Abstract:1867 Objectives: An impediment to antisense (AS) imaging is poor delivery of “naked” DNAs across tumor cell membranes. Numerous carriers are under study to improve transport, often involving covalent conjugation to DNA. To avoid complex and inefficient conjugation, this study was conducted to explore the use of streptavidin as a linker between biotinylated DNAs and three biotinylated carriers as models. Methods: The two cationic peptides TAT and polyarginine and the lipophilic cholesterol have been reported to improve cellular transport of DNAs. The biotinylated peptides were purchased while the biotinylated cholesterol was synthesized in house. The 18 mer AS phosphorothioate DNA against the mdr1 mRNA of Pgp was purchased in biotinylated form with a primary amine group on the opposite end. Since it was considered important to place the radiolabel on the DNA, the DNA was conjugated for radiolabeling with 99mTc using MAG3. Preparation thereafter of the 1 : 1 carrier : DNA constructs followed simple mixing in a solution of 0.20 M ammonium acetate, 0.5 M NaCl, pH 7.0, by adding very slowly and with agitation the carrier to the steptavidin or the radiolabeled DNA to the streptavidin-peptides/cholesterol construct at a 1 :1 molar ratio. Preliminary studies of cellular accumulations were then performed at 50 nM concentration, 37°C in KB-31 (Pgp+), KB-G2 (Pgp++) and TCO-1 (Pgp++) cells in FCS-free media for 24 h and the results compared to identical studies performed simultaneously with the naked DNA as control. Results: Size exclusion HPLC by UV and radioactivity detection showed only a single peak to demonstrate that the 1 : 1 construct was successfully prepared in each case. In the cell studies, no evidence of cellular toxicity by growth delay was observed in any study. The cellular accumulations of the TAT and cholesterol constructs increased in the approximate order of increasing mdr1 expression TCO-1>KB-G2>KB-31 going from 37, 21 and 14% for TAT and 35, 15 and 10% for cholesterol. Furthermore, the ratio of cellular accumulation of each radiolabeled construct compared to naked increased in all cases reaching values of 1.3 to 1.5. Conclusions: One advantage to the use of (strept)avidin as linker is in the simplicity of preparation and therefore the ease with which different constructs may be made and tested. The results of this study indicated that (strept)avidin appears to be a useful method of linking antisense oligomers and their carriers as an alternative to covalent conjugation. It remains to be determined how the pharmacokinetics of radiolabeled DNAs are influenced by this method of attachment.
