Abstract:The reactions between cytochrome c and [Pt-dien]2+ under different conditions are reported. It is found that the yield of modified protein derivatives as well as the likely binding sites for platinum complex in cytochrome c are sensitive to the reaction conditions. It is first reported that transition-metal complex [Pt-dien]2+ can bind to Met 80 residue under mild conditions, which causes the FeS bond of ferricytochrome c to be broken. The binding sites of the platinum complex in the modified protein derivatives were identified by analysis of the molar ratio of the two metal atoms (Pt and Fe), 1H NMR spectra, and UV-vis spectra.

Abstract:The reaction between cytochrome c and trans-[PtCl2(NH3)(2)] has been studied under different conditions. The complex can not only bind to a Met-80 residue, which causes the Fe-S bond of ferricytochrome c to be broken, but can also induce a facile conversation of low-spin iron(III) cytochrome c to the high-spin form under mild conditions. Cytochrome c can also be cross-linked selectively by trans-[PtCl2(NH3)(2)]. The binding sites of the platinum complex in the protein derivatives were identified by analysis of the molar ratio of the two metals (Pt and Fe), H-1 NMR and UV/VIS spectroscopy and differential-pulse voltammetry.

Abstract:Proton-coupled electron transfer (PCET) was examined in a series of biomimetic, covalently linked Ru(II)(bpy)(3)-tyrosine complexes where the phenolic proton was H-bonded to an internal base (a benzimidazyl or pyridyl group). Photooxidation in laser flash/quench experiments generated the Ru(III) species, which triggered long-range electron transfer from the tyrosine group concerted with short-range proton transfer to the base. The results give an experimental demonstration of the strong dependence of the rate constant and kinetic isotope effect for this intramolecular PCET reaction on the effective proton transfer distance, as reflected by the experimentally determined proton donor-acceptor distance.
Electron Transport Through a Coordination Junction Formed by Carboxy Thiophenols and Bivalent Metal Ions

Abstract:Electron transport through a series of molecular junctions (CTP-M) composed of two carboxyl thiophenols (CTPs) on each gold electrode coordinating with a divalent metallic ion (M) in the center has been investigated using the density functional theory combined with nonequilibrium Green's function (DFT-NEGF) approach. The electronic structure, I-V characteristics, density of states (DOS), and transmission spectra of all CTP-M models have been calculated and compared to the junction without the metallic ion (CTP-H). While strong electronic coupling is established in the coordination structure, the transportation is efficient, showing higher junction conductance as compared to CTP-H. On the contrary, weak electronic coupling is formed between the carboxyl groups and the alkali metal ions (CTP-Mg and CTP-Ca) due to a twisted configuration, exhibiting smaller junction conductance. The strong dependence of the current on the coordination complex has been interpreted with the fine structure of the tunneling barrier. H-bonds result in a bump in the tunneling barrier profile, impeding the electron transfer. When the transition metal ions were incorporated into the molecular junction, the central part of the tunneling barrier turns into an energy well, facilitating the electron transfer. This study provides an effective method to control the interface electron transport required in the construction of flexible wearable electronic devices and functional electronic molecular devices.
Mapping the Electron Transfer Interface Between Cytochrome B5 and Cytochrome C

Abstract:To characterize the cytochrome b(5) (Cyt b(5))-cytochrome c (Cyt c) interactions during electron transfer, variants of Cyt b(5) have been employed to assess the contributions of electrostatic interactions (substitution of surface charged residues Glu44, Glu48, Glu56, and Asp60 and heme propionate), hydrophobic interactions, and the thermodynamic driving forces (substitutions for hydrophobic residues in heme pocket residues Phe35, Pro40, Val45, Phe58, and Val61). The electrostatic interactions play an important role in maintaining the stability and specificity of the Cyt b(5)-Cyt c complex that is formed. There is no essential effect on the intraprotein complex electron transfer even if most of the involved negatively charged residues on the surface of Cyt b(5) have been removed. The results support a dynamic docking paradigm for Cyt b(5)-Cyt c interactions. The orientation that is optimal for binding may not be optimal form for electron transfer. Substitution of hydrophobic residues does not have a significant effect on the binding between Cyt b(5) and Cyt c; rather, it regulates the electron transfer rates via changes in the driving force. Combining the electron transfer studies of the Cyt b(5)-Cyt c system and the Cyt b(5)-Zn-Cyt c system, we obtain the reorganization energy (0.6 eV) at an ionic strength of 150 mM.
Influence Of Electrode-Molecule Interface On Electron Transport

Abstract:We have studied the metal-molecule interface linkage effects of metal-molecule-metal systems by first principles method, which is based on density functional theory (DFT) and nonequilibrium Green's functions (NEGF) to calculate the electron transport of open metal-molecule-metal systems. Metal electrodes are described through 3-D atomic model instead of a non-atomic (like Jelhum model) description. Several open systems are constructed, optimized and simulated. Sulphur atom (S) and cyano-group (CN) are employed to connect An electrodes and molecule borazine/benzene. The density of states (DOS) and the transmission functions (M of constructed systems are investigated. Results show that DOS of the systems are affected little but transmission properties a lot by terminal groups CN and S. The peaks in the TF mismatch with those in DOS and the mismatch resulting from terminal group CN is larger than it from terminal atom S. Calculated transmission functions show that the systems with terminal group CN present better conductance at lower bias and this effect is much more significant in the borazine systems than that in the benzene systems.

Abstract:Electron transfer (ET) between redox proteins is an essential process in the respiratory and photosynthetic transport chains. While intra-protein ET is well characterized, the experimental methods to investigate inter-protein ET are limited by the presence of the solvent and by the transient nature of the protein-protein interaction and ET event, which are averaged in protein ensembles. Wiring precisely oriented redox protein partners to the nanoscale electrodes of an electrochemical scanning tunneling microscope allows to record the time- and distance-dependence of the current flowing between them. These methods have revealed that the current flowing between individual protein pairs extends much beyond tunneling distances and that it is electrochemically gated. However, the mechanism of this process, and especially the identity of the charge carriers in the aqueous solution, remains to be elucidated. To determine the species involved in long distance charge transport between the redox partner proteins Cc and Cc1 of the respiratory chain, we have performed recordings as a function of pH, in heavy water solutions, and in degassed solutions. We observe that the spatial span and electrochemical gating of long-distance currents are reduced at high pH. Currents are also less extended in heavy water and at low oxygen concentration. Using the experimental structures of oxidized and reduced Cc and pKa calculations, we identify residues that can switch their availability to form hydrogen bonds with water and/or engage in proton-coupled electron transfer reactions. Overall, our results show that long distance currents between cytochrome c (Cc) and cytochrome c1 (Cc1) through the aqueous solution are assisted by superoxide anions and by protons, and we discuss possible molecular mechanisms and their relevance for interprotein ET.

Abstract:Proton-coupled electron transfer (PCET) underpins energy conversion in biology. PCET may occur with the unidirectional or bidirectional transfer of a proton and electron and may proceed synchronously or asynchronously. To illustrate the role of PCET in biology, this review presents complementary biological and model systems that explore PCET in electron transfer (ET) through hydrogen bonds [azurin as compared to donor-acceptor (D-A) hydrogen-bonded networks], the activation of C-H bonds [alcohol dehydrogenase and soybean lipoxygenase (SLO) as compared to Fe(III) metal complexes], and the generation and transport of amino acid radicals [photosystem II (PSII) and ribonucleotide reductase (RNR) as compared to tyrosine-modified photoactive Re(I) and Ru(II) complexes]. In providing these comparisons, the fundamental principles of PCET in biology are illustrated in a tangible way.

Abstract:In this work, we reported the investigation on the interaction between DNAstrands self-assembled at gold electrodes and an electron transfer protein, cytochrome c. Weobserved that cytochrome c exhibited well-defined electrochemistry in both double-strandedand single-stranded DNA films. This suggested that the electron transfer reaction ofcytochrome c arose possibly due to the electron hopping along DNA strands rather thanwiring along the double helix. We also compared the heterogeneous electron transfer rate ofcytochrome c with that of a ruthenium complex, which further confirmed this mechanism.

Abstract:This work used molecular modeling and experimental measurements to explore electron transfer between bacterial cytochromes and nanomaterials. The results highlight the importance of outer‐membrane protein configuration and active center microenvironment in governing electron transfer processes. Site‐directed mutagenesis can adjust electron transfer rates, enhancing the understanding of microbe‐nanostructure interactions for improved biotic‐linked‐abiotic bioelectrocatalytic system design. Electroactive microbial cells have evolved unique extracellular electron transfer to conduct the reactions via redox outer‐membrane (OM) proteins. However, the electron transfer mechanism at the interface of OM proteins and nanomaterial remains unclear. In this study, the mechanism for the electron transfer at biological/inorganic interface is investigated by integrating molecular modeling with electrochemical and spectroscopic measurements. For this purpose, a model system composed of OmcA, a typical OM protein, and the hexagonal tungsten trioxide (h‐WO3) with good biocompatibility is selected. The interfacial electron transfer is dependent mainly on the special molecular configuration of OmcA and the microenvironment of the solvent exposed active center. Also, the apparent electron transfer rate can be tuned by site‐directed mutagenesis at the axial ligand of the active center. Furthermore, the equilibrium state of the OmcA/h‐WO3 systems suggests that their attachment is attributed to the limited number of residues. The electrochemical analysis of OmcA and its variants reveals that the wild type exhibits the fastest electron transfer rate, and the transient absorption spectroscopy further shows that the axial histidine plays an important role in the interfacial electron transfer process. This study provides a useful approach to promote the site‐directed mutagenesis and nanomaterial design for bioelectrocatalytic applications.

Abstract:The function of protein in long-range biological electron transfer is a question of debate. We report some preliminary results in femtosecond spectroscopic study of photosynthetic bacterial light-harvesting antenna complex assembled onto TiO2 nanoparticle with an average size of 8 nm in diameter. Crystal structure shows that photosynthetic bacterial antenna complex LH2 has a ring-like structure composed by α- and β-apoprotein helices. The α- and β-transmembrance helices construct two concentric cylinders with pigments bacteriochlorophyll a (Bchl a) and carotenoid (Car) buried inside the protein. We attempt to insert TiO2 nanoparticle into the cavity of the inner cylindrical hollow of LH2 to investigate the nature of the electron transfer between the excited-state Bchl a and the TiO2 nanoparticle. A significant decrease in the ground state bleaching recovery time constant for Bchl a at 850 nm (B850) in respect to that of the Bchl a in free LH2 has been observed. By using the relation of distance-dependent long-range electron transfer rate in protein, the distance between the donor B850 and the acceptor TiO2 nanoparticle has been estimated, which is about 0.6 nm. The proposed method of assembling proteins onto wide-gap semiconductor nanoparticle can be a promising way to determine the role of the protein playing in biological electron transfer processes.

Abstract:To investigate the electron transfer properties between metal centers and its influencing factors in binuclear and trinuclear cyanido-bridged mixed valence complexes with cis/trans-configuration, binuclear cis-[Cp(dppe)Fe(μ-NC)Ru(5,5'-dmbpy)2(μ-CN)][PF6]n (cis-1[PF6]n, n = 1, 2) and trinuclear cis/trans-[Cp(dppe)Fe(μ-NC)Ru(5,5'-dmbpy)2(μ-CN)Fe(dppe)Cp][PF6]n (cis/trans-2[PF6]n, n = 2, 3, 4) (Cp = 1,3-cyclopentadiene, dppe = 1,2-bis(diphenylphosphino)ethane, 5,5'-dmbpy = 5,5'-dimethyl-2,2'-bipyridyl) cyanido-bridged complexes were synthesized and well characterized. The experimental results indicate that the presence of the other terminal fragment Cp(dppe)FeIII results in the higher MMCT energy in cis-2[PF6]4 than in cis-1[PF6]2. In addition, the trans-configuration are conductive to electron transfer between metal centers than the cis-configuration in trinuclear cyanido-bridged mixed valence complexes. Moreover, these mixed valence complexes cis-1[PF6]2 and cis/trans-2[PF6]n (n = 3, 4) could be assigned to Class II systems according to Robin and Day.

Abstract:The electrochemical proton reactivity of transition metal complexes receives significant attentions. A thorough understanding of proton-coupled electron transfer (PCET) pathways is essential for elucidating the mechanism behind a proton reduction reaction, and controlling the pathway is a key focus in the field of the catalyst development. Spin interactions within complexes, which arise during electron transfer, can affect significantly the PCET pathway. Herein, we explore the phenomenon of spin rearrangement during the electrochemical reorganization of high-spin cobalt complexes. Our findings reveal that opposing spin interactions, induced by different coordination environments, can alter the PCET pathway. Finally, detailed analysis of the PCET pathway allows us to propose mechanisms for proton reduction in high-spin cobalt complexes.

Abstract:Distinguishing proton-coupled electron transfer (PCET) from uncoupled electron transfer (ET) in proteins can be challenging. A recent investigation [J. C. Koone, M. Simmang, D. L. Saenger, M. L. Hunsicker-Wang and B. F. Shaw, J. Am. Chem. Soc. , 145 , 16488–16497] reported that protein charge ladders and capillary electrophoresis can distinguish between single ET, PCET, and two-proton coupled ET (2PCET) by directly measuring the change in protein net charge upon reduction/oxidation (Δ Z ET ). The current study used similar methods to assess PCET in zinc-free, "double copper" superoxide dismutase-1 (4Cu-SOD1), where one copper is bound at the copper site of each monomer and one copper is bound at the bridging zinc site, resulting in a quasi-type III Cu center. At pH 7.4, the net charge ( Z ) of the 4Cu-SOD1 dimer was unaffected by reduction of all four Cu 2+ ions, i.e. , Δ Z 4ET = −0.09 ± 0.05 per dimer (−0.02 ± 0.01 per copper atom). These values suggest that PCET is taking place at all four Cu atoms of the homodimer. Molecular dynamics and Poisson–Boltzmann calculations suggest that a metal-coordinating histidine at the zinc site (His71) is the proton acceptor. These data show how ligands of a naturally occurring zinc site can help facilitate PCET when the right redox metal is bound.

Abstract:Two recent investigations into electron trans fer reactions are described. The first study probes solvent effects on intramolecular electron transfer of donor-bridge-acceptor systems. The study compares a molecule which possesses a molecular cleft between the donor and acceptor moiety to one without such a cleft. The influence on the electronic coupling of a solvent molecule between the donor and acceptor is discussed. The second study concerns heterogeneous electron transfer through self-assembled monolayer films on InP electrodes. The data show that the film inhibits oxidation of the semiconductor. Electron tunneling through the film is explored, and the chemical functionalization of the film is shown to modify the charge recombination processes in the electrode.
Influence of the Interdomain Interface on Structural and Redox Properties of Multiheme Proteins

Abstract:Multiheme proteins are important in energy conversion and biogeochemical cycles of nitrogen and sulfur. A diheme cytochrome c4 (c4) was used as a model to elucidate roles of the interdomain interface on properties of iron centers in its hemes A and B. Isolated monoheme domains c4-A and c4-B, together with the full-length diheme c4 and its Met-to-His ligand variants, were characterized by a variety of spectroscopic and stability measurements. In both isolated domains, the heme iron is Met/His-ligated at pH 5.0, as in the full-length c4, but becomes His/His-ligated in c4-B at higher pH. Intradomain contacts in c4-A are minimally affected by the separation of c4-A and c4-B domains, and isolated c4-A is folded. In contrast, the isolated c4-B is partially unfolded, and the interface with c4-A guides folding of this domain. The c4-A and c4-B domains have the propensity to interact even without the polypeptide linker. Thermodynamic cycles have revealed properties of monomeric folded isolated domains, suggesting that ferrous (FeII), but not ferric (FeIII) c4-A and c4-B, is stabilized by the interface. This study illustrates the effects of the interface on tuning structural and redox properties of multiheme proteins and enriches our understanding of redox-dependent complexation.

Abstract:The physiological electron-transfer (ET) partners, cytochrome c peroxidase (CcP) and cytochrome c (Cc)1, can be modified to exhibit photoinitiated ET through substitution of Zn (or Mg) for Fe in either partner. Laser excitation of the Zn-porphyrin (ZnP) to its triplet excited state (3ZnP) initiates direct heme-heme ET to the ferriheme center of its partner across the protein-protein interface. This photoinitiated ET produces the charge-separated intermediate, I = [ZnP+CcP, Fe2+Cc], with a metalloporphyrin pi-cation radical (ZnP+) in the donor protein and a ferroheme acceptor protein. I, in general, is thought to return to the ground state by a thermal ET process that involves direct heme-heme back-ET to complete a simple photocycle. We here contrast intracomplex ET between yeast iso-1 Cc and ZnCcP(WT) (wild-type) with that for two ZnCcP(X) variants: X = W191F, with redox-active W191 replaced by Phe; WYM4, a W191F mutant with further replacement of four other potentially redox-active sites (W51F, Y187F, Y229F, and Y236F). The results show that W191 acts as an ET mediator, which "short-circuits" the direct heme-heme back-ET through a two-step, hopping process in which the ZnP+ cation radical formed by photoinitiated ET rapidly oxidizes W191, and the resultant W191+, in turn, rapidly oxidizes Fe2+Cc.

Abstract:Electron transfer through organic molecules is affected by several factors. The influence of electrodes, the interface between the organic molecule and electrodes, the environment and the molecular structure should be considered. In this paper, we reviewed theoretical and experimental studies of molecular electron transfer, such as the molecular dynamics simulations on the formation of the metallic electrodes, quantum chemistry studies on molecular conformation and conductance, and experimental studies of electron transfer in molecular junction using scanning probe microscopy and electrochemistry. Among the experimental methods, microscopic techniques such as scanning probe microscopy have been developed to study molecular electron transfer. Alternatively, macroscopic measurements have also been instructive to obtain the microscopic structural information.

Abstract:Decades of research describe myriad redox enzymes that contain cofactors arranged in tightly packed chains facilitating rapid and controlled intra‐protein electron transfer. Many such enzymes participate in extracellular electron transfer (EET), a process which allows microorganisms to conserve energy in anoxic environments by exploiting mineral oxides and other extracellular substrates as terminal electron acceptors. In this work, we describe the properties of the triheme cytochrome PgcA from Geobacter sulfurreducens. PgcA has been shown to play an important role in EET but is unusual in containing three CXXCH heme binding motifs that are separated by repeated (PT)x motifs, suggested to enhance binding to mineral surfaces. Using a combination of structural, electrochemical, and biophysical techniques, we experimentally demonstrate that PgcA adopts numerous conformations stretching as far as 180 Å between the ends of domains I and III, without a tightly packed cofactor chain. Furthermore, we demonstrate a distinct role for its domain III as a mineral reductase that is recharged by domains I and II. These findings show PgcA to be the first of a new class of electron transfer proteins, with redox centers separated by some nanometers but tethered together by flexible linkers, facilitating electron transfer through a tethered diffusion mechanism rather than a fixed, closely packed electron transfer chain.
