Abstract:Converting common biomass materials to high-performance biomedical products could not only reduce the environmental pressure associated with the large-scale use of synthetic materials, but also increase the economic value. Chitosan as a very promising candidate has drawn considerable attention owing to its abundant sources and remarkable bioactivities. However, pure chitosan materials usually exhibit insufficient mechanical properties and excessive swelling ratio, which seriously affected their in vivo stability and integrity when applied as tissue engineering scaffolds. Thus, simultaneously improving the mechanical strength and biological compatibility of pure chitosan (CS) scaffolds becomes very important. Here, inspired by the fiber-reinforced construction of natural extracellular matrix and the porous structure of cancellous bone, we built silk microfibers/chitosan composite scaffolds via ice-templating technique. This biomimetic strategy achieved 500% of mechanical improvement to pure chitosan, and meanwhile still maintaining high porosity (> 87%). In addition, the increased roughness of chitosan pore walls by embedded silk microfibers significantly promoted cell adhesion and proliferation. More importantly, after subcutaneous implantation in mice for four weeks, the composite scaffold showed greater structural integrity, as well as better collagenation, angiogenesis, and osteogenesis abilities, suggesting its great potential in biomedicine.
Preparation and Characterization of Bionic Bone Structure Chitosan/hydroxyapatite Scaffold for Bone Tissue Engineering.

Abstract:Three-dimensional oriented chitosan (CS)/hydroxyapatite (HA) scaffolds were prepared via in situ precipitation method in this research. Scanning electron microscopy (SEM) images indicated that the scaffolds with acicular nano-HA had the spoke-like, multilayer and porous structure. The SEM of osteoblasts which were polygonal or spindle-shaped on the composite scaffolds after seven-day cell culture showed that the cells grew, adhered, and spread well. The results of X-ray powder diffractometer and Fourier transform infrared spectrometer showed that the mineral particles deposited in the scaffold had phase structure similar to natural bone and confirmed that particles were exactly HA. In vitro biocompatibility evaluation indicated the composite scaffolds showed a higher degree of proliferation of MC3T3-E1 cell compared with the pure CS scaffolds and the CS/HA10 scaffold was the highest one. The CS/HA scaffold also had a higher ratio of adhesion and alkaline phosphate activity value of osteoblasts compared with the pure CS scaffold, and the ratio increased with the increase of HA content. The ALP activity value of composite scaffolds was at least six times of the pure CS scaffolds. The results suggested that the composite scaffolds possessed good biocompatibility. The compressive strength of CS/HA15 increased by 33.07% compared with the pure CS scaffold. This novel porous scaffold with three-dimensional oriented structure might have a potential application in bone tissue engineering.
Engineering Scaffolds Integrated with Calcium Sulfate and Oyster Shell for Enhanced Bone Tissue Regeneration

Abstract:Engineering scaffolds combinging natural biomineral and artificially synthesized material hold promising potential for bone tissue regeneration. In this study, novel bioactive calcium sulfate/oyster shell (CS/OS) composites were prepared. Comparing to CS scaffold, the CS/OS composites with a controllable degradation rate displayed enhanced mineral nodule formation, higher alkaline phosphate (ALP) activity and increased proliferation rate while treated osteocytes. In CS/OS composites group, elevated mRNA levels of key osteogenic genes including bone morphogenetic protein-2 (BMP-2), runt-related transcription factor 2 (Runx2), osterix (Osx), and osteocalcin (OCN) were observed. Furthermore, The up-regulation of BMP-2 and type I collagen (COL-I) was observed for CS/OS composites relative to a CS group. Scaffolds were implanted into critical-sized femur cavity defects in rabbits to investigate the osteogenic capacity of the composites in vivo. The CS/OS scaffolds with proper suitable times and mechanical strength strongly promoted osteogenic tissue regeneration relative to the regeneration capacity of CS scaffolds, as indicated by the results of histological staining. These results suggest that the OS-modified CS engineering scaffolds with improved mechanical properties and bioactivity would facilitate the development of a new strategy for clinic bone defect regeneration.
Chitosan/alginate bionanocomposites adorned with mesoporous silica nanoparticles for bone tissue engineering

Abstract:The regeneration of oral and craniofacial bone defects ranging from minor periodontal and peri-implant defects to large and critical lesions imposes a substantial global health burden. Conventional therapies are associated with several limitations, highlighting the development of a unique treatment strategy, such as tissue engineering. A well-designed scaffold for bone tissue engineering should possess biocompatibility, biodegradability, mechanical strength, and osteoconductivity. For this purpose, mesoporous silica nanoparticles (MSNs) were synthesized and incorporated at different ratios (10, 20, and 30%) into alginate/chitosan (Alg/Chit)-based porous composite scaffolds fabricated through the freeze-drying method. The MSN incorporation significantly improved the mechanical strength of the scaffolds while showing a negligible decreasing effect on the porosity. All of the samples showed desirable swelling behaviors, which is beneficial for cell attachment and proliferation. The MSN-containing scaffolds indicated a decreased hydrolytic degradation in an MSN percentage-dependent manner. The fabricated scaffolds did not depict cytotoxic characteristics. The Alg/Chit/MSN30 scaffolds not only showed noncytotoxic properties, but also increased the cell viability significantly compared to the control group. The biomineralization properties of the MSN-containing nanocomposite scaffolds were significantly higher than the Alg/Chit composite, suggesting the potential of these nanoparticles for bone tissue engineering applications. Taken together, it is concluded that the Alg/Chit/MSN30 scaffolds are considerable substances for bone tissue regeneration, and MSN has a great tissue engineering potential in addition to its extensive biomedical applications.Graphical abstract

Abstract:Chitin nanocrystals (CNCs) with length and width of 300 and 20nm were uniformly dispersed in chitosan (CS) solution. The CS/CNCs composite scaffolds prepared utilizing a dispersion-based freeze dry approach exhibit significant enhancement in compressive strength and modulus compared with pure CS scaffold both in dry and wet state. A well-interconnected porous structure with size in the range of 100-200μm and over 80% porosity are found in the composite scaffolds. The crystal structure of CNCs is retained in the composite scaffolds. The incorporation of CNCs leads to increase in the scaffold density and decrease in the water swelling ratio. Moreover, the composite scaffolds are successfully applied as scaffolds for MC3T3-E1 osteoblast cells, showing their excellent biocompatibility and low cytotoxicity. The results of fluorescent micrographs images reveal that CNCs can markedly promote the cell adhesion and proliferation of the osteoblast on CS. The biocompatible composite scaffolds with enhanced mechanical properties have potential application in bone tissue engineering.

Abstract:Inspired by the natural extracellular matrix, the organic-inorganic composite nanofibers are promising scaffolds for bone tissue engineering. Chitosan-based nanofibers are widely used as bone tissue engineering scaffolds with good biocompatibility but pungent solvents are frequently used for its processing. Carboxymethyl chitosan (CMCS), a water-soluble derivative of chitosan, has better biodegradability and bioactivity which allows CMCS to chelate Ca2+ and induce the deposition of apatite. Moreover, with water as solvent, CMCS nanofibers avoid the acidic salt removal comparing to electrospun-chitosan. In this study, we successfully prepared uniform CMCS nanofibers with the aid of polyethylene oxide (PEO) and obtained the optimized conditions with a voltage of 25 kV and PEO of molecular weight 1000 kDa. We further prepared hydroxyapatite (HA) coated electrospun CMCS nanofibers by biomimetic mineralization using 5 times simulated body fluid. The promotion of osteogenic differentiation of mouse bone marrow stromal cells (mBMSCs) in vitro was evaluated on the nanofibers scaffolds. Cell experiments revealed that CMCS-HA composite nanofibers increased the ALP activity. The gene expression level of Runx2 and ALP were about 1.6 and 4.3 folds at the 7 days, and 5.1 and 10 folds at the 14 days on CMCS-HA nanofibrous membranes than that on CMCS alone samples. The level of OCN increased by 24 and 1.5 times on the CMCS-HA scaffolds than CMCS scaffolds at the 14 and 21 days. In vivo new bone formation by nanofiber scaffolds was investigated in a critical-size rat calvarial bone defect model. Micro-CT results showed that the whole defect was covered by new bone after CMCS-HA filling the defect for 12 weeks. The results of H&E staining and Masson's trichrome staining on histological sections further confirmed that composite nanofibers promoted new bone formation and maturation.
Biomineralized poly (l-lactic-co-glycolic acid)-tussah silk fibroin nanofiber fabric with hierarchical architecture as a scaffold for bone tissue engineering

Abstract:In bone tissue engineering, the fabrication of a scaffold with a hierarchical architecture, excellent mechanical properties, and good biocompatibility remains a challenge. Here, a solution of polylactic acid (PLA) and Tussah silk fibroin (TSF) was electrospun into nanofiber yarns and woven into multilayer fabrics. Then, composite scaffolds were obtained by mineralization in simulated body fluid (SBF) using the multilayer fabrics as a template. The structure and related properties of the composite scaffolds were characterized using different techniques. PLA/TSF (mass ratio, 9:1) nanofiber yarns with uniform diameters of 72±9μm were obtained by conjugated electrospinning; the presence of 10wt% TSF accelerated the nucleation and growth of hydroxyapatite on the surface of the composite scaffolds in SBF. Furthermore, the compressive mechanical properties of the PLA/TSF multilayer nanofiber fabrics were improved after mineralization; the compressive modulus and stress of the mineralized composite scaffolds were 32.8 and 3.0 times higher than that of the composite scaffolds without mineralization, respectively. Interestingly, these values were higher than those of scaffolds containing random nanofibers. Biological assay results showed that the mineralization and multilayer fabric structure of the composite nanofiber scaffolds significantly increased cell adhesion and proliferation and enhanced the mesenchymal stem cell differentiation toward osteoblasts. Our results indicated that the mineralized nanofiber scaffolds with multilayer fabrics possessed excellent cytocompatibility and good osteogenic activity, making them versatile biocompatible scaffolds for bone tissue engineering.
Hybrid Nanostructured Hydroxyapatite-Chitosan Composite Scaffold: Bioinspired Fabrication, Mechanical Properties and Biological Properties.

Abstract:The fabrication of bone scaffolds with interconnected porous structure, adequate mechanical properties, excellent biocompatibility and osteoinductivity presents a great challenge. Herein, a hybrid nanostructured hydroxyapatite-chitosan (HA-CS) composite scaffold has been fabricated according to the following steps: (i) the deposition of brushite-CS on a CS fibre porous scaffold by a dip-coating method; and (ii) the formation of a hybrid nanostructured HA-CS composite scaffold by the in situ conversion of brushite to HA using a bioinspired mineralization process. The hybrid HA-CS composite scaffold possesses three-dimensional (3D) interconnected pores with pore sizes of 30-80 μm. The HA rods with a length of ∼200 nm and width of ∼50 nm are perpendicularly oriented to the CS fibres. Interestingly, the abovementioned HA rods are composed of many smaller nanorods with a length of ∼40 nm and width of ∼10 nm oriented along the c-axis. The hybrid nanostructured HA-CS composite scaffold exhibits good mechanical properties with a compression strength of 9.41 ± 1.63 MPa and an elastic modulus of 0.17 ± 0.02 GPa, which are well-matched to those of trabecular bone. The influences of the hybrid HA-CS composite scaffold on cells have been investigated using human bone marrow stem cells (hBMSCs) as cell model and the CS fibre porous scaffold as the control sample. The hybrid HA-CS composite scaffold not only supports the adhesion and proliferation of hBMSCs, but also improves the osteoinductivity. The alkaline phosphatase activity and mineralization deposition on the hybrid HA-CS composite scaffold are higher than those on the CS fibre porous scaffold. Moreover, the hybrid HA-CS composite scaffold can promote the formation of new bone in rat calvarial defects as compared with the CS fibre porous scaffold. The excellent biocompatibility, osteoinductivity and mechanical properties suggest that the hybrid nanostructured HA-CS composite scaffold has great potential for bone tissue engineering.

Abstract:In this study, we report the physico-chemical and biological properties of a novel biodegradable composite scaffold made of nano-hydroxyapatite and natural derived polymers of chitosan and carboxymethyl cellulose, namely, n-HA/CS/CMC, which was prepared by freeze-drying method. The physico-chemical properties of n-HA/CS/CMC scaffold were tested by infrared absorption spectra (IR), transmission electron microscope(TEM), scanning electron microscope(SEM), universal material testing machine and phosphate buffer solution (PBS) soaking experiment. Besides, the biological properties were evaluated by MG63 cells and Mesenchymal stem cells (MSCs) culture experiment in vitro and a short period implantation study in vivo. The results show that the composite scaffold is mainly formed through the ionic crossing-linking of the two polyions between CS and CMC, and n-HA is incorporated into the polyelectrolyte matrix of CS-CMC without agglomeration, which endows the scaffold with good physico-chemical properties such as highly interconnected porous structure, high compressive strength and good structural stability and degradation. More important, the results of cells attached, proliferated on the scaffold indicate that the scaffold is non-toxic and has good cell biocompatibility, and the results of implantation experiment in vivo further confirm that the scaffold has good tissue biocompatibility. All the above results suggest that the novel degradable n-HA/CS/CMC composite scaffold has a great potential to be used as bone tissue engineering material.

Abstract:The fabrication of bone scaffolds with interconnected porous structure, adequate mechanical properties and excellent biocompatibility presents a great challenge. Herein, a hybrid nanostructured chitosan/chondroitin sulfate/hydroxyapatite (ChS/CSA/HAP) in situ composite scaffold was prepared by in situ fabrication and freeze-drying technique. The composition and morphology of scaffold were characterized by Fourier transform infrared (FT-IR) spectroscopy, X-ray diffraction (XRD) and scanning electron microscopy (SEM). It proved that the low crystallinity of HAP crystals were uniformly distributed in ChS/CSA organic matrix and the nanostructured hybrid scaffold exhibited good mechanical property. The biocompatibility and in vitro bioactivity were detected by MTT-assay, maturation (alkaline phosphatase (ALP) activity), Hoechst 33258 and PI fluorescence staining. In vitro tests indicated that the hybrid scaffold not only promoted the adhesion and proliferation of osteoblasts, but also improved the growth of the osteoblasts. Therefore, it is promising for bone repair application in bone tissue engineering.

Abstract:Carboxymethyl cellulose was first incorporated into nano-hydroxyapatite/chitosan to obtain a novel composite of nano-hydroxyapatite/chitosan/carboxymethyl cellulose (n-HA/CS/CMC) as a three-dimensional scaffolds by freeze-drying. The surface morphology and properties of the scaffold were investigated by infrared absorption spectra (IR), X-ray diffraction (XRD), scanning electron microscope (SEM), mechanical testing and soaking in simulated body fluids (SBF) soaking. The results showed that strong chemical interactions were formed between the three phases. Moreover, the n-HA/CS/CMC composite scaffold with 30wt% CMC had the most ideal porous structure with a pore size ranging from 100 to 500μm and a porosity 77.8%, and the highest compressive strength of 3.54MPa. In addition, the SBF soaking experiment showed the scaffold of 30wt% CMC had an acceptable degradation rate and good bioactivity in vitro. All the results suggest that n-HA/CS/CMC composite scaffolds have potential as bone tissue engineering materials.
A novel nanocomposite for bone tissue engineering based on chitosan–silk sericin/hydroxyapatite: biomimetic synthesis and its cytocompatibility

Abstract:A simple and effective approach was developed to synthesize chitosan-silk sericin/hydroxyapatite nanocomposites by in situ precipitation and two methods of alkali diffusion were carried out in this study. The objective of this paper was to investigate the different properties of the nanocomposites. SEM showed that the rod-like hydroxyapatite particles with a diameter of 20-50 nm were distributed homogeneously within the chitosan-silk sericin matrix, and the formation mechanism was also investigated. The results of FTIR and XRD indicated that the inorganic phase in the nanocomposite was carbonate-substituted hydroxyapatite with low crystallinity. In terms of mechanical properties, chitosan-silk sericin/hydroxyapatite nanocomposites exhibited a higher elastic modulus and compressive strength than that of the chitosan/hydroxyapatite nanocomposites. In vitro cytocompatibility of the nanocomposite was evaluated by CCK-8 assay and SEM through MG63 osteoblast cells cultured on the samples, which demonstrated that they are non-toxic and support cell growth. These results suggest that the chitosan-silk sericin/hydroxyapatite nanocomposites are promising biomaterials for bone tissue engineering.
Nanocomposite Methacrylated Silk Fibroin-Based Scaffolds for Bone Tissue Engineering

Abstract:The treatment of bone defects is a clinical challenge. Bone tissue engineering is gaining interest as an alternative to current treatments, with the development of 3D porous structures (scaffolds) helpful in promoting bone regeneration by ensuring temporary functional support. In this work, methacrylated silk fibroin (SilMA) sponges were investigated as scaffolds for bone tissue engineering by exploiting the combination of physical (induced by NaCl salt during particulate leaching) and chemical crosslinking (induced by UV-light exposure) techniques. A biomimetic approach was adopted to better simulate the extracellular matrix of the bone by introducing either natural (mussel shell-derived) or synthetic-origin hydroxyapatite nanoparticles into the SilMA sponges. The obtained materials were characterized in terms of pore size, water absorption capability and mechanical properties to understand both the effect of the inclusion of the two different types of nanoparticles and the effect of the photocrosslinking. Moreover, the SilMA sponges were tested for their bioactivity and suitability for bone tissue engineering purposes by using osteosarcoma cells, studying their metabolism by an AlamarBlue assay and their morphology by scanning electron microscopy. Results indicate that photocrosslinking helps in obtaining more regular structures with bimodal pore size distributions and in enhancing the stability of the constructs in water. Moreover, the addition of naturally derived hydroxyapatite was observed to be more effective at activating osteosarcoma cell metabolism than synthetic hydroxyapatite, showing a statistically significant difference in the AlamarBlue measurement on day 7 after seeding. The methacrylated silk fibroin/hydroxyapatite nanocomposite sponges developed in this work were found to be promising tools for targeting bone regeneration with a sustainable approach.
Characterization of Silk Fibroin/Chitosan 3D Porous Scaffold and In Vitro Cytology

Abstract:Bone tissue engineering is a powerful tool to treat bone defects caused by trauma, infection, tumors and other factors. Both silk fibroin (SF) and chitosan (CS) are non-toxic and have good biocompatibility, but are poor biological scaffolds when used alone. In this study, the microscopic structure and related properties of SF/CS composite scaffolds with different component ratios were examined. The scaffold material most suitable for osteoblast growth was determined, and these results offer an experimental basis for the future reconstruction of bone defects. First, via freeze-drying and chemical crosslinking methods, SF/CS composites with different component ratios were prepared and their structure was characterized. Changes in the internal structure of the SF and CS mixture were observed, confirming that the mutual modification between the two components was complete and stable. The internal structure of the composite material was porous and three-dimensional with a porosity above 90%. We next studied the pore size, swelling ratio, water absorption ratio, degradation and in vitro cell proliferation. For the 40% SF-60% CS group, the pore size of the scaffold was suitable for the growth of osteoblasts, and the rate of degradation was steady. This favors the early adhesion, growth and proliferation of MG-63 cells. In addition to good biocompatibility and satisfactory cell affinity, this material promotes the secretion of extracellular matrix materials by osteoblasts. Thus, 40% SF-60% CS is a good material for bone tissue engineering.

Abstract:Good biocompatibility and osteogenesis of three-dimensional porous scaffolds are critical for bone tissue engineering. In this work, biomimetic hydroxyapatite/gelatin-chitosan core-shell nanofibers composite scaffolds have been fabricated to mimic both the specific structure and the chemical composition of natural bone. The coaxial electrospinning technique was introduced to prepare gelatin-chitosan core-shell structured nanofibers mat which formed three-dimensional porous structure for promoting cells growth. The gelatin-chitosan core-shell nanofibers formed Arginine-Glycine-Aspartic acid (RGD)-like structure to mimic the organic component of natural bone extracellular matrix. Hydroxyapatite (Ca10(PO4)6(OH)2, HAP), as the major mineral constituent of native bone, was then deposited onto the surface of gelatin-chitosan core-shell structured nanofibers by a wet chemical method. Compared with chitosan nanofibers, gelatin nanofibers and chitosan-gelatin composite nanofibers, gelatin-chitosan core-shell structured nanofibers improved the mineralization efficiency of hydroxyapatite and formed a homogeneous HAP deposit. When Human osteoblast like cell line (MG-63) were cultured on the materials, the results showed that hydroxyapatite deposited on the gelatin-chitosan core-shell structured nanofibers could further enhance osteoblast cell proliferation. The biomimetic composite scaffolds could be suggested as a promising material to promote osteoblast cell growth in bone tissue engineering.
Porous Nano-Minerals Substituted Apatite/chitin/pectin Nanocomposites Scaffolds for Bone Tissue Engineering

Abstract:In the current study, a porous 3D scaffold using Gallium-Apatite/chitin/pectin (Ga-HA/C/P) nanocomposites scaffolds (NCS) were fabricated by freeze-drying process with applications in orthopedics (bone tissue engineering). Various NCSs (0%, 30%, 50 and 70%) were prepared and characterized for its chemical structure, crystalline phase, surface texture by using various techniques such as FT-IR, XRD and SEM-EDX, respectively. The analyses of physicochemical properties proved that the formulated scaffolds were highly porous, and mechanically stable with superior density. The nanocomposite scaffolds also presented with increased swelling ability, lower biodegradation rate and higher mechanical strength. Further, biocompatibility and cytotoxicity of Ga-HA/C/P nanocomposite scaffolds were studied using NIH3T3 cells and MG-63 cells revealed no toxicity and cells attached and proliferated on scaffolds. Further implantation of prepared NCS showed mature bone formation through formation of new bone cells and osteoblast differentiation. Also, Ga-HA/C/P nanocomposites scaffolds proved to be more effective than chitin-pectin composite scaffolds. Taking results together it can be inferred that the prepared nanocomposite scaffolds possesses the prerequisites and showed great potential for treating orthopedic applications.
Nanosilica-Anchored Polycaprolactone/Chitosan Nanofibrous Bioscaffold to Boost Osteogenesis for Bone Tissue Engineering

Abstract:The strategy of incorporating bioactive inorganic nanomaterials without side effects as osteoinductive supplements is promising for bone regeneration. In this work, a novel biomass nanofibrous scaffold synthesized by electrospinning silica (SiO2) nanoparticles into polycaprolactone/chitosan (PCL/CS) nanofibers was reported for bone tissue engineering. The nanosilica-anchored PCL/CS nanofibrous bioscaffold (PCL/CS/SiO2) exhibited an interlinked continuous fibers framework with SiO2 nanoparticles embedded in the fibers. Compact bone-derived cells (CBDCs), the stem cells derived from the bone cortex of the mouse, were seeded to the nanofibrous bioscaffolds. Scanning electron microscopy and cell counting were used to observe the cell adhesion. The Counting Kit-8 (CCK-8) assay was used. Alkaline phosphatase (ALP), Alizarin red staining, real-time Polymerase Chain Reaction and Western blot tests were performed to confirm the osteogenesis of the CBDCs on the bioscaffolds. The research results demonstrated that the mechanical property of the PCL together with the antibacterial and hydrophilic properties of the CS are conducive to promoting cell adhesion, growth, migration, proliferation and differentiation. SiO2 nanoparticles, serving as bone induction factors, effectively promote the osteoblast differentiation and bone regeneration. This novel SiO2-anchored nanofibrous bioscaffold with superior bone induction activity provides a better way for bone tissue regeneration.

Abstract:A ternary composite scaffold of poly(L-lactide)/hydroxyapatite/chitosan fibers was fabricated for bone tissue engineering. Scanning electron microscopy analysis showed that hydroxyapatite particles and chitosan fibers in microscale uniformly distributing in poly(L-lactide) matrix not only increased the mechanical property but also prevented the crack and platelet formation on the pore surface of poly(L-lactide) matrix at cooling. The X-ray diffraction peak intensity of poly(L-lactide) component decrease may be due to the presence of hydroxyapatite particles and chitosan fibers as the crystal nuclei that led to the crystallinity decrease of poly(L-lactide). The in vitro degradation test showed that the degradation of poly(L-lactide) was resisted by enhancing the chitosan fiber content. The bone mesenchymal stem cell culture indicated that the composite scaffolds promoted the attachment and proliferation of cells, which would likely attach to and stretch along the chitosan fibers. Therefore, this ternary composite scaffold will be a promising matrix material for bone tissue engineering.

Abstract:Currently, great efforts have been made to enhance the mechanical strength of bone tissue engineering (BTE) scaffolds, which are composed of biopolymeric matrices and inorganic nano-fillers. But the tunability of mechanical strength in a wide range for BTE scaffolds has seldom been investigated in spite of the great importance of this performance. In this work, a chitosan-tussah silk fibroin/hydroxyapatite (CS-TSF/HAp) hydrogel was synthesized by using a novel in situ precipitation method. Through in situ inducing the conformation transition of TSF in the CS-TSF/HAp hydrogel, which could be monitored by XRD, FT-IR, TGA, and DTA, the elastic modulus and fracture strength of the final CS-TSF/HAp composite could be tailored in a wide range without changing its composition, morphology, roughness, and crystal structures. The elastic modulus of the CS-TSF/HAp composite ranged from ∼250 to ∼400MPa while its fracture strength ranged from ∼45 to ∼100MPa. In order to clarify the rationale behind this process, a speculative explanation was provided. In vitro cell culture indicated that MC3T3-E1 cells cultured on the CS-TSF/HAp composite had positive adhesion, proliferation, and differentiation potential. We believed that the CS-TSF/HAp composite could be used as an ideal scaffold platform for cell culture and implantation of bone reconstruction.

Abstract:Chitosan-based nanocomplexes with various forms were prepared by ionically crosslinking with tripolyphosphate (TPP) in different acidic media under mild conditions. It was found that the self-assembly and ionic interactions of chitosan and TPP were greatly affected by reaction media, and chitosan-based nanofibers could be obtained in adipic acid medium while nanoparticles were formed in acetic acid medium. Using bovine serum albumin (BSA) as a macromolecular model-drug, in vitro drug release studies indicated that chitosan-based nanofibers and nanoparticles exhibited a similar prolonged release profile. In addition, the bioinspired mineralization of both chitosan-based nanofibers and nanoparticles was carried out by soaking them in synthetic body fluids (SBF). Transmission electron microscopy (TEM) and X-ray Diffraction (XRD) results indicated that chitosan-based nanofibers have better inductivity for nano-hydroxyapatite formation than chitosan-based nanoparticles. The results suggested that biomimetic chitosan-based nanofibers with controlled release capacity of bioactive factors may be of use in bone tissue engineering for enhancing the bioactivity and bone inductivity.
