Abstract:Integrating multiple functionalities into individual nanoscale complexes is of tremendous importance in biomedicine, expanding the capabilities of nanoscale structures to perform multiple parallel tasks. Here, the ability to enhance two different imaging technologies simultaneously-fluorescence optical imaging and magnetic resonance imaging-with antibody targeting and photothermal therapeutic actuation is combined all within the same nanoshell-based complex. The nanocomplexes are constructed by coating a gold nanoshell with a silica epilayer doped with Fe3O4 and the fluorophore ICG, which results in a high T-2 relaxivity (390 mM(-1) s(-1)) and 45X fluorescence enhancement of ICG. Bioconjugate nanocomplexes target HER2+ cells and induce photothermal cell death upon near-IR illumination.
Simultaneous Excitation and Emission Enhancement of Fluorescence Assisted by Double Plasmon Modes of Gold Nanorods

Abstract:Here we propose a scheme utilizing the double plasmon modes of gold nanorod (GNR) to efficiently enhance the fluorescence of surrounding emitters. The transversal and longitudinal surface plasmon resonance (TSPR and LSPR, respectively) modes of GNR are simultaneously utilized to enhance the excitation and emission efficiency, respectively. To demonstrate the idea, GNRs coated with an Oxazine-725 dye molecule-doped silica shell are employed. For comparison, gold nanospheres with the same shell are also studied, of which the single plasmon resonance mode matches only with the excitation wavelength of Oxazine-725. The experimental results, in agreement with theoretical simulations using the discrete dipole approximation method, successfully demonstrate the efficient excitation and emission enhancement of fluorescence assisted by the double SPRs of GNRs.

Abstract:Light-mediated therapy has many unique merits but monotherapy strategies rarely completely inhibit tumor growth because resistance often develops. Combination therapy is a promising strategy in oncology and has demonstrated superior safety and efficacy over monotherapy. Here, we conjugated a scintillator complex and gold nanorod nanosensitizer for dual-modal image-guided photothermal and X-ray-induced photodynamic therapy (PDT). Lanthanide complexes were successfully conjugated and offer excellent X-ray-excited optical luminescence for PDT effects. The strong near-infrared (NIR) light and X-ray absorption abilities of gold nanorods make the nanosensitizer function as both a photothermal agent for photothermal therapy and a radiosensitizer for enhanced radiotherapy. The studies in vitro and in vivo demonstrated that the nanosensitizer offers good dual-modal imaging capability and significantly suppresses tumor progression under NIR light and X-ray irradiation. This work shows the great potential of conjugating scintillator lanthanide complexes and gold nanosensitizers for multimodal image-guided therapy of deep-seated tumors.
In situ crystal growth of gold nanocrystals on upconversion nanoparticles for synergistic chemo-photothermal therapy.

Abstract:A multifunctional cancer therapy nanocomposite was proposed and synthesized by linking the pH-responsive SH-PEG-DOX prodrug onto gold nanocrystals that were grown in situ on the surface of upconversion nanoparticles (UCNPs). In the structure of the SH-PEG-DOX prodrug, a hydrazone bond was utilized for subsequent pH-responsive drug release in the intracellular acidic microenvironment of cancer cells. This innovative assembly method is facile and mild, and can be used to obtain nanocomposites of UCNPs and gold, which show excellent photostability and biocompatibility. The final UCNPs@Au-DOX nanocomposites offer efficient treatment effects in vitro under irradiation with an 808 nm laser due to the synergistic effect of chemotherapy and photothermal therapy. In addition, the UCNPs@Au-DOX nanocomposites show excellent intracellular locating ability via upconversion luminescence (UCL) imaging with Er3+ ions and magnetic resonance imaging (MRI) with Gd3+ ions, indicating that they have potential as a visual tracking agent in cancer treatment. Therefore, the presented bioimaging-guided multifunctional synergistic therapy nanocomposites are promising tools for imaging-guided cancer therapy.
Photothermal conversion of SiO2@Au nanoparticles mediated by surface morphology of gold cluster layer

Abstract:Photothermal effects in SiO2@Au core-shell nanoparticles have demonstrated great potential in various applications for drug delivery, thermo-photovoltaics and photothermal cancer therapy, etc. However, the photothermal conversion of SiO2@Au nanoparticles partially covered by disconnected gold clusters has rarely been investigated systematically. Here, we control the surface morphology of gold clusters on the photothermal conversion performance of SiO2@Au core-shell nanoparticles by means of chemically adjusting the synthesis parameters, including amounts of gold salts, pH value and reducing agent. The macroscopic variations of the photothermal heating of different nanoparticle dispersions are significantly influenced by the nanoscale differences of gold cluster morphologies on the silica core. The temperature rise can be enhanced by the strong near-field coupling and collective heating among gold clusters with a relatively uniform distribution on the silica core. A numerical model of the simplified photothermal system is formulated to interpret the physical mechanism of the experimental observation, and shows a similar trend of temperature rise implying a reasonably good agreement with experimental data. Our work opens new possibilities for manipulating the light-to-heat conversion performance of SiO2@Au core-shell nanoparticles and potential applications of heat delivery with spatial resolution on the nanoscale.

Abstract:With the increasing need for multi-purpose analysis in the biomedical field, traditional single diagnosis methods cannot meet the requirements. Therefore new multifunctional technologies and materials for the integration of sample collection, sensing and imaging are in great demand. Core-shell nanoparticles offer a unique platform to combine multifunctions in a single particle. In this work, we have constructed a novel type of core-shell superparamagnetic nanoshell (Fe₃O₄@SiO₂@Au), composed of a Fe₃O₄ cluster core, a thin Au shell and a SiO₂ layer in between. The obtained multifunctional nanoparticles combine the magnetic properties and plasmonic optical properties effectively, which were well investigated by a number of experimental characterization methods and theoretical simulations. We have demonstrated that Fe₃O₄@SiO₂@Au nanoparticles can be utilized for two-photon luminescence (TPL) imaging, near-infrared surface-enhanced Raman scattering (NIR SERS) and cell collection by magnetic separation. The TPL intensity could be further greatly enhanced through the plasmon coupling effect in the self-assembled nanoparticle chains, which were triggered by an external magnetic field. In addition, Fe₃O₄@SiO₂@Au nanoparticles may have great potential applications such as enhanced magnetic resonance imaging (MRI) and photo-thermotherapy. Successful combination of multifunctions including magnetic response, biosensing and bioimaging in single nanoparticles allows further manipulation, real-time tracking, and intracellular molecule analysis of live cells at a single-cell level.
Combined near-IR photothermal therapy and chemotherapy using gold-nanorod/chitosan hybrid nanospheres to enhance the antitumor effect

Abstract:Motivated by the enhanced cytotoxicity of some chemotherapeutic agents at temperature rise, multifunctional chitosan nanospheres which co-carried gold nanorods and cisplatin were prepared. These hybrid nanospheres were characterized by dynamic light scattering, zeta potential, UV-Vis absorption spectra and transmission electron microscopy. It was demonstrated that these 120 nm-sized hybrid nanospheres could be selectively accumulated at the tumor site with about 11% injection dose per g of tumor, and produced local hyperthermia to an average temperature of 49 degrees C in tumor tissue after near-IR irradiation for 10 min. Due to the enhanced cytotoxicity of cisplatin at elevated temperatures, cisplatin-loaded hybrid nanospheres showed about one-second lower IC50 values than hybrid nanospheres alone in vitro and almost complete tumor growth inhibition in vivo. Compared with chemotherapy or photothermal treatment alone, the combined photothermal therapy and chemotherapy had a significantly synergistic effect and improved the therapeutic efficacy, which was supported by immunofluorescence staining and in vivo apoptosis imaging.

Abstract:A facile process for the preparation of multifunctional nanospheres combining several advantages of mesoporous channels, up-conversion (UC) luminescence, and photothermal responses into one single entity is reported. First, Gd2O3:Yb/Er assembled mesoporous silica with 2D hexagonal (MCM-41) and 3D cubic (MCM-48) network have been prepared via a one-step procedure. Then, gold nanocrystals with diameter of 5 nm are integrated with the amino group functionalized nanocomposites. Upon 980 nm near infrared (NIR) laser irradiation, a wavelength-dependent enhancement of the UC intensities is observed due to the surface plasmon resonance (SPR) effect of attached gold nanoparticles. These composites have good biocompatibility and sustained anticancer drug (doxorubicin, DOX) release properties, making it a promising candidate for drug delivery. Particularly, under 980 nm NIR laser irradiation, the green UC emission overlaps the SPR band of gold nanocrystals, which causes a photothermal effect of gold nanocrystals and induces a rapid DOX release from the Au hybrid materials. This DOX loaded multifunctional system has an obvious cytotoxic effect and photothermally killing enhanced effect on SKOV3 ovarian cancer cells. The endocytosis process was also demonstrated through confocal laser scanning microscope (CLSM) images. Such novel multifunctional anticancer drug delivery systems, which combine hyperthermia with the chemotherapeutic drugs by synergistic effect, should be of high potential in cancer therapy.

Abstract:Multifunctional nanocarriers based on chitosan/gold nanorod (CS-AuNR) hybrid nanospheres have been successfully fabricated by a simple nonsolvent-aided counterion complexation method. Anticancer drug cisplatin was subsequently loaded into the obtained hybrid nanospheres, utilizing the loading space provided by the chitosan spherical matrix. In vitro cell experiments demonstrated that the CS-AuNR hybrid nanospheres can not only be utilized as contrast agents for real-time cell imaging but also serve as a near-infrared (NIR) thermotherapy nanodevice to achieve irradiation-induced cancer cell death owing to the unique optical properties endowed by the encapsulated gold nanorods. In addition, an effective attack on the cancer cells by the loaded anticancer drug cisplatin has also been observed, rendering the obtained nanocarriers an all-in-one system possessing drug delivery, cell imaging, and photothermal therapy functionalities.

Abstract:<p>The unsymmetrical morphology and unique properties of Janus nanoparticles (JNPs) provide superior performances for biomedical applications. In this work, a general and facile strategy is developed to construct a series of symmetrical and unsymmetrical chitosan/gold nanoparticles. Taking advantage of the active motion derived from Janus structure, selective surface functionalization of polysaccharide domain, and photothermal effect of gold nanorods, Janus chitosan/gold nanoparticles (J‐Au‐CS) are selected as a model system to construct Janus‐structured chitosan/gold nanohybrids (J‐ACP). Near‐infrared (NIR)‐responsive J‐ACP composed of polycationic chitosan nanospheres and PEGylated gold nanorods hold great potential to realize photoacoustic (PA) imaging‐guided complementary photothermal therapy (PTT)/gene therapy for breast cancer. The morphology effect of chitosan/gold nanostructures on enhanced PTT, cellular uptake, and gene transfection is investigated. The feasibility of PA imaging to track the accumulation of J‐ACP and guide PTT is also explored. Notably, synergistic therapy is achieved based on PTT‐enhanced gene therapy. In addition, the loading function of chitosan/gold nanoparticles for fluorescence imaging is demonstrated. The current work extends the application of JNPs for imaging‐guided synergistic cancer therapy and provides flexible candidates with distinct structures for diverse biomedical applications.</p>

AuNPs Decorated PLA Stereocomplex Micelles for Synergetic Photothermal and Chemotherapy

Abstract:A unique platform for combined photothermal and chemotherapy using PLA stereocomplex (PLA SC) micelles-induced hybrid gold nanocarriers is designed. The PLA SC micelles, made from the self-assembly of poly(ethylene glycol)-block-poly(l-lactide) (PEG-PLLA) and poly(2-(dimethylamino) ethyl methacrylate)-block-poly(d-lactide) (PDMAEMA-PDLA), for the first time are used as a template to fabricate the hybrid PLA SC@Au core-shell nanocarriers, in which the anticancer drugs are encapsulated within the core, while the Au nanoparticles are tethered in the shell via the in situ reduction of AuCl4- by PDMAEMA. The obtained PLA SC@Au hybrid nanocarriers exhibit low toxicity and remarkable photothermal effect. Upon near-infrared laser irradiation, the on-site photothermal therapy can further induce an accelerated drug release from the hybrid nanocarrier reservoir via hyperthermia heating of the nanocarriers, thus leading to a synergistic photothermal and chemotherapy toward a significantly improved efficacy in tumor shrinkage. The as-designed PLA SC@Au hybrid nanocarriers, with their biocompatible compositions, dual-drug delivery characteristics, and combined photothermal/chemotherapy, show high potential as a novel platform for cancer treatment.

Abstract:Phototherapeutics is gaining momentum as a mainstream treatment for cancer, with gold–semiconductor nanocomposites showing promise as potent phototherapeutic agents due to their structural tunability, biocompatibility and functional diversity. Such nanohybrids possess plasmonic characteristics in the presence of gold and the catalytic nature of semiconductor units, as well as the unexpected physicochemical properties arising from the contact interface. This perspective provides an overview of the latest research on gold–semiconductor nanocomposites for photodynamic, photothermal and photocatalytic therapy. The relationship between the spatial configuration of these nanohybrids and their practical performance was explored to deliver comprehensive insights and guidance for the design and fabrication of novel composite nanoplatforms to enhance the efficiency of phototherapeutics, promoting the development of nanotechnology-based advanced biomedical applications.
Gold-nanobranched-shell based drug vehicles with ultrahigh photothermal efficiency for chemo-photothermal therapy

Abstract:Development of combined chemo-photothermal nanoplatform is of great interest for enhancing antitumor efficacy. Herein, a multifunctional drug delivery system was synthesized based on gold-nanobranched coated betulinic acid liposomes (GNBS-BA-Lips) for chemo-photothermal synergistic therapy. In this system, GNBS-BA-Lips exhibited broad near-infrared (NIR) absorption, preferable photothermal response and good photostability under NIR irradiation. Importantly, the gold-nanobranched nanostructure possessed high photothermal conversion efficiency (η = 55.7%), and the temperature change (ΔT) reached 43.2 °C after laser irradiation for 5 min. Upon NIR irradiation, the nanocarriers apparently endowed higher cell uptake, resulting in an enhanced intracellular drug accumulation. Furthermore, the tumor growth inhibition ratio achieved from chemo-photothermal therapy of GNBS-BA-Lips was 86.9 ± 1.1%, which was higher than that of the chemotherapy or photothermal therapy alone, showing an outstanding synergistic anticancer effect. Our data suggested that the nanoplatform should be considered as a critical platform in the development of cancer multi-mode therapies.

Abstract:Two key concerns exist in contemporary cancer chemotherapy in clinics: limited therapeutic efficiency and substantial side effects in patients. In recent years, researchers have been investigating revolutionary cancer treatment techniques and photo-thermal therapy (PTT) has been proposed by many scholars. A drug for photothermal cancer treatment was synthesized using the hydrothermal method, which has a high light-to-heat conversion efficiency. It may also be utilized as a clear multi-modality bioimaging platform for photoacoustic imaging (PAI), computed tomography (CT), and magnetic resonance imaging (MRI). When compared to single-modality imaging, multi-modality imaging delivers far more thorough and precise details for cancer diagnosis. Furthermore, gold-doped upconverting nanoparticles (UCNPs) have an exceptionally high target recognition for tumor cells. The gold-doped UCNPs, in particular, are non-toxic to normal tissues, endowing the as-prepared medications with outstanding therapeutic efficacy but exceptionally low side effects. These findings may encourage the creation of fresh effective imaging-guided approaches to meet the goal of photothermal cancer therapy.

Abstract:Core-shell metal nanostructures with versatile functions have attracted extensive attention and are highly desirable for imaging and therapeutic purposes. Among them, gold and silver nanomaterials are widely explored for biological applications due to their unique properties. Despite a wide range of applications, limited enhancement ability and insufficient photothermal performance have hampered their further development. In this work, a novel multifunctional nanoprobe, a Au@Ag nanoplate (NP), is fabricated with a biocompatible surface in the aqueous phase. The as-obtained nanocomposite possesses a unique core-shell triangular configuration, sharp apexes, and a large specific surface area, exhibiting strong absorption at 780 nm. PEG-Au@Ag NPs depict highly sensitive photoacoustic imaging (PAI) capacity and extraordinary photothermal conversion efficiency (η = 73%) under 808 nm laser irradiation. Raman signals are multiplied benefitting from the enhanced surface plasma resonance contributed by the silver layer and sharp spears. PAI provides deeper pathological information while Raman detection presents superficial optical properties. Their union forms comprehensive scale coverage for disease imaging and localization. Outstanding photothermal therapy and antibacterial efficacy are observed on animal disease models. This novel multifunctional nanocomposite not only holds great potential as an excellent contrast agent for the combination of PAI and Raman evaluation, but also allows tumor and infection therapy as well as the corresponding therapeutic monitoring.

Abstract:Although platinum-based anticancer drugs have prevailed in traditional chemotherapy, the severe side effects and drug resistance hampered their clinical applications. Combinational therapies have exhibited promises in antitumor treatments. However, the integration of different anticancer modalities is often challenging because of the conflicts between maximal synergy of therapeutic effects and reduction of side effects. Here, we report a multifunctional platform composed of metallacycle-based star polymer and gold nanorods for synergistic photochemotherapy. We applied the Pt-based metallacycle-cored star polymers as both a photodynamic photosensitizer and a Pt-based anticancer drug. The gold nanorods play the dual role of photothermal converter and energy acceptor of fluorescence resonance energy transfer effect from the star polymers. The photothermal effect enhanced tumor permeability of reactive oxygen species and Pt-based drug molecules and the photodynamic effect destructed heat shock proteins to prevent the mechanism of cellular self-protection response to heat. This mutually beneficial strategy results in high therapeutic efficacy and negligible side effects verified by both in vitro and in vivo studies. Rattle-Type Gold Nanorods/Porous-SiO2 Nanocomposites as Near-Infrared Light-Activated Drug Delivery Systems for Cancer Combined Chemo-Photothermal Therapy

Abstract:Rattle-type nanostructures with movable cores, porous shells, and hollow interiors have become attractive nanoplatforms in the field of nanomedicine, especially for targeted and stimuli-responsive drug delivery. In this work, rattle-type gold nanorods@void@porous-SiO2 (GVPS) nanocomposites were fabricated facilely using the surface-protecting etching method and exhibited high photothermal conversion efficiency. Taking advantage of the porous shell and hollow interior, the nanocomposites have abundant space for drug loading and successfully improved the drug loading capacity up to ∼19.6%. To construct a multifunctional drug delivery system, GVPS was further functionalized with polyethylene glycol (PEG) and cyclic RGD peptides to improve biocompatibility as well as selectivity toward the targeted cancer cells. Besides, to achieve precise regulation and near-infrared laser activation of the drug release, a phase-changing material, 1-tetradecanol (1-TD, Tm: 39 °C), was employed as gatekeepers in this system. After incubation with an αVβ3 integrin receptor-overexpressed cell line, the as-prepared GVPSPR-DOX/TD nanocomposites exhibited great performances in combined photothermal therapy and chemotherapy. It is worth noting that the combined therapy showed superior efficiency in cancer cell killing to chemotherapy or photothermal therapy alone.
Synthesis of Structure-Adjustable R-Au/Pt-CdS Nanohybrids with Strong Plasmon Coupling and Improved Photothermal Conversion Performance

Abstract:Noble metal nanomaterials with a localized surface plasmon resonance effect exhibit outstanding advantages in areas such as photothermal therapy and photocatalysis. As a unique plasmonic metal nanostructure, gold nanobipyramids have been attracting much interest due to their strong specific local electric field intensity, large optical cross sections, and high refractive index sensitivity. In this study, we propose a novel three-component hetero-nanostructure composed of rough gold nanobipyramids (R-Au NBPs), Pt, and CdS. Initially, purified gold nanobipyramids are regrown to form R-Au NBPs that have a certain degree of roughness. These R-Au NBP substrates with a rough surface provide more hotspots and strengthen the intensity of localized electric fields. Subsequently, Pt and CdS nanoparticles are selectively deposited onto the surface of R-Au NBPs. Pt nanoparticles can provide more active sites. Each component of this hetero-nanostructure directly contacts others, creating multiple electron transfer channels. This novel design allows for tunable localized plasmon resonance wavelengths ranging from the visible to near-infrared regions. These factors contribute to the final superior photothermal conversion performance of the R-Au/Pt-CdS nanohybrids. Under the irradiation of near-infrared light (1064 nm), the photothermal conversion efficiency of R-Au/Pt-CdS reached 38.88%, which is 4.49, 1.5, and 1.22 times higher than that of Au NBPs, R-Au NBPs, and R-Au NBPs/Pt, respectively.

Abstract:In an attempt to integrate photodynamic therapy (PDT) with photothermal therapy and chemotherapy for enhanced anticancer activity, we have rationally synthesized a multifunctional upconversion nanoplatform using NaYF4:Yb/Tm/Er/Fe nanoparticles (NPs) as the core and NaYbF4:1% Tm as a shell. The as-synthesized core-shell upconversion (CSU) NPs exhibited diverse and enhanced photoluminescence emissions in a wide range (UV to NIR) consequent upon Fe3+ doping in the core and fabrication of an active shell. Subsequently, CSU was first decorated with titania NPs as photosensitizers. Next, the mesoporous silica (MS) shell loaded with doxorubicin (DOX) via a photocleavable Ru complex as the gating molecule was developed around titania-containing CSU. Finally, gold nanorods (GNRs) with localized surface plasmon resonance (LSPR) at 800 nm were incorporated around the MS layer to obtain the multifunctional nanoplatform. We demonstrated that the UV, blue, and NIR emissions from the CSU produced ROS-mediated PDT through titania activation, induced DOX release through photocleavage of the Ru complex, and generated hyperthermia by LSPR activity of GNRs, respectively, upon a single NIR excitation through FRET. The therapeutic efficacy was validated on HeLa cell lines in vitro by various microscopic and biochemical studies under a significantly milder NIR irradiation and lower dosage of the nanoplatforms, which have been further demonstrated as diagnostic nanoprobes for cell imaging.
