Abstract:BACKGROUND:The constitutively existed tumor stem cells in the microenvironment of bone marrow changed the characteristics of marrow stromal cells and extracellular matrix,and involved in the malignancy of hematopoietic stem cells.OBJECTIVE:To investigate the matrix metalloproteinase-9(MMP-9) expression in bone marrow mesenchymal stem cells(BMSCs) in chronic myelocytic leukemia patients and its adhesive effects on hematopoietic stem cells.DESIGN,TIME AND SETTING:The cytology in vitro controlled study was performed at the Basic College,Peking Union Medical College from September 2007 and February 2008.MATERIALS:Bone marrow samples,which were collected from 4 chronic myelocytic leukemia patients and 4 healthy volunteers,were obtained from the Department of Hematology,the 309 Hospital of Chinese PLA.Umbilical cord blood was provided by Beijing Umbilical Cord Blood Hematopoietic Stem Cell Pool.293T cells were presented by Professor Kong from Nankai University.METHODS:Bone marrow was harvested from healthy volunteers and chronic myelocytic leukemia patients.BMSCs were collected by the Ficoll method in vitro.Umbilical cord blood was separately packed,centrifuged to remove the supernatant.The intermedium was taken.Umbilical cord blood mononuclear cells were isolated and obtained by the same method.BMSCs in the logarithmic phase were incubated in a 96-well culture plate at 1×107/L per well.When 100% cells were confluent,umbilical cord blood mononuclear cells were incubated in the same culture plate at 1×105 per well for 1 hour.Following non-adherent cells were removed,adherent rate was calculated.At the second passage,BMSCs were harvested.Mass concentration of soluble intercellular adhesion molecule-1 in the supernatant was measured by enzyme linked immunosorbent assay.MPP-9 siRNA interference vector was constructed.293T cells were transfected and encased by the liposome method.BMSCs were transfected following virus amplification.MAIN OUTCOME MEASURES:Five indexes were compared between healthy donors and chronic myelocytic leukemia patients.Three indexes were compared among chronic myelocytic leukemia patients before and after virus transfection.RESULTS:Compared with the healthy donors,MMP-9 mRNA and protein levels in BMSCs were significantly increased in the chronic myelocytic leukemia patients,and no significant differences were detected in intercellular adhesion molecule-1,and mass concentration of soluble intercellular adhesion molecule-1 was significantly increased in the supernatant(P < 0.05).Adherent rate of umbilical cord blood mononuclear cells were significant decreased(P < 0.05).Compared with the non-transfection BMSCs,MMP-9 mRNA expression was significant reduced following virus transfection,and intercellular adhesion molecule-1 protein expression was not affected,and the adherent rate of umbilical cord blood mononuclear cells was significantly increased(P < 0.05).CONCLUSION:The action mechanism of MMP-9-splited intercellular adhesion molecule-1 exists in BMSCs from chronic myelocytic leukemia patients.It is hypothesized that free soluble intercellular adhesion molecule-1 blocks the binding site of LFA-1,and finally affects the adherence of BMSCs on hematopoietic stem cells.
