Freeze-Thawed Combined Optimized Chemically Extracted Allograft for the Repair of Sciatic Nerve Gap in Rats

Abstract:This study examined sciatic nerve regeneration and functional recovery in rats following repair with autografted nerve, freeze-thawed nerve (FTN), optimized chemically extracted acellular nerve (OAN) or freeze-thawed combined optimized chemically extracted acellular nerve (FTOAN). Twelve weeks following replacement with 10 mm nerve segments, motor recovery was assessed using a sciatic nerve function index, electrophysiology test, and measurement of the humid weight of the gastrocnemius muscle. Nerve regeneration was assessed by analysis of hematoxylin and eosin staining, toluidine blue staining, and transmission electron microscopy. The results indicate that sciatic nerve regeneration and functional recovery from repair with FTOAN segments was significantly better than with FTN or OAN segments, and equivalent to autografts. Furthermore, the FTOAN segments were not rejected or absorbed by the host after repairing sciatic nerve gap of rats. Histopathological analysis demonstrated that complete removal of cellular and myelin debris from nerve segments was achieved with freeze-thawing and mild chemical extraction, which promoted the recovery of axon diameter, myelin sheath thickness and reduced gastrocnemius muscle atrophy. Therefore, the FTOAN method is a promising alternative to promote successful peripheral nerve damage repair.
