Abstract:Significant progress has been made on understanding the critical role of organic components in directing the collagen mineralization.We hypothesize that the inorganic trace elements might also play important role in the mineralization of collagenous matrix.To this aim,we systematically compared the in-vitro biomineralization behaviors of gelatin,gelatin-HA and gelatin-SiHA electrospun membranes.The results indicated that the presence of Si ions played a striking influence on the nucleation behaviors and mineralized structures.The gelatin-SiHA samples demonstrated more homogeneous nucleation within the gelatin fiber and growth along the fiber direction,in comparison with the heterogeneous nucleation and growth of spherulitic clusters on top of the nanofiber surface,i.e.extrafibrillar mineralization.The likely shift of the nucleation mode to the intrafibrillar mineralization in the presence of Si ions led to good alignment of apatite c-axis with the long axis of the nanofiber,resulting in a mineralization process and microstructure that were closer to those in natural bone.Cellular response analysis indicated that Si incorporation improved the MSC attachment and cytoskeleton organization.Such findings might have important implication in both understanding the complex mechanisms involved in collagen mineralization and optimal designing of advanced bio-inspired materials with potential superior mechanical and biological properties.

Abstract:Gelatin methacryloyl (GelMA) has become a popular biomaterial in the field of bioprinting. The derivation of this material is gelatin, which is hydrolyzed from mammal collagen. Thus, the arginine-glycine-aspartic acid (RGD) sequences and target motifs of matrix metalloproteinase (MMP) remain on the molecular chains, which help achieve cell attachment and degradation. Furthermore, formation properties of GelMA are versatile. The methacrylamide groups allow a material to become rapidly crosslinked under light irradiation in the presence of a photoinitiator. Therefore, it makes great sense to establish suitable methods for synthesizing three-dimensional (3D) structures with this promising material. However, its low viscosity restricts GelMA's printability. Presented here are methods to carry out 3D bioprinting of GelMA hydrogels, namely the fabrication of GelMA microspheres, GelMA fibers, GelMA complex structures, and GelMA-based microfluidic chips. The resulting structures and biocompatibility of the materials as well as the printing methods are discussed. It is believed that this protocol may serve as a bridge between previously applied biomaterials and GelMA as well as contribute to the establishment of GelMA-based 3D architectures for biomedical applications.
Engineering the Viscoelasticity of Gelatin Methacryloyl (gelma) Hydrogels Via Small “dynamic Bridges” to Regulate BMSC Behaviors for Osteochondral Regeneration

Abstract:The dynamic extracellular matrix (ECM) constantly affects the behaviors of cells. To mimic the dynamics of ECM with controllable stiffness and energy dissipation, this study proposes a strategy in which a small molecule, 3,4-dihydroxybenzaldehyde (DB), was used as fast “dynamic bridges'’ to construct viscoelastic gelatin methacryloyl (GelMA)-based hydrogels. The storage modulus and loss modulus of hydrogels were independently adjusted by the covalent crosslinking density and by the number of dynamic bonds. The hydrogels exhibited self-healing property, injectability, excellent adhesion and mechanical properties. Moreover, the in vitro results revealed that the viscous dissipation of hydrogels favored the spreading, proliferation, osteogenesis and chondrogenesis of bone marrow mesenchymal stem cells (BMSCs), but suppressed their adipogenesis. RNA-sequencing and immunofluorescence suggested that the viscous dissipation of hydrogels activated Yes-associated protein (YAP) by stabilizing integrin β1, and further promoted nuclear translocation of smad2/3 and β-catenin to enhance chondrogenesis and osteogenesis. As a result, the viscoelastic GelMA hydrogels with highest loss modulus showed best effect in cartilage and subchondral bone repair. Taken together, findings from this study reveal an effective strategy to fabricate viscoelastic hydrogels for modulating the interactions between cells and dynamic ECM to promote tissue regeneration.

Abstract:This study develops a novel gelatin hydrogel through microstructural optimization. The resulting hydrogel exhibits superior mechanical properties compared to conventional methacrylated gelatin (GelMA) gels while preserving key benefits like rapid gelation, cell adhesion, and complete biodegradability. These combined properties significantly enhance the precision and complexity of gelatin hydrogels for 3D printing and their applications in biomedicine. Gelatin hydrogels (e.g., methacrylated gelatin gel, abbreviated GelMA gel) have garnered significant attention in tissue engineering and therapeutic drug and cell delivery due to their complete degradability and intrinsic ability to support cell adhesion. However, their practical applications are often constrained by their poor mechanical performance, which stems from their single network structure. This limitation poses significant challenges in load‐bearing scenarios and restricts their use in advanced biofabrication technologies, where robust mechanical properties are essential. Here a hydrogel is developed composed entirely of gelatin using a phototriggered transient‐radical and persistent‐radical coupling (PTPC) reaction to achieve an optimized microstructure. This hydrogel features a phase‐separated structure with enhanced interfacial bonding, significantly improving mechanical performance compared to conventional GelMA gels. Notably, this approach preserves the inherent properties of gelatin, including biocompatibility, cell adhesion, and degradability, thereby extending its applicability in the biomedical field, particularly in advanced biofabrication methods such as 3D printing. This approach offers a superior solution to meet the complex demands of sophisticated biomanufacturing technologies, expanding the potential applications of gelatin hydrogels in the biomedical field.

Rheological Properties of Coordinated Physical Gelation and Chemical Crosslinking in Gelatin Methacryloyl (GelMA) Hydrogels.

Abstract:Synthetically modified proteins, such as gelatin methacryloyl (GelMA), are growing in popularity for bioprinting and biofabrication. GelMA is a photocurable macromer that can rapidly form hydrogels, while also presenting bioactive peptide sequences for cellular adhesion and proliferation. The mechanical properties of GelMA are highly tunable by modifying the degree of substitution via synthesis conditions, though the effects of source material and thermal gelation have not been comprehensively characterized for lower concentration gels. Herein, the effects of animal source and processing sequence are investigated on scaffold mechanical properties. Hydrogels of 4-6 wt% are characterized. Depending on the temperature at crosslinking, the storage moduli for GelMA derived from pigs, cows, and cold-water fish range from 723 to 7340 Pa, 516 to 3484 Pa, and 294 to 464 Pa, respectively. The maximum storage moduli are achieved only by coordinated physical gelation and chemical crosslinking. In this method, the classic thermo-reversible gelation of gelatin occurs when GelMA is cooled below a thermal transition temperature, which is subsequently "locked in" by chemical crosslinking via photocuring. The effects of coordinated physical gelation and chemical crosslinking are demonstrated by precise photopatterning of cell-laden microstructures, inducing different cellular behavior depending on the selected mechanical properties of GelMA.

Seeded Mineralization In Silk Fibroin Hydrogel Matrices Leads To Continuous Rhombohedral Caco3 Films

Abstract:As many biominerals are formed in gel-like media, hydrogel-mediated mineralization is deemed as paradigms of biomineralization and ideal approaches to synthetic minerals with hierarchical architectures and related functions. Nevertheless, the long diffusion distance in hydrogels makes mineralization a diffusion-limited process, leading to isolated crystals instead of uniform hierarchical architectures. In the current study, seeded mineralization in silk fibroin hydrogel matrices is successful in delivering continuous rhombohedral CaCO3 films. Though the coverage of hydrogel matrices makes mineralization a diffusion-limited process, the presence of seed layers promotes the growth of uniform overlayers in proper conditions. The regulation of the solid content of hydrogels provides a rational route to rhombohedral architectures with tunable morphologies and thickness. In the course of mineralization, the hydrogel matrices are partially occluded in rhombohedral films as inter- and intra-crystalline constituents, as confirmed by scanning and transmission electron microscopy. Our study confirms the availability of synthesizing continuous mineralized films with hierarchical architectures and the structural gradient in hydrogel matrices via self-organized mineralization. These films with the occlusion of hydrogel constituents may exhibit significant strength and resilience, and their formation can deepen our mechanistic understanding of biomineralization proceeding in gel-like media.
Stiff and Tough Hydrogels Prepared Through Integration of Ionic Cross-linking and Enzymatic Mineralization

Abstract:Enzymatic mineralization has become an effective approach to enhancing the stiffness of hydrogels for bone tissue engineering, but generally with limited toughness. On the other hand, double network cross-linking provides hydrogel with enhanced toughness. In this study, we integrated double cross-linking method with enzymatic mineralization to synthesize stiff and tough hydrogels. We have synthesized three kinds of sodium alginate-polyacrylamide (SA-PAM) double-network hydrogels and systematically compared the composition and structure differences, mechanical properties, and biological properties of the different hydrogels in the absence and presence of mineralization. In particular, we examined the role of specific cross-linking ions, i.e., calcium, zinc and strontium ions, in modulating the mineralization process. Synergistic effect of ionic cross-linking and enzymatic mineralization was clearly observed with dramatic increase in compressive modulus. In particular, mineralized hydrogel cross-linked with Sr2+ showed the highest compressive Young's modulus of 17.28 ± 3.56 MPa, which was 37 times of that of the original hydrogel. In addition, it had the highest tensile Young's modulus at 2.60 ± 0.25 MPa and 84 ± 5.5% elongation at break. Such synergistic effect from Sr2+ was attributed to a more uniformed mineralization process due to the early initiation of a more homogeneous nucleation process and subsequent denser mineralized structure. Cellular study also suggested that cell proliferation, adhesion and osteogenic differentiation were improved as a result of enzymatic mineralization. Our results provided an effective way for the preparation of stiff and tough hydrogels with osteogenesis, and demonstrated potential in bone tissue engineering applications.Statement of significanceHydrogels with excellent stiffness, stability and biocompatibility have attracted significant attentions in the bone tissue engineering applications. Our results suggested that the synergistic effect of ionic cross-linking and enzymatic mineralization rendered more enhancement of the compressive and tensile stiffness of SA-PAM DN hydrogels, as well as the toughness, swelling stability and cellular response. In particular, mineralized hydrogel cross-linked with Sr2+ showed the highest compressive Young's modulus of 17.28 ± 3.56MPa, which was 37 times of that of the original hydrogel. Such synergistic effect from Sr2+ was attributed to a more uniformed mineralization process. Abstract:Biomineralization of calcium carbonate, an important inorganic biomaterial, has been extensively studied. In this paper, calcium carbonate was synthesized by biomineralization using gelatin as the matrix. The structural and morphological properties of the resulting CaCO3 materials were characterized by field emission scanning electron microscopy (FESEM), X-ray diffraction (XRD), and Fourier-transform infrared (FT-IR) spectroscopy. The effects of varying the concentration of gelatin, the concentration of calcium ions and the reaction time were investigated. The results showed that the structures of the product are controlled by the ratio of the concentration of gelatin to calcium ions. As the ratio of the concentration of gelatin to calcium ions was gradually decreased, the compact spherical CaCO3 morphology changed to double-snail-like, spherical-like and finally loose spherical CaCO3 aggregates. At higher gelatin to calcium ion rations, vaterite was the stable phase, and the formation of CaCO3 with concave holes was accompanied by a phase transition from vaterite to calcite at lower ratios.

Abstract:Biomineralization approaches have been increasingly adopted to synthesizing advanced materials with superior properties. Nevertheless, the potential influence of inorganic trace elements on the mineralization process of collagen has been rarely reported, despite of the significant progress achieved on exploiting the critical roles of organic polymers in regulating the collagen mineralization. To this aim, the potential roles of Si, Zn and Sr in regulating the mineralization of gelatin-hydroxyapatite (HA) composite fibers have been examined in this study. The results indicated that the incorporation of trace elements not only promoted the biomineralization of gelatin, but also led to drastic change in the mineralization behavior. In particular, the gelatin-SiHA sample showed uniform mineralization predominantly inside the fibers, with nucleation and growth directions along the c-axis of the gelatin fibers. On the contrary, the gelatin-HA sample showed nucleation outside the fibers and spherical mineral crystals on top of fibers, typical structure for heterogeneous nucleation. As the mineralization process proceeded, the gelatin-ZnHA and gelatin-SrHA samples evolved into having similar structure as the gelatin-SiHA sample, despite of showing totally different mineralization behaviors at early time. Overall, the incorporation of trace elements seemed to lower the nucleation barriers, led to a more homogeneous mineralization mode within the fiber region and formation of mineralized structures closer to those in natural bone. Moreover, mineralized samples with trace elements demonstrated improved adhesion and cytoskeleton organization of osteoblastic cells. Such finding would provide important insight for understanding the mineralization process and the optimal design of advanced biological materials.
Templated mineralization of synthetic hydrogels for bone-like composite materials: role of matrix hydrophobicity.

Abstract:Bone-mimetic mineral-polymer composite materials have several applications ranging from artificial bone grafts to scaffolds for bone tissue engineering; templated mineralization is an effective approach to fabricate such composites. In this study, we synthesized bone-like composites using synthetic hydrogels having pendant side chains terminating with carboxyl groups as a template for mineralization. The role of matrix hydrophobicity on mineralization was examined using poly(ethylene glycol) hydrogels modified with varying lengths of anionic pendant side chains (CH2=CHCONH(CH2)(n)COOH, where n = 1, 3, 5, and 7). The ability of these hydrogels to undergo templated mineralization was found to be strongly dependent upon the length of the pendant side chain as is evident from the extent of calcification and morphology of the minerals. Moreover, mineralized phases formed on the hydrogels were confirmed to resemble apatite-like structures. In addition to demonstrating the importance of material hydrophobicity as a design parameter for the development of bone-like synthetic materials, our study also provides a potential explanation for the in vitro differences between the apatite-nucleating capacity of aspartate-rich osteopontin and glutamate-rich bone sialoprotein.
Biomimetic Mineralized Hydroxyapatite Nanofiber-Incorporated Methacrylated Gelatin Hydrogel with Improved Mechanical and Osteoinductive Performances for Bone Regeneration

Abstract:Purpose: Methacrylic anhydride-modified gelatin (GelMA) hydrogels exhibit many beneficial biological features and are widely studied for bone tissue regeneration. However, deficiencies in the mechanical strength, osteogenic factors and mineral ions limit their application in bone defect regeneration. Incorporation of inorganic fillers into GelMA to improve its mechanical properties and bone regenerative ability has been one of the research hotspots.Methods: In this work, hydroxyapatite nanofibers (HANFs) were prepared and mineralized in a simulated body fluid to make their components and structure more similar to those of natural bone apatite, and then different amounts of mineralized HANFs (m-HANFs) were incorporated into the GelMA hydrogel to form m-HANFs/GelMA composite hydrogels. The physicochemical properties, biocompatibility and bone regenerative ability of m-HANFs/GelMA were determined in vitro and in vivo.Results: The results indicated that m-HANFs with high aspect ratio presented rough and porous surfaces coated with bone-like apatite crystals. The incorporation of biomimetic m-HANFs improved the biocompatibility, mechanical, swelling, degradation and bone regenerative performances of GelMA. However, the improvement in the performance of the composite hydrogel did not continuously increase as the amount of added m-HANFs increased, and the 15m-HANFs/GelMA group exhibited the best swelling and degradation performances and the best bone repair effect in vivo among all the groups.Conclusion: The biomimetic m-HANFs/GelMA composite hydrogel can provide a novel option for bone tissue engineering in the future; however, it needs further investigations to optimize the proportions of m-HANFs and GelMA for improving the bone repair effect.

Abstract:At present, the treatment of bone defect is one of the most concerned problems in biomedical fields. Despite the wide variety of scaffolds, there is a challenge to select materials that can mimic the structural integrity and biocompatibility of natural bone. In our study, gelatin methacryloyl (GelMA) and sodium alginate (Alg) were used to prepare three-dimensional (3D) GelMA/Alg hybrid hydrogel, which can simulate the structure and biological function of natural extracellular matrix due to their high water content and porous structure. The interconnected and homogeneous pores of the scaffold facilitate the transport of nutrients during the bone regeneration. Then hydroxyapatite (HA) coated GelMA/Alg (GelMA/Alg-HA) hydrogel was obtained by sequential mineralization. The mineralized hydrogel was obtained by immersing hydrogel alternately in a solution of calcium and phosphorus at 37 °C. The hydrogel was modified with a coating of HA under a mild condition. The calcium crosslinked Alg could provide nucleation sites for HA crystals. And the sequential mineralization will improve the physical properties and osteoinductivity of the hydrogels by introducing HA, which is similar to the mineral component of natural bone. Analytical results confirmed that the HA particles were uniformly distributed in the surface of the hydrogels and the mineral contents were about 40% after three cycles. The compressive strength was improved from 22.43 ± 6.39 to 131.03 ± 9.26 kPa. In addition, MC3T3-E1 cell co-culture experiments shown that the mineralized GelMA/Alg-HA hybrid hydrogel possess good biocompatibility, which is conducive to the growth of new bone tissue and bone repair.
Biomimetically-mineralized Composite Coatings on Titanium Functionalized with Gelatin Methacrylate Hydrogels

Abstract:Immobilizing organic-inorganic hybrid composites onto the implant surface is a promising strategy to improve host acceptance of the implant. The objective of this present study was to obtain a unique macroporous titanium-surface with the organic-mineral composite coatings consisting of gelatin methacrylate hydrogel (GelMA) and hydroxyapatite (HA). A 3-(trimethoxysilyl) propyl methacrylate (TMSPMA) layer was first coated onto the titanium surface, and surface was then covalently functionalized with GelMA using a photochemical method. Mineralization of the GelMA coating on the titanium surface was subsequently carried out by a biomimetic method. After 3-day mineralization, a large number of mineral phases comprising spherical amorphous nanoparticles were found randomly deposited inside GelMA matrix. The resulting mineralized hydrogel composites exhibited a unique rough surface of macroporous structure. The structure of the prepared GelMA/HA composite coating was studied by field emission scanning electron microscopy (FESEM), energy dispersive X-ray spectra (EDS), attenuated total refraction Fourier transform infrared spectroscopy (ATR-FTIR), X-ray photoelectron spectroscopy (XPS) and X-ray diffraction (XRD). Water contact angle measurement revealed the hydrophilicity properties of composite coatings. GelMA/HA on titanium after the TMSPMA treatment is very stable when tested in vitro with a PBS solution at 37 degrees C, due to the role of TMSPMA as a molecular bridge. It was expected that the macroporous GelMA/HA composite coatings might potentially promote and accelerate titanium (Ti)-based implants osseointegration for bone repair and regeneration. Abstract:Natural polymers and synthetic polymers have been extensively studied as scaffold materials, with the former offering advantages such as biocompatibility, biodegradability, and structural similarity to the natural extracellular matrix (ECM). However, the use of natural polymers in extrusion-based 3D printing has been limited by their poor mechanical properties and challenging rheological properties. In this study, gelatin and sodium alginate were utilized as scaffold materials, with the addition of Ca2+ and Mg2+ components to enhance their physical and chemical properties, and influence early cell behavior. Subsequently, these materials were fabricated into scaffolds using 3D printing. Our results demonstrated that the addition of Ca2+ and Mg2+ could improve the compactness of the 3D network structure, mechanical strength, swelling properties and degradation properties of methacrylated gelatin/methacrylated sodium alginate (GelMA/SAMA) composite hydrogel. In vitro cell tests revealed that the GelMA/SAMA composite hydrogel exhibited negligible cytotoxicity and promoted early cell viability, particularly with the higher concentration of Mg2+ in the material. Notably, the extrusion 3D printing process successfully produced GelMA/SAMA scaffolds. These results collectively indicate that GelMA/SAMA composite scaffolds hold promise as potential biomaterials for tissue engineering applications.
Biomineralized Hydrogel with Enhanced Toughness by Chemical Bonding of Alkaline Phosphatase and Vinylphosphonic Acid in Collagen Framework

Abstract:Alkaline phosphatase (ALP) and phosphoprotein participation in collagen mineralization is one of the most important physiological processes of bone formation. Simulating the natural mineralization process with the involvement of ALP and phosphoproteins is a powerful tool for the preparation of bone repair scaffolds. Searching for compatible approaches to chemically bond ALP with collagen molecules and introducing phosphoprotein-like molecules into a collagen network is the challenge of the day. Here, we synthesized alkaline phosphatase methacrylamide (ALP-MA) via amidation reaction to enable ALP to be grafted uniformly into the photo-cross-linking collagen gel, achieving homogeneous enzymatic mineralization. Furthermore, vinylphosphonic acid (VAP), a phosphoprotein-like molecule containing phosphonate groups, was successfully introduced on collagen molecules through photo cross-linking, playing the role of a phosphoprotein for inducing mineralized CaP clusters deposited on the collagen backbone. Hence, a hydrogel termed CAV was synthesized via photochemical reaction among collagen methacrylamide (Col-MA), ALP MA, and VAP (active mineral bonding site) for in situ mineralization. We found that the binding between the collagen network and CaP clusters would lead to the generation of mechanically enhanced mineralized collagen hydrogel. Encapsulated bone marrow stromal cells (BMSCs) exhibited good growth and proliferation in the in situ enzymatic mineralization process. Additionally, the simulated mineralization system is highly favorable for micropatterned structure construction and 3D bioprinting. In brief, we designed a novel approach to effectively simulate the physiological mineralization process of bone formation. The approach, incorporating great photo-cross-linking performance and enzymatic mineralization activity, was beneficial for in situ cell encapsulation and excellent cell-compatible 3D printing, holding great promise for bone tissue engineering research.

Abstract:Gelatin methacryloyl (GelMA) is widely used for the formulation of hydrogels in diverse biotechnological applications. After the derivatization of raw gelatin, the degree of functionalization (DoF) is an attribute of particular interest as the functional residues are necessary for crosslinking. Despite progress in the optimization of the process found in the literature, a comparison of the effect of raw gelatin on the functionalization is challenging as various approaches are employed. In this work, the modification of gelatin was performed at room temperature (RT), and eight different gelatin products were employed. The DoF proved to be affected by the bloom strength and by the species of gelatin at an equal reactant ratio. Furthermore, batch-to-batch variability of the same gelatin source had an effect on the produced GelMA. Moreover, the elasticity of GelMA hydrogels depended on the DoF of the protein as well as on bloom strength and source of the raw material. Additionally, GelMA solutions were used for the microfluidic production of droplets and subsequent crosslinking to hydrogel. This process was developed as a single pipeline at RT using protein concentrations up to 20% (w/v). Droplet size was controlled by the ratio of the continuous to dispersed phase. The swelling behavior of hydrogel particles depended on the GelMA concentration.
On the Structure Alteration of Crosslinkable Gelatin Coupled with Methacrylic Acid and Its Hydrogel

Abstract:Physical structures of a crosslinkable gelatin derivative (GM) were studied in terms of alteration of apparent molecular weight, triple helix content and mechanical strength. The GM with a substitution degree (DS) of 49% and 79% was prepared by grafting mechacrylic acid (MA), which was able to form injectable hydrogel by photoinitiating polymerization. The zeta potential was increased along the increase of DS. After modification, the apparent number-average molecular weight (M n ) detected by gel permeation chromatography was decreased to about 2/3 of gelatin, while the apparent weight-average molecular weight (M w ) was changed within a small range. Differential scanning calorimetry and circular dichroism (CD) revealed that ability of triple-helix formation of GM was decreased along with the increase of DS and decrease of GM concentration. After photocrosslinking, the sol-gel transition of GM49 physical-chemical hydrogel still existed, but completely disappeared for its chemical hydrogel. The physical-chemical hydrogel showed a larger storage modulus at 20°C than at 37°C as a result of additional physical crosslinking.

Abstract:Based on the basic principles of biomineralization, the CaCO3/gelatin composite can be synthesized by the biomimetic synthesis under dynamic conditions, using gelatin as the matrix. The results of scanning electron microscopy with an energy dispersive X-ray analysis system (SEM-EDAX) showed that the synthesized composite, which has special microstructure and morphology and certain orientation, was different completely from the calcium carbonate that was formed in the water. The morphology, orientation and contents of the main elements (calcium, oxygen and nitrogen) of crystal are quite different for various content of the gelatin.

Abstract:Mineralization of hydrogels, desirable for bone regeneration applications, may be achieved enzymatically by incorporation of alkaline phosphatase (ALP). ALP-loaded gellan gum (GG) hydrogels were mineralized by incubation in mineralization media containing calcium and/or magnesium glycerophosphate (CaGP, MgGP). Mineralization media with CaGP:MgGP concentrations 0.1:0, 0.075:0.025, 0.05:0.05, 0.025:0.075 and 0:0.1 (all values mol/dm3 , denoted A, B, C, D and E, respectively) were compared. Mineral formation was confirmed by IR and Raman, SEM, ICP-OES, XRD, TEM, SAED, TGA and increases in the the mass fraction of the hydrogel not consisting of water. Ca was incorporated into mineral to a greater extent than Mg in samples mineralized in media A-D. Mg content and amorphicity of mineral formed increased in the order A < B < C < D. Mineral formed in media A and B was calcium-deficient hydroxyapatite (CDHA). Mineral formed in medium C was a combination of CDHA and an amorphous phase. Mineral formed in medium D was an amorphous phase. Mineral formed in medium E was a combination of crystalline and amorphous MgP. Young's moduli and storage moduli decreased in dependence of mineralization medium in the order A > B > C > D, but were significantly higher for samples mineralized in medium E. The attachment and vitality of osteoblastic MC3T3-E1 cells were higher on samples mineralized in media B-E (containing Mg) than in those mineralized in medium A (not containing Mg). All samples underwent degradation and supported the adhesion of RAW 264.7 monocytic cells, and samples mineralized in media A and B supported osteoclast-like cell formation. Comparison of globular albumin methacryloyl and random-coil gelatin methacryloyl: Preparation, hydrogel properties, cell behaviors, and mineralization

Abstract:Bovine serum albumin methacryloyl (BSAMA) is a newly emerging photocurable globular protein-based material whereas gelatin methacryloyl (GelMA) is one of the most popular photocurable fibrous protein-based materials. So far, the influence of their different structural conformations as building blocks on hydrogel properties and mineral deposition has not been investigated. Here, we compared their differences in structures, gelation kinetics, hydrogel properties, mineralization, and cell behaviors. BSAMA maintained a stable globular structure while GelMA exhibited temperature-sensitive conformations (4 - 37 °C). BSAMA displayed slower gelation kinetics and much more retarded enzymatic degradation compared to GelMA. Photocurable BSAMA (6.41 - 390.95 kPa) and GelMA hydrogels (36.09 - 199.70 kPa) exhibited tunable mechanical properties depending on their concentrations (10 - 20%). Interestingly, BSAMA hydrogels mineralized needle-like apatite (Ca/P: 1.409) with higher crystallinity compared to GelMA hydrogels (Ca/P: 1.344). BSAMA and GelMA supported satisfactory cell (MC3T3-L1) viability of 99.43 ± 0.57% and 97.14 ± 0.69%, respectively. However, BSAMA gels were less favorable to cell proliferation and migration than GelMA gels. In serum-free environments, cells on GelMA displayed a higher amount of attachment, a more elongated shape, and a longer protrusion compared to those on BSAMA (p < 0.01) during the early adhesion.
