Abstract:Gelatin methacrylate (GelMA)-based hydrogels are gaining a great deal of attention as potentially implantable materials in tissue engineering applications because of their biofunctionality and mechanical tenability. Since different natural tissues respond differently to mechanical stresses, an ideal implanted material would closely match the mechanical properties of the target tissue. In this regard, applications employing GelMA hydrogels are currently limited by the low mechanical strength and biocompatibility of GelMA. Therefore, this review focuses on modifications made to GelMA hydrogels to make them more suitable for tissue engineering applications. A large number of reports detail rational synthetic processes for GelMA or describe the incorporation of various biomaterials into GelMA hydrogels to tune their various properties, e.g., physical strength, chemical properties, conductivity, and porosity, and to promote cell loading and accelerate tissue repair. A novel strategy for repairing tissue injuries, based on the transplantation of cell-loaded GelMA scaffolds, is examined and its advantages and challenges are summarized. GelMA-cell combinations play a critical and pioneering role in this process and could potentially accelerate the development of clinically relevant applications.

Abstract:Softness and firmness are seemingly incompatible traits that synergize to create the unique soft-yet-firm tactility of living tissues pursued in soft robotics, wearable electronics, and plastic surgery. This dichotomy is particularly pronounced in tissues such as fat that are known to be both ultrasoft and ultrafirm. However, synthetically replicating this mechanical response remains elusive since ubiquitously employed soft gels are unable to concurrently reproduce tissue firmness. We have addressed the tissue challenge through the self-assembly of linear-bottlebrush-linear (LBL) block copolymers into thermoplastic elastomers. This hybrid molecular architecture delivers a hierarchical network organization with a cascade of deformation mechanisms responsible for initially low moduli followed by intense strain-stiffening. By bridging the firmness gap between gels and tissues, we have replicated the mechanics of fat, fetal membrane, spinal cord, and brain tissues. These solvent-free, nonleachable, and tissue-mimetic elastomers also show enhanced biocompatibility as demonstrated by cell proliferation studies, all of which are vital for the safety and longevity of future biomedical devices.

Abstract:Soft materials featuring dynamic networks represent a burgeoning frontier in materials science, offering multifaceted applications spanning soft robotics, biomaterials, and flexible electronics. Unraveling the time-dependent constitutive behavior of these materials, rooted in dynamic networks, stands as a pivotal pursuit for engineering advancements. Herein, we fabricate a tough and extreme-temperature-tolerant glycerogel with a polymer network crosslinked by metal-coordination crosslinkers and conduct a thorough analysis of its intricate mechanical responses across monotonic loading, relaxation, creep, and cyclic tests. We then develop a physically grounded constitutive model integrating the dynamics of crosslinker association/dissociation and polymer chain diffusion, furnishing a holistic framework to elucidate their interplay. We employ a statistical description, using density functions of chains in terms of end-to-end vectors, to characterize network reconfiguration. The evolution of chain density under external load, mediated by crosslinker kinetics and chain diffusion in a viscous medium, leads to intriguing variations in elastic energy and stress responses. Through meticulous experimental validation and numerical simulations, we demonstrate the efficacy of the model in forecasting the mechanical behavior of dynamic polymer networks under diverse loading scenarios, encompassing strain rate effects, stress relaxation, Mullins effect, and self-recovery phenomena. Our findings provide valuable insights into the design and optimization of dynamic network-based materials for diverse applications in biomedical and engineering fields.
Mechanically Resilient, Injectable, and Bioadhesive Supramolecular Gelatin Hydrogels Crosslinked by Weak Host-Guest Interactions Assist Cell Infiltration and in Situ Tissue Regeneration

Abstract:Although considered promising materials for assisting organ regeneration, few hydrogels meet the stringent requirements of clinical translation on the preparation, application, mechanical property, bioadhesion, and biocompatibility of the hydrogels. Herein, we describe a facile supramolecular approach for preparing gelatin hydrogels with a wide array of desirable properties. Briefly, we first prepare a supramolecular gelatin macromer via the efficient host-guest complexation between the aromatic residues of gelatin and free diffusing photo-crosslinkable acrylated β-cyclodextrin (β-CD) monomers. The subsequent crosslinking of the macromers produces highly resilient supramolecular gelatin hydrogels that are solely crosslinked by the weak host-guest interactions between the gelatinous aromatic residues and β-cyclodextrin (β-CD). The obtained hydrogels are capable of sustaining excessive compressive and tensile strain, and they are capable of quick self healing after mechanical disruption. These hydrogels can be injected in the gelation state through surgical needles and re-molded to the targeted geometries while protecting the encapsulated cells. Moreover, the weak host-guest crosslinking likely facilitate the infiltration and migration of cells into the hydrogels. The excess β-CDs in the hydrogels enable the hydrogel-tissue adhesion and enhance the loading and sustained delivery of hydrophobic drugs. The cell and animal studies show that such hydrogels support cell recruitment, differentiation, and bone regeneration, making them promising carrier biomaterials of therapeutic cells and drugs via minimally invasive procedures.

Abstract:The mechanical properties of polymer gels devote to emerging devices and machines in fields such as biomedical engineering, flexible bioelectronics, biomimetic actuators, and energy harvesters. Coupling network architectures and interactions has been explored to regulate supportive mechanical characteristics of polymer gels; however, systematic reviews correlating mechanics to interaction forces at the molecular and structural levels remain absent in the field. This review highlights the molecular engineering and structural engineering of polymer gel mechanics and a comprehensive mechanistic understanding of mechanical regulation. Molecular engineering alters molecular architecture and manipulates functional groups/moieties at the molecular level, introducing various interactions and permanent or reversible dynamic bonds as the dissipative energy. Molecular engineering usually uses monomers, cross-linkers, chains, and other additives. Structural engineering utilizes casting methods, solvent phase regulation, mechanochemistry, macromolecule chemical reactions, and biomanufacturing technology to construct and tailor the topological network structures, or heterogeneous modulus compositions. We envision that the perfect combination of molecular and structural engineering may provide a fresh view to extend exciting new perspectives of this burgeoning field. This review also summarizes recent representative applications of polymer gels with excellent mechanical properties. Conclusions and perspectives are also provided from five aspects of concise summary, mechanical mechanism, biofabrication methods, upgraded applications, and synergistic methodology.
Ligament-Inspired Tough and Anisotropic Fibrous Gel Belt with Programed Shape Deformations via Dynamic Stretching

Abstract:Nature provides perpetual inspiration for exploring anisotropic materials to implement complex functions and motions like biological organisms. In particular, fibrous hydrogel-based anisotropic aggregates have attracted tremendous interest as fantastic materials for development into artificial ligaments or muscles. Such aggregates combine the structural anisotropy and macroscopic flexibility of fiber materials, with the intelligence, softness, and wetness of hydrogel materials. However, controlled fabrication of such hydrogels with aligned microstructures, even in a macroscopic level, remains a challenge. Here, a facile and general strategy was proposed to develop ligament-inspired multistructural (mono/bilayer) gel belts via dynamic stretching of multistrand pregels, accompanied by the simultaneous assembly of hydrogel fibers. The resultant gel belts evolved into anisotropic and aligned micro- and macrostructures, exhibiting high elastic moduli (0.01-23.5 MPa) and unique anisotropic swelling behaviors. Through further physical and chemical structure design, bioinspired multiple fibrous gel-based actuators were developed to achieve anisotropic, relatively fast (within 60 s), and delicate macroscopic shape deformations. This work provides a great platform for the design and construction of next-generation soft materials for biomimetic tissues.

Designing Viscoelastic Gelatin-PEG Macroporous Hybrid Hydrogel with Anisotropic Morphology and Mechanical Properties for Tissue Engineering Application

Abstract:The mechanical properties of scaffolds play a vital role in regulating key cellular processes in tissue development and regeneration in the field of tissue engineering. Recently, scaffolding material design strategies leverage viscoelasticity to guide stem cells toward specific tissue regeneration. Herein, we designed and developed a viscoelastic Gel-PEG hybrid hydrogel with anisotropic morphology and mechanical properties using a gelatin and functionalized PEG (as a crosslinker) under a benign condition for tissue engineering application. The chemical crosslinking/grafting reaction was mainly involved between epoxide groups of PEG and available functional groups of gelatin. FTIR spectra revealed the hybrid nature of Gel-PEG hydrogel. The hybrid hydrogel showed good swelling behavior (water content > 600%), high porosity and pore interconnectivity suitable for tissue engineering application. Simple unidirectional freezing followed by a freeze-drying technique allowed the creation of structurally stable 3D anisotropic macroporous architecture that showed tissue-like elasticity and was capable of withstanding high deformation (50% strain) without being damaged. The tensile and compressive modulus of Gel-PEG hybrid hydrogel were found to be 0.863 MPa and 0.330 MPa, respectively, which are within the range of normal human articular cartilage. In-depth mechanical characterizations showed that the Gel-PEG hybrid hydrogel possessed natural-tissue-like mechanics such as non-linear and J-shaped stress-strain curves, stress softening effect, high fatigue resistance and stress relaxation response. A month-long hydrolytic degradation test revealed that the hydrogel gradually degraded in a homogeneous manner over time but maintained its structural stability and anisotropic mechanics. Overall, all these interesting features provide a potential opportunity for Gel-PEG hybrid hydrogel as a scaffold in a wide range of tissue engineering applications.
Skin-Inspired Gels with Toughness, Antifreezing, Conductivity, and Remoldability

Abstract:In recent years, nature-inspired conductive hydrogels have become ideal materials for the design of bioactuators, healthcare monitoring sensors, and flexible wearable devices. However, conductive hydrogels are often hindered by problems such as the poor mechanical property, nonreusability, and narrow operating temperature range. Here, a novel skin-inspired gel is prepared via one step of blending polyvinyl alcohol, gelatin, and glycerin. Due to their dermis-mimicking structure, the obtained gels possess high mechanical properties (fracture stress of 1044 kPa, fracture strain of 715%, Young's modulus of 157 kPa, and toughness of 3605 kJ m-3). Especially, the gels exhibit outstanding strain-sensitive electric behavior as biosensors to monitor routine movement signals of the human body. Moreover, the gels with low temperature tolerance can maintain good conductivity and flexibility at -20 °C. Interestingly, the gels are capable of being recovered and reused by heating injection, cooling molding, and freezing-thawing cycles. Thus, as bionic materials, the gels have fascinating potential applications in various fields, such as human-machine interfaces, biosensors, and wearable devices.

Abstract:Engineering living tissues that simulate their natural counterparts is a dynamic area of research. Among the various models of biological tissues being developed, fiber‐shaped cellular architectures, which can be used as artificial blood vessels or muscle fibers, have drawn particular attention. However, the fabrication of continuous microfiber substrates for culturing cells is still limited to a restricted number of polymers (e.g., alginate) having easy processability but poor cell–material interaction properties. Moreover, the typical smooth surface of a synthetic fiber does not replicate the micro‐ and nanofeatures observed in vivo, which guide and regulate cell behavior. In this study, a method to fabricate photocrosslinkable cell‐responsive methacrylamide‐modified gelatin (GelMA) fibers with exquisite microstructured surfaces by using a microfluidic device is developed. These hydrogel fibers with microgrooved surfaces efficiently promote cell encapsulation and adhesion. GelMA fibers significantly promote the viability of cells encapsulated in/or grown on the fibers compared with similar grooved alginate fibers used as controls. Importantly, the grooves engraved on the GelMA fibers induce cell alignment. Furthermore, the GelMA fibers exhibit excellent processability and could be wound into various shapes. These microstructured GelMA fibers have great potential as templates for the creation of fiber‐shaped tissues or tissue microstructures.

Abstract:Naturally sourced gels from food biopolymers have advanced in recent decades to compare favourably in performance and breadth of application to their synthetic counterparts. Here, we comprehensively review the constitutive nature, gelling mechanisms, design approaches, and structural and mechanical properties of food gels. We then consider how these food gel design principles alter rheological and tribological properties for food quality improvement, nutrient-modification of foods while preserving sensory perception, and targeted delivery of drugs and bioactives within the gastrointestinal tract. We propose that food gels may offer advantages over their synthetic counterparts owing to their source renewability, low cost, biocompatibility and biodegradability. We also identify emerging approaches and trends that may improve and expand the current scope, properties and functionalities of food gels and inspire new applications.
An organo-hydrogel with extreme mechanical performance and tolerance beyond skin

Abstract:Soft polymer gels encounter difficulties in meeting the demanding requirements of real-life applications due to the long-standing challenge to reproduce the excellent mechanical properties and multifunctionalities observed in natural soft tissues. Here, a skin-like hierarchically structured organo-hydrogel with an all-around performance that matches and outperforms mammalian skin is reported, including high stretchability of 2227% strain, remarkable strength of 20.78 MPa, record-breaking fracture toughness up to 260 MJ/m 3 , together with excellent resistance to fracture and fatigue (fatigue threshold over 30.4 kJ/m 2 ), superior long-term stability and tolerance to freezing and elevated temperatures. It also shows high conductivity and excellent electrical sensing capability. The underlying synergistic multi-scale reinforcement mechanisms of the organo-hydrogel are also validated via molecular dynamics simulations and experimental results. Hence, this work formulates a model design applicable to various polymer gels to expand their potential in applications including flexible electronics, multi-functional bionic sensors, energy storage devices and soft robotics.

Abstract:Gelatin methacryloyl (GelMA), a kind of modified biomaterials with adjustable degradability from gelatin, can composite with various composite materials and therapeutic substances including cells through various physiochemical direct/indirect interactions to achieve treatment effect. Furthermore, owing to that the GelMA possesses photocrosslinking ability and excellent rheology properties, GelMA itself and its composite materials have the potential to be manufactured to function in diversified forms with 2D or 3D structures as required, such as thin films, microswimmers, biomimetic scaffolds, etc. Combined with the excellent biocompatibility from gelatin, GelMA composite hydrogels have been widely applied in biomedical applications. Unlike published reviews concentrating on a specific application fields or construction strategies of GelMA, this review summarized the multiscale construction strategies and available forms of GelMA complex hydrogel step by step based on its excellent physical and chemical properties. These strategies could be integrated to perform more desirable effects. In the end, the relevant biomedical applications in combination with the repair requirements of different tissues are briefly discussed. This systematic review can provide assistance for the rational application of GelMA in the field of biomedical.

Solvent-free, supersoft and superelastic bottlebrush melts and networks

Abstract:Polymer gels are the only viable class of synthetic materials with a Young's modulus below 100 kPa conforming to biological applications, yet those gel properties require a solvent fraction. The presence of a solvent can lead to phase separation, evaporation and leakage on deformation, diminishing gel elasticity and eliciting inflammatory responses in any surrounding tissues. Here, we report solvent-free, supersoft and superelastic polymer melts and networks prepared from bottlebrush macromolecules. The brush-like architecture expands the diameter of the polymer chains, diluting their entanglements without markedly increasing stiffness. This adjustable interplay between chain diameter and stiffness makes it possible to tailor the network's elastic modulus and extensibility without the complications associated with a swollen gel. The bottlebrush melts and elastomers exhibit an unprecedented combination of low modulus (∼100 Pa), high strain at break (∼1,000%), and extraordinary elasticity, properties that are on par with those of designer gels.

Abstract:Hydrogels have attracted increasing attention in recent years for diverse biomedical applications. In situ forming hydrogels provide additional advantages over conventional hydrogels, such as ease of administration and minimally invasive treatment, which are convenient for in vivo surgical operation and improve patient compliance. Therefore, they have been extensively investigated as scaffolds or carriers of drugs, cells, and bioactive molecules. Recently, a variety of in situ forming gels have been developed through physical and/or chemical cross-linking reactions, which can respond to external stimuli, such as light, temperature, and pH. In this chapter, we present an overview of the versatile crosslinking strategies for in situ forming hydrogel fabrication and discuss their current advancements and future perspectives.

Abstract:Fibrin gels, prepared from fibrinogen and thrombin, the key proteins involved in blood clotting, were among the first biomaterials used to prevent bleeding and promote wound healing. The unique polymerization mechanism of fibrin, which allows control of gelation times and network architecture by variation in reaction conditions, allows formation of a wide array of soft substrates under physiological conditions. Fibrin gels have been extensively studied rheologically in part because their nonlinear elasticity, characterized by soft compliance at small strains and impressive stiffening to resist larger deformations, appears essential for their function as haemostatic plugs and as matrices for cell migration and wound healing. The filaments forming a fibrin network are among the softest in nature, allowing them to deform to large extents and stiffen but not break. The biochemical and mechanical properties of fibrin have recently been exploited in numerous studies that suggest its potential for applications in medicine and bioengineering.

Abstract:Gelatin methacryloyl (GelMA) hydrogels have been widely used for various biomedical applications due to their suitable biological properties and tunable physical characteristics. GelMA hydrogels closely resemble some essential properties of native extracellular matrix (ECM) due to the presence of cell-attaching and matrix metalloproteinase responsive peptide motifs, which allow cells to proliferate and spread in GelMA-based scaffolds. GelMA is also versatile from a processing perspective. It crosslinks when exposed to light irradiation to form hydrogels with tunable mechanical properties. It can also be microfabricated using different methodologies including micromolding, photomasking, bioprinting, self-assembly, and microfluidic techniques to generate constructs with controlled architectures. Hybrid hydrogel systems can also be formed by mixing GelMA with nanoparticles such as carbon nanotubes and graphene oxide, and other polymers to form networks with desired combined properties and characteristics for specific biological applications. Recent research has demonstrated the proficiency of GelMA-based hydrogels in a wide range of tissue engineering applications including engineering of bone, cartilage, cardiac, and vascular tissues, among others. Other applications of GelMA hydrogels, besides tissue engineering, include fundamental cell research, cell signaling, drug and gene delivery, and bio-sensing.

This manuscript explores the advancements and future directions of injectable biomimetic gels in biomedical applications and highlights the significant potential of hydrogels in wound healing, tissue regeneration, and controlled drug delivery due to their enhanced biocompatibility, multifunctionality, and mechanical properties. Despite these advancements, challenges such as mechanical resilience, controlled degradation rates, and scalable manufacturing remain. This manuscript discusses ongoing research to optimize these properties, develop cost-effective production techniques, and integrate emerging technologies like 3D bioprinting and nanotechnology. Addressing these challenges through collaborative efforts is essential for unlocking the full potential of injectable biomimetic gels in tissue engineering and regenerative medicine.

Abstract:The field of polymeric biomaterials has received much attention in recent years due to its potential for enhancing the biocompatibility of systems and devices applied to drug delivery and tissue engineering. Such applications continually push the definition of biocompatibility from relatively straightforward issues such as cytotoxicity to significantly more complex processes such as reducing foreign body responses or even promoting/recapitulating natural body functions. Hydrogels and their colloidal analogues, microgels, have been and continue to be heavily investigated as viable materials for biological applications because they offer numerous, facile avenues in tailoring chemical and physical properties to approach biologically harmonious integration. Mechanical properties in particular are recently coming into focus as an important manner in which biological responses can be altered. In this Account, we trace how mechanical properties of microgels have moved into the spotlight of research efforts with the realization of their potential impact in biologically integrative systems. We discuss early experiments in our lab and in others focused on synthetic modulation of particle structure at a rudimentary level for fundamental drug delivery studies. These experiments elucidated that microgel mechanics are a consequence of polymer network distribution, which can be controlled by chemical composition or particle architecture. The degree of deformability designed into the microgel allows for a defined response to an imposed external force. We have studied deformation in packed colloidal phases and in translocation events through confined pores; in all circumstances, microgels exhibit impressive deformability in response to their environmental constraints. Microgels further translate their mechanical properties when assembled in films to the properties of the bulk material. In particular, microgel films have been a large focus in our lab as building blocks for self-healing materials. We have shown that their ability to heal after damage arises from polymer mobility during hydration. Furthermore, we have shown film mobility dictates cell adhesion and spreading in a manner that is fundamentally different from previous work on mechanotransduction. In total, we hope that this Account presents a broad introduction to microgel research that intersects polymer chemistry, physics, and regenerative medicine. We expect that research intersection will continue to expand as we fill the knowledge gaps associated with soft materials in biological milieu.

Abstract:Given the creation of materials based on nanoscale science, nanotechnology must be combined with other disciplines. This role is played by the new concept of nanoarchitectonics, the process of constructing functional materials from nanocomponents. Nanoarchitectonics may be highly compatible with applications in biological systems. Conversely, it would be meaningful to consider nanoarchitectonics research oriented toward biological applications with a focus on material systems. Perhaps, hydrogels are promising as a model medium to realize nanoarchitectonics in biofunctional materials science. In this review, we will provide an overview of some of the defined targets, especially for tissue engineering. Specifically, we will discuss (i) hydrogel bio-ink for 3D bioprinting, (ii) dynamic hydrogel as an artificial extracellular matrix (ECM), and (iii) topographical hydrogel for tissue organization. Based on these backgrounds and conceptual evolutions, the construction strategies and functions of bio-gel nanoarchitectonics in medical applications and tissue engineering will be discussed.

Abstract:Gels are an increasingly important class of soft materials with applications ranging from regenerative medicine to commodity materials. A major drawback of gels is their relative mechanical weakness, which worsens further under strain. We report a new class of responsive gels with latent crosslinking moieties that exhibit strain-stiffening behavior. This property results from the lability of disulfides, initially isolated in a protected state, then activated to crosslink on-demand. The active thiol groups are induced to form inter-chain crosslinks when subjected to mechanical compression, resulting in a gel that strengthens under strain. Molecular shielding design elements regulate the strain-sensitivity and spontaneous crosslinking tendencies of the polymer network. These strain-responsive gels represent a rational design of new advanced materials with on-demand stiffening properties with potential applications in elastomers, adhesives, foams, films, and fibers.
