Synthesis and Evaluation of 2 Beta-(N-Halopyridylaminomethyl)-3 Beta-(4-Chlorophenyl)Tropanes As Potential Pet Ligands for Dopamine Transporter

Abstract:299 Objectives Changes in dopamine transporters (DAT) have been observed in several central nervous system disorders, such as Parkinson’s disease. PET imaging of DAT density in the brain is a potentially valuable tool for studying the etiopathology of degenerative brain disorders. The goal of this study was to further develop the 18F-radiolabeled tropane derivatives as DAT imaging agents. A series of 2β-(N-halopyridylaminomethyl)-3β-(4-chlorophenyl)tropane derivatives were synthesized and evaluated. Methods 2β-(N-bromo/fluoropyridylaminomethyl)-3β-(4-chlorophenyl)tropane derivatives were synthesized from a tropane ester derivative via a four step process. The binding abilities of the compounds were measured and compared to that of the reference ligand [125I]IPT to rat striatum homogenates and also with the known compound 2β-carbomethoxy-3β-(4-chlorophenyl)tropane (β-CCT). Radio-synthesis was performed by nucleophilic substitution of Boc-protected bromo precursor with [18F]F- in DMSO, followed by deprotection. In vivo biodistribution studies of 18F-labeled compound was evaluated in male Sprague-Dawley rats at 2 min and 60 min post injection. Results One of the best compounds in the series was 2β-(N-6-fluoropyridylaminomethyl)-3β-(4-chlorophenyl)tropane (6-FPAMCPT) that possessed a Ki value of 10.3nM (β-CCT, Ki=13.5nM), which was selected for further examination. 18F labeled 6-FPAMCPT had a radiochemical purity greater than 98%. Biodistribution studies showed that 6-18FPAMCPT had brain uptake at 2 min of 0.40%ID/g and possessed target-to-nontarget (striatum-to-cerebellum) ratio of 2.02 at 60 min after injection. Conclusions The results showed that 6-FPAMCPT may be useful for DAT imaging
