Relation of MAGE Gene Promoter Demethylation and MAGE Mrna Expression in Colorectal Carcinoma

Abstract:Objective To observe the relation between MAGE gene promoter region demethylation and MAGE mRNA expression in colorectal carcinoma .Methods The expression and demethylation of MAGE-1, -3 genes from 65 samples with human colorectal carcinomas and corresponding adjacent colonic mucosa specimens were detected by reverse transcrip -tion polymerase chain reaction(RT-PCR) and methylation-specific PCR.The association between the expression and pro-moter region demethylation of the genes was analyzed .Results Expression rate of MAGE-1, -3 genes was 18.5%(12/65) and 33.9%(22/65), respectively, however, none of the corresponding adjacent non-tumorous tissues expressed any MAGE mRNA.But the positive rates of promoter demethylation of the two genes were as follows : 29.2%(19/65) and 47.7%(31/65) in cancerous tissues , the corresponding adjacent non-tumorous 1.5%(1/65) and 4.6%(3/65) respec-tively.MAGE-1,-3 genes mRNA expression had relation with its promoter demethylaton (all P<0.05).Conclusions Demethylaton in promoter regions of MAGE-1,-3 genes may be the critical mechanism of mRNA expression , and may pro-vide information of early diagnosis and prognosis in colorectal cancer patients .
