Abstract:First Ni(II) ternary complex using the quinolone antibacterial agent enoxacin (HEn) as ligand and 1,10-phenanthroline as co-ligand has been synthesized and characterized. It is a mononuclear structure, in which enoxacin acts as a bidentate ligand bound to the metal through the ketone oxygen and a carboxylate oxygen atom. The complex exhibited good binding propensity to human and bovine serum albumin proteins having relatively high binding constants (6.40x10(4) and 7.12x10(4), respectively). The investigation of the interaction of the complex with calf-thymus (CT) DNA has been performed with UV and circular dichroism (CD) spectroscopies, indicating that they bind to CT DNA probably by the intercalative binding mode. The binding constant (K-b) of the complex with CT DNA calculated with UV is 2.03x10(5), which is higher than that of free enoxacin drug (2.09x10(4)) and even higher than that of typical intercalation indicator (1.23x10(5)) of ethidium bromide (EB). Fluorescence competitive studies with EB have revealed that the complex exhibited the ability to displace the DNA-bound EB using the intercalative binding site. In addition, the antimicrobial activity showed that the complex exhibited a little bit good inhibition (MIC=1.843 (gmL(-1)) against B. subtilis than free HEn.

Abstract:Novel metal complexes of the second-generation quinolone antibacterial agent enrofloxacin with copper(II) and neutral bidentate ligands have been prepared and characterized with elemental analysis reflectance, IR and mass spectroscopy. Complexes have been screened for their in-vitro antibacterial activity against two Gram(+ve) Staphylococcus aureus, Bacillus subtilis, and three Gram((-ve)) Serratia marcescens, Escherichia coli and Pseudomonas aeruginosa organisms using the double dilution technique. The binding of this complex with CT-DNA has been investigated by absorption titration, salt effect and viscosity measurements. Binding constant is ranging from 1.3×10(4)-3.7×10(4). The cleavage ability of complexes has been assessed by gel electrophoresis using pUC19 DNA. The catalytic activity of the copper(II) complexes towards the superoxide anion (O2.-) dismutation was assayed by their ability to inhibit the reduction of nitroblue tetrazolium (NBT).

Abstract:Four erbium(III) complexes with the fluoroquinolones enrofloxacin, levofloxacin, flumequine and sparfloxacin as ligands were synthesized and were characterized by a wide range of physicochemical and spectroscopic techniques as well as single–crystal X–ray crystallography. The compounds were evaluated for their activity against the bacterial strains Staphylococcus aureus, Bacillus subtilis, Escherichia coli and Xanthomonas campestris , which was higher than that of corresponding free quinolones. The interaction mode of the complexes with calf–thymus DNA is via intercalation, as suggested by diverse studies such as UV–vis spectroscopy, DNA–viscosity measurements and competitive studies with ethidium bromide. Fluorescence emission spectroscopy revealed the high affinity of the complexes for bovine and human serum albumin and the determined binding constants suggested a tight and reversible binding of the compounds with both albumins.

Abstract:A series of dinuclear paddle-wheel like transition metal complexes based on 2-phenylquinoline-4-carboxylic derivative L have been synthesized and characterized by IR, elemental analysis, and X-ray diffraction single crystal analysis. The biological activities of L and its complexes were evaluated as assayed antibacterial activities, including Escherichia coli, Pseudomonas aeruginosa, Bacillus subtilis and Staphylococcus aureus. The results indicated that these complexes showed better antibacterial activities than the free ligand or metal salts alone. Among them, the Zn(II) and Cd(II) complexes with IC50 of 0.57 µg/mL and 0.51 µg/mL, respectively, showed excellent antibacterial activity against Staphylococcus aureus.

Abstract:A new binuclear copper (II) complex with 1, 10-phen and fluoroquinolone ligands, [Cu-2(cfc)(2)(phen)(2) (pip)] (NO3)(2)(CO)(2), was synthesized and its crystal structure was solved. The formula of the complex is C28H24ClCuFN5O7, and the crystallography data are as follows, triclinic, space group P (1) over bar, a = 0.80372 (6) nm, b = 0.94712 (7) nm, c = 1.67826(13) nm, alpha = 88.5650(10)degrees, beta = 80.9920(10)degrees, gamma = 70.2780(10)degrees, V = 1.18722(6) nm(3), Z = 2, D = 1.848g . cm(-3), F(000) = 676. R-1 = 0.0627 and omegaR(2) 0.1684 ( I > 2sigma (I)) The copper atom is five-coordinate with square-based pyramidal geometry and involves two nitrogen atoms from phen, two oxygen atoms from cfc and one nitrogen atom from pip anion. The stacking effect between cfc and phen ligands from two neighboring molecules was observed. FT-IR spectrum of the complex was also discussed. Both ligand and complex were assayed against gram-positive and gram-negative bacteria by doubling dilutions method; the complex shows the same minimal inhibitory concentration (MIC) as the corresponding ligand against S. Aureus, E. Coli and M. Lutens bacteria. The inhibitory effect of the ligand and complex on leukemia HL-60 cell line and liver cancer BEL-7402 cell line have been measured by using MTT (Methyl-Thiazol-Tetrozolium) and SRB (Sulphurhodamin B) assay methods, respectively. The results indicated that the complex has strong inhibitory effect on HL-60 and BEL-7402 cell line.

Abstract:Two novel coordination polymer {[Cu(Cip)]·2H2O}n (1) and {[Cu2(Cip)2ClO4(CH3OH)]·0.5H2O}n (2) (HCip=ciprofloxacin, Cip=deprotonated ciprofloxacin) have been synthesized and characterized by elemental analysis, infrared spectra and single-crystal X-ray diffraction. Both complexes 1 and 2 exhibit good binding propensity to human or bovine serum albumin protein (HSA or BSA) with relatively high binding constant values. UV and circular dichroism (CD) spectroscopies studies of the interaction of ciprofloxacin and its complexes with calf-thymus DNA (CT DNA) show that complex 2 exhibits higher binding constant to CT DNA (Kb=3.32×106) and the results indicate they interact with DNA mainly via intercalation. Fluorescence competitive studies with ethidium bromide (EB) reveal that the complexes could compete with EB and displace it to bind to DNA using the intercalative binding site.

Abstract:Enrofloxacin (EFX) is a third-generation fluoroquinolone drug especially used for animals world wide, which shows broad-spectrum and significant antibacterial activities. In this study, EFX was selected as the ligand to synthesize two transition metal complexes, EFX-Mn and EFX-Co, in which the bio-active metal ions, manganese(II) and cobalt(II), was set to be the active metal center. Their crystal structures were reported and characterized by X-ray single crystal diffraction analysis. Both the crystal structures of EFX-Mn and EFX-Co belong to the same monoclinic crystal system with P2 1 /c space group. Viewed from the crystal structure of EFX-Mn or EFX-Co, the central Mn(II)/Co(II) adapted a six-coordinated octahedral configuration, respectively by two EFX ligands and two H 2 O co-ligands. The characteristic UV-Vis absorption peaks of both complexes were examined in tris-saline buffer solution in a time-dependent mode in 24 h, which suggested that the coordination status of EFX-Mn and EFX-Co in the solid state still maintained under near-physiological condition. In vitro antibacterial activity tests demonstrated that EFX-Mn and EFX-Co showed similar and significant growth inhibitory effects towards five typical pathogenic bacteria, compared with EFX itself. While for two gram-negative bacterial strains, E. coli and S. typhi , EFX-Mn and EFX-Co showed higher bactericidal effects than EFX, the bactericidal concentration of which was only a half as low as that of EFX.

Abstract:Three new Co(II) complexes, [Co4(L)2(μ3-CrO4)2](ClO4)2·2CH3CN (1), [Co2(L)(μ2-na)(H2O)](ClO4)2 (2) and [Co2(L)(μ2-ba)](ClO4)2·0.5CH3CN (3) (Hna=nicotinic acid, Hba=benzoic acid, HL=N,N,N',N'-tetrakis (2-quinolylmethyl)-1,3-diaminopropan-2-ol), have been synthesized and characterized by various physicochemical techniques. The Co(II) centers are connected by endogenous alkoxy bridge from L(-) and various extrinsic auxiliary linkers, some of which display coordination number asymmetry (5, 6-coordinated for 1 and 2; 5, 5-coordinated for 3). It is worth mentioning that complex 1 contains two rare reported μ3-η(1), η(1), η(1)-CrO4(2-) moieties. Susceptibility data of three complexes indicated intramolecular antiferromagnetic coupling of high-spin Co(II) atoms with exchange integral values (J) -14.94 cm(-1), -11.26 cm(-1) and -13.66 cm(-1) for 1, 2 and 3, respectively. Interaction of compounds with calf thymus DNA (CT-DNA) have been investigated by absorption spectral titration, ethidium bromide (EB) displacement assay and viscosity measurement, which revealed that compounds bound to CT-DNA with a moderate intercalative mode, accompanied the affinities order: 1>2≈3. Three complexes exhibit oxidative cleavage of pBR322 plasmid DNA including a reliance on H2O2 as the activator. Compound 1 demonstrates an increased DNA cleavage activity as compared with 2 and 3, which could degrade super coiled DNA (SC DNA) into nicked coiled DNA (NC DNA) in lower concentration (5 μM). Moreover, all compounds could quench the intrinsic fluorescence of bovine serum albumin (BSA) in a static quenching process. Complex 1 also shows higher anticancer activity than cisplatin with lower IC50 value of incubation for both 24 h and 48 h.

Abstract:New Cu(II) and Ni(II) complexes of acetyl coumarin derivatives with gatifloxacin were synthesized. Characterization of the complexes was carried out by elemental analysis, FT-IR, FAB-MS, electronic spectra and magnetic measurement along with thermo gravimetric studies. Thermal study reveals the absence of coordinated water molecule in case of Cu(II) complexes. All the Cu(II) complexes were found more potent antibacterial activity compared to Ni(II) complexes. The antituberculosis data have shown that copper conjugation may be advantageous in designing highly effective drugs for antitubercular therapy. These complexes were also found to be potent antioxidizing agents comprising radical scavenging capability and ferric reducing abilities.

Abstract:Two new mononuclear complexes, [Ni(cepha)(2)]center dot 6H(2)O (1) and [Zn(cepha)(2)]center dot 6H(2)O (2) (cepha=cephalosporoate) have been synthesized and characterized by elemental analysis, infrared spectroscopy, thermogravimetric analysis and X-ray powder diffraction analysis. The structures of complexes have also been determined by X-ray single crystal diffraction analysis, and the results show that the complexes 1 and 2 are both in orthorhombic system with space group P22(1)2(1). The cephalexin hydrolyzes during the coordination process to produce cephalosporin intermediate, cephalosporoate (cepha), which participates in coordination. Metal ion acts as six-coordinated center forming a distorted octahedral geometry, and the complexes are further extended to a three-dimensional network via intermolecular hydrogen bonds. The antitumor activity in vitro of cephalexin, metal acetate and complexes were carried out by MTT assay. The results reveal that complexes 1 and 2 have a certain inhibiting activity against MCF-7 and HepG-2 cell lines. CCDC: 1497051, 1; 1497052, 2.

Abstract:The influence of copper(II) ions on the binding of Pazufloxacin (PFLX), which is a forth generation quinolone antimicrobial agent, to calf thymus DNA (CTDNA) has been investigated by fluorescence, absorption spectroscopy and viscosity measurement. The results show that PFLX and Cu2+ can form binary complex with mole ratio 2:1, PFLX and CTDNA can form complex mediated by Cu2+. A ternary system of PFLX–Cu2+–DNA were proposed. The experimental results show that the PFLX binds to CTDNA by two kinds of binding modes, intercalative mode and groove binding mode.

Abstract:“Drug repositioning” is a current trend which proved useful in the search for new applications for existing, failed, no longer in use or abandoned drugs, particularly when addressing issues such as bacterial or cancer cells resistance to current therapeutic approaches. In this context, six new complexes of the first-generation quinolone oxolinic acid with rare-earth metal cations (Y3+, La3+, Sm3+, Eu3+, Gd3+, Tb3+) have been synthesized and characterized. The experimental data suggest that the quinolone acts as a bidentate ligand, binding to the metal ion via the keto and carboxylate oxygen atoms; these findings are supported by DFT (density functional theory) calculations for the Sm3+ complex. The cytotoxic activity of the complexes, as well as the ligand, has been studied on MDA-MB 231 (human breast adenocarcinoma), LoVo (human colon adenocarcinoma) and HUVEC (normal human umbilical vein endothelial cells) cell lines. UV-Vis spectroscopy and competitive binding studies show that the complexes display binding affinities (Kb) towards double stranded DNA in the range of 9.33 × 104 − 10.72 × 105. Major and minor groove-binding most likely play a significant role in the interactions of the complexes with DNA. Moreover, the complexes bind human serum albumin more avidly than apo-transferrin.

Metal Complexes of C-Functionalized Macrocyclic Dioxotetraamines Bearing 8-Hydroxyquinoline: Stability in Aqueous Solutions

Abstract:Two novel 3-hydroxyquinoline connected dioxotetraamines have been designed and synthesized, and have been characterized by elemental analysis, IR, mass spectra and H-1 NMR. The two ligands L-1 and L-2 have two chelating groups. Each can react with a transition metal ion forming complexes. Potentiometric titrations have been performed in 0.1 mol/L NaNO3 at 25 degreesC giving the 1:1 stability constants. Coupled with UV spectroscopy the affinity of 8-hydroxyquinoline and dioxotetraamines to transition metal ions were compared and the possible structure of the metal complex species in solution was discussed. The results show that as to Mn2+, Zn2+ and Co2+, 8-hydroxyquinoline is a stronger chelating reagent than dioxo[13] or [14]tetraamine macrocycles and at 1:1 molar ratio (M:L), the former binds in 100% to the metal ions, while the dioxotetraamines can be only partially or not coordinated.

Abstract:Two new complexes, [CuLCl]ClO4 (1) and [Zn2L2SO4(H2O)2](ClO4)2 (2) [L=N,N-bis(quinolin-2-ylmethyl)quinolin-8-amine], have been synthesized and structurally characterized. The interactions of two complexes with CT-DNA have been investigated by UV absorption, fluorescence spectroscopy, viscosity measurements and gel electrophoresis under physiological conditions. Results show that the complexes bind to CT-DNA with a moderate intercalative mode and exhibit efficient DNA cleavage activity on UV-A light of 365 nm. Furthermore, two complexes could quench the intrinsic fluorescence of BSA in a static quenching process based on BSA binding experiments. Notably, in vitro cytotoxicity study of two complexes on four human tumor cells lines (7404, HeLa, MCF-7, and HepG-2) indicate that both of them have the potential to act as effective anticancer drugs with low IC50 values.

Abstract:The synthesis, physicochemical characterizations, and biological investigations on the chelation of the antibiotic medication enoxacin (EXN) with certain vital metal ions, such as platinum(IV), ruthenium(III), and iridium(III) are discussed in this study. These compounds structures were interpreted based on elemental analysis and spectral measurements (FTIR, 1H-NMR, UV-Vis electronic, and XRD). The findings show that there is same coordination pathway for the chelation behavior of enoxacin complexes: a monodentate manner. The Pt(IV), Ru(III), and Ir(III) complexes are coordinated through the N atom of the piperazine ring, which is an unusual mode of coordination for this class of compounds. The dissolved complexes in DMSO were found to have non-electrolyte nature based on their molar conductance measurements. The formula for these complexes is [Pt(EXN)3(H2O)2Cl] (1), [Ru(EXN)3(H2O)3] (2), and [Ir(EXN)3(H2O)3] (3). Using scanning electron microscope (SEM) analysis and X-ray powder diffraction (XRD), the nano-scale range of the Pt(IV), Ru(III), and Ir(III) complexes has been determined. Results of in vitro cytotoxicity were examined using MCF-7 cells (human breast cancer cell line) and HepG-2 cells (human hepatocellular carcinoma) that had been grown. The cytotoxic activity of the metal complexes demonstrated a cytotoxic effect against the growth of human breast and liver cancer cell lines. Abstract:Coordination compounds of Co(II) , Ni(II), Cu(II) and Zn(II) of the type [M(L1-2)2(H2O)2] were obtained from Schiff base ligands [HL1-2] by the condensation reaction of 5-bromosalicyldehyde/5-chlorosalicyldehyde with 4-amino quinoline using methanol as a solvent in (1:1) molar ratio. The metal complexes were synthesized by reacting the aqueous methanolic solution of metal acetates M(CH3COO)2xH2O with the hot methanolic solution of their respective Schiff base ligands in (1:2) molar ratio. Various physicochemical techniques i.e. elemental analysis, molar conductance measurements, magnetic moment studies and different spectral studies (1H NMR, 13C NMR, UV–Vis, FT-IR, ESR) and mass spectrometry were used for characterization of compounds. The spectral data confirmed the bidentate nature (NO) of Schiff base ligands coordinating through nitrogen atom of azomethine group and oxygen atom of hydroxyl group resulting in the formation of octahedral geometry of all metal complexes. The Schiff base ligands and their transition metal complexes were screened against two gram-positive bacterial species i.e. (S. aureus, B. subtilis), two-gram negative bacterial species i.e. (E. coli, P. aeruginosa) and two fungal species (A. niger and C. albicans) using serial dilution method with ciprofloxacin and fluconazole as reference drug. The results obtained from antimicrobial studies showed that metal complexes are more active than their respective Schiff base ligands. Complex 5 and 6 are potent antimicrobial agent.

Abstract:Two Schiff base complexes [Cu(L)(2)] (1) and [Zn(L)(2)]center dot 3H(2)O (2) of (E)-4-fluoro-2-((3-morpholino propylimino)methyl)phenol (HL) have been synthesized and characterized by elemental analysis and single-crystal X-ray diffraction. Crystal data for 1: triclinic, space group P (1) over bar with a = 4.8000(10), b = 11.109(2), c = 13.921(3) angstrom, alpha = 66.55(3), beta = 80.35(3), gamma = 81.09(3)degrees, V = 668.2(2) angstrom(3), Z = 1, D-c = 1.477 g/cm(3), F(000) = 311, and the final R = 0.0744 and wR = 0.1601 for 2301 observed reflections with I > 2 sigma(I). And those for 2: monoclinic, space group P2(1)/c with a = 14.789(3), b = 11.713(2), c = 18.846(4) angstrom, beta = 107.85(3)degrees, V = 3107.4(11) angstrom(3), Z = 4, D-c = 1.389 g/cm(3), F(000) = 1368, and the final R = 0.0749 and wR = 0.1348 for 5423 observed reflections with I > 2 sigma(I). Both 1 and 2 are four-coordinated complexes, with a square-planar geometry of copper(II) in 1 and a slightly distorted tetrahedral geometry of zinc(II) in 2. An asymmetric unit consists of only one half of a Cu ion and one L ligand in 1, while an asymmetric unit is composed of one Zn ion, two L ligands and three water molecules in 2. Non-classical C-H center dot center dot center dot O and C-H center dot center dot center dot F hydrogen bonds play important roles in the formation of a 1D chain of 1, but O-H center dot center dot center dot O and O-H center dot center dot center dot N hydrogen bonds play dominant roles in the self-assembly of a 3D network of 2. The antibacterial activities of 1 and 2 against B. subtilis, S. aureus, S. faecalis, P. aeruginosa, E. coli and E. cloacae have been evaluated by MTT method.

Abstract:The interactions of cobalt sulphate and nickel acetate with ofloxacin (ofo, I), a 4-quinoline derivative, were studied. The hydrothermal technique was adopted in this work. The isolated solid complexes were characterized by elemental analysis, infrared spectra, electronic spectra, fluorescence spectra and thermal analysis. The results support the formation of complexes of the formula Co(ofo)(2) center dot 4H(2)O (II) and Ni(ofo)(2) center dot 4H(2)O (III). The infrared spectra of the isolated solid complexes suggested that ofo acts as bidentate ligands through one of the oxygen atoms of the carboxylic g roup and the ring carbonyl oxygen atom. Thermogravimetric (TG) and its differential (DTG) were carried out for the complexes. The data obtained indicated that the thermal decomposition of the two complexes in inert atmosphere proceeded approximately with two main degradation steps. I showed a intense fluorescence in solid state at lambda(ex)/lambda(em) = 365 nm/461 rim, and two complexes displayed weakly similar emission maximum at 470 nm in the powder samples at ambient temperature, while the emission of II and III may be mainly originated from the intraligand excited states of I. I, II and III were assayed against two kinds of gram-positive and two kinds of gram-negative bacteria by in vitro doubling dilutions method. The results indicated that II and III have the similar minimal inhibitory concentration (MIC) as the I against S. Aureus, M. Lutens, E. Coli and P. Aeruginosa. The inhibitory effect of I, II and III on leukemia HL-60 cell line has been measured by using MTT (Methyl-Thiazol-Tetrozolium) method and that on liver cancer BEL-7402 cell line measured by SRB (Sulphurhodamin B) method. The results indicated that the complexes in the high concentration have inhibitory effect on HL-60 cell line.
