Abstract:5-Hydroxymethyl-2′-deoxycytidine (5hmdC) phosphoramidite and triphosphate are important building blocks in 5hmdC-containing DNA synthesis for epigenetic studies. However, efficient and practical methods for the synthesis of these compounds are still limited. The current research provides an intensively improved synthetic method that enables the preparation of commercially available cyanoethyl-protected 5hmdC phosphoramidite with an overall yield of 39% on 5 g scale. On the basis of facile and efficient accesses to cyanoethyl protected-5hmdU and 5hmdC intermediates, two efficient synthetic routes for 5hmdC triphosphate were also developed.

Abstract:Nucleotide analogues empower chemical biologists to investigate the relationship between RNA structure and function on a level deeper than would be possible using only the four natural nucleotides. N-4-Methylcytidine and N-4-hydroxycytidine represent two cytidine analogues whose physicochemical properties enable biochemical tests for specific aspects of cytidine function within RNA. Here, we describe an efficient synthesis of N-4-methylcytidine and N-4-hydroxycytidine derivatives, and their corresponding phosphoramidites in 40.7% and 25.2% yield, respectively, starting from uridine. These phosphoramidites couple efficiently during solid-phase synthesis, although N-4-hydroxycytidine reverts partially to cytidine (5-10%) during oligonucleotide synthesis and deprotection.

Abstract:New cADPR mimic 5 that integrates the simplified nucleobase and ribose was designed and synthesized by using Cu(Ⅰ)-catalyzed Huisgen [3+2] cycloaddition to build the simplified triazole nucleobase and microwave-assisted reaction to carry out intramolecular pyrophosphorylation.Primary biological investigation showed that the introduction of an ether strand to replace northern and southern riboses could retain calcium inducing activity,but the introduction of a carbon strand to replace the northern ribose could lead to decreased activity.

Abstract:The utilization of 5-aminopropargyl nucleotide serves as a versatile molecular biology tool for the introduction of functional groups into a nucleic acid target of interest by using in-vitro enzymatic incorporation method. This article describes a simple, reliable, general, and efficient two-step chemical method for the synthesis of 5-(3-aminopropargyl)-2'-deoxycytidine-5'-O-triphosphate, 5-(3-aminopropargyl)-cytidine-5'-O-triphosphate, 5-(3-aminopropargyl)-2'-deoxyuridine-5'-O-triphosphate, and 5-(3-aminopropargyl)-uridine-5'-O-triphosphate, starting from the corresponding pyrimidine triphosphate. The first step involves regioselective C-5 iodination of pyrimidine triphosphate using N-iodosuccinimide and sodium azide. In the second step, propargylamine is coupled to the iodo-pyrimidine using the palladium-catalyzed Sonogashira reaction, producing good yields of highly pure (>99.5% HPLC) 5-aminopropargyl-pyrimidine-5'-O-triphosphate. In this approach, the palladium-catalyzed Sonogashira coupling reaction is highly chemoselective and does not involve protection and deprotection. © 2019 by John Wiley & Sons, Inc.

Abstract:A series of novel 1'-O-alpha-methyl isonucleosides were efficiently and stereoselectively synthesized with 1,2,3,5-tetra-O-acetyl-D-ribofuranose as the starting material. The key steps include regioselective and stereoselective deprotection of the 2,4-dichlorobenzyl group at C2, triflation, and substitution with appropriate nucleobases using cesium carbonate as the base. Removal of the residual 2,4-dichlorobenzyl groups and subsequent transformation afforded the title compounds in 30-37% overall yield. The products represent a new type of isonucleosides with 1'-O-substitution.

Abstract:L‐2'‐deoxythreose nucleoside phosphonates PMDTA and PMDTT possess potent anti‐HIV activity. Herein, a novel class of 3'‐C‐branched‐l ‐threose nucleoside phosphonate analogs, 5'‐deoxy‐3'‐C‐hydroxymethyl‐3'‐O‐phosphonomethyl‐d ‐xylose nucleosides, were synthesized and biologically evaluated. The key sugar intermediate 3‐C‐benzyloxymethyl‐3‐O‐diethylphosphonomethyl‐1,2‐O‐isopropylidene‐α‐d ‐5‐deoxyxylose (8 ) was firstly synthesized, which may be an interesting scaffold for access to diverse 3'‐C‐branched l ‐threosyl nucleoside phosphonate derivatives. And the key synthesis involved Wittig olefination of 1,2‐O‐isopropylidene‐3‐oxo‐α‐d ‐5‐deoxyxylose, stereoselective dihydroxylation of alkenes by aqueous KMnO4, selective benzylation of hydroxymethyl group under activation of dibutyltin oxide, and introduction of phosphonate group by nucleophilic substitution. Eventually, glycosylation under Vorbrüggen conditions provided 3'‐C‐hydroxymethyl‐3'‐O‐phosphonomethyl‐β‐d ‐5'‐deoxyxylose nucleoside analogs in satisfying yield.

Abstract:Nucleoside 5'-triphosphates (NTPs) play key roles in biology and medicine. However, these compounds are notoriously difficult to synthesize. We describe a one-pot method to prepare NTPs from nucleoside 5'-H-phosphonate monoesters via pyridinium phosphoramidates, and we used this approach to synthesize ATP, UTP, GTP, CTP, ribavirin-TP, and 6-methylpurine ribonucleoside-TP (6MePTP). Poliovirus RNA-dependent RNA polymerase efficiently employed 6MePTP as a substrate, suggesting that the cognate nucleoside, a poorly understood antiviral agent, may damage viral RNA.

Abstract:The chemical synthesis of phosphoramidite derivatives of all four 5'-deoxy-5'-thioribonucleosides is described. These phosphoramidites contained trityl (A, G, C, and U), dimethoxytrityl (A and G), or tert-butyldisulfanyl (G) as the 5'-S-protecting group. The application of several of these phosphoramidites for solid-phase synthesis of oligoribonucleotides containing a 2'-O-photocaged 5'-S-phosphorothiolate linkage or 5'-thiol-labeled RNAs is also further investigated.

Abstract:A general approach is presented for synthesizing alkyne-modified nucleoside triphosphates via the Sonogashira cross-coupling reaction of unprotected halogenated 2ʹ-deoxynucleoside, followed by monophosphorylation and the reaction of the corresponding phosphoromorpholidate with tributylammonium pyrophosphate. A highly efficient approach for the milligram-scale synthesis of base-modified nucleoside triphosphates with an amino acid-like side chain was developed. The present chemical method outweighs the other reported methods of a base-modified nucleoside triphosphates synthesis in terms of it being a protection-free strategy, the shortening of reaction steps, and increased yields (about 70%). The resulting 8-alkynylated dATP was tested as a substrate for DNA polymerases in a primer extension reaction.

Abstract:In the structures of the HDV ribozyme a cytosine nucleobase resides at the active site poised to participate directly in catalysis Defining the functional role of the nucleobase requires nucleoside analogues that perturb the functional groups in a strategic manner. Herein, we have developed efficient methods for the synthesis of five C-nucleoside phosphoramidite derivatives that, when used in combination, provide strategies for probing the potential functional role of cytosine's keto group and imino nitrogen. Phosphoramidites 15a and 15b were synthesized in 11 steps starting from 2-amino-5-bromopyrimidine (1a) and 2-amino-5-bromopyridine (1b), respectively, with overall yields of 10.8% and 6.6%, respectively. Phosphoramidite 21 was prepared from intermediate 11b in seven steps with an overall yield of 33.7%. Phosphoramidites 23 and 25 were prepared from 2,4-diamino-5-(beta-D-ribofuranosyl)-1,3-pyrimidine (22) and pseudoisocytidine (24), respectively, with in overall yield of 15.9% (six steps) and 37.9% (four steps), respectively. These phosphoramidites were incorporated into oligonucleotides by solid-phase synthesis.

Abstract:OBJECTIVE To find a commercially useful route for the preparation of 1-(2′-bromo-2′-deoxy-3′,5′-di-O-propionyl-β-D-ribosyl)thymine. METHOD To uses thymine and tetra-O-acylribofuranose as starting materials. The process involved the following steps: silylanization, condensation, deacetylation and halogenation. RESULTS The procedure was simple and the yield of this route was higher than other synthetic routes by using cheaper reagents.CONCLUSION The improved synthetic process is practical.

Abstract:Fifteen new 3-[(4-hydroxy-6-methly-2(1H)-pyridinones)-3-yl]-4-substituted- (1H)-1,2,4-triazole-5-thione compounds were synthesized from methyl 4-hydroxy-6- methyl-2-oxo-1,2-dihydropyridine-3-carboxylate in good overall yields by four step reactions. Molecular structure of these compounds was confirmed by IR, NMR, MS, elemental analysis and X-ray diffraction. Their antitumor activities were evaluated in vitro by the SRB and MTT methods. The results show that there are two compounds which are highly effective against KB cells and one compound which is highly effective against Bel-7402 cells.

Abstract:5-azacitidine is a prescription injectable drug used for the treatment of myelodysplastic syndromes (MDS) and acute myelogenous leukemia (AML). In this paper, we would like to report a new approach to 5-azacitidine analogs from dicyanodiamine and formic acid or benzoic acid through several efficient reactions, such as cyclization reaction, Vorbr��ggen coupling, and so on. Among those compounds, two azacytosine analogs and two 5-azacitidine analogs have not been reported yet. All products and important intermediates were characterized by 1HNMR and MS spectrums. ? Abstract:An optimized approach for the synthesis of N1-ethoxymethyl-substituted cyclic inosine diphosphoribose (cIDPRE),an analogue of cyclic adenosine diphosphoribose (cADPR),has been developed via microwave-assisted intramolecular cyclization.The target compound has been successfully obtained through N1-substitution,phosphorylation,cyclization and deprotection.By using this method,8-amino and bromo-substituted cIDPRE analogues were successfully obtained in good yield.The new approach has greatly shortened the synthetic route and enhanced the overall efficiency.
