Abstract:OBJECTIVE:To investigate the value of detecting cytokeratin 19(CK19) and thyroperoxidase (TPO) expression in the diagnosis of thyroid diseases including thyroid carcinoma, multinodular goiter, adenoma and Hashimoto thyroiditis.METHODS:SP immunohistochemistry was used to detect the expressions of CK19 and TPO in paraffin-embedded thyroid tissue specimens obtained from 62 patients with thyroid carcinoma (30 with papillary carcinomas, 22 with follicular variant of papillary carcinomas, and 10 with follicular carcinomas) and 44 with benign thyroid diseases (including 22 with multinodular goiters, 14 with adenoma, and 8 with Hashimoto thyroiditis).RESULTS:CK19 expression was detected in 96.8% of the thyroid carcinomas and in 4.5% of benign thyroid diseases, demonstrating a significant difference in CK19 expression between the two thyroid diseases (P<0.01). TPO expression was found in 100% of benign thyroid disease and in 3.2% of thyroid carcinoma, showing also a significant difference between them (P<0.01).CONCLUSION:CK19 and TPO can be important molecular markers for diagnosing thyroid carcinoma.
Immunohistochemistry related to the diagnosis and metastasis of papillary thyroid carcinoma

Abstract:OBJECTIVE: To estimate the clinical value of CK19, p53, and p63 on distinguishing benign and malignant thyroid diseases and get further knowledge about the relationship between p53, p63, cerbB-2, nm23 and papillary thyroid carcinoma (PTC) metastasis. METHODS: By the immunohistochemical method called Elivision, CK19, p53, p63, cerbB-2 and nm23 were examined respectively in 57 PTC and 57 thyroid benign disease patients. RESULTS: CK19 expression rates in PTC and benign disease of thyroid were 91.2% and 1.76%, respectively; p53 expressions in PTC and benign disease were 89.5% and 17.5%, respectively. The p63 positive rates were 22.8% and 12.3% in PTC and benign disease, respectively. There were no statistical differences in the expressions of p53, p63, cerbB-2 and nm23 between the metastasis group and non-metastasis group(P>0.05). The positive rates of cerbB-2 and nm23 in PTC were 84.2% and 91.2%, respectively. CONCLUSIONS: CK19 and p53 are valuable assistant diagnostic points for PTC pathological diagnosis. p63 has no explicit value for distinguishing diagnosis of benign or malignant diseases in thyoid. p53, p63, cerbB-2 and nm23 have no relationship with metastasis of PTC. cerbB-2 expression degrees are positively related to nm23.
Expression of Glucose Transporter-1, Hypoxia Inducible Factor-1Α and Beclin-1 in Head and Neck Cancer and Their Implication.

Abstract:Previous studies have demonstrated that oncogenes, tumor suppressor genes, and hypoxic factors are involved in the pathogenesis, development, and progression of head and neck cancer. In this study, we investigated the expression of HIF-1α, GLUT-1, and beclin-1 in head and neck cancers and analyzed the relationship between these markers and clinicopathologic factors. 44 paraffin-embedded samples of head and neck cancer specimens were collected. The expression of HIF-1α, GLUT-1, Ki 67, and beclin-1 was detected by immunohistochemical technique. The expression of HIF-1α, GLUT-1, Ki 67, and beclin-1 in head and neck cancers were higher than those for the corresponding paracancerous tissues. Stratification analysis revealed a significant difference between GLUT-1 expression in older patients with laryngeal/hypopharyngeal SCC and younger patients with laryngeal/hypopharyngeal SCC. No significant differences in GLUT-1 or beclin-1 or HIF-1α or Ki 67 expression were found between clinicopathologic characteristics, including lymph node metastasis, T stage, clinical stage, and location, for any of the cancer types studied. Pearson analysis revealed that there was a negative correlation between beclin-1 and HIF-1α (r=0.482, P=0.001), and between beclin-1 and Ki-67 (r=-0.366, P=0.0151). Whether beclin-1 plays a role in carcinogenesis in head and neck cancers should be further studied.
Diagnostic Significance of Ck19, Ret, Galectin-3 and Hbme-1 Expression for Papillary Thyroid Carcinoma

Abstract:Aims To evaluate CK19, RET, galectin-3 and HBME-1 expression in papillary thyroid carcinoma (PTC) and to evaluate their diagnostic significance.Methods 155 PTC specimens and 83 other diseased-thyroid specimens were collected. Immunohistochemistry for CK19, RET, galectin-3 and HBME-1 was performed.Results The 155 PTC cases were classified into eight variants according to the WHO classification, including 74 cases of classic PTC, 40 cases of papillary microcarcinoma, and rare variants. CK19, RET, galectin-3 and HBME-1 expression was 87.1% (135/155), 71.0% (110/155), 91.6% (142/155), and 95.5% (148/155), respectively, for the PTC group; expression of all these markers was much higher than that in the control group (p<0.05). However, the expression of these markers did not differ among the variants (p>0.05). The expression of these markers, particularly CK19 and RET, was diffuse and strong in the papillary structure of PTC, but weak and focal in the papilla of tissue with benign disease. The expression of CK19 in follicular PTC was significantly higher than in follicular thyroid carcinoma (FTC) (p<0.05).Conclusions CK19, RET, galectin-3 and HBME-1 expression in PTC was higher than that in benign disease cases, but these were not specific markers for PTC. In summary, combining markers can increase the reliability and differential diagnosis of PTC. It is also worth noting that CK19 was very useful not only for the differentiation of benign and malignant papillary structure but also for the differential diagnosis of follicular PTC and FTC.
CD56 expression in benign and malignant thyroid lesions

Abstract:Introduction: Thyroid cancer is the most common endocrine malignancy with more than 95% originating from follicular epithelial cells. Diagnostic dilemma may arise in occasional cases such as when an encapsulated nodule with a follicular growth pattern exhibits clear nuclei with grooves making it difficult to distinguish a follicular adenoma from encapsulated follicular variant papillary thyroid carcinoma. This study aimed to evaluate the diagnostic utility of an immunohistochemical marker, CD56, to distinguish between benign and malignant thyroid lesions. Materials and methods: We retrospectively studied CD56 expression in 54 benign and 54 malignant thyroid lesions using archival formalin fixed paraffin-embedded tissue blocks for the study period from January 2010 to December 2015, diagnosed in a tertiary hospital. Results: CD56 was expressed in 52/54 (96.3%) of benign specimens and only 24/54 (44.4%) of malignant ones. The malignant specimens comprised 31 (57.4%) papillary thyroid carcinomas (PTC), 11 (20.3%) follicular carcinomas (FC), seven (13%) medullary thyroid carcinomas (MC), one (1.9%) poorly differentiated carcinoma (PC) and four (7.4%) anaplastic carcinomas (AC). CD56 was not expressed in 28/31 (90.3%) of the PTCs, 1/11 (9.1%) FCs, 1/4 (25%) of ACs while all MCs and the PD were positive. The benign group comprised nodular hyperplasias (29/54), lymphocytic thyroiditis (10/54), follicular adenomas (FA) (14/54) and one hyalinising trabecular tumour. CD56 was expressed in all the benign cases except one FA and one nodular hyperplasia. Thirteen of the 14 FAs were CD56 positive. The difference in expression between benign and malignant tumours was statistically significant as the p value was <0.01. Conclusion: CD56 is a potentially good immunohistochemical marker for differentiating papillary thyroid carcinoma from other benign follicular lesions of the thyroid especially in differentiating follicular variant PTC from FA in equivocal cases.

Abstract:OBJECTIVE:To study immunohistochemical expression of cytokeratin19 (CK19), galectin-3 (Gal-3) and HBME-1 in thyroid lesions and to assess their usefulness as markers in the differential diagnoses of thyroid nodular lesions.METHODS:Immunohistochemical staining was performed on formalin-fixed paraffin-embedded tissue of 21 cases of nodular goiters, 14 cases of toxic goiters, 15 cases of follicular adenomas (FA), 13 cases of follicular carcinomas (FC), 13 cases of follicular variant papillary carcinomas (FVPC) and 48 cases of classic papillary carcinomas (CPC).RESULTS:All three markers were expressed in the cytoplasm with no or weak expression in benign lesions and diffuse and strong in malignant cases. Positive expressions of CK19, Gal-3 and HBME-1 were present in 11of 21, two of 21, four of 21 in nodular goiters, seven of 14, one of 14, one of 14 in toxic goiters, nine of 15, two of 15, two of 15 in FA, 10 of 13, eight of 13, seven of 13 in FC, 13 of 13, 11 of 13, 12 of 13 in FVPC, and 48 of 48, 45 of 48, 46 of 48 in CPC. The expression rates of the three markers between benign lesions (nodular goiters, toxic goiters and FA) and malignant lesions (FA, FVPC and CPC) were statistically significant. Among the three follicular lesions (FA, FC and FVPC), the differences were statistically significant as well. Nine, seven and six cases were negative for all three markers in nodular goiters, toxic goiters and FA, respectively. Only one case in FC was negative for all three markers, no case was all negative in FVPC and CPC; the rate of one case with two or more positive marker expression in nodular goiters, toxic goiters, FA, FC, FVPC and PC was 14.2% (3/21), 21.43% (3/14), 20.0% (3/15), 69.2% (9/13), 92.3% (12/13), 100.0% (48/48), the differences between benign lesions and malignant lesions and between FA, FC and FVPC were also statistically significant.CONCLUSIONS:Immunohistochemical stains of CK19, Gal-3 and HBME-1, especially when used in combination, can be an important adjunct to the histopathological diagnoses of thyroid lesions.
Abnormal expression of Thy-1 as a novel tumor marker in lung cancer and its prognostic significance]

Abstract:The purpose of this study was to investigate the expression of Thy-1 immunohistochemically in different lung tumors and its prognostic significance in non-small cell lung cancer (NSCLC) cases. We also evaluate relationship between Thy-1 and p53 expression status so as to find any clue about the mechanism.In this study, we used anti-Thy-1/CD90 antibody to detect the expression pattern of Thy-1 in different lung tumor sections, which were embedded in paraffin blocks. The expressions of Thy-1 in 175 lung tissue cases, including different pathological types, were analyzed as tissue array form. We also detect expression status in 91 NSCLC among these cases and analyze the relationship between Thy-1 and p53. The relationship between Thy-1 expression and patients' survival was studied.We first found that anti-Thy-1 antibody can strongly stain a nuclear molecule in different type of lung cancer cells. Among lung cancer cases, 89 (56.7%) cases showed strong nuclear staining for Thy-1 specially. In univariate and multivariate analysis for 91 NSCLC patients we found TNM staging, lymph node status and Thy-1 overexpression in nuclei were independent factors to affect the prognosis of NSCLC patients. In lymph node non metastasis subgroup cases, Thy-1 negative patients had significant longer survival than Thy-1 positive cases (mean survival: 46.42 mons vs 38.56 mons, P = 0.0207). There was a significant association between Thy-1 and p53 expression (P < 0.0001).There is a significant overexpressed Thy-1 located in lung cancer cell nucleus as compared to the normal tissue or benign tumor cells of lung, and it is one of the factors effected on the prognosis of NSCLC patients. This finding suggests that Thy-1 maybe a novel latent malignant marker in the lung cancer pathology. The association between Thy-1 and p53 expression in nucleus suggests that p53 protein and Thy-1 may have some interaction.

Abstract:Although most differentiated thyroid carcinoma has a clinically favorable prognosis, some of specific types of thyroid cancer (such as anaplastic thyroid carcinoma and advanced papillary thyroid carcinoma) show fatal outcomes and require novel treatments. Immunotherapy is a promising avenue for the treatment of advanced thyroid carcinoma. B7-H3 (B7 homolog 3 protein) and ICAM-1 (intercellular adhesion molecule 1), as two important immune checkpoints (ICPs), is becoming hopeful target spots for immunotherapy. A growing amount of evidence has suggested that B7-H3 and ICAM-1 are upregulated in papillary thyroid carcinoma. However, their expression level in specific types of thyroid cancer remains largely unclear. In the present study, we explored the expression level of B7-H3 and ICAM-1 in different types of thyroid carcinoma. In the groups of the TCGA cohort, both B7-H3 and ICAM-1 mRNA were highly expressed in thyroid carcinoma. Furthermore, the patients with Stage2, 61-80y, Follicular thyroid papillary carcinoma and N0 had lower B7-H3 and ICAM-1 mRNA expression. In the groups of our cohort, PTCs and ATCs showed frequently moderate to strong expression of B7-H3 and ICAM-1 protein expression. The significant relevance of B7-H3 staining score with ICAM-1 staining score was observed in TCGA database and our cohort, which might open avenues for the combination therapy in advanced thyroid cancer.

Clinical Significances of the TSHR and MMP-9 Expressions in Papillary Thyroid Carcinoma

Abstract:[Objective] To evaluate the expressions of thyroid stimulating hormone receptor(TSHR) and matrix metalloproteinase-9(MMP-9) in papillary thyroid carcinoma(PTC) and their clinical significances.[Method] The SP immunohistochemistry was used to detect the expressions of TSHR and MMP-9 protein in 56 cases of PTC,30 cases of nodular goiter and 11 cases of normal thyroid tissue.[Results] The positive expression rate of TSHR in PTC was significantly higher than those of nodular goiter and normal thyroid tissue(P0.05).and the rates in PTC with lymph node metastasis and without were 76.92% and 95.35% respectively(P0.05),The rates in stage Ⅲ-Ⅳand stageⅠ-Ⅱwere 89.66 % and 92.60 % respectively(P0.05);The positive expression rate of MMP-9 protein in PTC was significantly higher than those of nodular goiter and normal thyroid tissue(P0.05).and the rates in PTC with lymph node metastasis and without were 92.31 % and 79.07 % respectively(P0.05),and the rates in stage Ⅲ-Ⅳand stageⅠ-Ⅱwere 89.66 % and 74.07 % respectively(P0.05).Their expressions of both TSHR and MMP-9 were not related to sex,age and tumor size of the patient(P0.05).[Conclusion] Over-expression of TSHR might be related to the development of PTC and become a early marker of the thyroid carcinoma.The over-expression of MMP-9 could be related to invasion and metastasis.
Analysis of the Clinical Value of hsa_circ_0001955 in Papillary Thyroid Cancer Treated with 131 Iodine

Abstract:The prevalent histological variant within differentiated thyroid carcinoma is papillary thyroid carcinoma, also known as PTC. The study investigated the clinical performance of serum hsa_circ_0001955 in predicting the prognosis of PTC treated with radical thyroidectomy and iodine 131 nail clearance. The relative expression of serum circ_0001955 of PTC patients was detected before and after accepting radical thyroidectomy combined with 131I thyroid remnant ablation by RT-qPCR. Serum thyroglobulin (Tg) and thyroglobulin antibody (TgAb) levels were quantified by an automatic chemiluminescence immunoassay analyzer. Multivariate logistic regression analysis was employed to investigate the risk factors associated with the prognosis of PTC patients with postoperative 131I therapy. The serum circ_0001955 levels in 127 PTC patients were higher than that in 96 multinodular goiter patients and 110 healthy controls before treatment and had diagnostic values for PTC patients. After 131I treatment, serum circ_0001955 levels and Tg value have a correlation with potential recurrence (WBS positive). Serum circ_0001955, Tg, and TgAb value, and their combination may have diagnostic value in predicting recurrence. Serum circ_0001955 levels in patients with PTC after radical thyroidectomy and iodine 131 thyroidectomy may help predict recurrence.

Abstract:Purpose To explore the expression and significance of Galectin-3,CK19,HBME-1 and CD56 in nodular goiter combined with papillary thyroid microcarcinoma.Methods The immunohistochemical staining of Galectin-3,CK19,HBME-1 and CD56 was performed in ten patients with nodular goiter combined with papillary thyroid microcarcinoma.Results The moderate to strong staining for Galectin-3,CK19,HBME-1 was observed in papillary thyroid microcarcinoma but negative to mild in nodular goiter.Only one case of nodular goiter showed moderate staining for CK19.Eight cases of ten cases of papillary thyroid microcarcinoma,however,showed negative for CD56 staining and the two cases revealed positive.In the nodular goiter,nine cases showed moderate to strong staining for CD56 and only one case was mild staining.Conclusions The combined detection with Galectin-3,CK19,HBME-1 and CD56 will further improve the accuracy of papillary thyroid microcarcinoma.
Expression of Epidermal Growth Factor Receptor and Cell Proliferative Protein in Thymic Carcinoma and Type-B3 Thymoma

Abstract:Objective:To investigate the expression of epidermal growth factor receptor(EGFR),survivin,P53 protein and proliferating cell nuclear antigen(PCNA) in thymic carcinoma and type-B3 thymoma. Methods:The clinicopathological changes of thymic carcinoma in 9 cases and of type-B3 thymoma(well differentiated thymic carcinoma) in 21 cases were examined by microscopic observation of immunohistochemical stained EGFR,survivin,P53 protein and PCNA,respectively. Results:Clinically,the median age of the patients with the tumor was 43.5 years old.All the tumors displayed a pattern of infiltration growth.Immunohistochemical staining of the tumors indicated that the expressions of EGFR,survivin,P53 protein and PCNA in thymic carcinoma were 77.8%(7/9),88.8%(8/9),77.8%(7/9) and 100%(9/9),respectively.Meanwhile,in type-B3 thymoma,they were 67.7%(14/21),71.4%(15/21),33.3%(7/21) and 85.7%(18/21),respectively.It was statistically significant that the immunohistochemical phenotypes of the tumor displayed a different reaction for P53 protein. Conclusion:The thymic carcinoma and type-B3 thymoma may possess a complete invasive characteristic,clinically.To discriminate thymic carcinoma and type-B3 thymoma,the expression of P53 protein is a more valuable index than EGFR,survivin or PCNA.
Special Expression of Thy-1 in Different Malignant Tumors

Abstract:To investigate the expression status of Thy-1 in malignant cancerous tissues and to evaluate whether Thy-1 could be a potential tumor marker.Immunohistochemistry method was used to examine Thy-1 expressions in different malignant tumor tissues. We used a monoclonal antibody specific for Thy-1 and the indirect Catalysis Signal Amplification method. With the help of tissue microarray (TMA), we examined expression the status of Thy-1 in 1451 different types of malignant tumor tissues, 144 normal tissues and benign lesions.All the malignant tumor cells were grouped as one group (malignant cells group) and normal or benign tumor tissue cells as another group (normal cells group). Among the entire 1451 malignant cases group, positive Thy-1 expression with diffuse and strong staining was observed in 734 (734/1451; 50.59%) cases. In the benign cases only 6 chronic cervicitis cases showed weak staining for Thy-1. All the normal tissue showed negative staining. One-way ANVOA analysis showed F value between these two groups was 147.229 (P < 0.0001).A significantly stronger expression of Thy-1 in malignant tumors was observed. Special overexpression of Thy-1 in malignant tumors suggests that Thy-1 might be a potential novel tumor marker with respect to cancer progression. Chronic cervicitis has some relationship with cervix carcinoma. When it develops into atypical hyperplasia, it will be a precancerous lesion for cervix carcinoma. In this research we found weak staining for Thy-1 in chronic cervicitis lesion, so Thy-1 may play a crucial role in carcinogenesis for cervix carcinoma. The research about the relationship between Thy-1 expressions in cancer cells and in precancerous lesion will provide some clues to understand the mechanism for carcinogenesis process.
Clinical value of Cyclin D1 and P21 in the differential diagnosis of papillary thyroid carcinoma

Abstract:Abstract Background With the continuous discovery of new borderline thyroid lesions and benign and malignant “gray areas”, coupled with the limitations of traditional immune indicators, the differential diagnosis of papillary thyroid carcinoma (PTC) has become more difficult. Cyclin D1 and P21 are cell cycle regulators involved in the occurrence and metastasis of multiple tumors, including PTC, but their specific functions are unclear. Methods In our study, immunohistochemical staining was used to explore the expression of Cyclin D1 and P21 in PTC, paracancerous tissue, follicular adenoma (FA) and papillary thyroid hyperplasia. In addition, their relationship with the clinicopathological features of PTC and their differential diagnostic value in distinguishing between intralymph node PTC metastases and intralymph node ectopic thyroid tissue were studied. Results Among 200 primary PTC lesions, Cyclin D1 and P21 were found to be expressed in 186 (93.00%) and 177 (88.50%), respectively, and their expression levels were significantly higher in PTC tissue than in adjacent tissue, FA tissue and papillary thyroid hyperplasia tissue ( P < 0.05). The expression levels of Cyclin D1 and P21 were positively correlated with tumor size and lymph node metastasis ( P < 0.05) but not with sex, age, number of tumor lesions, histological subtype, chronic lymphocytic thyroiditis or TNM stage ( P < 0.05). The expression levels of Cyclin D1 and P21 were significantly correlated ( P < 0.05). The positivity rates of Cyclin D1 and P21 in intralymph node PTC metastases were 97.96% (48/49) and 89.80% (44/49), respectively, which were significantly higher than those in intralymph node ectopic thyroid tissue ( P < 0.05). The sensitivity (Se) and negative predictive value (NPV) of Cyclin D1 and P21 detection alone or in combination were higher than those of the combined detection of the classical antibody markers CK19, HBME-1 and Galectin-3. Besides, the Se, Sp, PPV and NPV of Cyclin D1 and P21 in differentiating intralymph node PTC metastases and intralymph node ectopic thyroid tissue were higher. Conclusions The results of our study show that Cyclin D1 and P21 are highly sensitive and specific markers for the diagnosis of PTC that are superior to traditional classical antibodies. And, these two markers are of great value in the differential diagnosis of intralymph node PTC metastases and intralymph node ectopic thyroid tissue.

Abstract:Background: TP53 gene plays a major role in the negative control of cell proliferation and in the regulation of signaling cascades. TP53 mutation may have a relevant role in the malignant transformation of thyroid cells as well as thyroid tumor progression. TP53 mutation has been detected only in few well differentiated thyroid carcinomas and is absent in benign conditions. Methods: A total of 162 prospective thyroid cytology and corresponding histological samples diagnosed from atypia of indeterminate significance (AUS) to malignant, were studied via immunocytochemistry for p53. Hence, 50 benign lesions (B) were used as negative control. Molecular analysis for p53 only was performed. Results: The cytology resulted in 50 B, 48 AUS, 40 follicular neoplasms (FNs), 23 suspicious for malignancy (SFM), and 1 malignant (M) case. The authors reported 102 negative and 60 positive p53 cases. The 60 positive cases included 27 cases with weak and/or focal cytoplasmic positivity (+1) and 33 with cases moderate (2+) to strong (3+) cytoplasmic and/or nuclear expression. Overall, 71 cases had histology (2 B, 11 AUS, 37 FN, 20 SFM, and 1 M) including 61.7% benign and 38.2% malignant diagnoses. Only 16 of 71 (5 FN, 10 SFM, and 1 M) were p53-positive. Furthermore, 100% AUS and 86.5% FN cases were p53-negative, none of which had malignant histology. All p53-positive cases were associated with a larger nodule size, tall-cell variant subtype, multifocality, extra thyroidal infiltration, and nodal metastases. Noninvasive follicular thyroid neoplasm with papillary like nuclear features were negative for p53. Few discrepancies in p53 intensity were observed on histology; there were no differences with the molecular testing. Conclusions: p53 might be useful in discriminating thyroid follicular lesions. p53 is likely to be a useful diagnostic marker in recognizing indeterminate lesions that are well-differentiated thyroid cancers.

Abstract:Purpose To investigate the expression of PPM1D in papillary carcinoma of thyroid(PTC) and its relationship with clinicopathologic features and p53.Methods The expression of PPM1D protein was detected in 55 cases of PTC tissues compared with normal tumor-adjacent thyroid tissues by immunohistochemistry.The data was analyzed statistically to evaluate the relationship between the expression of PPM1D,p53 and the clinicopathologic features.Results The expression of PPM1D was much higher in PTC(74.5%,41/55) than that in normal tumor-adjacent thyroid tissues(10.9%,6/55).The difference was statistically significant(P<0.05).PPM1D was found high-expressed in PTC with interstitial cirrhosis(P<0.05).PPM1D also showed negative correlation with p53(r=-0.339,P<0.05) in PTC.Conclusions Increased expression of PPM1D is associated with thyroid papillary carcinoma and the potential coordination between PPM1D and p53 may play an important role in the process of tumor genesis and development.

Abstract:OBJECTIVE:To detect and identify the potential specific serum biomarkers for diagnosis of papillary thyroid cancer.METHODS:Samples of 35 patients with papillary thyroid carcinoma, 40 patients with benign thyroid nodule and 34 healthy individuals were analyzed using the SELDI-TOF ProteinChip System and bioinfomation technology to find the differential peaks which were separated by HPLC and then further analyzed by LC-MS/MS. The protein sequences were analyzed by SEQUEST software and searched in Bioworks database.RESULTS:The top six mass-to-charge ratio (M/Z) peaks with the smallest P value were 6651, 6452, 7653, 7932, 15 106 and 15 848 Da, respectively. The 6651 and 6452 Da proteins were weakly expressed in papillary thyroid carcinoma but highly expressed in benign thyroid nodules and healthy individuals. The differences had statistical significance (P < 0.01). The 7653, 7932, 15 106, 15 848 Da proteins were highly expressed in papillary thyroid carcinoma but weakly expressed in benign thyroid nodules and healthy individuals. The differences were statistically significant (P < 0.01). Combination of these six proteins, using the method of leave-one-out to make crossing detection, the specificity of discriminating papillary thyroid carcinoma and non-cancer was 88.0%, and its sensitivity was 92.5%. The 6651 and 6452 Da proteins were identified as apolipoprotein C-I and apolipoprotein C-III, respectively. The 7653 and 15 106 Da proteins were identified as the same protein-alpha-globin, and the 7932 and 15,848 Da proteins were identified as the same protein-beta-globin.CONCLUSION:The detection of differentially expressed apolipoprotein C-I, apolipoprotein C-III, alpha-globin, and beta-globin may have utility for diagnosis of papillary thyroid carcinoma and are worthy of further investigation.
C-erbB-2 and P-53 Expressions in Benign and Malignant Liver Diseases

Abstract:AIM To study the expression of oncogene C erbB 2 and tumor suppressor gene p 53 and their relationship with benign and malignant liver diseases. METHODS In situ hybridization (ISH) and Immunohistochemistry (IHC) were performed in subjects with 5 normal liver, 11 of small liver cell dysplasia (SLCD), 22 liver cirrhosis and 11 well differenciated hepatocellular carcinoma (HCC) and 24 cases of poorly differenciated HCC with the cDNA probes and specific antibodies respectively. RESULTS The expression rates of C erbB 2 mRNA in normal liver, SLCD, liver cirrohosis, well differenciated HCC and poorly differenciated HCC were 0, 73%, 64%, 37% and 25% respectively, those of C erbB 2 protein in these five types of tissues were 0, 100%, 82%, 45% and 29% respectively. The expression rates of P53 mRNA in normal liver, SLCD, liver cirrohosis, well differenciated HCC and poorly_differenciated HCC were 0, 73%, 68%, 36% and 25% respectively while that of p 53 protein in these five types of tissues were 0, 0, 0, 27% and 42% respectively. CONCLUSION C erbB 2 may play a role in the initial stage of hepatocarcinogenesis. Key points are SLCD proliferate liver cells and bile duct cell. Wild type p 53 has antitumor effect whereas mutant p 53 play a role in the later stage of hepatocarcinogenesis.

Abstract:Background Thyroid carcinoma is the most common endocrine malignancy and a common cancer among the malignancies of head and neck. Noninvasive and convenient biomarkers for diagnosis of papillary thyroid carcinoma (PTC) as early as possible remain an urgent need. The aim of this study was to discover and identify potential protein biomarkers for PTC specifically. Methods Two hundred and twenty four (224) serum samples with 108 PTC and 116 controls were randomly divided into a training set and a blind testing set. Serum proteomic profiles were analyzed using SELDI-TOF-MS. Candidate biomarkers were purified by HPLC, identified by LC-MS/MS and validated using ProteinChip immunoassays. Results A total of 3 peaks ( m/z with 9190, 6631 and 8697 Da) were screened out by support vector machine (SVM) to construct the classification model with high discriminatory power in the training set. The sensitivity and specificity of the model were 95.15% and 93.97% respectively in the blind testing set. The candidate biomarker with m/z of 9190 Da was found to be up-regulated in PTC patients, and was identified as haptoglobin alpha-1 chain. Another two candidate biomarkers (6631, 8697 Da) were found down-regulated in PTC and identified as apolipoprotein C-I and apolipoprotein C-III, respectively. In addition, the level of haptoglobin alpha-1 chain (9190 Da) progressively increased with the clinical stage I, II, III and IV, and the expression of apolipoprotein C-I and apolipoprotein C-III (6631, 8697 Da) gradually decreased in higher stages. Conclusion We have identified a set of biomarkers that could discriminate PTC from non-cancer controls. An efficient strategy, including SELDI-TOF-MS analysis, HPLC purification, MALDI-TOF-MS trace and LC-MS/MS identification, has been proved successful.

Abstract:Objective:To determine the role and interaction of the cell cycle regulator cyclin D1 and p27 in thyroid cancer and to assess their clinical value.Methods:We studied the expression of cyclins Dl and the p27 by immunohistochemistry in 65 cases,including 30 cases of thyroid adenoma(TA)and 35 cases of papillary carcinoma(PC).Results:The results showed that cyclin D1 expression was seen in 22 cases(63%)of PC and 18 cases(60%) of TA.No significant difference in the intensity of cyclin Dl(P0.05) staining was seen between PC and TA;p27 expression was seen in all the TA and 83%(29/35) PC,and the difference in the intensity of p27 staining between them was statistically significant(P﹤0.05).In PC,There was no correlation between the expression cyclin D1 and p27;meanwhile,the grades of cyclin D1 and p27 staining had nothing to do with the stage of pTNM and lymph node metastasis of the PC(P﹥0.05).Conclusion:The decrease in p27 expression plays a more important role than overexpression of cyclins D1 in thyroid carcinogenesis and p27 immunostaining may benefit to the diagnosis of PC.
