Genetic Component Involved in Nasopharyngeal Carcinoma Development

Abstract:Nasopharyngeal carcinoma (NPC) is a malignancy with striking geographic and ethnic distribution, and familial clustering of NPC is frequently observed. Emigration and dialect studies also suggest the involvement of genetic factors in the development of NPC. Therefore, many investigations on susceptibility genes have been carried out and polymorphism or specific allele types of HLA, TCR, GSTM1, PIGR and cytochrome P450 were reported to be related to high risk of NPC. More importantly, a pedigree-based study provided evidence of linkage of NPC to chromosome 4p15.1-q12. Somatic genetic changes, such as allelic loss, chromosomal gains or losses, specific gene alterations, were reported to occur frequently in NPC. These findings mentioned above were summarized in this paper and a carcinogenesis model for NPC is proposed.
