Abstract:Multimodal imaging technique is an alternative approach to improve sensitivity of early cancer diagnosis. In this study, highly fluorescent and strong X-ray absorption coefficient gold nanoclusters (Au NCs) are synthesized as dual-modality imaging contrast agents (CAs) for fluorescent and X-ray dual-modality imaging. The experimental results show that the as-prepared Au NCs are well constructed with ultrasmall sizes, reliable fluorescent emission, high computed tomography (CT) value and fine biocompatibility. In vivo imaging results indicate that the obtained Au NCs are capable of fluorescent and X-ray enhanced imaging.

Abstract:Gold nanoclusters (AuNCs) with fluorescence in the Near Infrared (NIR) by both one- and two-photon electronic excitation were incorporated in mesoporous silica nanoparticles (MSNs) using a novel one-pot synthesis procedure where the condensation polymerization of alkoxysilane monomers in the presence of the AuNCs and a surfactant produced hybrid MSNs of 49 nm diameter. This method was further developed to prepare 30 nm diameter nanocomposite particles with simultaneous NIR fluorescence and superparamagnetic properties, with a core composed of superparamagnetic manganese (II) ferrite nanoparticles (MnFe2O4) coated with a thin silica layer, and a shell of mesoporous silica decorated with AuNCs. The nanocomposite particles feature NIR-photoluminescence with 0.6% quantum yield and large Stokes shift (290 nm), and superparamagnetic response at 300 K, with a saturation magnetization of 13.4 emu g-1. The conjugation of NIR photoluminescence and superparamagnetic properties in the biocompatible nanocomposite has high potential for application in multimodal bioimaging.

Abstract:The development of a multifunctional nanoprobe capable of non-invasive multimodal imaging is crucial for precise tumour diagnosis. Herein, we report a facile polymer-assisted method to produce Au-Fe3O4 nanocomposites (NCPs) for the dual-modal magnetic resonance (MR) and X-ray computed tomography (CT) imaging of tumours. In this approach, amino-functionalized Au nanospheres were first obtained by surface modification of the bifunctional polymer SH-PEG-NH2. Hydrophilic and carboxyl-functionalized Fe3O4 nanoparticles were produced by phase transfer of reverse micelle oxidation in our previous work. The Au nanoparticles were conjugated with hydrophilic Fe3O4 nanoparticles through an amide reaction. The obtained Au-Fe3O4 nanocomposites display a high r2 relativity (157.92 mM−1 s−1) and a Hounsfield units (HU) value (270 HU) at Au concentration of 8 mg/mL and could be applied as nanoprobes for the dual-modal MR/CT imaging of a xenografted tumour model. Our work provides a facile method to prepare Au-Fe3O4 nanocomposites for dual-modal MR/CT imaging, and this method can be extended to prepare other multifunctional nanoparticles for multimodal bioimaging.

Abstract:A simple seed-mediated growth route is developed to fabricate monodisperse, uniform superparamagnetic Fe(3)O(4) core/gold shell structured nanocomposites with tunable sizes and optical properties, in which gold seed formation and attachment onto the core surface via S-Au covalent bonding proceeds almost simultaneously in the one-pot synthesis. The as-prepared nanocomposite is demonstrated to have a great potential for magnetic resonance imaging (MRI)-guided, focused photothermal tumor therapy under near-IR laser radiation.

Abstract:Multicomponent Ni7Co3 alloy-Au nanorings can be facilely synthesized at room temperature using metallic Ni7Co3 alloy nanorings as both reducing agent and sacrificial template in water. These novel alloy-Au hybrid nanorings are well biocompatible due to coating with poly(vinylpyrrolidone) and gold nanoparticles. When increasing the molar ratio of alloy to gold salt from 1:1 to 5:1, the number of the gold nanoparticles loaded on the surface of alloy nanorings were found to be decreased, and their morphology was transformed from rod to particle, remaining the ring-like assemblies. Their saturation magnetization increased due to the ring-like assemblies, resulting in the enhancement of the signal intensity in Spin-spin relaxation time (T-2) weighted magnetic resonance imaging measurement. These alloy-Au hybrid nanocomposites can perform as multimodal imaging contrast agents for cancer cell diagnosis by two-photon fluorescent imaging and T-2-weighted magnetic resonance imaging. In addition, their potential application for photothermal therapy was preliminarily investigated.

Abstract:Multimodal imaging based on complementary detection principles has great potential for improving the accuracy of tumor diagnosis. Fe3O4 core/Au shell nanoparticles (Fe3O4@Au NPs) can be used as an effective multimodal contrast agent due to combining the advantageous and highly complementary features of the Fe3O4 core and gold shell. In the present work, we have developed a novel Fe3O4@Au NP based probe for targeting and multimodal imaging of cancer cells. The prepared Fe3O4@Au NPs have been characterized by transmission electron microscopy (TEM), visible spectra and SERS. The potential use of the prepared Fe3O4@Au NPs as a multimodal contrast probe for magnetic resonance imaging (MRI), microwave-induced thermoacoustic imaging and photoacoustic imaging was then evaluated. Importantly, when conjugated with a cancer cell targeted molecular and fluorescent dye, the Fe3O4@Au NPs could be internalized by the corresponding cancer cells selectively and sensitively, and fluorescence imaging was realized at the same time. The bio-modified Fe3O4@Au NPs incorporating multiple functionalities into one single nano-structured system can be effectively used for targeting and multimodal imaging of cancer cells simultaneously.
Porous Gold Nanocluster-Decorated Manganese Monoxide Nanocomposites for Microenvironment-Activatable MR/photoacoustic/CT Tumor Imaging

Abstract:Stimuli-responsive nanoprobes that integrate multi-modal imaging capacities are highly desirable for precise tumor visualization. Herein, novel porous gold nanocluster-decorated manganese monoxide nanocomposites (MnO@Au NCs) were synthesized via a facile approach. The porous gold nanocluster layer was germinated on the surface of the as-prepared MnO@DMSA NPs through simple reduction of chloroauric acid in the presence of hydroxylamine hydrochloride. The MnO@Au NCs could be effectively internalized by tumor cells and slowly release Mn2+ ions within the acidic tumor microenvironment, improving the visualization of the tumor morphology. Benefitting from the porous architecture, the enhanced accessibility of Mn centers to proximal water molecules greatly augmented T-1-weighted MRI contrast capacity. Compared with the conventional Mn-based contrast agents, the porous Au nanoclusters on MnO@Au NCs could delay the release of Mn2+ ions and thus effectively prolong the diagnostic time window. The broad near-infrared absorption of MnO@Au NCs features a high photoacoustic imaging depth than that of conventional gold nano-spheres. Moreover, the Au nanoclusters exhibited desirable X-ray computed tomography contrast and rapid clearance from the living body. The as-prepared MnO@Au NCs hold great potential for accurate tumor imaging.

Abstract:Cancer treatment has a far greater chance of success if the neoplasm is diagnosed before the onset of metastasis to vital organs. Hence, cancer early diagnosis is extremely important and remains a major challenge in modern therapeutics. In this contribution, facile and new method for rapid multimodal tumor bioimaging is reported by using biosynthesized iron complexes and gold nanoclusters via simple introduction of AuCl4- and Fe2+ ions. The observations demonstrate that the biosynthesized Au nanoclusters may act as fluorescent and computed tomography probes for cancer bioimaging while the iron complexes behave as effective contrast agent for magnetic resonance imaging. The biosynthesized iron complexes and gold nanoclusters are found biocompatible in vitro (MTT (3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide) assay) and in vivo for all the vital organs of circulatory and excretory system. These observations raise the possibility that the biosynthesized probes may find applications in future clinical diagnosis for deep seated early neoplasms by multimodal imaging.

Abstract:Nanoparticles with a gold shell and iron core have unique optical and magnetic properties that can be utilized for simultaneous detection and treatment strategies. Several nanoparticles have been synthesized and show an ability to mediate a variety of potential applications in biomedicine including cancer molecular optical and magnetic resonance imaging, controlled drug delivery, and photothermal ablation therapy. However, to be effective these nanoparticles must be delivered efficiently into their targets. In this review, we will provide an updated summary of the gold-shelled magnetic nanoparticles that have been synthesized, methods for characterization, and their potential for cancer diagnosis and treatment. We will also discuss the biological barriers that need to be overcome for the effective delivery of these nanoparticles. The desired nanoparticle characteristics needed to evade these biological barriers, such as size shape. Surface charge, and surface coating are also explained.

Abstract:This paper reports a type of multifunctional hybrid nanoparticle (NP) composed of gold nanocrystals coated on and/or embedded in a magnetite-fluorescent porous carbon core-shell NP template (Fe3O4@PC-CDs-Au) for biomedical applications, including magnetic/NIR-responsive drug release, multicolor cell imaging, and enhanced photothermal therapy. The synthesis of the Fe3O4@PC-CDs-Au NPs firstly involves the preparation of core-shell template NPs with magnetite nanocrystals clustered in the cores and fluorescent carbon dots (CDs) embedded in a porous carbon shell, followed by an in situ reduction of silver ions (Ag+) loaded in the porous carbon shell and a subsequent replacement of Ag NPs with Au NPs through a galvanic replacement reaction using HAuCl4 as a precursor. The Fe3O4@PC-CDsAu NPs can enter the intracellular region and light up mouse melanoma B16F10 cells in multicolor mode. The porous carbon shell, anchored with hydrophilic hydroxyl/carboxyl groups, endows the Fe3O4@PC-CDs-Au NPs with excellent stability in the aqueous phase and a high loading capacity (719 mg g(-1)) for the anti-cancer drug doxorubicin (DOX). The superparamagnetic Fe3O4@PC-CDs-Au NPs with a saturation magnetization of 23.26 emu g(-1) produce localized heat under an alternating magnetic field, which triggers the release of the loaded drug. The combined photothermal effects of the Au nanocrystals and the CDs on/in the carbon shell can not only regulate the release rate of the loaded drug, but also efficiently kill tumor cells under NIR irradiation. Benefitting from their excellent optical properties, their magnetic field and NIR light-responsive drug release capabilities and their enhanced photothermal effect, such nanostructured Fe3O4@PC-CDs-Au hybrid NPs are very promising for simultaneous imaging diagnostics and high efficacy therapy.

Abstract:New multifunctional nanospheres have been designed and synthesized through a green and facile strategy, which could be readily used in multi-modality tumor imaging through near-infrared fluorescence (NIRF) imaging, magnetic resonance imaging (MRI) and computed tomography (CT) imaging. Such nanospheres are made of porous superparamagnetic Zn1/3Fe8/3O4 nanosphere cores covered by a thin layer shell of Ag clusters. While the shell of Ag clusters provides efficient NIRF imaging, the Zn1/3Fe8/3O4 nanosphere cores allow external magnetic manipulation and readily facilitates their utilization for MRI and CT imaging.

Abstract:Multifunctional nanocomposites combining imaging and therapeutic functions have great potential for cancer diagnosis and therapy. In this work, we developed a novel theranostic agent based on hollow gold nanospheres (HGNs) and superparamagnetic iron oxide nanoparticles (SPIO). Taking advantage of the excellent magnetic properties of SPIO and strong near-infrared (NIR) absorption property of HGNs, such nanocomposites were applied to targeted magnetic resonance imaging (MRI) and photoacoustic imaging (PAI) of cancer cells. In vitro results demonstrated they displayed significant contrast enhancement for T2-weighted MRI and strong PAI signal enhancement. Simultaneously, the nanocomposites exhibited a high photothermal effect under the irradiation of the near-infrared laser and can be used as efficient photothermal therapy (PTT) agents for selective killing of cancer cells. All these results indicated that such nanocomposites combined with MRI-PAI and PTT functionality can have great potential for effective cancer diagnosis and therapy.

Abstract:Multifunctional hybrid nanoparticles (NPs, ∼100 nm) that combine magnetic Fe3O4 nanocrystals and fluorescent carbon dots (CDs) in porous carbon (C) were successfully synthesized using a one-pot solvothermal method by simply increasing the H2O2 concentration. The resultant Fe3O4@C-CDs hybrid NPs not only demonstrate excellent magnetic responsive properties (Ms = 32.5 emu g-1) and magnetic resonance imaging ability (r = 674.4 mM-1 s-1) from the Fe3O4 nanocrystal core, but also exhibit intriguing photoluminescent (quantum yield ∼6.8%) properties including upconversion fluorescence and excellent photostability from the CDs produced in the porous carbon. The hybrid NPs can enter the intracellular region and illuminate mouse melanoma B16F10 cells under different excitation wavelengths. Meanwhile, the mesoporous carbon shell and hydrophilic surface functional groups endow the hybrid NPs with high loading capacity (835 mg g-1) for the anti-cancer drug doxorubicin and excellent stability in aqueous solutions. More importantly, the hybrid NPs can absorb and convert near-infrared (NIR) light to heat due to the existence of CDs, and thus, can realise NIR-controlled drug release and combined photothermo/chemotherapy for high therapeutic efficacy. Such nanostructured Fe3O4@C-CDs hybrid NPs demonstrate great promise towards advanced nanoplatforms for simultaneous imaging diagnostics and high efficacy therapy.

Abstract:This study reports magnetic targeting guided magnetic (MR)/photoacoustic (PA) dual-modal imaging by core/shell Fe3O4/Au nanoparticles. In this work, MR imaging provides time-dependent tumor location, and PA imaging reveals high resolution vasculatures inside the tumor. It is noted that the synthesized Fe3O4/Au nanoparticles exhibited higher r2 value up to 329 mM-1 s-1 than previously reported T2 contrast agents. Furthermore, the Fe3O4/Au NPs are applied as a promising candidate for in vivo MR/PA imaging of tumors by intravenously injection into LNCaP tumor-beared mice. The MR/PA imaging results show a significantly enhanced MR/PA images in the tumor site. The prepared core/shell Fe3O4/Au nanoparticles will be widely applicable in multi-modal imaging.

Abstract:It is essential to develop a simple synthetic strategy to improve the quality of multifunctional contrast agents for cancer diagnosis. Herein, we report a time-saving method for gadolinium (Gd3+) ions-mediated self-assembly of gold nanoclusters (GNCs) into monodisperse spherical nanoparticles (GNCNs) under mild conditions. The monodisperse, regular and colloidal stable GNCNs were formed via selectively inducing electrostatic interactions between negatively-charged carboxylic groups of gold nanoclusters and trivalent cations of gadolinium in aqueous solution. In this way, the Gd3+ ions were chelated into GNCNs without the use of molecular gadolinium chelates. With the co-existence of GNCs and Gd3+ ions, the formed GNCNs exhibit significant luminescence intensity enhancement for near-infrared fluorescence (NIRF) imaging, high X-ray attenuation for computed tomography (CT) imaging and reasonable r1 relaxivity for magnetic resonance (MR) imaging. The excellent biocompatibility of the GNCNs was proved both in vitro and in vivo. Meanwhile, the GNCNs also possess unique NIRF/CT/MR imaging ability in A549 tumor-bearing mice. In a nutshell, the simple and safe GNCNs hold great potential for tumor multi-modality clinical diagnosis.

Abstract:Designing nanomaterials with advanced functions and physical properties to improve cancer diagnosis and treatment has been an enormous challenge. In this work, we report the synthesis of magnetic gold nanowreaths (AuNWs) by combining wet-chemical synthesis with layer-by-layer self-assembly. The presence of Au branches, small junctions, and central holes in AuNWs led to improved photothermal effect compared with Au nanoring seeds and thick Au nanoring with smooth surface. The self-assembly of exceedingly small magnetic iron oxide nanoparticles (ES-MIONs) on the surfaces of AuNWs not only effectively quenched the T1-weighted magnetic resonance imaging (MRI) ability due to the enhanced T2 decaying effect, but also provided the responsiveness to glutathione (GSH). After intravenous injection, the T1 signal of magnetic AuNWs initially in the "OFF" state can be intelligently switched on in response to the relatively high GSH concentration in tumor. And the formation of larger assemblies of ES-MIONs improved their tumor delivery compared to ES-MIONs themselves. Thus, the magnetic AuNWs showed higher MRI contrast than ES-MIONs or commercial Magneveist in T1-weighted MR imaging of tumor. Furthermore, the magnetic AuNWs have absorption in near infrared range, leading to strong photoacoustic signal and effective photoablation of tumor. Therefore, our GSH-responsive self-assembled magnetic AuNWs could enhance T1-weighted MRI and photoacoustic imaging of tumor and be used for imaging-guided photothermal therapy.

Abstract:Multicomponent Ni7Co3 alloy–Au nanorings can be facilely synthesized at room temperature using metallic Ni7Co3 alloy nanorings as both reducing agent and sacrificial template in water. These novel alloy–Au hybrid nanorings are well biocompatible due to coating with poly(vinylpyrrolidone) and gold nanoparticles. When increasing the molar ratio of alloy to gold salt from 1:1 to 5:1, the number of the gold nanoparticles loaded on the surface of alloy nanorings were found to be decreased, and their morphology was transformed from rod to particle, remaining the ring-like assemblies. Their saturation magnetization increased due to the ring-like assemblies, resulting in the enhancement of the signal intensity in Spin–spin relaxation time (T2) weighted magnetic resonance imaging measurement. These alloy–Au hybrid nanocomposites can perform as multimodal imaging contrast agents for cancer cell diagnosis by two-photon fluorescent imaging and T2-weighted magnetic resonance imaging. In addition, their potential application for photothermal therapy was preliminarily investigated.

Abstract:We present the synthesis and application of a new type of dual magnetic and plasmonic nanostructures for magnetic-field-guided drug delivery and combined photothermal and photodynamic cancer therapy. Near-infrared-absorbing gold nanopopcorns containing a self-assembled iron oxide cluster core were prepared via a seed-mediated growth method. The hybrid nanostructures are superparamagnetic and show great photothermal conversion efficiency (η=61%) under near-infrared irradiation. Compact and stable nanocomplexes for photothermal-photodynamic therapy were formed by coating the nanoparticles with near-infrared-absorbing photosensitizer silicon 2,3-naphthalocyannie dihydroxide and stabilization with poly(ethylene glycol) linked with 11-mercaptoundecanoic acid. The nanocomplex showed enhanced release and cellular uptake of the photosensitizer with the use of a gradient magnetic field. In vitro studies using two different cell lines showed that the dual mode photothermal and photodynamic therapy with the assistance of magnetic-field-guided drug delivery dramatically improved the therapeutic efficacy of cancer cells as compared to the combination treatment without using a magnetic field and the two treatments alone. The "three-in-one" nanocomplex has the potential to carry therapeutic agents deep into a tumor through magnetic manipulation and to completely eradicate tumors by subsequent photothermal and photodynamic therapies without systemic toxicity.
