for future applications in bone tissue engineering.

Abstract:Some Ca–Mg-silicate ceramics have been widely investigated to be highly bioactive and biodegradable, whereas their osteogenic potential and especially biomechanical response in the early stage in vivo are scarcely demonstrated. Herein, the osteogenesis capacity and mechanical evolution of the akermanite (Ca 2 MgSi 2 O 7 ) porous materials manufactured by ceramic ink writing three-dimensional printing technique were investigated systematically in a critical size femur defect model, in comparison with the clinically available β-tricalcium phosphate porous bioceramic. Such three-dimensional printed akermanite scaffolds possess fully interconnected pores of ∼280 × 280 μm in size and over 50% porosity with appreciable compressive strength (∼71 MPa), that is 7-fold higher than that of the β-tricalcium phosphate porous bioceramics (∼10 MPa). After 6 weeks and 12 weeks of implantation, the percentage of newly formed bone and more new bone was observed in the akermanite group as compared with the β-tricalcium phosphate group ( p < 0.01). Moreover, significant higher mRNA expressions of osteogenic genes were detected in the akermanite group by PCR analysis ( p < 0.01). The in vivo mechanical strength decreased during the process of implantation, but maintained a relative high level (∼14 MPa) which was still higher than that of the host cancellous bone (5–10 MPa) at 12 weeks post-implantation. On the contrary, the β-tricalcium phosphate scaffold always exhibited a very low mechanical strength (∼8 MPa). These results suggest that the three-dimensional printed akermanite scaffolds are promising for the bone tissue regeneration and repair of load-bearing bone defects.

Abstract:Preparation of 3D scaffolds for bone tissue engineering (BTE) is a challenging task as it required appropriate pore size and porosities, excellent mechanical properties, controlled rate of bioerosion. The focus of this work is the fabrication of silk fibroin (SF) 3D microparticle scaffolds with the incorporation of hydroxyapatite (HA) and calcium sulfate (CaS) as bioceramics. Physicochemical characterization shows ∽30% filler loading, ∽ 40% optimum porosity with >100 µm pore size for these filled/unfilled scaffolds. Nanoindentation studies show improved Young's modulus at microparticle level with the incorporation of bioceramics. There is a three‐fold increase in Young's Modulus in the case of SF‐HA while two‐fold increase for SF‐CaS. In‐vitro bioerosion study results in early bioerosion with SF‐CaS scaffold while prolonged bioerosion with SF‐HA scaffold. In‐vitro osteo‐regenerative potential is analyzed by estimating ALP, BMP‐2 and OCN. SF‐CaS supports the early stage differentiation as compared to SF‐50%HA scaffolds, which predominantly support at late stage. The expression of TNF‐α, suggesting a reduced risk of immune rejection. This work, therefore, concludes that although SF support bone tissue regeneration, the choice of bioceramic enhances the applicability in various clinical scenarios by providing a controlled rate of bioerosion, tunable speed of osteo‐regeneration and improved load bearing capacities.This article is protected by copyright. Abstract:Desirable scaffolds for tissue engineering should be biodegradable carriers to supply suitable microenvironments mimicked the extracellular matrices for desired cellular interactions and to provide supports for the formation of new tissues. In this work, a kind of slightly soluble bioactive ceramic akermanite (AKT) powders were aboratively selected and introduced in the PLGA matrix, a novel l-lactide modified AKT/poly (lactic-<i>co</i>-glycolic acid) (m-AKT/PLGA) composite scaffold was fabricated via a solvent casting-particulate leaching method improved by solvent self-proliferating process. The effects of m-AKT contents on properties of composite scaffolds and on MC3T3-E1 cellular behaviors <i>in vitro</i> have been primarily investigated. The fabricated scaffolds exhibited three-dimensional porous networks, in which homogenously distributed cavities in size of 300-400 μm were interconnected by some smaller holes in a size of 100-200 μm. Meanwhile, the mechanical structure of scaffolds was reinforced by the introduction of m-AKT. Moreover, alkaline ionic products released by m-AKT could neutralize the acidic degradation products of PLGA, and the apatite-mineralization ability of scaffolds could be largely improved. More valuably, significant promotions on adhesion, proliferation, and differentiation of MC3T3-E1 have been observed, which implied the calcium, magnesium and especially silidous ions released sustainably from composite scaffolds could regulate the behaviors of osteogenesis-related cells.

Abstract:Porous bioceramic scaffolds were obtained by the 3D printing technique starting from a mixture of hydroxypropyl methyl cellulose and a powder obtained by sol-gel method which contains merwinite, monticellite, pseudowolastonie and periclase.The scaffolds were thermally treated at 1370 °C for 3 h and the main mineralogical compound assessed by XRD was akermanite. The obtained scaffolds have adequate mechanical and biological properties thus a great potential for applications in hard tissue engineering. The positive results obtained for this type of scaffolds are due to the precision of 3D printing technique, i.e. ability to control shape and size of both scaffolds and pores, as well as the high reproducibility. The porous bioactive ceramic scaffolds have adequate compressive strength (up to 3.5 MPa) and exhibit mineralization capacity and potential in stimulating cell adhesion and proliferation as well as bacteriostatic properties.

Abstract:The aggregation of low-dimensional nanofillers in a matrix has long been an unsolved problem in the fabrication of high-performance ceramic composites. Here, we demonstrated for the first time that a combined nanostructure was constructed by two-dimensional graphene and one-dimensional boron nitride nanotubes (BNNTs), in which the long and straight BNNTs sandwiched the broad and thin graphene sheets, thereby efficiently inhibiting the aggregation of graphene. The combined nanostructure was then used to reinforce akermanite bone scaffolds fabricated using selective laser sintering. The results showed remarkable improvements in their dispersion and reinforcing efficiency in ceramic bone scaffolds. Moreover, the compressive strength and fracture toughness were maximally improved by 207% and 33% with a graphene-BNNT ratio of 1:3 and total nanofiller content of 3.6 wt%, respectively. It was attributed to the efficient load transfer and energy-absorbing by graphene and BNNTs. Furthermore, the reinforced scaffolds also showed favorable biocompatibility and bioactivity. A novel porous titanium with engineered surface for bone defect repair in load‐bearing position

Abstract:Porous titanium exhibits low elastic modulus and porous structure is thought to be a promising implant in bone defect repair. However, the bioinert and low mechanical strength of porous titanium have limited its clinical application, especially in load‐bearing bone defect repair. Our previous study has reported an infiltration casting and acid corrosion (IC‐AC) method to fabricate a novel porous titanium (pTi) with 40% porosity and 0.4 mm pore diameter, which exerts mechanical property matching with cortical bone and interconnected channels. In this study, we introduced a nanoporous coating and incorporated an osteogenic element strontium (Sr) on the surface of porous titanium (named as Sr‐micro arch oxidation [MAO]) to improve the osteogenic ability of the pTi by MAO. Better biocompatibility of Sr‐MAO was verified by cell adhesion experiment and cell counting kit‐8 (CCK‐8) test. The in vitro osteogenic‐related tests such as immunofluorescence staining, alkaline phosphatase staining and real‐time polymerase chain reaction (RT‐PCR) demonstrated better osteogenic ability of Sr‐MAO. Femoral bone defect repair model was employed to evaluate the osseointegration of samples in vivo. Results of micro‐CT scanning, sequential fluorochrome labeling and Van Gieson staining suggested that Sr‐MAO showed better in vivo osteogenic ability than other groups. Taking results of both in vitro and in vivo experiment together, this study indicated the Sr‐MAO porous titanium could be a promising implant load‐bearing bone defect.

Abstract:This study investigated the effects of a calcium magnesium silicate bioceramic (akermanite) for bone regeneration in vitro and in vivo, with β-tricalcium phosphate (β-TCP) as a control. In vitro, the human bone marrow-derived mesenchymal stromal cells (hBMSCs) were cultured in an osteogenic medium supplemented with a certain concentration of two bioceramics' extracts for 20 days. An MTT assay showed that akermanite extract promoted proliferation of hBMSC significantly more than did β-TCP extract. The results of alkaline phosphatase (ALP) activity test and the expression of osteogenic marker genes such as ALP, osteopontin (OPN), osteocalcin (OCN) and bone sialoprotein (BSP) demonstrated that the osteogenic differentiation of hBMSC was enhanced more by akermanite extract than by β-TCP extract. In vivo, a histomorphology analysis and histomorphometry of the two porous bioceramics implants in rabbit femur defect models indicated that both in early- and late-stage implantations, akermanite promoted more osteogenesis and biodegradation than did β-TCP; and in late-stage implantations, the rate of new bone formation was faster in akermanite than in β-TCP. These results suggest that akermanite might be a potential and attractive bioceramic for tissue engineering.
Evaluation of Mechanical and Biological Properties of Akermanite/poly-Ether-etherketone Composite Fabricated by Hightemperature Laser Powder Bed Fusion

Abstract:High-temperature laser bed powder fusion (HT-LPBF) technology is an ideal method for processing poly-ether-ether-ketone (PEEK) implants with personalized bionic structures, but the biological inertia of PEEK limits its medical applications. In this study, we evaluated the mechanical and biological properties of a novel akermanite (AKM)/PEEK composite for HT-LPBF. The results showed that tiny AKM particles are evenly attached to the surface of the PEEK particle. The delayed peak crystallization temperature and stable sintering window ensure the processing feasibility of the AKM/PEEK composites. The tensile strength and Young's modulus are in the range of 30.83-98.73 MPa and 2.27-3.71 GPa, respectively, which can match the properties of cancellous bones and meet their implanting requirement. The CCK-8 experiments demonstrated the biocompatibility of the composites and the good proliferation of bone marrow stromal cells. The dense hydroxyapatite network layer and petal-like hydroxyapatite demonstrates biological activity, indicating that the composite has a good potential in the orthopedics fields.
Preparation, Mechanical Properties and in Vitro Degradability of Wollastonite/tricalcium Phosphate Macroporous Scaffolds from Nanocomposite Powders

Abstract:A new class of scaffolds with a gain size of 200 nm was prepared from wollastonite/tricalcium phosphate (WT) nanocomposite powders (termed “nano-sintered scaffolds”) through a two-step chemical precipitation and porogen burnout techniques. For a comparison, WT scaffolds with a grain size of 2 μm were also fabricated from submicron composite powders (termed “submicron-sintered scaffolds”) under the same condition. The resultant scaffolds showed porosities between 50 ± 1.0% and 65 ± 1.0% with a pore size ranging from 100 μm to 300 μm. The WT nano-sintered scaffolds exhibited compressive strength and elastic modulus values that were about twice that of their submicron-sintered counterparts. The in vitro degradation tests demonstrated that the degradability could be regulated by the grain size of bioceramics. The decreased specific surface area of pores in the nano-sintered scaffolds led to their reduced degradation rate. The mechanical properties of the nano-sintered scaffolds exhibited less strength loss during the degradation process. The WT macroporous nano-sintered scaffolds are a promising and potential candidate for bone reconstruction applications.
Akermanite bioceramics promote osteogenesis, angiogenesis and suppress osteoclastogenesis for osteoporotic bone regeneration

Abstract:AbstractIt is a big challenge for bone healing under osteoporotic pathological condition with impaired angiogenesis, osteogenesis and remodeling. In the present study, the effect of Ca, Mg, Si containing akermanite bioceramics (Ca2MgSi2O7) extract on cell proliferation, osteogenic differentiation and angiogenic factor expression of BMSCs derived from ovariectomized rats (BMSCs-OVX) as well as the expression of osteoclastogenic factors was evaluated. The results showed that akermanite could enhance cell proliferation, ALP activity, expression of Runx2, BMP-2, BSP, OPN, OCN, OPG and angiogenic factors including VEGF and ANG-1. Meanwhile, akermanite could repress expression of osteoclastogenic factors including RANKL and TNF-α. Moreover, akermanite could activate ERK, P38, AKT and STAT3 signaling pathways, while crosstalk among these signaling pathways was evident. More importantly, the effect of akermanite extract on RANKL-induced osteoclastogenesis was evaluated by TRAP staining and real-time PCR assay. The results showed that akermanite could suppress osteoclast formation and expression of TRAP, cathepsin K and NFATc1. Thein vivoexperiments revealed that akermanite bioceramics dramatically stimulated osteogenesis and angiogenesis in an OVX rat critical-sized calvarial defect model. All these results suggest that akermanite bioceramics with the effects of Mg and Si ions on osteogenesis, angiogenesis and osteoclastogenesis are promising biomaterials for osteoporotic bone regeneration.

Abstract:The high porosity and interconnectivity of scaffolds are critical for nutrient transmission in bone tis-sue engineering but usually lead to poor mechanical properties. Herein, a novel method that combines acid etching (AE) with selective laser sintering (SLS) and reaction bonding (RB) of Al particles is pro-posed to realize highly improved porosity, interconnectivity, mechanical strength, and in vitro bioactivity in 3D Al2O3 scaffolds. By controlling the oxidation and etching behaviors of Al particles, a tunable hol-low spherical feature can be obtained, which brings about the distinction in compressive response and fracture path. The prevention of microcrack propagation on the in situ formed hollow spheres results in unique near elastic buckling rather than traditional brittle fracture, allowing an unparalleled compressive strength of 3.72 +/- 0.17 MPa at a high porosity of 87.7% +/- 0.4% and pore interconnectivity of 94.7% +/- 0.4%. Furthermore, scaffolds with an optimized pore structure and superhydrophilic surface show excellent cell proliferation and adhesion properties. Abstract:In recent years, the demand for clinical bone grafting has increased. As a new solution for orthopedic implants, polyether ether ketone (PEEK, crystalline PAEK) has excellent comprehensive performance and has been practically applied in clinic. In this research, a noteworthy elevated scheme to enhance the performance of PEEK scaffolds was presented. The amorphous aggregated poly (aryl ether ketone) (PAEK) resin was prepared as the matrix material, which maintained high mechanical strength and could be processed through solution. So, the tissue engineering scaffolds with multilevel pores could be printed by low-temperature deposited manufacturing (LDM) to improve biologically inert scaffolds with smooth surfaces. Also, the feature of PAEK's solution processing is profitable to uniformly add the functional components for bone repair. Ultimately, A system of orthopedic implantable PAEK material based on intermolecular interactions, surface topology, and surface modification was established. The specific steps include synthesizing PAEK that contain polar carboxyl structures, preparing bioinks and fabricating scaffolds by LDM, preparation of scaffolds with strontium-doped mineralized coatings, evaluation of their osteogenic properties in vitro and in vivo, and investigation on the effect and mechanism of scaffolds in promoting osteogenic differentiation. Tailoring mechanical and in vitro biological properties of calcium‒silicate based bioceramic through iron doping in developing future material

Abstract:Dense iron-doped akermanite ceramics with 0.3, 0.6 and 0.9 mol% of Fe3+ were synthesized via high-speed planetary ball milling and subsequently subjected to sintering at 1200 and 1250 °C. The aim of the current work was to investigate the effect of trivalent iron (Fe3+) in tuning the physicomechanical and in vitro biological properties of akermanite. The incorporation of Fe3+ into akermanite host and sintering at a high temperature of 1200 °C resulted in a synergistic effect in enhancing the sinterability and densification of akermanite ceramics. Although varying the Fe3+ content, it was found that similar densification and mechanical properties (i.e., diametral tensile strength, Vickers microhardness and fracture toughness) were observed for the doped ceramics at 1250 °C, indicating that this newly developed formulation is temperature-dependent. Fe3+-doped akermanite ceramics revealed greater in vitro bioactivity as compared to undoped akermanite, demonstrated by better coverage of needle-like apatite precipitates after 21 days of immersion in simulated body fluid. Additionally, Rat-1 cells cultured in direct contact with Fe3+-doped akermanite ceramics showed almost double levels of cell proliferation than their undoped counterpart on both 3 and 7 days of culture. Our finding suggests that 0.9Fe-AK ceramic is a suitable formulation to be considered for future bone substitute material as it provides sufficient mechanical strength as well as good bioactivity and the ability to encourage cell proliferation.
