Abstract:This paper reviews the recent achievements in the enhanced production of volatile fatty acids (VFAs) from waste activated sludge (WAS). The enhanced strategies are divided into two approaches. The first strategy focuses on the regulation of carbon-to-nitrogen (C/N) ratio by co-digestion of WAS with carbon-rich substrates, including municipal solid wastes (MSW), marine algae, agricultural residues, and animal manures. The other strategy is to enhance the solubilization and hydrolysis of WAS or inhibit the methanogenesis by applying various pretreatments, such as mechanical, chemical, enzymatic, and thermal pretreatment. Finally, the applications of WAS-derived VFAs are discussed. The future researches in enhancing VFAs production and wide application of the VFAs from both technical and economic perspectives are proposed.

Abstract:Abstract Due to the problems of the hydrolysis of wasted activated sludge (WAS), fermentation stage takes a long time and volatile fatty acids (VFAs) output efficiency is low. This study investigated the effect of ultrasound combined with Fenton (US/Fenton) pretreatment on the sludge disintegration and anaerobic fermentation. The sludge was treated by three different pretreatments respectively: ultrasound alone, Fenton alone and US/Fenton. Results show that after three pretreatments, sludge floc structure was disintegrated and oxidized, the sludge particle morphology after US/Fenton pretreatment was changed the most obviously. After US/Fenton, specific surface area was the largest and the particle size reduction was remarkable from 50.24 μm to22.44 μm. The SCOD concentration after US/Fenton was 3461.72 mg COD L-1, which was raised 1.73 times, 2.68 times and13.29 times as against that after ultrasound, Fenton and the untreated. The degree of disintegration (DDCOD) of the US/Fenton pretreatment was 15.4% which was greater than ultrasound pretreatment of 9.97% and Fenton pretreatment of 3.18%. The VFAs concentration after US/Fenton pretreatment reached a maximum value of 4594 mg COD L-1 at the eighth day, it was raised 31.8% and 70.15% as separately compared with the Fenton and ultrasound pretreatments. The pretreatments changed the VFAs composition and proportion. The highest HAC concentration after US/Fenton was 2039 mg COD L-1, which was 1.72 and 1.78 times of that in Fenton and ultrasound, respectively. This study showed that the US/Fenton pretreatment had a certain synergistic promotion effect on the sludge anaerobic fermentation. The ultrasound and Fenton co-work, the ability of the VFAs production is further improved.
Continuous volatile fatty acid production from waste activated sludge hydrolyzed at pH 12

Abstract:This study adopted rapid alkaline treatment at pH 12 to hydrolyze 66% of total chemical oxygen demands. Then the hydrolyzed liquor was fermented in a continuous-flow stirred reactor to produce volatile fatty acids (VFAs) at 8-h hydraulic retention time and at 35 °C. The maximum VFA productivity reached 365 mg VFAs g(-1) volatile suspended solids in a 45-d operation, with most produced VFAs being acetate and propionate, principally produced by protein degradation. The Bacteroidia, ε-proteobacteria and the Clostridia were identified to be the classes correlating with the fermentation processes. The fermented liquor was applied to denitrifying phosphorus removal process as alternative carbon source after excess phosphorus and nitrogen being recycled via struvite precipitation. Fermented liquors from alkaline hydrolysis-acid fermentation on waste activated sludge are a potential renewable resource for applications that need organic carbons.
Role of pretreatment type and microbial mechanisms on enhancing volatile fatty acids production during anaerobic fermentation of refinery waste activated sludge

Abstract:This study compared the effects of alkaline, thermal, thermal-peroxymonosulfate (PMS), and alkyl polyglucose (APG) pretreatments on volatile fatty acids (VFAs) production from refinery waste activated sludge (RWAS), including VFAs yield, composition, organics components, microbial communities, and the potential improvement of mechanisms. All pretreatments effectively enhanced the bioconversion of RWAS and consequently promoted the hydrolysis process, which inhibited the methanogenesis process. However, the release of lignin/carboxyl-rich alicyclic molecules (CRAM)-like compounds and tannin substances in Thermal-PMS and APG groups significantly influenced the acidogenesis and acetogenesis processes. Among all pretreatments, alkaline pretreatment showed the highest VFAs yield of 95.06 mg/g volatile solids (VS) and VS removal of 17%. This result could be associated with the enrichment of functional hydrolytic-acidification bacteria, such as Planococcus and Soehngenia, and increased metabolism of amino acids, carbohydrates, and nucleotides. By considering an economical and efficient perspective, this study recommended the alkaline pretreatment for the anaerobic fermentation of RWAS.

Abstract:In this paper, the synthesis of polyhydroxyalkanoates (PHAs) by activated sludge with aerobic dynamic feeding process was conducted in a sequencing batch reactor by using food wastes and excess sludge fermentation liquid as the carbon source. The volatile fatty acids (VFAs) in the fermentation liquid were divided into even-numbered (acetic and butyric acid) and odd-numbered (propionic and valeric acid). The experiments conducted by central-composite design (CCD) showed that the pH could significantly affect the ratio of even-numbered to odd-numbered VFAs. Statistical analysis indicated a positive correlation (R2=0.97, P<0.05) between the consumption of even-numbered VFAs and the synthesized of PHB, while the consumption of odd-numbered VFAs were correlated with the synthesized PHV. By controlling the ratio of even-numbered to odd-numbered VFAs, the contents of PHV in the PHAs could be controlled within the range of 22–30%. When fermentative VFAs were used as the substrate for the synthesis of PHAs, the microbial synthesis of PHA and biomass was higher than that mixture of analytically pure acids was used. These results are of vital significance for the comprehensive utilization of solid wastes.
Enhanced hydrolysis of waste activated sludge and recovery of volatile fatty acids: Performance and mechanistic analysis of synergistic treatment with sodium citrate and calcium oxide

Abstract:Anaerobic fermentation has emerged as a promising method of transforming waste activated sludge into high-value products (e.g., volatile fatty acids (VFAs)). This work developed sodium citrate (SC)-calcium oxide (CaO) pretreatment to accelerate the production of VFAs by enhancing sludge solubilization and disintegration of extracellular polymeric substances. The results showed that co-pretreatment with 0.25 g/g TSS of SC and 0.05 g/g TSS of CaO effectively boosted VFAs accumulation (5823.3 mg COD/L), which was 12.2 times higher than the Control group. SC-CaO pretreatment enhanced hydrolysis and acidogenesis by providing ample organic substrates, thereby promoting the growth of hydrolytic and acidogenic bacteria. Additionally, the fermentation broth resulting from co-pretreatment exhibited lower phosphorus concentration and higher biodegradability. Economic analysis confirmed that the combined pretreatment is cost-effective. This work provides a viable strategy for enhancing high-value product recovery from sludge.
Upgrading VFAs bioproduction from waste activated sludge via co-fermentation with soy sauce residue

Abstract:Conditioning of extra carbon sources has been widely reported to facilitate fermentation of waste activated sludge (WAS). Soy sauce residue (SSR) was a relatively untapped carbon source for sludge conditioning. This batch study aimed to evaluate the possible implementation of SSR for volatile fatty acids (VFAs) production from WAS. To upgrade the bioavailability of feedstock, three typical pretreatment methods were conducted, i.e., ammonium hydroxide (AH), sulfuric acids (SA) and thermal assisted alkaline (TA). AH pretreated test (AH-PT) outperformed due to a relatively strong structure decomposition of cellulosic materials as revealed by infrared spectroscopic analysis and crystal index. As a result, performed a high hydrolysis rate of 4449 mg COD/d, 1.12-1.23-fold higher than that in TA and SA pretreated tests (TA-PTand SA-PT), and 7.8-fold higher than that in the Control test. Meanwhile, a volatile fatty acids (VFAs) contribution of 401.2 mg COD/g SSR∙L and a maximum acidification rate of 3.59 d–1 was recorded, with a high sum proportion of mall molecular acetic and propionic 82.2%, 11%–70% increase over the other three tests. Besides, speciation process characterized with functional genus differentiation was identified by microbial diversity and distribution investigation and canonical correspondence analysis (CCA). Finally, a potential market value of 0.49–0.65 Billion €/year was preliminary estimated, showing promise of resource recovery from both WAS and SSR instead of extensive disposal.

Abstract:The study evaluated influences of sludge concentration, temperature and solids retention time (SRT) for the hydrolysis of waste activated sludge (WAS) in anaerobic digesters. The results indicated that volatile fatty acids (VFA) production increased when the concentration of mixed liquor volatile suspended solids (MLVSS) was higher. When SRT was 48 h, VFA concentration increased 8.43 times from 4.57 to 23.78 gMLVSS/L. VFA generation was significantly affected with different temperature and SRT. When the temperature changed from 40 to 50°C, it induced 1.65-fold increase in VFA yield. The optimal SRT was 48 h. As VFA concentration decreased only 1.31 times compared with maximum VFA production at SRT of 120 h. Iso-valeric acid, acetic acid and n-butyric acid were the dominant VFA produced and would improve ployhydroxyvalerate proportion in polymer. The feasibility of nitrogen and phosphorus recovery and the risk of metal ion released depended on the nature of WAS.

Abstract:The combined fermentation of sludge from the secondary sedimentation tank of municipal wastewater treatment plants and food waste enables not only waste reduction, but also the acquisition of volatile fatty acids (VFAs) for the biosynthesis of polyhydroxyalkanoates (PHAs). To better understand variables influencing the production of VFAs and PHAs, this study considered anaerobic fermentation of VFAs under different pH conditions. The production of VFA was the highest at an initial pH of 9.0, reaching 25,934 ± 1,485 mg COD/L and a VFAs/S COD ratio of 0.61 ± 0.04. When the fermentation liquid was used as a carbon source for PHA biosynthesis, continuous pulsed feeding resulted in the highest PHA synthesis rate of 64.5 ± 1.8%, while the culture receiving a one-time feeding had the lowest rate of only 51.5 ± 2.0%.

Abstract:Anaerobic fermentation is widely installed to recovery valuable resources and energy as CH4 from waste activated sludge (WAS), and its implementation in developing countries is largely restricted by the slow hydrolysis, poor efficiency, and complicate inert components therein. In this study, enzyme-enhanced fermentation was conducted to improve sludge solubilization from 283 to 7728 mg COD/L and to enhance volatile fatty acids (VFAs) yield by 58.6 % as compared to the conventional fermentation. The rapid release of organic carbon species, especially for tryptophan- and tyrosine-like compounds, to outer layer of extracellular polymeric substance (EPS) occurred to reduce the structural complexity and improve the sludge biodegradability towards VFAs production. Besides, upon enzymatic pretreatment the simultaneous exposure of hydrophilic and hydrophobic groups on sludge surfaces increased the interfacial hydrophilicity. By quantitative analysis via interfacial thermodynamics and XDLVO theory, it was confirmed that the stronger hydrophilic repulsion and energy barriers in particle interface enhanced interfacial mass transfer and reactions involved in acidogenic fermentation. Meanwhile, these effects stimulate the fermentation functional microflora and predominant microorganism, and the enrichment of the hydrolytic and acid-producing bacteria in metaphase and the proliferation of acetogenic bacteria, e.g., Rubrivivax (+9.4 %), in anaphase also benefits VFAs formation. This study is practically valuable to recovery valuable VFAs as carbon sources and platform chemicals from WAS and agriculture wastes.

Abstract:In this study the combination pretreatment of sodium citrate (SC) and heating was investigated to enhance the volatile fatty acid (VFA) production during anaerobic fermentation of waste activated sludge (WAS). The results showed that the combined pretreatment (0.3 g-SC/g-TSS + at 121 °C for 30 min) resulted to the maximum VFA yield of 354.5 mg-COD/g-VS, which was 16.4, 6.0 and 2.0-fold of that of control, with sole SC and sole heat pretreatment, respectively. Meanwhile, the combined pretreatment greatly improved the proportion of acetate and butyrate, while the production of propionate significantly decreased. Mechanism analysis indicated that the combined pretreatment effectively promoted WAS disintegration and solubilization. Moreover, the heat pretreatment exhibited superior performances to the SC pretreatment on release of organic matters and enhancement of VFA production in the combined pretreatment. The microbial analysis revealed that with the combined pretreatment the microbial community shifted toward hydrolysis-acidification, such as Enterobacter and Macellibacteroides.

Abstract:Central-composite design (CCD) and response surface methodology (RSM) were used to optimize the parameters of volatile fatty acid (VFA) production from food wastes and dewatered excess sludge in a semi-continuous process. The effects of four variables (food wastes composition in the co-substrate of food wastes and excess sludge, hydraulic retention time (HRT), organic loading rate (OLR), and pH) on acidogenesis were evaluated individually and interactively. The optimum condition derived via RSM was food wastes composition, 88.03%; HRT, 8.92days; OLR, 8.31gVSS/ld; and pH 6.99. The experimental VFA concentration was 29,099mg/l under this optimum condition, which was well in agreement with the predicted value of 28,000mg/l.

Abstract:Volatile fatty acids (VFAs) production from anaerobic digestion of waste activated sludge (WAS) is often limited by the slow hydrolysis and/or poor substrate availability. Increased attention has been given to enhance the hydrolysis and acidification of WAS recently. This study presented an efficient and green strategy based on the combined use of nitrite pretreatment and alkaline pH to stimulate hydrolysis and VFA accumulation from WAS. Results showed that both proteins and polysaccharides increased in the presence of nitrite, indicating the enhancement of sludge solubilization and hydrolysis processes. Mechanism investigations showed that nitrite pretreatment could disintegrate the sludge particle and disperse extracellular polymeric substances (EPS). Then, anaerobic digestion tests demonstrated VFA production increased with nitrite treatment. The maximal VFA accumulation was achieved with 0.1 g N/L nitrite dosage and pH 10.0 at a sludge retention time (SRT) of 7 days, which was much higher VFA production in comparison with the blank, sole nitrite pretreatment, or sole pH 10. The potential analysis suggested that the combined nitrite pretreatment and alkaline pH is capable of enhancing WAS digestion with a great benefit for biological nutrient removal (BNR).

Effect of composition of volatile fatty acids on yield of polyhydroxyalkanoates and mechanisms of bioconversion from activated sludge

Abstract:Polyhydroxyalkanoates (PHA) is green biodegradable natural polymer. Here PHA production from volatile fatty acids (VFAs) was investigated in sequential batch reactors inoculated with activated sludge. Single or mixed VFAs ranging from acetate to valerate were evaluated, and the dominant VFA concentration was 2 times of that of the others in the tests. Results showed that mixed substrates achieved about 1.6 times higher yield of PHA production than single substrate. The butyrate-dominated substrates maximized PHA content at 72.08% of VSS, and the valerate-dominated substrates were followed with PHA content at 61.57%. Metabolic flux analysis showed the presence of valerate in the substrates caused a more robust PHA production. There was at least 20% of 3-hydroxyvalerate in the polymer. Hydrogenophaga and Comamonas were the main PHA producers. As VFAs could be produced in anaerobic digestion of organic wastes, the methods and data here could be referred for efficient green bioconversion of PHA.

Abstract:The utilization of waste activated sludge (WAS) to recover energy in the form of medium-chain fatty acids (MCFAs) is increasingly attractive due to the high energy density of products. However, the anaerobic fermentation process of WAS would be restricted by the poor degradability of WAS. Herein, we propose an effective approach to improving MCFA production from anaerobic fermentation of WAS using free ammonia (FA) pretreatment. The maximum MCFA production was achieved at 8.3 g of COD/L when the pretreated FA concentration was 255 mg/L, increased by 4.8-fold compared with the control (1.4 g COD/L). In addition, FA pretreatment shortened the lag phase of MCFAs production and promoted the WAS degradation compared to the control without FA pretreatment. Mechanism analysis showed that FA improved the solubilization process, which should be attributed to FA pretreatment altering the WAS properties, that is, smaller particle size and lower hydrophobicity. Moreover, FA-pretreated sludge as feedstock also promoted hydrolysis, acidification, and chain elongation processes for MCFA production, indicating that the activity of related microorganisms has been improved, which was also strongly confirmed by the microbial analysis results. On the basis of these results, a "circle economy" hypothesis that combines FA pretreatment strategy with wastewater treatment plants (WWTPs) by using easily recovered FA from anaerobic fermentation liquor was finally proposed with potential environmental and economic benefits.

Abstract:Thermal hydrolysis has been widely applied to improve organics bioconversion during sludge anaerobic treatment currently, based on which, liquid fermentation to produce volatile fatty acids (VFAs) with high concentration and good purity has been successfully developed by only using hydrolysate as the substrate to avoid the interference of "useless" residual solids. Therefore, obtaining high-quality hydrolysate is the prerequisite for VFAs production via liquid fermentation. However, previous studies on sludge thermal hydrolysis either only focused on organics release or only on sludge dewatering. Actually, the quantity, quality and recovery of the carbon released from sludge are equally important, and a balance between them should be established. Results in the present study indicated that organics concentration in sludge hydrolysate could not be arbitrarily enhanced by increasing thermal hydrolysis intensity or sludge concentration, and interestingly there seemed a threshold of around 32 g/L that the highest concentration the sludge hydrolysate could reach. Moreover, with the increase of hydrolysis intensity, the bioavailability of sludge could be promoted but reached the maximum with BOD 5 /COD of around 0.44 at 180 °C, while sludge dewaterability could be improved but also trended to stable after 160 °C. The findings of this study demonstrate that excessively high hydrolysis intensity would not only waste energy but also induce forms of non-biodegradable organics. The performances of sludge liquid fermentation, including VFAs production and sludge reduce, were closely related to hydrolysis intensity, the choice of which should be based on the balance of the quantity, quality and recovery of the released carbon.

Abstract:The application of chemical surfactants in the production of volatile fatty acids (VFAs) during sludge anaerobic fermentation has drawn wide attentions. Compared with chemosynthetic surfactants, bio-surfactants are readily biodegradable and thus have a low negative impact on the environment. This paper studied the effects of three types of bio-surfactants (surfactin, rhamnolipid and saponin) on VFA production during sludge anaerobic fermentation. The release of nitrogen and phosphorus was also observed. Dissolution of organic matters, fluorescence spectra of filtrates, key enzyme activities, surface activity and biodegradation of the bio-surfactants were analyzed to reveal the mechanisms. The results indicated that WA production was increased to approximately 4-fold versus the blank (75 mg COD/g VSS) using surfactin, rhamnolipid and saponin at the dosage of 0.05, 0.05, 0.10 g/g DS, respectively. However, some differences existed in their mechanisms of improved acid production. Surfactin mainly increased the dissolution of organic matters to reach a high VFA accumulation. Rhamnolipid also enhanced the dissolution but inhibited the methanogenesis. In addition, its effects on microbial activities limited the further improvement of VFA production. Despite a lower surface activity than the other two bio-surfactants, saponin showed a similar improvement of VFA production through its self-degradation. (C) 2014 Elsevier B.V. All rights reserved.

Abstract:Crawfish shell wastes (CSW) are abundant with valuable substrates, which possess great resource potentials. This work has revealed the feasibility of CSW reutilization for effective volatile fatty acids (VFAs) generation with waste active sludge (WAS) via anaerobic fermentation. The maximum VFAs generation reached 17963 mg COD/L when the CSW/WAS mass ratio was 3.25:1, while it was 784 mg COD/L in the control reactor. Meanwhile, each gram of CSW could be transformed into 264 mg COD equivalent VFAs. The feed of CSW brought in massive organic substrates, which significantly promoted the solubilization, hydrolysis and acidification processes in the CSW/WAS co-fermentation systems simultaneously. The metabolism of carbohydrates took priority over proteins in contributing to the VFAs promotion. More importantly, the functional anaerobes (i.e., Paludicola sp. and Macellibacteroides sp.), which participated in the main substrates decomposition with the VFAs as the primary metabolites, were evidently enriched in the presence of CSW. The critical hydrolases and acid-forming enzymes relevant to VFAs biosynthesis were all enhanced. Moreover, the key genes encoding for the main substrates metabolisms (i.e., chitin) and VFAs production were remarkably up-regulated. The evident promotion of microbial activities and metabolic functions was mainly ascribed to the favorable fermentation conditions (i.e., pH, ORP and C/N) induced by CSW conditioning. This study proposed an innovative strategy to recover high-value products from CSW, and in-depth insights of microbial traits and genetic functions in CSW/WAS co-fermentation systems.

Abstract:Anaerobic acidogenesis of waste activated sludge (WAS) presents significant potential for resource recovery and waste treatment. However, the slow hydrolysis of WAS limits the efficiency of this approach. In this study, we applied urea hydrogen peroxide (UHP) pretreatment to enhance WAS hydrolysis and investigated the effects of operating parameters on volatile fatty acid (VFA) production and the associated mechanisms. Results demonstrated that UHP significantly improved WAS hydrolysis and VFA production, with a three-fold increase in soluble chemical oxygen demand (SCOD) compared to the control group. UHP dosage emerged as the most critical factor for VFA production, with the maximum VFA concentration increasing from 1127.6 to 8800.9 mg COD per L as UHP dosage ranged from 0 to 6 mmol g −1 VSS (Volatile suspended solids). At an optimal UHP dosage of 4 mmol g −1 VSS, both the unit oxidant promotion efficiency (ΔVFAs/ΔUHP) and the maximum VFA concentration reached relatively high levels, at 35.3 mg COD per mmol and 7527.3 mg COD per L, respectively. UHP pretreatment generated alkaline conditions, H 2 O 2 , ·OH and free ammonia, which collectively disrupted the extracellular polymeric substances (EPS) structure, transforming unextractable EPS into extractable forms and promoting the release of organic matter during both the pretreatment and fermentation stages. Excitation-emission matrix (EEM) analysis revealed that UHP increased the concentration of easily utilizable organic matter, providing more substrates for acidogenic bacteria and enhancing VFA production. Furthermore, weak alkaline conditions and high free ammonia concentrations in the UHP group facilitated VFA accumulation by preventing rapid acidification and suppressing methanogen activity. This study offers valuable insights into the potential of UHP pretreatment for enhancing WAS hydrolysis and VFA production, with promising applications in wastewater treatment and resource recovery.
