Abstract:Optical second harmonic generation (SHG) together with surface tension measurements are used to investigate soluble amphiphilic molecules adsorbed at the air/water interface from solution. It is found that the adsorbed molecules form a single polar-oriented monolayer which, at saturation, resembles a close-packed monolayer of insoluble molecules. SHG is particularly sensitive to the noninversion symmetry of the interfacial region and therefore provides an inherent discrimination against isotropically oriented molecules in the subsurface and the solution.

Abstract:Optical second harmonic generation is employed to investigate the adsorption of soluble naphthalene sulfonates from the bulk solution to the air/water interface in the presence of excess counter ions. Both the surface density of surfactant molecules and the surface pressure were measured as functions of surfactant concentration in solution to yield the adsorption isotherm and the surface pressure/area isotherm. The system investigated shows nonideal gas behavior. The ratio of the activity coefficients at the surface and in the bulk is not unity; however both appear to be constant over the concentration range probed.

Abstract:Insoluble molecular monolayers at gas-liquid interfaces provide an insight to the understanding of surfactants, wetting, micromulsions and membrane structures,and offer a possibility to study the rich world of 2-dimensional phase-transitions. In the interpretation of the observed properties of these systems various assumptions about the molecular orientation are often made, but so far few clear experimental data exist [1]. In this paper we will show how optical second harmonic generation (SHG) can be used to measure the molecular orientation of monolayers of surfactant molecules at water-air interfaces. By simultaneously measuring the surface pressure versus surface molecular area we can show for the first time that the observed liquid condensed-liquid expanded transition is an orientational phase-transition.

Abstract:Lipopeptide surfactants are composed of amino acids and fatty acids and carry the inherent benefits of biocompatibility, degradability, functionality and easiness for structural design. In this work, the interfacial adsorption of a novel lipopeptide surfactant C14K1 has been examined by combining the measurements of spectroscopic ellipsometry (SE) and neutron reflection (NR). As for other model surfactants, the silicon oxide/water interface has been adopted to undertake the interfacial screening due to its readiness to facilitate the parallel measurements. SE revealed that the dynamic interfacial adsorption was characterized by an initial fast step within the first 2-3 min, followed by a slow relaxation step over the subsequent 30 min or so. Majority of the lipopeptide was found to adsorb with the fast initial step but the subsequent structural relaxation and adjustment led to the adsorption plateau. Interestingly, this pattern of dynamic adsorption is very different from amphiphilic peptides such as V6K1 and V6K2, but is similar to diblock copolymers bearing a hydrophilic MPC block and a pH responsive DEA block comprised of tertiary amine groups. Subsequent NR, in conjunction with deuterium labeling, revealed the formation of bilayer structure over a wide concentration and pH range, with the hydrophobic tails sandwiched in the middle and hydrophilic charged head groups projected outwards. The fully packed bilayer is some 40 angstrom thick and changes in pH did not affect layer thickness much but did alter the packing density due to the different electrostatic repulsion associated with the charge dissociation of the head groups. Whilst surface adsorption of C14K1 behaved in many aspects like nonionic C12E6 (surface tension, salt effect and bilayer packing), detailed structural determination revealed little intermixing between the heads and tails, an observation different from C12E6.

Abstract:Lipopeptides are an important group of biosurfactants expressed by microorganisms. Because they are well-known for being biocompatible, biodegradable, and highly surface active, they are attractive for a wide range of applications. Natural lipopeptide surfactants are however impure; it is hence difficult to use them for exploring the structure-function relation. In this work, a series of cationic lipopeptide surfactants, C(14)K(n) (n = 1-4), where C denotes the myristic acyl chain and K denotes lysine (Lys), have been synthesized, and their interfacial behavior has been characterized by studying their adsorption at the silicon/water interface (bearing a thin native oxide layer) using spectroscopic ellipsometry and neutron reflection (NR). The dynamic adsorption was marked by an initial fast step within the first 2-3 min followed by a slow molecular relaxation process over the subsequent 20-30 min. The initial rate of time-dependent adsorption and the equilibrated adsorbed amount showed a steady decrease with increasing n, indicating the impact of the molecular size, structure, and charge. NR revealed the formation of sandwiched bilayers from C(14)K(n), similar to conventional surfactants such as nonionic C(12)E(6) and cationic C(16)TAB. However, the electrostatic attraction between K and the silica surface caused confinement of the K groups, forcing the head segments into a predominantly flat-on conformation. This characteristic structural feature was confirmed by the almost constant thickness of the headgroup regions ranging from 8 to 11 Å as determined from NR combined with partial deuterium labeling to the acyl tail. An increase in area per molecular pair with n resulted directly from increasing the footprint. As a result, the hydrophobic back-to-back tail mixing and acyl chain tilting rose with n. The extent of chain-head intermixing became so intensified that the C(14)K(4) bilayer could be approximated to a uniform layer distribution.

Abstract:As an intrinsically surface-specific technique, Second Harmonic Generation (SHG) is widely used in the study of interface in recent years. The SHG signals from the air/liquid interface of Rhodamine B and Sodium Dodecyl Benzene Sulfonate (SDBS) aqueous solutions were obtained and analyzed, which demonstrate that the SHG signal intensity of Rhodamine B is stronger than that of SDBS. Compared with one single solution, the SHG signal intensity of the mixed aqueous solution of Rhodamine B and SDBS decreases. From the UV-VIS absorption spectrum of the two aqueous solutions, it can be seen that Rhodamine B has an absorption peak closer to the second harmonic frequency. Therefore the resonance of the second harmonic frequency with the frequency of the dipole transitions of the chromophore considerably enhances the signal intensity. Furthermore, the hyperpolarizabilities of the molecules of Rhodamine B and SDBS are calculated from first-principles, which reveal that the hyperpolarizability of Rhodamine B molecule is greater than that of SDBS molecule. When they are mixed, molecules of Rhodamine B and SDBS gather together because Rhadamine B molecule carries positive surface charge and SDBS is anionic surfactant, causing the decrease of the SHG intensity of the mixed solution.

Abstract:The array behaviour of anionic surfactant sodium dodecylbenzenesulfonate (SDBS) on the air/water interface was investigated using non-resonant surface second harmonic generation (SHG) and molecular simulation. It was found that the SHG intensities changed with surfactant concentrations non-monotonously under Critical Micelle Concentration (CMC). The mean orientational angle of SDBS molecule at the air/liquid interface was calculated, and the molecular array behaviour was analyzed by molecular dynamics (MD) simulation. The simulated results agreed with the experimental results very well, which provided the detailed information about the interfacial molecular configuration of surfactant, not only indicating the SHG intensity related to the Ns and r(θ), but also demonstrates that the microscopic information presented by the MD simulation method is reliable. According to the results, the surfactant molecules get clustered even at very low concentration far lower than the CMC.

Abstract:Chemistry at the surface of solid particles in colloidal dispersions is important in the description of such diverse topics as nutrient and contaminant flow in the environment and the creation of better paints and cosmetics. Particles in these systems are often characterized by molecular-level, surface heterogeneity. This molecular heterogeneity influences many aspects of the chemical processes of interest, among the simplest of which is physisorption. In this study we construct adsorption isotherms for the triphenylmethane dye Malachite Green on various latex particles using the flow through Second Harmonic Generation (ftSHG) technique. This technique makes it possible to construct significantly higher resolution isotherms than conventional separation methods. Here we chose both particles that we expect to be chemically homogeneous (plain polystyrene) and particles whose surface we expect to have molecular level heterogeneity (carboxylated and hydroxylated polystyrene). We find that, using the ftSHG isotherms, we are able to quantify the adsorption energies and site densities of multiple adsorption sites on the carboxylated and hydroxylated polystyrene particles but find only a single type of adsorption site on the plain polystyrene. We expect the ability to measure adsorption site energy and density on chemically heterogeneous latex colloids will be useful in the creation of better biosensors and that the application of ftSHG to other chemically heterogeneous colloids should provide insight into a variety of natural and engineered processes.
Surface-induced Monolayer Alignment Probed by Second Harmonic Generation

Abstract:Surface-induced alignment of molecules by a substrate is a fascinating problem of both basic and applied interest. As an example, it is well known that the bulk alignment of a liquid crystal cell can be controlled by the surface treatment, but the mechanism is hardly understood. Rubbing of polymer or surfactant coated substrates is a technique commonly used to create bulk homogeneous alignment (average molecular orientation parallel to the surface). Both short range interaction at the molecular level and a bulk elastic interaction of a macroscopic nature have been proposed as possible alignment mechanisms. We have used optical second harmonic generation (SHG) to study the effect of interfacial interaction on the orientational distribution of a surface monolayer of molecules and learn how this interaction can induce the bulk alignment.

Abstract:Surface second harmonic generation (SHG) was used to probe orientation of cavitand molecules deposited on water as Langmuir films and on fused silica substrates either bare or functionalized with a surfactant. Measurements performed during compression of the Langmuir films reveal that while the surface density may vary, molecules always appear more or less oriented with their long axis perpendicular to the water surface. Phase measurements indicate that the vase-like heads of the molecules cover the water for Langmuir films, but face the air for monolayers spread on surfactant-coated substrates.

Abstract:Second harmonic generation (SHG) has now been well established as a versatile surface-sensitive probe [1]. It has been used to study electrochemical processes at electrode surfaces, molecular adsorption and desorption at metal and semiconductor surfaces, orientational phase transition of molecular monolayers on water, surface reconstruction and epitaxial growth, and so on. More recently, it has been employed as a tool to monitor monolayer polymerization and other surface reactions [2], to probe polar order of molecules at interfaces [3], and to measure molecular nonlinearity [4]. While most surface techniques are restricted to the solid/vacuum environment, SHG is applicable to nearly all interfaces as long as the interfaces are accessible by light. In addition, SHG has the advantages of being capable of in-situ measurements with high temporal, spatial, and spectral resolutions.
SHG Investigation of Liquid Crystal-Silane Interfaces

Abstract:Using optical second harmonic generation, we have studied the interaction between 4′-n-octyl-4-cyanobiphenyl (8CB) liquid crystal molecular monolayers and glass substrates treated with three types of silane coupling agent: MAP, DMOAP, and OTS. On adsorption from the gaseous phase at atmospheric pressure, we measure the coverage, orientation, and binding energy of monolayers on these substrates and find that, although the bulk alignment differs from different silanes, the alignment of the monolayer is unaffected. We also find a polar layer of liquid crystal at the surface of MAP and DMOAP, unlike the dimerized layers that are found in the bulk liquid crystal. The number of binding sites on these silanes is approximately equal and less than the number of sites on clean glass. Their binding energies are also slightly below that on glass. On OTS, no polar layer formed, although a dimerized bilayer was present.

Abstract:Emulsions are widely used in industrial and environmental remediation applications. The breaking and reformulation of emulsions, which occur during their use, lead to changes in their surface composition as well as their physical and chemical properties. Hence, a fundamental understanding of the transfer of surfactant molecules between emulsion particles is required for optimization of their applications. However, such an understanding remains elusive because of the lack of in situ and real-time surface-specific techniques. To address this, we designed and synthesized the surfactant probe molecules MG-butyl-1 (2) and MG-octyl-1 (3), which contain an n-butyl and an n-octyl chain, respectively, and a charged headgroup similar to that in malachite green (MG, 1). MG is known to be effective in generating second harmonic generation (SHG) signals when adsorbed onto surfaces of colloidal microparticles. Making use of the coherent nature of SHG, we monitored in real-time the transfer of 2 and 3 between oil-in-water emulsion particles with diameters of ~220 nm. We found that 3 is transferred ~600 times slower than 2, suggesting that an increase in the hydrophobic chain length decreases the transfer rate. Our results show that SHG combined with molecular design and synthesis of surfactant probe molecules can be used to measure the rate of surfactant transfer between emulsion particles. This method provides an experimental framework for examining the factors controlling the kinetics of surfactant transfer between emulsion particles, which cannot be readily investigated in situ and in real-time using conventional methods.

Abstract:Infrared-visible sum-frequency (SFG) vibrational spectroscopy is used to study chain conformation of cationic surfactant monolayers (dioctadecyl dimethyl ammonium chloride, DOAC), adsorbed at various quartz/liquid interfaces at low coverage. The results show that the chains can assume many different conformations: an all-trans form if the liquid is alkane with a similarly long chain length, a bent form with significant trans-gauche defects if the liquid is CCL4 or CDCL3 or short-chain alkanes, and a highly contracted form if the liquid is water or alcohol.

Abstract:The surface properties of carbon black microparticles in aqueous solution are characterized through adsorption of a hydrophobic ion (malachite green) monitored by optical second-harmonic generation (SHG). Carbon black particle surfaces with 14 different O/C ratios (from 0.028 to 0.270) have been examined. The adsorption isotherm of MG on the surface of carbon black particles weighing as little as 0.010 g can be measured using SHG measured during continuous titration of the MG molecule into the solution. The SHG data allows determination of the adsorption free energy and reveals coherent interference between the SH fields from the surface-adsorbed MG molecules and the carbon black particle. The change in the coherent interference implies a change of adsorption orientation of MG on surfaces with different O/C ratios. There is strong correlation found between the observed adsorption characteristics and the O/C ratio of the surfaces. Furthermore, MG adsorption isotherms of surfaces with higher O/C ratios appear similar to the ones of surfaces preadsorbed with surfactants that can increase surface hydrophilicity and dispersion in aqueous solution. All observations indicate that particle surfaces with higher O/C ratios appear more hydrophilic. This study demonstrates once again the many advantages of SHG as an effective surface analytical technique of colloidal particles.

Abstract:Phospholipids are ubiquitous components of biomembranes and common biomaterials used in many bioengineering applications. Understanding adsorption of phospholipids at the air-water surface plays an important role in the study of pulmonary surfactants and cell membranes. To date, however, the biophysical mechanisms of phospholipid adsorption are still unknown. It is challenging to reveal the molecular structure of adsorbed phospholipid films. Using combined experiments with constrained drop surfactometry and molecular dynamics simulations, here, we studied the biophysical mechanisms of dipalmitoylphosphatidylcholine (DPPC) adsorption at the air-water surface. It was found that the DPPC film adsorbed from vesicles showed distinct equilibrium surface tensions from the DPPC monolayer spread via organic solvents. Our simulations revealed that only the outer leaflet of the DPPC vesicle is capable of unzipping and spreading at the air-water surface, whereas the inner leaflet remains intact and forms an inverted micelle to the interfacial monolayer. This inverted micelle increases the local curvature of the monolayer, thus leading to a loosely packed monolayer at the air-water surface and hence a higher equilibrium surface tension. These findings provide novel insights, to our knowledge, into the mechanism of the phospholipid and pulmonary surfactant adsorption and may help understand the structure-function correlation in biomembranes.

Abstract:A single molecular layer is generally sufficient to produce observable optical second harmonic generation (SHG). Furthermore, the selection rules governing this process make the SHG from a single monolayer often stronger than that from the medium supporting the monolayer. We have studied SHG from various Langmuir-type monolayers (i.e. monolayers spread on a water surface) in the following contexts: (1) Study of chemical reactions (e.g. polymerization) and two-dimensional phase transitions in molecular monolayers on water. (2) Development of a new technique to evaluate optical nonlinear coefficients of organic molecules, and their relationship to the molecular structure.
