Abstract:A new Schiff-base fluorescence probe CS containing benzimidazole and coumarin fluorophores was synthesized and characterized by NMR, IR and ESI-MS. Fluorescence and absorption spectra show the unique changes in the presence of Hg2+ and Cu2+. Hg2+ induces a remarkable fluorescence enhancement and results in mercury-promoted hydrolysis of a mercury-CS complex to convert the imine group to the aldehyde group. Cu2+ leads to a colorimetric change from colorless to pale yellow and causes formation of a copper-CS complex. Thus, CS can act as a fluorescent chemodosimeter for Hg2+ and a naked-eye chemosensor Cu2+. The cell imaging experiments for Hg2+ show a slow enhancement of a bright blue fluorescence with increasing incubation time, while Cu2+ instantly quenches the fluorescence and then the fluorescence intensity has no further change by extending the incubation time. Therefore, the probe CS shows its potential application for simultaneous differential detection of Hg2+ and Cu2+ in living cells based on the fluorescence response time. (C) 2016 Elsevier B.V. Abstract:Metal ions have a significant impact on the environment and the growth and development of organism,therefore,the recognition and detection of metal ions in the environment and organism is gaining widespread attention. Among different analytical methods,fluorescence analysis has the advantages of high sensitivity,good selectivity,real-time and in situ detection,etc.,which is a good tool to achieve the recognition and detection of metal ions in the environment and organism. This paper is aimed to review the coumarin-based fluorescent sensors for the recognition and detection of heavy metal ions (Hg2+、Pb2+、Cd2+、Ni2+、Ag+),biologically important transition metal ions (Cu2+、Zn2+、Fe3+) and their applications in recent five years,focusing on the design and synthesis of sensors, recognition mechanism,sensing characteristics and applications. With the higher requirement for metal ions detection , the design of coumarin-based fluorescent sensors will be developed for better performance with higher sensitivity,selectivity and stronger anti-interference in the future. In addition, applications of coumarin-based fluorescent sensors in biological detection are expected to be further developed.

Abstract:A novel chemosensor 1 (CS1) bearing one coumarin and two carbodithioate groups was synthesized and its fluorescent sensing behavior toward metal ions was investigated. Ag+ addition to a CH3COCH3/H2O (3:7, v:v) solution of CS1 gave a significantly quenched fluorescence. Other ions including Pb2+, Zn2+, Cu2+, Ca2+, Cd2+, Co2+, Mg2+, Mn2+, Hg2+, Ag+, Ni2+ induced no or much smaller spectral changes. This constitutes an ON–OFF Ag+-selective fluorescent chemosensor.

Abstract:A nonfluorescent coumarin derivative (1) was synthesized as an efficient "turn-on" fluorescent chemodosimeter for Cu(2+) in water. Mechanism studies suggested that 1 formed a complex with Cu(2+) in a 1:2 metal-to-ligand ratio, and a 50-fold fluorescence enhancement was observed when the complex simultaneously underwent Cu(2+)-promoted hydrolysis at ambient temperature.

Abstract:Isatin-3-(7′-Methoxychromone-3′-methylidene) hydrazone ( L ) was synthesized based on chromone schiff base, and used to construct a novel sensor to detect Cr 3+ . Fluorescence detection experiments were carried out for a range of different concentrations of Cr 3+ in aqueous solutions. A concentration calculation model was built on the basis of eliminating interference of excitation spectrum in the fluorescence spectra with mathematical method. Results showed that probe L displayed a 70 -fold fluorescence enhancement upon the addition of Cr 3+ due to the photo-induced electron transfer (PET) effect. On the other hand, metal ions except Cr 3+ did not cause significant change in either the absorption or the fluorescence spectrum of L. In addition, L showed a good selectivity to Cr 3+ over other metal cations, especially Al 3+ and Cu 2+ . The probe L can detect Cr 3+ highly and selectively by the direct chelation enhanced fluorescence with a detection limit of 3.14 × 10 −6 M. Furthermore, benefiting from their good water solubility and biocompatibility, cell imaging and real-time monitoring of Cr 3+ in living HepG2 cells were successfully achieved.

Abstract:In this work, a hydrophilic naphthalimide-based fluorescence chemosensor (sensor 1) was synthesized for Cu2+ recognition, in which 2-(2-aminoethoxy)ethanol was introduced to improve the hydrophily and Schiff base acted as the multidentate ligand for Cu2+. The effect factors, sensing mechanism and regenerability of sensor 1 for Cu2+ sensing were systematically investigated. It was found that sensor 1 displayed a long emission wavelength of 532 nm upon excited in visible light region (436 nm), and the good water solubility made it utilized in aqueous media. It could selectively react with Cu2+ over other common metal ions to form a 2:1 complex within 1 min and result in significant fluorescence quench. The fluorescence change was linear to 0.5-10.0 μmol L-1 of Cu2+ with a low detection limit of 3.74 × 10-8 mol L-1. Sensor 1 has been successfully utilized for analyzing Cu2+ in water samples as well as imaging cellular Cu2+. Moreover, in view of fluorescence "on-off-on" switches of sensor 1 induced by Cu2+ and EDTA, an IMPLICATION logic gate was constructed based on fluorescence mode with Cu2+ and EDTA as inputs.

Abstract:Copper ion (Cu2+) plays important role in life system and industry, therefore, efficient detection of Cu2+ is very important. To improve the selectivity and sensitivity of Cu2+, herein, quinoline and coumarin groups were rationally employed into a novel fluorescent sensor CQ to efficiently sensing Cu2+ through the collaboration of these two groups. Interestingly, the CQ possesses aggregation‐induced emission (AIE) properties and shows nice selective and sensitive fluorescent response to Cu2+. The lowest limit of detection (LOD) for Cu2+ is 2.81×10‐8 M. Moreover, the Cu2+ sensing process has good anti‐interference properties. The possible recognition mechanism was investigated by experiments and theoretical calculations, which supplied a deep insight not only on the collaboration of the quinoline and coumarin groups in the Cu2+ recognition process, but also on the charge transfer induced fluorescent response mechanism. The results shown that the CQ could specifically detected Cu2+ via the collaboration of the quinoline and coumarin groups through multiple coordination interactions. Importantly, test strips based on CQ were fabricated, which could act as test kits for convenient and efficient detection of Cu2+.

Abstract:A new ratiometric fluorescent sensor for Cu(2+), WLN, has been developed via integrating a 1,8-naphthalimide fluorophore with 8-aminoquinoline. WLN exhibits a highly selective ratiometric response to Cu(2+) over other transition metal ions in aqueous media. Moreover, its practical ratiometric imaging ability for intracellular Cu(2+) has been confirmed in human breast adenocarcinoma cells (MCF-7 cells) using a confocal microscope.

Abstract:(E)-4-Chloro-3-[{2-(4-nitrophenyl)hydrazono}methyl]-2H-chromen-2-one (C), a coumarin derivative has been studied toward its ion sensing properties for F-, CN- and Cu2+. A proton-transfer mechanism for F- sensing has been deduced with the help of 1H NMR titration alongwith from the changes in the absorption and emission spectra of C in the presence of F-. C formed 1:1 stoichiometric complex with each of these analytes. Sensing of C toward Cu2+ is poor, but interestingly in the presence of F-or CN- the sensing ability of Cu2+ gets enhanced many folds, and C can act as F-or CN- mediated off-on sensor for Cu2+. Moreover, colorimetric strip (pre-coated with the coumarin derived compound) tests for F-and CN- from their DMSO solution at high temperature (~100°C) opens up the door for easiest naked eye recognition and distinction of these ions, and also for naked-eye detection of F- and CN- from its aqueous solution at high temperature (~100°C).

Abstract:A rhodamine B derivative 4 containing a highly electron-rich S atom has been synthesized as a fluorescence turn-on chemodosimeter for Cu2+. Following Cu2+-promoted ring-opening, redox and hydrolysis reactions, comparable amplifications of absorption and fluorescence signals were observed upon addition of Cu2+ this Suggests that chemodosimeter 4 effectively avoided the fluorescence quenching caused by the paramagnetic nature of Cu2+. Importantly, 4 can selectively recognize Cu2+ in aqueous media in the presence of other trace metal ions in organisms (Such as Fe3+ Fe2+, Cu+, Zn2+, Cr3+, Mn2+, Co2+, and Ni2+), abundant cellular cations (such as Na+, K+, Mg2+ and Ca2+), and the prevalent toxic metal ions in the environment (Such as Pb2+ and Cd2+) Will, high sensitivity (detection limit <= 10 ppb) and a rapid response time (<= 1 min). Moreover, by virtue of the chemodosimeter as fluorescent probe for Cu2+, confocal and two-photon microscopy experiments revealed a significant increase of intracellular Cu2+ concentration and the subcellular distribution of Cu2+. which was internalized into the living HeLa cells upon incubation in growth medium supplemented with 50 mu M CuCl2 for 20 h.

Abstract:A new fluorescent chemosensor P from 5-(thiophen-2-yl)oxazole-4-carbohydrazide and 4-diethylaminosalicylaldehyde was designed and synthesized for sequential detection of In3+ and Cr3+ ions. P exhibits strong fluorescence enhancement at 512 nm upon binding to In3+. The green fluorescence of complex P-In3+ was selectively quenched by Cr3+. The complex stoichiometry of P with In3+ and Cr3+ was identified as 1:2 for both in conjunction with Job’s Plot analysis, ESI-MS titration and theoretical calculations. Moreover, P was successfully applied to monitor target ions in actual water samples, and a portable paper sensor for the detection of In3+ was investigated to evaluate the versatility application.

Abstract:A novel fluorescent sensor for the detection of Cu 2+ was developed based on carbazole derivatives. After the addition of Cu 2+ , the sensor exhibited obvious fluorescence quenching phenomenon, and the optical signal variation also enabled the sensor to quantitatively analyze Cu 2+ due to the formation of a stable 1:1 metal-ligand complex in a short time. In addition, the sensor possessed chemical reversibility and pH stability. The cell imaging and zebra fish experiments also verified its application value in biological system.

Abstract:The induced quenching in fluorescence upon addition of copper (Cu 2+ ) ions and subsequent regain with aqueous hydrogen sulphide (HS − ) was studied with a series of coumarin di(picolyl)amine-based compounds, with varying electron withdrawing/donating abilities. The coumarin analogue with an amino group at the 7-position was the most emissive, followed by the methoxy-variant with both the t -butyl- and difluoro-variants being weakly emissive. The amino-variant was also most responsive to quenching upon the addition of Cu 2+ ions and was the most responsive to the subsequent addition of HS − ions. All compounds were shown to form predominately a 1:1 copper(II) complex, supported by mass spectral studies and X-ray crystallography. This investigation sheds light on the structural aspects of the copper-sensor interactions and subsequent copper sequestration, contributing to the understanding of their fluorescence properties and potential applications in sensing and imaging.

Abstract:A novel fluorescence chemical sensor-based probe 1-{[(E)-(2-aminophenyl)azanylidene]methyl}naphthalen-2-ol (AMN) was designed and synthesized, which performed a "naked eye" detection ability toward Cu2+ and Co2+ based on aggregation-induced emission (AIE) fluorescence strategy. It has sensitive detection ability for Cu2+ and Co2+. In addition, the color changed from yellow-green to orange under the sunlight, realizing the rapid identification of Cu2+/Co2+, which has the potential of on-site visual detection under the "naked eye". Moreover, different "on" and "off" fluorescence expressions were exhibited under excessive glutathione (GSH) in AMN-Cu2+ and AMN-Co2+ systems, which could be employed to distinguish Cu2+ from Co2+. The detection limits for Cu2+ and Co2+ were measured to be 8.29 × 10−8 M and 9.13 × 10−8 M, respectively. The binding mode of AMN was calculated to be 2:1 by Jobs' plot method analysis. Ultimately, the new fluorescence sensor was applied to detect Cu2+ and Co2+ in real samples (tap water, river water, and yellow croaker), and the results were satisfying. Therefore, this high-efficiency bifunctional chemical sensor platform based on "on–off" fluorescence detection will provide significant guidance for the advance development of single-molecule sensors for multi-ion detection.

Abstract:A multisignal sensor based on rhodamine B with a ferrocene substituent (FD7) has been developed as a highly sensitive chemosensor for Cr3+. A prominent fluorescence enhancement was found in the presence of Cr3+, which was accompanied by changes in the absorption spectrum and the electrochemical parameters. Furthermore, by means of confocal laser scanning microscopy experiments, it has been demonstrated that FD7 can be used as a fluorescent probe for monitoring Cr3+ in living cells.

Abstract:Cr3+ is one of the most essential trace elements in living organisms and plays a vital role in human metabolism. However, both deficiency and excess intake of Cr3+ can be harmful to the human body. Therefore, the quantitative determination of Cr3+ is of great significance in the field of life science. Based on this, in this study, a g-CNQDs@p-acetaminophenol fluorescence sensing system was developed for the quantitative detection of Cr3+ in actual complex samples. G-CNQDs were synthesized with sodium citrate and urea as precursors. The fluorescence signal was enhanced by the synergistic effect between p-acetaminophenol (APAP) and g-CNQDs. The fluorescence quenching phenomenon can be produced when Cr3+ is introduced into the fluorescence-enhanced g-CNQDs@p-acetaminophenol system. An "on-off" fluorescence sensing system was constructed based on g-CNQDs@p-acetaminophenol for the quantitative detection of Cr3+. The experimental data showed a wide linear region in the concentration range of 0.64-63.0 μM, and the detection limit was as low as 0.23 μM. The construction of the sensor system broadens the research field for the practical application of Cr3+.

Abstract:A novel seminaphthorhodafluor-based fluorescent probe was designed and prepared. It exhibited an outstanding off-on fluorescence response (about 45-fold enhancement) to Cu2+ because the structure changed from a spirocyclic form to a ring-open state. It showed satisfactory detection performances for Cu2+ with fast response speed, high selectivity and anti-interference, and a low detection limit (22.9 nM) due to its long emission wavelength and off-on fluorescence response mode. This probe was also satisfactorily applied for the monitoring of Cu2+ in living cells and zebrafish.
