Abstract:Polymeric porous microspheres can be used as functional vehicles in drug delivery and cell culture. However, the conventional porous microspheres are somewhat complicated with respect to preparation, and limited in composition and controllability. Here we report the preparation of poly(lactic-co-glycolic acid)/calcium carbonate (PLGA/CC) composite microspheres with uniform superficial macropores through a facile single-step method, where CC acts simultaneously as in situ pore-forming agent and reinforcing phase. The SEM images and quantified results from mercury intrusion porosimetry indicated that the size and density of the superficial macropores were highly controllable via changing the starting parameters such as size and content of CC particles and concentration of PLGA. Mouse bone mesenchymal stem cells were cultured on microspheres of different topographies to investigate the cell-substrate interaction. The results showed that cells adhered and grew well on all microspheres, while the topography with smaller and more discrete macropores exhibited the highest proliferation, indicating that cells responded to the topography of the microsphere. This work provides a novel approach to obtain diverse porous composite microspheres designed for tissue repair and study of cell-substrate interaction.
Effect of Foreign Ion Substitution and Micropore Tuning in Robocasting Single-Phase Bioceramic Scaffolds on the Physicochemical Property and Vascularization

Abstract:The inorganic powder slurry extrusion printing technique known as robocasting is an interesting method to fabricate complex porous architectures whereby feedstocks containing organic binders and powders are printed and the resulting scaffolds are subjected to sintering. A major limiting factor of this technique is the simultaneous tailoring of vascularization efficacy and osteogenic activity, usually done by adding the secondary phase in the organic slurry before the writing step. Mechanical mixing of biphasic powders is required to avoid compromising the biological performance and physical defects caused by significantly different physicochemical properties. This study addresses this issue by developing a selective ion doping and microstructure tuning for the production of bioceramic scaffolds with a binozzle robocasting process. Different metal ions (Sr2+, Mg2+) were doped into wollastonite (CaSiO3; CSi) powders considering the mechanical stability and bioactive enhancement of the bioceramic scaffolds. Subsequently, the Mg-doped CSi slurries were used as shell-nozzle feedstocks added with 5, 10, and 15 μm diameter polystyrene microbeads that allowed shell-layer micropore production in pore struts during sintering. Finally, the most promising pore-strut microstructures and mechanical evolution of scaffolds were evaluated, and especially the enhanced fibrovascularization potential was confirmed in dorsal muscle embedding model in rabbits. This study may open an avenue to designing multiproperty-tuned macro- and microporous bioceramics for bone regenerative medicine, especially in challenging bone defect conditions.

Abstract:Hollow polymer microspheres have attracted great attention in tissue regeneration as cell carrier. In this study, hollow poly(lactic-co-glycolic acid)/hydroxyapatite/calcium carbonate (PLGA/HA/CC) microspheres with porous surface were successfully fabricated using bioactive minerals as in situ pore-forming agents. No templates were introduced in this facile approach. The capability of HA to hold water contributed to the formation of the porous surface and the hollow structure. Furthermore, the gas-releasing effect originating from CC decomposition facilitated the generation of opened superficial macropores. Porcine mesenchymal stem cells (PMSCs) were cultured on the microspheres and the results highlighted a better proliferation on microspheres with opened macropores. Therefore, this subtly designed microsphere shows a potential application in cell delivery.
Degradation pattern of porous CaCO3 and hydroxyapatite microspheres in vitro and in vivo for potential application in bone tissue engineering

Abstract:Despite superior clinical handling, excellent biocompatibility, biodegradation property of calcium phosphate needs to be improved to coincide with the rate of new bone formation. In this study, spherical CaCO3 are fabricated in the presence of the silk sericin and then transformed into porous hydroxyapatite (HAP) microspheres via hydrothermal method. The degradation behavior of obtained CaCO3, HAP and their mixture is first investigated in vitro. The result demonstrates that the weight loss of HAP microspheres are almost 24.3% after immersing in pH 7.40 Tris-HCl buffer solution for 12 weeks, which is far slower than that of spherical CaCO3 (97.5%). The degradation speed of the mixtures depends on the proportion of CaCO3 and HAP. The mixture with higher content of CaCO3 possesses a quicker degradation speed. The obtained CaCO3 and HAP microspheres are injected into subcutaneous tissue of ICR mice with the assistance of sodium alginate. The result in vivo also shows an obvious difference of degradation speed between the obtained CaCO3 and HAP microspheres, implying it is feasible to modulate the degradation property of the mixture through changing the proportion of CaCO3 and HAP The good cytocompatibility of the two kinds of microspheres is proved and a mild inflammation response is observed only at early stage of implantation. The job offers a simple method to modify the degradation properties of biomaterial for potential use in bone tissue engineering.

Abstract:This paper reports polymeric scaffolds with spherical internal macropores and relatively large external dimension. Paraffin spheres with the diameter of several hundred microns were prepared by a suspension technique. Particulate leaching technique based on this kind of spherical porogens was combined with room-temperature compression molding technique to fabricate biodegradable poly(D,L-lactic- co -glycolic acid) (PLGA) porous scaffolds potentially for tissue engineering or in situ tissue induction. The scaffolds exhibited ordered macropores with good pore interconnectivity. The porosity ranged from 80 to 97% adjusted simply by varying porogen content. The foams with porosity around 90% have compressive modulus over 3 MPa and compressive strength over 0.2 MPa. As preliminary cell experiments with 3T3 fibroblasts cultured on the porous scaffolds indicate, the processing procedure of the scaffolds has not brought with problem in cytotoxicity.
Fabrication, Characterization and Cellular Biocompatibility of Porous Biphasic Calcium Phosphate Bioceramic Scaffolds with Different Pore Sizes

Abstract:Facile wet-chemical methods are applied to synthesize hydroxyapatite and β-tricalcium phosphate nanoparticles, respectively. Porous biphasic calcium phosphate (BCP) bioceramic scaffolds are then fabricated using as-prepared HA and β-tricalcium phosphate nanoparticle powders. The macro pore diameter of BCP bioceramic scaffolds can be controlled by adjusting the amount of surfactants. The average diameter of the macro pores in BCP bioceramic scaffolds increases from 100 to 600µm with the decrease amount of sodium dodecyl sulfate from 0.8 to 0.5g, respectively. The BCP bioceramic scaffolds gradually degrade and the calcium-phosphate compounds fully deposit when soaking in simulated body fluid solution. Moreover, The BCP bioceramic scaffolds have outstanding biocompatibility to promote the cellular growth and proliferation of human dental pulp stem cells (hDPSCs). The hDPSCs also demonstrate favorable cellular adhering capacity on the pore surface of scaffolds, especially on the scaffolds with 100–200µm pore diameter. The porous BCP bioceramic scaffold with inter-connected pore structure, outstanding in vitro cellular biocompatibility, favorable cell viability and adhesion ability will be a promising biomaterial for bone or dentin tissue regeneration.

Abstract:Biodegradable polymer microspheres have been increasingly attracting attention in bone repair. In this work, superficially porous poly(lactic-co-glycolic acid)/calcium carbonate (PLGA/CC) composite microsphere is prepared by spontaneous pore-forming emulsion method. In addition to being the component that neutralizes the acidic products from PLGA degradation, calcium carbonate (CC) also constructs macropores on the surface of microsphere and thus exempts the use of extraneous porogens, which simplifies the preparation process and avoids problems related to extraneous porogen. It is found that the size of CC plays the most essential role in forming the superficial macropores because it determines the water absorption capacity of CC. Release of dexamethasone from the superficially porous composite microspheres shows a mild burst and the subsequent tri-phase profile. Meanwhile, the cell culture test demonstrates that MG-63 cells grow well on the superficially porous composite microsphere and proliferate better than those on pure PLGA microsphere. Therefore, this versatile composite microsphere has a promising application for bone repair.

Abstract:A regular hydroxyapatite (HAp) micropattern was prepared combining post-lift-off photolithography, surface grafting of negatively charged chemicals, and simulated body fluid incubation. The chemical micropatterning of the bioceramics on glass was thus successfully generated. The resulting regular HAp microarray was confirmed by scanning electron microscopy, transmission electron microscopy, energy dispersive X-ray spectroscopy, X-ray diffraction, Raman spectroscopy, and Fourier transform infrared spectroscopy. A chemical micropattern of cell-adhesive bioceramic microislands on a non-fouling background was obtained after the glass background was passivated by poly(ethylene glycol). Culture of mammal cells (MC3T3) on the micropattern illustrated that the cell localization on the HAp microislands was well achieved. The present work affords a methodology of micropatterning of bioceramics for the study of interactions between cells and bioceramics.

Abstract:Composite polymer devices have gained considerable interest in biomedical applications. In this work, a composite poly(lactic-co-glycolic acid)/poly(epsilon-caprolactone) (PLGA/PCL) microsphere with a layered architecture and an island-sea topography was developed. PCL was found to occupy the shell layer of the microsphere, while PLGA constituted surface islands and the internal core. This subtle structure was derived from the different external and internal rates of phase separation between PCL and PLGA. The existence of the PLGA islands enhanced the hydrophilicity of the PCL shell. Mouse mesenchymal stem cells (mMSCs) were cultured and they grew well on the PLGA/PCL microspheres. The surface of the microsphere was microscopically composed of PCL sea and PLGA islands, and the mMSCs showed a trend to attach to the more hydrophilic PLGA islands and bridged between adjacent microspheres. This paper provides a new alternative to design functional microspheres for potential application in tissue repair.

Abstract:Biodegradable microspheres have been widely applied as cell carriers for tissue engineering and regenerative medicine. However, most cell carriers only have a simple planar structure and show poor biological activity and cell adherence, resulting in low cell density and unfavorable application effect. How to develop size-controllable microspheres with an open-porous structure remains a challenge, and is a key factor to extend their employment as cell/drug delivery vehicles to boost regeneration of tissues (e.g., bone). Herein, well-defined open porous microspheres of poly(lactic-co-glycolic acid) (PLGA with good biocompatibility approved by the Food and Drug Administration (FDA)) were developed by using a gas-assisted-emulsion and surface-alkalization-treatment technology (GEST). The gas-assisted-emulsion strategy enables the formation of microspheres with a large size of 200-300 mu m, meanwhile, the microspheres have a large amount of micropores with diameter in the range of 10-60 mu m. The following alkalization-treatment on the surface makes the microspheres form a good porous interconnectivity throughout both the surface and the interior of the microspheres. The good porous interconnectivity endows the microspheres with a highly open pore structure and a large specific surface area for nutrient exchange and cell attachment, thus promoting cell proliferation and nutrient transportation, promising their potential as an ideal cell carrier to increase cell density and bioactivity for cell therapy-based tissue engineering.

Abstract:Microparticles with porous structure can be applied as microcarriers for both cell culture and tissue regeneration. While well-controlled pore structure represents a critical challenge to be achieved. In the present study, in order to develop microcarriers for cell culture, a series of poly(epsilon-caprolactone) microspheres were fabricated with varied macroporous structures. Poly(epsilon-caprolactone) microspheres were prepared via the integration of the emulsion/solvent evaporation and particle leaching mechanisms. Particularly, by adjusting poly(epsilon-caprolactone) concentration and the ratio between the porogen paraffin and poly(epsilon-caprolactone), the microspheres with the pore size of 25.6-84.0m and the porosity of 57.4-75.5% were obtained. Further, the microspheres were subjected to alkaline hydrolysis, followed by surface coating with hydroxyapatite. These porous poly(epsilon-caprolactone) microspheres with surface modification well supported the adhesion and growth of human fibroblasts. Together, bioactive poly(epsilon-caprolactone) microspheres with controlled pore structure are potential to be applied in cell culture and tissue regeneration.

Abstract:A facile technique is herein reported to fabricate three-dimensional (3D) polymeric porous scaffolds with interior surfaces of a topographic microstructure favorable for cell adhesion. As demonstration, a well-known biodegradable polymer poly(lactide-co-glycolide) (PLGA) was employed as matrix. Under the porogen-leaching strategy, the large and soft porogens of paraffin were modified by colliding with small and hard salt particles, which generated micropits on the surfaces of paraffin spheres. The eventual PLGA scaffolds after leaching the modified porogens had thus interior surfaces of microscale roughness imprinted by those micropits. The microrough scaffolds were confirmed to benefit adhesion of bone marrow stromal cells (BMSCs) of rats and meanwhile not to hamper the proliferation and osteogenic differentiation of the cells. The insight and technique might be helpful for biomaterial designing in tissue engineering and regenerative medicine.

Abstract:The purpose of this work was to incorporate amorphous calcium phosphate (ACP) into porous poly(L-lactic acid) (PLLA), because ACP is capable of fast phase transformation and morphological change in body fluid, such, a desired pore wall surface within bone tissue engineering scaffolds can be created. A highly porous ACP/PLLA composite was prepared by a thermally induced phase separation technique. The results showed that the composite had an interconnected pore structure with 100 mu m macropores and 10 mu m micropores, and 91% porosity; 40 mu m primary particles of ACP were agglomerated to 3 mu m aggregates, and the aggregates were homogeneously distributed in pore walls; These aggregates showed to be in situ transformed into bone-like apatite after 1 h soaking in phosphate buffered saline solution. Human osteoblast-like cell culture showed that the ACP/PLLA composite had better cell adhesion and alkaline phosphotase activity than pure PLLA. This study demonstrates that the ACP/PLLA composite can enhance cytocompatibility and could act as a promising scaffold for bone tissue engineering. (c) 2006 Wiley Periodicals, Inc.
Facile Fabrication of Poly(l-Lactic Acid) Microsphere-Incorporated Calcium Alginate/hydroxyapatite Porous Scaffolds Based on Pickering Emulsion Templates

Abstract:In this study, we develop a facile one-pot approach to the fabrication of poly(L-lactic acid) (PLLA) microsphere-incorporated calcium alginate (ALG-Ca)/hydroxyapatite (HAp) porous scaffolds based on HAp nanoparticle-stabilized oil-in-water Pickering emulsion templates, which contain alginate in the aqueous phase and PLLA in the oil phase. The emulsion aqueous phase is solidified by in situ gelation of alginate with Ca(2+) released from HAp by decreasing pH with slow hydrolysis of D-gluconic acid δ-lactone (GDL) to produce emulsion droplet-incorporated gels, followed by freeze-drying to form porous scaffolds containing microspheres. The pore structure of porous scaffolds can be adjusted by varying the HAp or GDL concentration. The compressive tests show that the increase of HAp or GDL concentration is beneficial to improve the compressive property of porous scaffolds, while the excessive HAp can lead to the decrease in compressive property. Moreover, the swelling behavior studies display that the swelling ratios of porous scaffolds reduce with increasing HAp or GDL concentration. Furthermore, hydrophobic drug ibuprofen (IBU) and hydrophilic drug bovine serum albumin (BSA) are loaded into the microspheres and scaffold matrix, respectively. In vitro drug release results indicate that BSA has a rapid release while IBU has a sustained release in the dual drug-loaded scaffolds. In vitro cell culture experiments verify that mouse bone mesenchymal stem cells can proliferate on the porous scaffolds well, indicating the good biocompatibility of porous scaffolds. All these results demonstrate that the PLLA microsphere-incorporated ALG-Ca/HAp porous scaffolds have a promising potential for tissue engineering and drug delivery applications.

Abstract:Patterned structured surfaces are very useful to control a cell's microenvironment and to modulate certain cellular responses, such as cell adhesion, migration, proliferation, and differentiation. Herein, ordered micropatterns constructed by a quadrate convex with different sizes were fabricated on a hydroxyapatite [Ca-10(PO4)(6)(OH)(2), HAp] bioceramic surface using an ordered micropatterned nylon sieve as templates. The height, width and space of the convex for the patterns could be facilely regulated via simply tailoring the meshes of the template. Compared with traditional samples with flat surfaces, the fabricated HAp bioceramics with micropatterned surfaces possessed better wettability and higher surface energy, which significantly promoted the adhesion, proliferation, and osteogenic differentiation of rat bone marrow stromal cells (bMSCs). Furthermore, using a pattern size close to that of the cell size showed a better stimulation of cell response compared with larger pattern sizes. Our study suggests that the fabrication of micropatterned structured HAp bioceramics is critical for designing optimal biomaterials for bone regeneration and cell culture substrate applications.
A Hierarchically Porous Bioactive Glass-Ceramic Microsphere with Enhanced Bioactivity for Bone Tissue Engineering

Abstract:A hierarchically porous bioactive glass-ceramic microsphere with enhanced bioactivity and good biocompatibility was fabricated via electro spraying technique assisted with non-solvent induced phase inversion. The electro sprayed bioactive glass (ESBG) microspheres were 20 ± 5 μm in diameter, and composed of four characteristic pores: <200 nm, 200–500 nm, 500–1000 nm and >1000 nm. The influences of sintering temperatures (600 °C-1000 °C) on the microstructure, crystalline phase and the related in vitro bioactivity of the samples were investigated. All the ESBG microspheres possessed bioactivity, and the earliest apatite deposition time (3 h) occurred in the samples sintered at 1000 °C owing to the highly porous structure and the formation of wollastonite. The in vitro cytotoxicity was evaluated via CCK-8 assay using MC3T3-E1 cells, revealing the good biocompatibility as well. These results suggest that the ESBG microsphere is a promising material for bone tissue engineering.
Macroporous and Nanofibrous Polymer Scaffolds and Polymer/Bone-Like Apatite Composite Scaffolds Generated by Sugar Spheres

Abstract:Scaffolds are crucial to tissue engineering/regeneration. In this work, a technique combining a unique phase-separation process with a novel sugar sphere template leaching process has been developed to produce three-dimensional scaffolds. The resulting scaffolds possess high porosities, well connected macropores, and nanofibrous pore walls. The technique advantageously controls macropore shape and size by sugar spheres, interpore opening size by assembly conditions (time and temperature of heat treatment), and pore wall morphology by phase-separation parameters. The bioactivity of a macroporous and nanofibrous poly(L-lactic acid) (PLLA) scaffold was demonstrated by the bone-like apatite deposition throughout the scaffold in a simulated body fluid (SBF). Preincorporation of nanosized hydroxyapatite eliminated the induction period and facilitated the apatite growth in the SBF. Interestingly, the apatite growth primarily occurred on the surface of the pores (internal and external) but not the interior of the nanofibrous network away from the pore surface. It was also noticed that the macropore size did not affect the apatite growth rate, while the interpore opening size did. The compressive modulus also increased substantially when a continuous apatite layer was formed on the pore walls of the scaffold. The resulting composite scaffold mimics natural bone matrix with the combination of an organic phase (a polymer such as PLLA) and an inorganic apatite phase. The demonstrated bioactivity of apatite layer, together with well-controlled macroporous and nanofibrous structures, makes the novel nanocomposite scaffolds desirable for bone tissue engineering.
Tailoring the Nanostructured Surfaces of Hydroxyapatite Bioceramics to Promote Protein Adsorption, Osteoblast Growth, and Osteogenic Differentiation

Abstract:To promote and understand the biological responses of the implant via nanostructured surface design is essential for the development of bioactive bone implants. However, the control of the surface topography of the bioceramics in nanoscale is a big challenge because of their brittle property. Herein, the hydroxyapatite (HAp) bioceramics with distinct nanostructured topographies were fabricated via hydrothermal treatment using α-tricalcium phosphate ceramic as hard-template under different reaction conditions. HAp bioceramics with nanosheet, nanorod and micro-nanohybrid structured surface in macroscopical size were obtained by controlling the composition of the reaction media. Comparing with the traditional sample with flat and dense surface, the fabricated HAp bioceramics with hierarchical 3D micro-nanotextured surfaces possessed higher specific surface area, which selectively enhanced adsorption of specific proteins including Fn and Vn in plasma, and stimulated osteoblast adhesion, growth, and osoteogenic differentiation. In particular, the biomimetic features of the hierarchical micro-nanohybrid surface resulted in the best ability for simultaneous enhancement of protein adsorption, osteoblast proliferation, and differentiation. The results suggest that the hierarchical micro-nanohybrid topography might be one of the critical factors to be considered in the design of functional bone grafts.

Abstract:Tissue engineering has shown great promise for creating biological alternatives for implants. In this approach, scaffolding plays a pivotal role. Hydroxyapatite mimics the natural bone mineral and has shown good bone-bonding properties. This paper describes the preparation and morphologies of three-dimensional porous composites from poly(L-lactic acid) (PLLA) or poly(D,L-lactic acid-co-glycolic acid) (PLGA) solution and hydroxyapatite (HAP). A thermally induced phase separation technique was used to create the highly porous composite scaffolds for bone-tissue engineering. Freeze drying of the phase-separated polymer/HAP/solvent mixtures produced hard and tough foams with a co-continuous structure of interconnected pores and a polymer/HAP composite skeleton. The microstructure of the pores and the walls was controlled by varying the polymer concentration, HAP content, quenching temperature, polymer, and solvent utilized. The porosity increased with decreasing polymer concentration and HAP content. Foams with porosity as high as 95% were achieved. Pore sizes ranging from several microns to a few hundred microns were obtained. The composite foams showed a significant improvement in mechanical properties over pure polymer foams. They are promising scaffolds for bone-tissue engineering.
Development of Hierarchical Porous Bioceramic Scaffolds with Controlled Micro/nano Surface Topography for Accelerating Bone Regeneration

Abstract:Mimicking hierarchical porous architecture of bone has been considered as a valid approach to promote bone regeneration. In this study, hierarchical porous β-tricalcium phosphate (β-TCP) scaffolds were constructed by combining digital light processing (DLP) printing technique and in situ growth crystal process. Macro/micro hierarchical scaffolds with designed macro pores for facilitating the ingrowth of bone tissue were fabricated by DLP printing. Three types of micro/nano surface topography were obtained by in situ growth crystal process to regulate stem cells behavior. The attachment and proliferation of rat bone marrow mesenchymal stem cells (rBMSCs) were strongly dependent on the surface roughness and the specific surface area. The micro/nano surface topography distinctly facilitated the differentiation of rBMSCs by targeting MAPK, STAT and AKT signaling pathways, in which the sodium hydroxide treatment group showed the highest promoting effect. Furthermore, in vivo results of skull defect repair model of rats indicated that hierarchical scaffolds with micro/nano topographies exhibited appealing bone regeneration capacity. The hierarchical porous bioceramic scaffolds constructed by integrating structural design and physical stimulation of the external surface topography have great potential for rapid bone repair via modulation of microenvironmental regulatory pathways at the bone defect site.
