Abstract:A supramolecular double network hydrogel is presented by physical interpenetration of DNA and cucurbit[8]uril networks. In addition to exhibiting an increase in strength and thermal stability, the double network hydrogel possesses excellent properties such as stretchability, ductility, shear-thinning, and thixotropy. Moreover, it is enzymatically responsive to both nuclease and cellulase, as well as small molecules, showing great potential as a new soft material scaffold.

Abstract:Architected hydrogels are widely used in biomedicine, soft robots, and flexible electronics while still possess big challenges in strong toughness, and shape modeling. Here, inspired with the universal hydrogen bonding interactions in biological systems, a strain-induced microphase separation path toward achieving the printable, tough supramolecular polymer hydrogels by hydrogen bond engineering is developed. Specifically, it subtly designs and fabricates the poly (N-acryloylsemicarbazide-co-acrylic acid) hydrogels with high hydrogen bond energy by phase conversion induced hydrogen bond reconstruction. The resultant hydrogels exhibited the unique strain-induced microphase separation behavior, resulting in the excellent strong toughness with, for example, an ultimate stress of 9.1 +/- 0.3 MPa, strain levels of 1020 +/- 126%, toughness of 33.7 +/- 6.6 MJ m(-3), and fracture energy of 171.1 +/- 34.3 kJ m(-2). More importantly, the hydrogen bond engineered supramolecular hydrogels possess dynamic shape memory character, i.e shape fixing at low temperature while recovery after heating. As the proof of concept, the tailored hydrogel stents are readily manufactured by 3D printing, which showed good biocompatibility, load-bearing and drug elution, being beneficial for the biomedical applications. It is believed that the present 3D printing of the architected dynamic hydrogels with ultrahigh toughness can broaden their applications.

Abstract:It is still a challenge to achieve both excellent mechanical strength and biocompatibility in hydrogels. In this study, we exploited two interactions to form a novel biocompatible, slicing-resistant, and self-healing hydrogel. The first was molecular host-guest recognition between a host (isocyanatoethyl acrylate modified -cyclodextrin) and a guest (2-(2-(2-(2-(adamantyl-1-oxy)ethoxy)ethoxy)ethoxy)ethanol acrylate) to form three-arm host-guest supramolecules (HGSMs), and the second was covalent bonding between HGSMs (achieved by UV-initiated polymerization) to form strong cross-links in the hydrogel. The host-guest interaction enabled the hydrogel to rapidly self-heal. When it was cut, fresh surfaces were formed with dangling host and guest molecules (due to the breaking of host-guest recognition), which rapidly recognized each other again to heal the hydrogel by recombination of the cut surfaces. The smart hydrogels hold promise for use as biomaterials for soft-tissue repair.

Abstract:Supramolecular hydrogels based on host-guest interactions have drawn considerable attention due to their unique properties and promising applications. However, it is still a great challenge to construct supramolecular hydrogels that simultaneously achieve mechanical strength, processability, and biocompatibility. Herein, we present a rational design of a "supramolecular crosslinker" approach to fabricate a new host-guest hydrogel with super-stretchability, self-healing, and injectable properties and excellent biocompatibility. The star-shaped supramolecular crosslinker is formed by the host-guest interactions between octa-cyclodextrin polyhedral oligomeric silsesquioxane (OCDPOSS) and acrylamide-modified adamantane (Ad-AAm). Supramolecular hydrogels can be briefly prepared by UV-initiated copolymerization of acrylamide and supramolecular crosslinkers. Supramolecular hydrogels present impressive mechanical properties due to rigid POSS as the core of the supramolecular crosslinker. Moreover, multivalent host-guest interactions improve the ductility, rapid self-healing and injectable ability of these hydrogels. Simultaneously, these supramolecular hydrogels possess good biocompatibility and can be utilized as carriers for the sustained release of hydrophobic drugs. Thus, such supramolecular hydrogels will have potential applications for tissue engineering and drug delivery systems.

Abstract:The recognition-directed host-guest interaction is recognized as a valuable tool for creating supramolecular polymers. Functional hydrogels constructed through the dynamic and reversible host-guest complexation are endowed with a great many appealing features, such as superior self-healing, injectability, flexibility, stimuli-responsiveness and biocompatibility, which are crucial for biological and medicinal applications. With numerous topological structures and host-guest combinations established previously, recent breakthroughs in this area mostly focus on further improvement and fine-tuning of various properties for practical utilizations. The current contribution provides a comprehensive overview of the latest developments in host-guest supramolecular hydrogels, with a particular emphasis on the innovative molecular-level design strategies and hydrogel formation methodologies targeting at a wide range of active biomedical domains, including drug delivery, 3D printing, wound healing, tissue engineering, artificial actuators, biosensors, etc. Furthermore, a brief conclusion and discussion on the steps forward to bring these smart hydrogels to clinical practice is also presented.

Abstract:The development of supramolecular nanocomposite hydrogels with good mechanical properties and multifunctional characteristics remains challenging. The reinforced role of interfacial weak interactions is important for the mechanical properties of nanocomposite hydrogels. Here, a dynamic host-guest inclusion complex from the host molecule CB[7] and guest units was employed to prepare Fe3O4 hybrid supramolecular nanocomposite hydrogels. The results show that the as-obtained hydrogel with a porous structure was prepared. The CB[7]-modified Fe3O4 (Fe3O4@CB[7]) nanoparticles severed as a cross-linker for fabricating the hydrogel's network. By changing the Fe3O4@CB[7] content, their tensile stress ranged from 0.102 to 0.403 MPa and their compression stress ranged (70% compression strain) from 0.059 to 0.775 MPa. By changing the guest units, their tensile stress ranged from 0.3 MPa to 0.403 MPa. The self-healing efficiency of the hydrogels was 99% after 48 h at room temperature. The as-obtained hydrogels with strain sensitivity can be applied for detecting the movement of an elbow and finger. The supramolecular hydrogel exhibits NIR responsiveness, self-healing, injectability, tunable mechanical strength and conductive ability, and can be used in flexible electronics.
Tough Supramolecular Hydrogels with Excellent Self-Recovery Behavior Mediated by Metal-Coordination Interaction

Abstract:Based on the strategy of dynamic metal-coordination for improving mechanical properties, a further ideal was taken to prepare the tough hydrogels with the combination of self-crosslinking monomer and metal-coordination complexes. Herein, a series of hydrogels of poly(acrylamide-co-acrylic acid-co-N-hydroxymethyl acrylamide) (P(AM-co-AAc-co-NMAM)) consisting the chemical crosslinking induced by NMAM units and the physical crosslinking derived from the coordination complexes of carboxyl-Fe3+ were prepared. The molecular structure was investigated with the ATR-FTIR, Raman and UV-vis spectra. These hydrogels with different water content of 57-93% possess good mechanical performances. The optimal hydrogels possess high tensile strength (8.56 MPa), prominent modulus (15.5 MPa), remarkable toughness (37.85 MJ/m(3)) and superb tearing energy (7062 J/m(2)). The tough hydrogels also display excellent self-recovery (95% toughness recovery within 50 min), pH-triggered healing, shape memory and plasticity abilities. These hydrogels having high strength and toughness may broaden range of potential applications in load-bearing soft actuators, flexible electronics, etc.

Abstract:Dynamic crosslinking based on host-guest interaction has shown a number of advantages in constructing injectable hydrogel to be used for biomacromolecules and cell delivery materials. However, it is difficult to dynamically adjust the performance of the hydrogel with only one kind of crosslinking point presented in the structure of the hydrogel. In this study, a two-component supramolecular dynamic cross-linking hydrogel system was designed, employing the host-guest interaction between cucurbit[8]uril (CB[8]) and its homodimer guests to form ternary complex at a ratio of 1/2. The azobenzene guest residues and the phenyl pyridinium guest motifs were introduced into the side chain of hydrogel backbone polymer, in which the azobenzene guest would weakly bind with CB[8] showing faster dynamics, and the phenyl pyridinium salt guest would tightly bind with CB[8] showing slower dynamics. A series of hydrogel materials with the same network structure but dynamically adjustable properties were prepared by adjusting the proportions of these two dynamic cross-linking points. The obtained hydrogel materials could continuously change their properties such as rheology mechanical properties, self-healing, shear thinning, stress relaxation behavior, and macromolecular drug release, all of which are essential in biomedical fields and thus showed great potential in relevant applications. Abstract:The CB[8] modified Fe3O4 nanoparticles as cross‐linker for fabricating the hydrogels, which exhibit multifunction, including self‐healing, photothermal responsive and strain sensitive. The development of multifunctional supramolecular nanocomposite hydrogels remains challenging. Here, the dynamic host–guest interactions involving the host molecule CB[8] and guest units were utilized to prepare Fe3O4 hybrid supramolecular nanocomposite hydrogels. The results showed that the hydrogels obtained possessed a porous structure. The CB[8]‐modified Fe3O4 (Fe3O4@CB[8]) nanoparticles served as cross‐linkers in forming the network of hydrogels. By adjusting the Fe3O4@CB[8] content, the mechanical properties of the hydrogels could be controlled. The tensile stress was measured at 160 kPa with a fracture strain of 1380%, while the compression stress was 230 kPa at 70% compression strain. The self‐healing efficiency of the hydrogels at room temperature reached 95% after 24 h. The as‐obtained hydrogels show strain sensitivity and have the potential for applications in detecting elbow and finger movements. Our supramolecular nanocomposite hydrogels exhibit multiple functions, including self‐healing, injectability, photothermal responsiveness, and conductivity, making them suitable for integration into flexible electronics. Abstract:Stimuli-responsive polymeric hydrogels based on host-guest interactions have demonstrated potential as smart drug delivery platforms. However, host-guest interactions alone have been repeatedly highlighted to provide insufficient mechanical strength for the formed hydrogels likely due to the generally low grafting degrees of functional groups and steric hindrance of polymer backbone and grafted functions. Herein, a combination strategy of reversible addition-fragmentation chain-transfer polymerization (RAFT) and dual crosslinking was employed to fabricate thermo-and pH-dual responsive hydrogels with enhanced mechanical properties. Spe-cifically, two kinds of poly (N-isopropyl acrylamide) (PNIPAM)-based well-defined thermo-sensitive polymers, denoted as PH and PG, consisting of beta-cyclodextrin (beta-CD) and adamantane (Ad) groups were synthesized via RAFT copolymerization. A facile mixing of PH and PG with commercially available adipic dihydrazide led to the construction of hydrogels via host-guest interactions and dynamic acylhydrazone bonding. The resulting hydrogels not only exhibited pH-switchable sol-gel transitions and temperature-dependent swelling behaviors, but also possessed good stretchability (fracture stress up to 39.97 kPa) and self-healing property (self-healing efficiency above 90%). Notably, the resulting hydrogels showed pH-responsive controlled spatiotemporal release administration of rhodamine B due to the presence of dynamic acylhydrazone links. Together with good cyto-compatibility, the hydrogels developed herein represent promising candidates for potential transdermal drug delivery.

Abstract:The development of multifunctional supramolecular hydrogels with high mechanical strength and multifunction is in high demand. In this work, the diblock copolymer poly(acrylamide‐co‐1‐benzyl‐3‐vinylimidazolium bromide)–block–polyAzobenzene was synthesized through reversible addition−fragmentation chain transfer polymerization. The dynamic host–guest interactions between the host molecule cucurbit [8] uril and guest units were used to fabricate a 3D network of supramolecular hydrogels. Investigations on the properties of the supramolecular hydrogels show that the tensile stress of the sample is 1.46 MPa, eight times higher than that of hydrogel without liquid‐crystalline block copolymer, and the self‐healing efficiency of the supramolecular hydrogels at room temperature is 88.3% (fracture stress) and 100% (fracture strain) after 24 h. Results show that microphase‐separated structure plays a key role in high strength hydrogel, whereas the host–guest interaction endows the hydrogel with self–healing property. The supramolecular hydrogels with high mechanical strength, photo‐responsivity, injectablility and biocompatiblility can be used in various potential applications. Abstract:Pentacyclic triterpenoids, a class of naturally bioactive products having multiple functional groups, unique chiral centers, rigid skeletons, and good biocompatibility, are ideal building blocks for fabricating versatile supramolecular structures. In this research, the natural pentacyclic triterpenoid glycyrrhetinic acid (GA) was used as a guest molecule for -cyclodextrin (-CD) to form a GA/-CD (1:1) inclusion complex. By means of GA and -CD pendant groups in N,N-dimethylacrylamide copolymers, a supramolecular polymer hydrogel can be physically cross-linked by host-guest interactions between GA and -CD moieties. Moreover, self-healing of this hydrogel was observed and confirmed by step-strain rheological measurements, whereby the maximum storage modulus occurred at a [GA]/[-CD] molar ratio of 1:1. Additionally, these polymers displayed outstanding biocompatibility. The introduction of a natural pentacyclic triterpenoid into a hydrogel system not only provides a biocompatible guest-host complementary GA/-CD pair, but also makes this hydrogel an attractive candidate for tissue engineering.

Abstract:Automatic repair: a polymer with pendent dibenzo[24]crown-8 units (purple in picture) was cross-linked by two bisammonium salts (green) to form two supramolecular gels based on host-guest interactions. These two gels are stimuli-responsive materials that respond to changes of the pH value and are also self-healing materials, as can be seen by eye and as evidenced by rheological data.

Abstract:We report here a supramolecular hydrogel displaying a wide array of dynamic desirable properties. The key is using an ABA triblock copolymer containing a central poly(ethylene oxide) block and terminal poly(N-isopropylacrylamide) (PNI-PAm) block with ureido pyrimidinone (UPy) moieties randomly incorporated. Rapid hydrogelation is triggered upon increasing temperature above the lower critical solution temperature (LCST) of the supramolecular copolymer, where PNIPAm segments dehydrate and assemble into micelles, which subsequently provide hydrophobic microenvironments promoting UPy dimerization to grab polymer chains, thus forming hydrogen-bonded cross-linking points. The supramolecular hydrogels demonstrate fascinating shear-thinning, self-healable, thermo-reversible, and injectable properties, which allow withstanding repeated deformations and 3D construction of complex objects. Mesenchymal stem cells mixed with the hydrogel and injected through needles remain highly viable (>90%) during the encapsulation and delivery process. With these attractive dynamic physical properties, the supramolecular hydrogel holds great promise to support cell or drug therapies.

Abstract:The increasing preference for minimally invasive surgery requires novel soft materials that are injectable, with rapid self-healing abilities, and biocompatible. Here, by utilizing the synergetic effect of hydrophobic interaction and quadruple hydrogen bonding, an injectable supramolecular hydrogel with excellent self-healing ability was synthesized. A unique ABA triblock copolymer was designed containing a central poly(ethylene oxide) block and terminal poly(methylmethacrylate) (PMMA) block, with ureido pyrimidinone (UPy) moieties randomly incorporated (termed MA-UPy-PEO-UPy-MA). The PMMA block could offer a hydrophobic microenvironment for UPy moieties in water and thus boost the corresponding quadruple hydrogen bonding interaction of Upy–Upy dimers. Owing to the synergetic effect of hydrophobicity and quadruple hydrogen bonding interaction, the obtained MA-UPy-PEO-UPy-MA hydrogel exhibited excellent self-healing properties, and injectable capability, as well as superior mechanical strength, and therefore, it holds great promise in tissue engineering applications, including in cell support and drug release.

Abstract:As quite generally stated in the literature, low-molecular-weight (MW) poly(ethylene glycol) (PEG) (M-n less than 2K) and alpha-cyclodextrin (alpha-CD) can lead to only crystalline precipitates (not hydrogels). However, in this study we found that: (1) adamantane monoend-functionalized low-MW PEG (Ada-PEG, M-n = 1.1 or 2K) and alpha-CD lead to hydrogels but not to crystalline precipitates and (2) beta-cyclodextrin (beta-CD) surface-functionalized silica nanoparticles (beta-CD-SiO2) can be well dispersed in low-MW Ada-PEG and alpha-CD aqueous mixtures, resulting in hybrid hydrogels. The hydrophobic aggregation of Ada-PEG in case I and the further functionalization of beta-CD-SiO2 with PEG chains due to the inclusion complexation between beta-CD and the Ada group attached to PEG in case 2 were found to play a key role as a supra-crosslink that promoted the gelation of the inclusion complexes of alpha-CD and the low-MW Ada-PEG. The resultant native and hybrid hydrogels retained the basic characteristics of the supramolecular physical hydrogels, especially the shear-thinning property. The mechanical strength and the viscosity of the hybrid hydrogel were greatly improved in comparison with those of the corresponding native hydrogels.

Abstract:Natural polymer hydrogels are one of the best biomaterials for soft tissue repair because of their excellent biocompatibility, biodegradability and low immune rejection. However, they lack mechanical strength matching that of natural tissue and desired functionality (e.g., self-healing and 3D-printability). To solve these problems, we developed a host-guest supramolecule (HGSM) with three arms covalently crosslinked with a natural polymer to construct a novel hydrogel with non-covalent bonds integrated into a covalently crosslinked network. This unique structure enabled the hydrogel to exhibit improved mechanical properties and show both self-healing and 3D printing capabilities. The three-armed HGSM was first prepared via efficient non-covalent host-guest inclusion interactions between isocyanatoethyl acrylate-modified -cyclodextrin (-CD-AOI(2)) and acryloylated tetra-ethylene glycol-modified adamantane (A-TEG-Ad). Subsequently, a host-guest supramolecular hydrogel (HGGelMA) was obtained through copolymerization between the arms of the HGSM and gelatin methacryloyl (GelMA) to form a covalently crosslinked network. The HGGelMA was robust, fatigue resistant, reproducible and rapidly self-healing. In the HGGelMA, the covalent crosslinking maintained its overall shape, whereas the weak reversible non-covalent host-guest interactions reinforced its mechanical properties and enabled it to rapidly self-heal upon fracturing. The reversible non-covalent interactions could be re-established upon breaking, so as to heal the hydrogel and dissipate energy to prevent catastrophic fracture propagation. Furthermore, the precursors of the HGGelMA were sufficiently viscous and could be rapidly photocrosslinked to produce a robust scaffold with an exquisite internal structure through 3D printing. The 3D-printed HGGelMA hydrogel scaffold was biocompatible, promoted cell adhesion and proliferation, and supported tissue in-growth. Our strategy of integrating a non-covalently linked HGSM into a covalently linked hydrogel network represents a new approach to the development of natural polymers into biocompatible hydrogels with improved strength as well as desired self-healing and 3D-printability.
Cyclodextrin-based host–guest supramolecular hydrogel and its application in biomedical fields

Abstract:The cyclodextrin (CD) based host–guest inclusion complexation between cyclodextrins (CDs) and guest moieties has inspired the fabrication of novel supramolecular hydrogels for biomedical applications. From a topology of view, this article reviews the recent developments of two kinds of CD-based supramolecular hydrogels and their applications in biomedical fields respectively. On one hand, supramolecular hydrogels derived from CD-based poly(pseudo)rotaxanes generally displayed thixotropic and reversible properties and have been extensively developed as injectable drug delivery systems. On the other hand, supramolecular hydrogels based on the host–guest interaction between CDs and small guest moieties generally exhibited stimuli-responsive behaviors, typically the release of therapeutic agents. The development of CD-based supramolecular hydrogels provides a new platform for the design of novel biomaterials.

Abstract:Injectable conducting hydrogels (ICHs) are promising conductive materials in biomedicine and bioengineering fields. However, the synthesis of ICHs in previous work involved chemical cross-linking, and this may result in biocompatibility problems of the hydrogels. We present the successful synthesis of ICHs via noncovalent host-guest interactions, avoiding the side effect of covalent chemical cross-linking. The ICHs are based on the γ-cyclodextrin dimer as the host molecule and tetraaniline and poly(ethylene glycol) as the guests in a synthetic well-defined hydrophilic copolymer. The sol-gel transition mechanism of the in situ hydrogel is thoroughly investigated. This novel synthesis approach of ICHs via supramolecular chemistry will lead to various new biomedical applications for conducting polymers.
