Abstract:Because of the inherent difficulty in differentiating two olefins, the development of metal-catalyzed asymmetric cyclization of 1,6-dienes remains challenging. Herein, we describe the first rhodium(III)-catalyzed asymmetric borylative cyclization of cyclohexadienone-tethered mono-, 1,1di-, and (E)-1,2-disubstituted alkenes (1,6-dienes), affording optically pure cis-bicyclic skeletons bearing three or four contiguous stereocenters with high yields (25-93%), and excellent diastereoselectivities (>20:1 dr) and enantioselectivities (90-99% ee). This mild catalytic approach is generally compatible with a wide range of functional groups, which allows several facile conversions of the cyclization products. Furthermore, on the basis of our SAESI-MS experiment and computational study, a Rh(I)/(III) catalytic cycle is proposed in this tandem reaction, and the Rh(I) active species catalyzes the overall transformation via sequential oxidative addition of B(2)pin(2), olefin insertion, cyclizing conjugate addition, and reductive elimination. The irreversible conjugate addition determines the overall regioselectivity of borylative cyclization, and the ring strain favors the formation of 5,6-bicyclic structure. This highlights the control of ring strain in diene cyclizations, which provides a useful basis for future reaction designs.

Abstract:A systematic theoretical study of rhodium-catalyzed allylic alkylation reactions using hard benzylzinc nucleophiles was performed. The mechanisms were investigated in detail together with the regioselectivity associated with the branched and linear pathways and the intriguing anti - (as opposed to syn -) diastereoselectivity of this process. The preferred mechanism was found to be initiated by the oxidative addition of the rhodium(I) complex followed by migration of the benzyl nucleophile to the Rh center. The subsequent nucleophilic addition of a benzyl group to the η 3 -allyl moiety takes place via the formation of a new C–C bond to afford the final allylic benzylation product. This proposed mechanism readily explains the experimental observation that the products obtained from such reactions are branched (as opposed to linear) with an anti -configuration (as opposed to a syn -configuration). This work also provides reasons for the lower regioselectivity observed for a OCO 2 t Bu leaving group or cyanophenyl/secondary amide substituents at the 4-position of the substrate. The present study is expected to assist in the rational design of potential substrates for analogous reactions.

Abstract:The mechanism and origin of stereoselectivity of rhodium(II)-catalyzed cyclopropanation reactions with diazooxindole and styrene has been studied using density functional theory calculations. The catalyzed reactions by achiral Rh-2(OAc)(4) and chiral Rh-2(S-PTTL)(4) as well as the uncatalyzed model were comparatively studied. The computational results indicate that the cyclopropanation step in both Rh-2(OAc)(4) and Rh-2(S-PTTL)(4) models is a single concerted but asynchronous process. The nitrogen extrusion step is found to be the rate-limiting step of the catalytic cycle, whereas the cyclopropanation step is the stereoselectivity-determining step. The diastereomeric ratios (dr) and the enantiomeric excess (ee) values are successfully predicted, which are in good agreement with the experimental values. The high trans-diastereoselectivity might be governed by the pi-pi interactions between the syn indole ring in carbenoid ligand and the phenyl group in styrene, whereas the good enantioselectivity can be ascribed to the steric interaction between the phenyl ring in styrene and the phthalimido group in the catalyst as well as the aromatic interactions (pi-pi and CH-pi) in the transition states. Additionally, the methodological study using different functionals demonstrated the importance of considering the dispersion interactions in the current reaction systems. This theoretical study will help in understanding the mechanism of the asymmetric cyclopropanations of olefins through carbene-transfer reactions.

Abstract:In contrast with abundant methods for the asymmetric functionalization of alkyl radicals to generate stereogenic centres at reaction sites, catalytic enantioselective desymmetrizing functionalization of alkyl radicals for forging multiple stereocentres—including positions that are remote from the reaction sites—with both high enantio- and diastereoselectivity remains largely unexplored. The major challenge for such reactions is the high reactivity of open-shell alkyl radicals. Here, we describe a strategy to address this challenge: the use of Cu( ii ) phosphate to immediately associate with the in situ-generated reactive alkyl radical species, creating a compact and confined chiral microenvironment for effective stereocontrol. With this strategy, we have developed a general and efficient catalytic enantioselective desymmetrizing functionalization of alkene-tethered 1,3-diols. It provides various tetrahydrofurans and analogues bearing multiple stereocentres with remarkably high levels of enantio- and diastereocontrol. Density functional theory calculations and mechanistic experiments revealed a reaction mechanism involving an enantiodetermining outer-sphere C–O bond formation step.

Abstract:Described herein is an enantioselective dirhodium(II)-catalyzed cycloisomerization of diynes achieved by the strategy of desymmetrization, which not only represents a new cycloisomerization reaction of diynes but also constitutes the first Rh(II)-catalyzed asymmetric intramolecular cycloisomerization of 1,6-diynes. This protocol provides a range of valuable furan-fused dihydropiperidine derivatives with an enantiomerically enriched alkynyl-substituted aza-quaternary stereocenter in high efficiency, complete atom economy, and excellent enantioselectivity (up to 98% ee). Besides, the highly functionalized products could be easily transformed into various synthetically useful building blocks and conjugated with a series of pharmaceutical molecules. The mechanism involving a concerted [3+2] cycloaddition/[1,2]-H shift of the Rh(II) carbenoid intermediate was elucidated by DFT calculations and mechanistic studies. More importantly, the first single crystal of alkyne-dirhodium(II) was obtained to show that a η2-coordinating activation of alkynal by dirhodium(II) was involved. Weak hydrogen bondings between the carboxylate ligands and alkynal were found, which probably made the well-defined paddlewheel-like dirhodium(II) distinctive from other metal complexes in catalyzing this transformation. Abstract:The Rh(I)-catalyzed reaction of siloxyvinylcyclopropanes and diazoesters leads to the formation of siloxyvinylcyclobutane and 1,4-diene derivatives. With [Rh(cod)Cl]2 as the catalyst, the formation of 1,4-diene was favored over the formation of siloxyvinylcyclobutane. By changing the catalyst to [Rh(cod)2OTf], siloxyvinylcyclobutane derivatives are formed with excellent chemoselectivities and in moderate to good yields. The alkene products are also obtained as single E configured isomers. A detailed mechanism for this transformation is proposed on the basis of mechanistic experiments and DFT calculations. The effect of catalysts on the chemoselectivity of these reactions is also examined computationally.

Abstract:A highly chemo- and enantioselective hydrogenation of β-diketones was achieved by using [Ru(benzene)(S)-SunPhosCl]Cl for consistency in THF. The neighboring heteroatoms played important roles in guaranteeing the reactivity and controlling the chemoselectivity. These results suggested a potential approach for the clean and facile synthesis of functionalized chiral β-hydroxy ketones, which could otherwise be prepared through much less step-economic transformations.

Abstract:Ring strain plays an important role in metal-catalyzed cyclization of 1,6-dienes. Herein, we report a rhodium(III)-catalyzed asymmetric reductive cyclization of cyclohexadienone-tethered α,β-unsaturated compounds (1,6-dienes), including α,β-unsaturated ketones, esters, amides, sulfone, and phosphonate. The reactions undergo an unusual anti-Michael/Michael addition process, affording cis-bicyclic frameworks with good to high yields and good diastereo- and enantioselectivities. Abstract:Understanding how catalytic asymmetric reactions with racemic starting materials can operate would enable new enantioselective cross-coupling reactions that give chiral products. Here we propose a catalytic cycle for the highly enantioselective Rh(i)-catalysed Suzuki–Miyaura coupling of boronic acids and racemic allyl halides. Natural abundance 13C kinetic isotope effects provide quantitative information about the transition-state structures of two key elementary steps in the catalytic cycle, transmetallation and oxidative addition. Experiments with configurationally stable, deuterium-labelled substrates revealed that oxidative addition can happen via syn- or anti-pathways, which control diastereoselectivity. Density functional theory calculations attribute the extremely high enantioselectivity to reductive elimination from a common Rh complex formed from both allyl halide enantiomers. Our conclusions are supported by analysis of the reaction kinetics. These insights into the sequence of bond-forming steps and their transition-state structures will contribute to our understanding of asymmetric Rh–allyl chemistry and enable the discovery and application of asymmetric reactions with racemic substrates.

Abstract:One of the most widely employed strategies to produce chiral molecules involves the asymmetric hydrogenation of functionalized olefins using rhodium catalysts. Despite their excellent performance, the exact identity of the active Rh species is still ambiguous as each site may plausibly feature one or two phosphorus ligands. In this work, we used a sequential postsynthetic modification approach to successfully incorporate single-site Rh species into a zirconium-based metal-organic framework comprised of chiral spinol-based ligands. These Rh species feature one phosphorus ligand per Rh, which contrasts with the molecular analogue that contains two phosphorus ligands per Rh site. Following extensive characterization of the Rh-monophosphorus material using techniques including solid-state NMR and extended X-ray absorption fine-structure (EXAFS) spectroscopy, we studied their catalytic performance in the asymmetric hydrogenations of enamides and α-dehydroamino acid esters and observed excellent yields and enantioselectivities (up to 99.9% ee). Notably, the Rh-monophosphorus catalyst is 5 times more active than the homogeneous Rh-biphosphorus control, which we attributed to the higher activity of the single-site Rh-monophosphorus species and the confined MOF cavities that can enrich reactants. In addition, we observed a unique topology-dependent behavior in which linker expansion leads to the formation of a novel Zr-MOF with a distinct 4,8-connected net that cannot be phosphorylated, presumably due to intense tensile strain and steric repulsion present within this framework. Finally, we demonstrate the utility of this single-site Rh-monophosphorus catalyst in the gram-scale synthesis of (R)-cinacalcet hydrochloride, a first-in-class drug in the therapy of secondary hyperparathyroidism and parathyroid carcinoma, with 99.1% ee.

Abstract:The chiral dirhodium(ii) tetracarboxylate-catalyzed enantioselective intramolecular Buchner reaction of donor/donor-carbenes was reported and a series of valuable chiral polycyclic products were synthesized. Both aryloxy enynones and diazo compounds were efficient carbene precursors for this reaction. Excellent yields (up to 99%) and outstanding enantioselectivities (up to >99% ee) were achieved under standard conditions. For furyl substituted chiral cyclohepta[b]benzofurans bearing a substituent at the C4 position on cycloheptatrienes, control reactions showed that the chiral Buchner products could slowly racemize either under dark or natural light conditions. A diradical-involved mechanism rather than a zwitterionic intermediate was proposed to explain the racemization. Furthermore, furyl substituted chiral fluorene derivatives were obtained via asymmetric aromatic substitution when biaryl enynones were employed as carbene precursors.
