Abstract:The authors report on a supercontinuum tweezer which combines tweezing with broadband (∼3000cm−1) coherent anti-Stokes Raman scattering (CARS) spectroscopy by taking advantage of the high spatial coherence and broad bandwidth of pulsed supercontinuum generated in a nonlinear photonic crystal fiber. Polarization-discriminated and time-resolved CARS is investigated to suppress the nonresonant four-wave-mixing background.

Abstract:Time‐resolved coherent anti‐Stokes Raman scattering (CARS) impulsively excited by a supercontinuum (SC) is investigated. We show that it is critical to optimize the temporal overlapping of the constituent solitons of a SC in order to impulsively excite vibrational modes over a broad frequency range. The cross‐correlation frequency‐resolved optical gating technique is utilized to retrieve the optical response function of molecules under SC impulsive excitation, and background‐free CARS with spectral resolution significantly better than the bandwidth of the probe pulses is achieved. Abstract:Stimulated Raman scattering (SRS) is widely accepted as free of the nonresonant background appearing in coherent antiStokes Raman scattering (CARS), which led to a common misunderstanding that SRS is background free. On the contrary, the phase-sensitive detection scheme of conventional SRS imaging is subject to quite a few competing nonRaman processes, including transient absorption, cross-phase modulation, photothermal lensing, and two-photon absorption, which can often overwhelm real Raman signal and should be considered as nonresonant background. Although high-NA collection and longer excitation wavelength can help reduce such background signals, it still imposes limitations to image quality thus broader applications of SRS imaging. In order to make SRS imaging truly background free, new strategies have been demonstrated based on the aspect of spectroscopy, i.e., Raman process differs from background signals fundamentally in physics, which can then be distinguished spectrally. This chapter focuses on the technological advances of background-free SRS imaging, followed by a brief introduction of the enabling applications.
Multimodal Coherent Anti-Stokes Raman Scattering Microscopy with a Supercontinuum All-Fiber Laser

Abstract:We demonstrate a compact supercontinuum (SC) all-fiber laser source for multimodal coherent anti-Stokes Raman scattering (CARS) microscopy based on an Er-fiber seed. It can be used in both high-speed narrowband CARS microscopy and broadband CARS spectroscopy at tens of milliseconds per pixel, by simply switching without realignment. The common pump pulses, generated from the SC in a highly nonlinear fiber (HNLF), are obtained after spectral filtering. The central wavelength of pump pulses is 1067.2 nm with a bandwidth of 0.8 nm, which provides a high-spectral resolution (<;10 cm-1). By combining with the narrowband Stokes beam, which is directly obtained from the Er-fiber oscillator around ~1550 nm, we can achieve a narrowband CARS microscopy in frequency shifts of ~2700 to ~3200 cm-1. The SC Stokes beam in a broadband CARS spectroscopy ranging from 1150 to beyond 1700 nm is generated in an HNLF, with vibrational spectra over a bandwidth of more than 2800 cm-1. We also analyzed the influences of different HNLFs on the SC generation both in simulation and experiment. The capability of our system in a multimodal CARS microscopy is demonstrated by the CARS detection of polystyrene and pork.

Abstract:Coherent anti-Stokes Raman scattering (CARS) spectroscopy with time-delayed ultrashort pulses and a single-pixel photodetector has shown great potential for spectroscopic imaging and transient studies in chemistry and biological research. However, those systems rely on mechanical delay lines or two asynchronous optical combs with inflexible repetition frequencies, technically limiting their acquisition speeds. Here, we demonstrate a hybrid dual-comb CARS system involving a broadband fiber laser and a highly-flexible, frequency-modulated electro-optic comb. We achieve multiplex CARS spectra (2800-3200 cm-1), with a moderate resolution (22 cm-1), at a maximum refresh rate of 1 MHz, limited by the radio-frequency synthesizer we use. Fast spectroscopic CARS imaging is demonstrated for liquid mixtures. Our system enables spectral measurements in the high-wavenumber C-H stretching region at a record speed that is an order of magnitude higher than state-of-the-art systems, which may open up new opportunities for fast chemical sensing and imaging.

Abstract:We propose and demonstrate a supercontinuum fiber laser source for broadband coherent anti-Stokes Raman scattering (BCARS) microspectroscopy, seeded by a Yb-doped dissipative soliton fiber laser. Smooth continuum Stokes pulses ranging from 1050 to 1312 nm at -20 dB bandwidth are generated in a highly nonlinear fiber from the compact fiber seed, and the corresponding pump pulses are obtained after a narrowband filter centered at 1036 nm. With this fiber laser source, we are able to obtain BCARS spectra in 700-1900 cm-1 spectral range simultaneously with resolution less than 10 cm-1, and show its application in analyzing complex mixture. We also demonstrate the capability of our system in chemical imaging by simultaneously obtaining two two-dimensional CARS images of retinoic acid crystal at Raman shifts of ~1574 and ~1162 cm-1, respectively.

Abstract:We present a systematic characterization of coherent anti-Stokes Raman scattering (CARS) microscopy. CARS signal generation in a heterogeneous sample under a tight-focusing condition is formulated by the Green’s function method. The CARS radiation pattern and the forward- and backward-detected CARS signals from a three-dimensional Raman scatterer are calculated. The coherent nature of CARS image formation and its consequences for image contrast and spatial resolution are investigated. Experimental implementations of CARS microscopy with collinearly copropagating and counterpropagating excitation beams, forward and backward data collection, and polarization-sensitive detection are described. Finally, CARS images of unstained live cells with forward detection, epidetection, and polarization-sensitive detection are presented and compared.

Abstract:Coherent anti-Stokes Raman scattering (CARS) microscopy permits vibrational imaging with high-sensitivity, high speed, and three-dimensional spatial resolution. We review recent advances in CARS microscopy, including experimental design, theoretical understanding of contrast mechanisms, and applications to chemical and biological systems. We also review the development of multiplex CARS microspectroscopy, which allows high-speed characterization of microscopic samples, and CARS correlation spectroscopy, which probes fast diffusion dynamics with vibrational selectivity.

Abstract:We demonstrate a multi-color background-free coherent anti-Stokes Raman scattering (CARS) imaging system, using a robust, all-fiber, low-cost, multi-wavelength time-lens source. The time-lens source generates picosecond pulse trains at three different wavelengths. The first is 1064.3 nm, the second is tunable between 1052 nm and 1055 nm, and the third is tunable between 1040 mu and 1050 mu. When the time-lens source is synchronized with a mode-locked Ti:Sa laser, two of the three wavelengths are used to detect different Raman frequencies for two-color on-resonance imaging, whereas the third wavelength is used to obtain the off-resonance image for nonresonant background subtraction. Mixed poly(methyl methacrylate) (PMMA) and polystyrene (PS) beads are used to demonstrate two-color background-free CARS imaging. The synchronized multi-wavelength time-lens source enables pixel-to-pixel wavelength-switching. We demonstrate simultaneous two-color CARS imaging of CH2 and CH3 stretching vibration modes with real-time background subtraction in ex vivo mouse tissue. Abstract:The ability to enhance resonant signals and eliminate the non-resonant background is analyzed for Coherent Anti-Stokes Raman Scattering (CARS). The analysis is done at a specific frequency as well as for broadband excitation using femtosecond pulse-shaping techniques. An appropriate objective functional is employed to balance resonant signal enhancement against non-resonant background suppression. Optimal enhancement of the signal and minimization of the background can be achieved by shaping the probe pulse alone while keeping the pump and Stokes pulses in transform-limited-form (TLF). In some cases analytical forms for the probe pulse can be found, and numerical simulations are carried out for other circumstances. It is found that a good approximate solution for the optimal pulse in the two-pulse CARS is a superposition of linear and arctangent type phases for the pump. The well-known probe delay method is shown to be a quasi-optimal scheme for background suppression. The results should provide a basis to improve the performance of CARS spectroscopy and microscopy.

Abstract:We demonstrate an ambient light coherent anti-Stokes Raman scattering microscope that allows CARS imaging to be operated under environmental light for field use. The CARS signal is modulated at megahertz frequency and detected by a photodiode equipped with a lab-built resonant amplifier, then extracted through a lock-in amplifier. The filters in both the spectral domain and the frequency domain effectively blocked the room light contamination of the CARS image. In situ hyperspectral CARS imaging of tumor tissue under ambient light is demonstrated.

Abstract:A fiber laser source based on a spectral-compressed dissipative soliton oscillator for broadband coherent anti-Stokes Raman scattering (BCARS) microspectroscopy is reported. The high-power narrowband picosecond pump pulses for BCARS are obtained by spectral compression in a cascaded fiber amplifier. BCARS signal with high spectral resolution is demonstrated by this fiber source.

Abstract:We have developed an interferometric implementation of coherent anti-Stokes Raman scattering (CARS) which enables broadband coherent Raman spectroscopy free from non-resonant background (NRB), with a signal strength proportional to concentration. Abstract:We propose an all-fiber-generated dual-soliton pulses based scheme for the background-free detection of coherent anti-Stokes Raman spectroscopy under the spectral focusing mechanism. Due to the strong birefringence and high nonlinearity of a polarization-maintaining photonic crystal fiber (PM-PCF), two redshifted soliton pulses can be simultaneously generated relying on high-order dispersion and nonlinear effects along two eigenpolarization axes in the anomalous dispersion regime, while allowing feasible tunability of the frequency distance and temporal interval between them. This proposed scheme, termed as DS-CARS, exploits a unique combination of slight frequency-shift and advisable temporal walk-off of this two soliton pulses to achieve robust and efficient suppression of nonresonant background with compact all-fiber coherent excitation source. Capability of the DS-CARS is experimentally demonstrated by the background-free CARS spectroscopy and unambiguous CARS microscopy of polymer beads in the fingerprint region.

Abstract:Coherent anti-Stokes Raman microscopy (CARS) is a quantitative, chemically specific, and label-free optical imaging technique for studying inhomogeneous systems. However, the complicating influence of the nonresonant response on the CARS signal severely limits its sensitivity and specificity and especially limits the extent to which CARS microscopy has been used as a fully quantitative imaging technique. On the basis of spectral focusing mechanism, we establish a dual-soliton Stokes based CARS microspectroscopy and microscopy scheme capable of quantifying the spatial information of densities and chemical composition within inhomogeneous samples, using a single fiber laser. Dual-soliton Stokes scheme not only removes the nonresonant background but also allows robust acquisition of multiple characteristic vibrational frequencies. This all-fiber based laser source can cover the entire fingerprint (800-2200 cm-1) region with a spectral resolution of 15 cm-1. We demonstrate that quantitative degree determination of lipid-chain unsaturation in the fatty acids mixture can be achieved by the characterization of C = C stretching and CH2 deformation vibrations. For microscopy purposes, we show that the spatially inhomogeneous distribution of lipid droplets can be further quantitatively visualized using this quantified degree of lipid unsaturation in the acyl chain for contrast in the hyperspectral CARS images. The combination of compact excitation source and background-free capability to facilitate extraction of quantitative composition information with multiplex spectral peaks will enable wider applications of quantitative chemical imaging in studying biological and material systems.

Abstract:Despite its success in many fields, the implementation of coherent anti-Stokes Raman spectroscopy (CARS) in tackling the problems at interfaces was hindered by the enormous resonant and nonresonant background from the bulk. In this work, we have developed a novel CARS scheme that can probe a buried interface via ≥105-fold suppression of the nonresonant and resonant bulk contribution. The method utilizes self-destructive interference between the forward and backward CARS generated in the bulk near the Brewster angle. As a result, we can resolve the vibrational spectrum of submonolayer interfacial polar/apolar species immersed in the surrounding medium with huge CARS responses. We expect that our approach opens up the opportunity to interrogate the interfaces involving apolar molecules and benefits other nonlinear optical spectroscopic techniques, e.g., sum-frequency spectroscopy and four-wave mixing spectroscopy in general, to promote the signal-to-background noise ratio.

Abstract:Coherent anti-Stokes Raman scattering (CARS) is a powerful nonlinear spectroscopy technique that is rapidly gaining recognition of different molecules. Unfortunately, molecular concentration information is generally not immediately accessible from the raw CARS signal due to the nonresonant background. In addition, mainstream biomedical applications of CARS are currently hampered by a complex and bulky excitation setup. Here, we establish a dual-soliton Stokes based CARS spectroscopy scheme capable of quantifying the sample molecular, using a single fiber laser. This dual-soliton CARS scheme takes advantage of a differential configuration to achieve efficient suppression of nonresonant background and therefore allows extraction of quantitative composition information. Besides, our all-fiber based excitation source can probe the most fingerprint region (1100–1800 cm−1) with a spectral resolution of 15 cm−1 under the spectral focusing mechanism, where is considerably more information contained throughout an entire spectrum than at just a single frequency within that spectrum. Systematic studies of the scope of application and several fundamental aspects are discussed. Quantitative capability is further experimentally demonstrated through the determination of oleic acid concentration based on the linear dependence of signal on different Raman vibration bands.
