Abstract:The increasing use of modified Fe3O4 magnetic microparticles has raised safety concerns regarding their use and effect on human health. This study assessed the in vivo biosafety, DNA, and chromosome damage of modified Fe3O4 microparticles such as Au@Fe3O4, Ag@Fe3O4, Cs@Fe3O4, Pt@Fe3O4, and CdS@Fe3O4, using spleen-deficient rats. Spleen-deficient rats treated with naked and modified (Au, Cs, Pt) Fe3O4 microparticles (5000 mg/kg) displayed low toxicity. Only treatment with Cds@Fe3O4 resulted in elevated toxicity and death in rats. Au-, Ag-, and Pt-modified Fe3O4 increased the rate of hemolysis in rats relative to treatment with naked Fe3O4. Despite this, Au- and Pt-modified Fe3O4 increased the biocompatibility and reduced DNA and chromosome damage in rats relative to naked Fe3O4. While Cs@Fe3O4 microparticles displayed a higher biocompatibility than naked Fe3O4, they displayed no significant reduction in DNA and chromosome damage. In summary, Au and Pt surface-modified Fe3O4 microparticles display elevated in vivo biosafety compared to unmodified particles. The precious metal material, with good biological compatibility, surface modification of Fe3O4 is an effective strategy to improve the overall safety and potential therapeutic utility of these magnetic materials.
In Vitro Toxicological Assessment of Cobalt Ferrite Nanoparticles in Several Mammalian Cell Types

Abstract:Nanoparticles have been widely used in various fields due to the superior physicochemical properties and functions. As a result, human exposure to nanoparticles increases dramatically. Previous researches have shown that nanoparticles could travel through the respiratory, digestive system, or skin into the blood and then to the secondary organs such as the brain, heart, and liver. Besides, the nanoparticle toxicity is controversial and dependent on the sensitivity of the cell type, route of exposure, and condition, as well as their characteristics. Similarly, cobalt ferrite nanoparticles (CoFe2O4-NPs) have been used in different industrial fields, and have also various application possibilities in medical and biomedical fields. CoFe2O4-NPs induce toxic responses in various organisms such as human, mice, and algae. However, there is a serious deficit of information concerning their effects on human health and the environment. We aimed to investigate the toxic effects of CoFe2O4-NPs on liver (HepG2), colon (Caco-2), lung (A549), and neuron (SH-SY5Y) cells, which reflect different exposure routes in vitro, by using various toxicological endpoints. The cytotoxicity, genotoxicity, oxidative damage, and apoptosis induction of CoFe2O4-NPs (39 ± 17 nm) were evaluated. After 24 h, the nanoparticles decreased cell viability at ≤100 μg/mL, while increasing viability at >100 μg/mL. CoFe2O4-NPs induced DNA and oxidative damage with increased malondialdehyde (MDA) and 8-hydroxy deoxyguanosine (8-OHdG) levels and decreased glutathione (GSH) levels with no change in protein carbonyl (PC) levels. CoFe2O4-NPs had apoptotic effect in HepG2 and Caco-2 cells in a concentration-dependent manner and necrotic effects on SH-SY5Y and A549 cells. Consequently, the adverse effects of CoFe2O4-NPs should raise concern about their safety in consumer products.
An in vivo evaluation of acute toxicity of cobalt ferrite (CoFe2O4) nanoparticles in larval-embryo Zebrafish (Danio rerio)

Abstract:The broad spectrum applications of CoFe2O4 NPs have attracted much interest in medicine, environment and industry, resulting in exceedingly higher exposures to humans and environmental systems in succeeding days. Their health effects and potential biological impacts need to be determined for risk assessment. Zebrafish (Danio rerio) embryos were exposed to environmentally relevant doses of nano-CoFe2O4 (mean diameter of 40nm) with a concentration range of 10-500μM for 96h. Acute toxic end points were evaluated by survival rate, malformation, hatching delay, heart dysfunction and tail flexure of larvae. Dose and time dependent developmental toxicity with severe cardiac edema, down regulation of metabolism, hatching delay and tail/spinal cord flexure and apoptosis was observed. The biochemical changes were evaluated by ROS, Catalase (CAT), Lipid peroxidation (LPO), Acid phophatase (AP) and Glutatione s- transferase (GST). An Agglomeration of NPs and dissolution of ions induces severe mechanical damage to membranes and oxidative stress. Severe apoptosis of cells in the head, heart and tail region with inhibition of catalase confirms ROS induced acute toxicity with increasing concentration. Increased activity of GST and AP at lower concentrations of CoFe2O4 NPs demonstrates the severe oxidative stress. Circular dichroism (CD) spectra indicated the weak interactions of NPs with BSA and slight changes in α-helix structure. In addition, CoFe2O4 NPs at lower concentrations do not show any considerable interference with assay components and analytical instruments. The results are possible elucidation of pathways of toxicity induced by these particles, as well as contributing in defining the protocols for risk assessment of these nanoparticles.

Abstract:Nanotechnology is developing at a rapid pace with promises of a brilliant socio-economic future. The apprehensions of vivid future involvement with nano technology make nanoobjects ubiquitous in the macroscopic world of humans. Nanotechnology helps us to visualize the new mysterious horizons in engineering, sophisticated electronics, environmental remediation, biosensing, and nanomedicine. In all these hotspots, cobalt ferrite (CoFe) nanoparticles (NPs) are outstanding contestants because of their astonishing controllable physicochemical and magnetic properties with ease of synthesis methods. The extensive use of CoFe NPs may result in CoFe NPs easily penetrating the human body unintentionally by ingestion, inhalation, adsorption, etc. and intentionally being instilled into the human body during biomedical diagnostics and treatment. After being housed in the human body, it might induce oxidative stress, cytotoxicity, genotoxicity, inflammation, apoptosis, and developmental, metabolic and hormonal abnormalities. In this review, we compiled the toxicity knowledge of CoFe NPs aimed to provide the safe usage of this breed of nanomaterials.

Abstract:Fascinating super paramagnetic uniqueness of iron oxide particles at nano-scale level make them extremely useful in the state of the art therapies, equipments, and techniques. Cobalt ferrite (CoFe2O4) magnetic nanoparticles (MNPs) are extensively used in nano-based medicine and electronics, results in extensive discharge and accumulation into the environment. However, very limited information is available for their endocrine disrupting potential in aquatic organisms. In this study, the thyroid endocrine disrupting ability of CoFe2O4 NPs in Zebrafish larvae for 168-h post fertilization (hpf) was evaluated. The results showed the elevated amounts of T4 and T3 hormones by malformation of hypothalamus pituitary axis in zebrafish larvae. These elevated levels of whole body THs leads to delayed hatching, head and eye malformation, arrested development, and alterations in metabolism. The influence of THs disruption on ROS production and change in activities of catalase (CAT), mu-glutathione s-transferase (muGST), and acid phosphatase (AP) were also studied. The production of significantly higher amounts of in vivo generation of ROS leads to membrane damage and oxidative stress. Presences of NPs and NPs agglomerates/aggregates were also the contributing factors in mechanical damaging the membranes and physiological structure of thyroid axis. The increased activities of CAT, mu-GST, and AP confirmed the increased oxidative stress, possible DNA, and metabolic alterations, respectively. The excessive production of in vivo ROS leads to severe apoptosis in head, eye, and heart region confirming that malformation leads to malfunctioning of hypothalamus pituitary axis. ROS-induced oxidative DNA damage by formation of 8-OHdG DNA adducts elaborates the genotoxicity potential of CoFe2O4 NPs. This study will help us to better understand the risk and assessment of endocrine disrupting potential of nanoparticles. (C) 2015 Wiley Periodicals, Inc.
Biocompatibility Study of Nanosized Mn-Zn Ferrite

Abstract:Objective: To evaluate the in viro and in vivo toxicity of self-prepared nanosized Mn-Zn ferrite, which has the potential implication in tumor hyperthermia. Methods: MTT assay was used to investigate the in vitro citoxicity and hemolysis test was carried out to estimate whether it had blood toxicity. Mn-Zn ferrite suspended in sterile 0.9%NaCl was intraperitoneally injected into KM mouse to calculate the LD50 and the most tolerance dose(MTD), micronucleus(MN) were reckoned to identify whether it was genotoxic. Results: The experiments showed that the toxicity of the material on mouse fibroblast(L-929) cell lines was between 0-1 degree and it had no hemolysis activity. LD50 could not be estimated after intraperitoneal injection of 1 ml suspension at the highest concentration and the MTD was 6 g/kg; Micronucleus test showed no genotoxic effects. Conclusion: The self-prepared nanosized Mn-Zn ferrite is a kind of high biocompatibility materials and is suitable for further application in tumor hyperthermia.
Comparative Study of Pulmonary Responses to Nano- and Submicron-Sized Ferric Oxide in Rats.

Abstract:Ferric oxide (Fe(2)O(3)) nanoparticles are of considerable interest for application in nanotechnology related fields. However, as iron being a highly redox-active transition metal, the safety of iron nanomaterials need to be further studied. In this study, the size, dose and time dependent of Fe(2)O(3) nanoparticle on pulmonary and coagulation system have been studied after intratracheal instillation. The Fe(2)O(3) nanoparticles with mean diameters of 22 and 280 nm, respectively, were intratracheally instilled to male Sprague Dawley rats at low (0.8 mg/kgbw) and high (20 mg/kgbw) doses. The toxic effects were monitored in the post-instilled 1, 7 and 30 days. Our results showed that the Fe(2)O(3) nanoparticle exposure could induce oxidative stress in lung. Alveolar macrophage (AM) over-loading of phagocytosed nanoparticle by high dose treatment had occurred, while the non-phagocytosed particles were found entering into alveolar epithelial in day 1 after exposure. Several inflammatory reactions including inflammatory and immune cells increase, clinical pathological changes: follicular hyperplasia, protein effusion, pulmonary capillary vessel hyperaemia and alveolar lipoproteinosis in lung were observed. The sustain burden of particles in AM and epithelium cells has caused lung emphysema and pro-sign of lung fibrosis. At the post-instilled day 30, the typical coagulation parameters, prothrombin time (PT) and activated partial thromboplastin time (APTT) in blood of low dose 22 nm-Fe(2)O(3) treated rats were significantly longer than the controls. We concluded that both of the two-sized Fe(2)O(3) particle intratracheal exposure could induce lung injury. Comparing with the submicron-sized Fe(2)O(3) particle, the nano-sized Fe(2)O(3) particle may increase microvascular permeability and cell lysis in lung epitheliums and disturb blood coagulation parameters significantly.
Size-dependent cytotoxicity of Fe3O4 nanoparticles induced by biphasic regulation of oxidative stress in different human hepatoma cells.

Abstract:The application of Fe3O4 nanoparticles (NPs) has made great progress in the diagnosis of disease and in the drug delivery system for cancer therapy, but the relative mechanisms of potential toxicity induced by Fe3O4 have not kept pace with its development in the application, which has hampered its further clinical application. In this article, we used two kinds of human hepatoma cell lines, SK-Hep-1 and Hep3B, to investigate the cytotoxic effects and the involved mechanisms of small Fe3O4 NPs with different diameters (6 nm, 9 nm, and 14 nm). Results showed that the size of NPs effectively influences the cytotoxicity of hepatoma cells: 6 nm Fe3O4 NPs exhibited negligible cytotoxicity and 9 nm Fe3O4 NPs affected cytotoxicity via cellular mitochondrial dysfunction and by inducing necrosis mediated through the mitochondria-dependent intracellular reactive oxygen species generation. Meanwhile, 14 nm Fe3O4 NPs induced cytotoxicity by impairing the integrity of plasma membrane and promoting massive lactate dehydrogenase leakage. These results explain the detailed mechanism of different diameters of small Fe3O4 NPs-induced cytotoxicity. We anticipate that this study will provide different insights into the cytotoxicity mechanism of Fe3O4 NPs, so as to make them safer to use in clinical application.
Zinc Ferrate Nanoparticles for Applications in Medicine: Synthesis, Physicochemical Properties, Regulation of Macrophage Functions, and in Vivo Safety Evaluation

Abstract:Zinc ferrate nanoparticles (ZnFe(2)O(4)NPs) have attracted enormous interest as potential nanomaterials. The purpose of this study was to examine thein vitromacrophages toxicity,in vivosafety, and immunogenicity. Three kinds of ZnFe(2)O(4)NPs with different shapes (round, litchi, and raspberry), nano-sizes, and pores were successfully prepared.In vitroexperiments showed that ZnFe(2)O(4)NPs caused no cytotoxicity against the RAW 264.7 cells up to administered dose of 200 mu g/mL, enhanced proinflammatory cytokine TNF-alpha, and costimulatory marker CD86 expression in the RAW 264.7 cells. Interestingly, ZnFe(2)O(4)NPs reduced ROS expression, which was inconsistent with common metal oxide NPs such as iron oxide (Fe3O4) NPs and zinc oxide (ZnO) NPs. ZnFe(2)O(4)NPs improved the RAW 264.7 cells phagocytosed more neutral red. There was no obvious difference in body weight, the number of immune cells, organ index, and expression of inflammatory factors in serum of rats administrated intravenously and subcutaneously on day 21 after treatment by ZnFe(2)O(4)NPs in comparison with the blank control. These results demonstrated that ZnFe(2)O(4)NPs slightly enhanced the function of the RAW 264.7 cellsin vitrobut caused no obvious toxicity to macrophages as well as rat blood cells, and low immunogenicity in rats, suggesting that ZnFe(2)O(4)NPs as a biocompatible nanomaterials achieved potential for bioapplication in the future.
Effect of Surface Modification on Cellular Internalization of Fe3O4 Nanoparticles in Strong Static Magnetic Field.

Abstract:Magnetic nanoparticles (MNPs) controlled by alternating mangetic field (AMF) are widely investigated in biomedical applications, while the effects of strong static magnetic field (SMFs) on mammalian cells with MNPs for drug-delivery, magnetic resource imaging and magnetofection have been evaluated poorly. Although surface modifications provide a suitable system for expanding the bioapplication of MNPs, the viability and the cellular internalization of modified MNPs which stands for their biocompatibility and efficiency in application need to be examined urgently. In present study, human lung cancer cells (A549), a well-known epithelial cell model for drug metabolism research, were used to evaluate the effects of strong SMFs on cellular internalization and cell viability of Fe3O4 MNPs modified by chitosan, dextran, polyacrylamide, polyethylene glycol, phosphatidylcholine, cationic-charged and anionic-charged. The cationic-charged and phosphatidylcholine-coated Fe3O4 MNPs could increase the cellular uptaken in a dose dependent manner and the particles caused a vacuolar appearance in A549 cells. With exposure to 8.5 T SMF, the assay of ATP content showed that anionic-charged and phosphatidylcholine-coated Fe3O4 MNPs changed the energy metabolism of A549 cells, which might be consistent with the observation in cellular internalization. The cell viability and proliferation of A549 cells were all slightly affected by various modified MNPs with or without 8.5 T SMF exposure.

Abstract:The toxicity of nanostructured materials and nanoparticles are very important considerations for many nanotechnology applications. Iron nanowires (NWs), as useful magnetic nanomaterials, may be good candidates for several biomedical applications. Here Fe NWs with an average diameter of about 50 nm were prepared by electrodeposition within the nanopores of anodic aluminum oxide (AAO) templates, and characterized by using scanning electron microcopy (SEM), transmission electron microscopy (TEM) and vibrating sample magnetometer (VSM). The cytotoxicity of the Fe NWs was studied at cell level. Specifically, the influence of concentration and size on the cytotoxicity of Fe NWs to HeLa cells was evaluated by MTT assay combined with direct morphology observations by TEM and phase contrast microscopy. The results clearly showed that the presence of Fe NWs had no significant effect on the cell proliferation and cell viability, even the HeLa cells exposed to Fe NWs at the high concentration of 10,000 per cell for 72 h still showed high cell viability of about 80%. From phase contrast microscopy, confocal laser scanning microscopy (CLSM) and TEM observations, it was found that Fe NWs were indeed internalized by HeLa cells. The cellular uptake process and results of Fe NWs demonstrated that as-prepared Fe NWs had not only a good biocompatibility but also a very low cytotoxicity.
The Biological Toxicity of Ferrite NanoNi-Zn Materials on Blood and Cells

Abstract:The influences of ferrite nanoNi-Zn powders on blood and cells of rats are studied by biological and physiological methods. The ferrite nanoNi-Zn powders are made by high-temperature oxidized method, its size is 50-80 nm obtained by Scanning Electron Microscope and X-ray diffraction image . From the experiment we find that the amount of leucocytes in the bloods in the experimental group is considerably large than that in the controlled group (p<;<;0.05) , but the amount of erythrocytes in the controlled group is considerably high than that in the Experimental group (p<;<;0.01) . The obvious differences show that the ferrite nanoNi-Zn powders have the evident biological effect and can change the biochemical and general indexes of blood of the rats. We measure and collect the infrared spectra of absorption of hemaglobins in the bloods after injection of ferrite nanoNi-Zn powders and find the variations of the strengths and wavenumbers of peaks in their infrared spectra relative to that in cotrolled group. This shows the molecular structure of hematoglobin changes in such a case. Just so, the above biological effects of ferrite nanoNi-Zn occur.

Methods MTT assay was used to evaluate the in vitro cyitoxicity; hemolysis test was carried out to estimate blood toxicity; Fe 2O 3 suspended in sterile 0.9% NaCl were intraperitoneally injected into KM mouse to calculate the LD 50; micronucleus (MN) were reckoned to identify genotoxicity. Results The experiments showed that the toxicity of the material on mouse fibroblast (L929) cell lines was between 0～1 degree;it had no hemolysis activity; LD 50 arrived at 5.45g·kg -1 after intraperitoneal injection of 1ml suspension; micronucleus test showed it had no genotoxic effects either. Conclusions Our results show that the selfprepared nanosized F 2O 3 is a kind of high biocompatibility materials and perhaps is suitable for further application in tumor hyperthermia.

Abstract:The environmental safety effect of nano-sized iron materials including Fe_3O_4,Fe_3O_4-zeolite composite and iron-doped zeolite was studied using a tetrahymena thermophila model. The magnetic nano-sized Fe_3O_4 is most sensitive to the suppression of the grouth of tetrahymena thermophila, as compared with the Fe_3O_4-zeolite composite and iron-doped zeolite. Also, the suppression was increased with the material concentration.

Abstract:Abstract In this study, we report the synthesis and characterization of NiFe 2 O 4 and CoFe 2 O 4 nanoparticles (NPs) which are widely used in the biomedical area. There is still limited knowledge how the properties of these materials are influenced by different chemical routes. In this work, we investigated the effect of heat treatment over cytotoxicity of cobalt and niquel ferrites NPs synthesized by sol-gel method. Then the samples were studied using transmission electron microscopy (TEM), X-ray diffraction (XRD), X-ray photoelectron spectroscopy (XPS), vibrating sample magnetometer (VSM), Fourier Transform Infrared Spectroscopy Analysis (FTIR), and X-ray fluorescence (XRF). The average crystallite sizes of the particles were found to be in the range of 20–35 nm. The hemocompatibility (erythrocytes and leukocytes) was checked. Cytotoxicity results were similar to those of the control test sample, therefore suggesting hemocompatibility of the tested materials.

Abstract:The ultrasmall nanoparticles easily lead to a more seriously response than larger nanoparticles because of their physicochemical features. It is essential to understand their cytotoxicity effects for their further application. Here, we used ultrasmall 9 nm Fe3O4 NPs to explore its cytotoxicity mechanism on breast cancer cells. We demonstrated 9 nm Fe3O4 NPs was effectively internalized into cells and located in nucleus, subsequently, it inhibited DNA synthesis through inducing S-phase arrest. Moreover, 9 nm Fe3O4 NPs induced ROS production and oxidative damage by disturbing the expression of antioxidant-related genes (HMOX-1, GCLC and GCLM), which resulted in the enhancement of cells apoptosis and inhibition of cell proliferation, suggesting its potential to be used as therapeutic drug.

Abstract:Ferroferric oxide nanoparticle (denoted as Nano-Fe3O4) has low toxicity and is biocompatible, with a small particle size and a relatively high surface area. It has a wide range of applications in many fields such as biology, chemistry, environmental science and medicine. Because of its superparamagnetic properties, easy modification and function, it has become an important material for addressing a number of specific tasks. For example, it includes targeted drug delivery nuclear magnetic resonance (NMR) imaging in biomedical applications and in environmental remediation of pollutants. Few articles describe the preparation and modification of Nano-Fe3O4 in detail. We present an evaluation of preparation methodologies, as the quality of material produced plays an important role in its successful application. For example, with modification of Nano-Fe3O4, the surface activation energy is reduced and good dispersion is obtained.
