Abstract:Scintillators are essential for high-energy radiation detection in a variety of potential applications. However, due to complex fabrication processes and nanocrystal homogeneity, conventional scintillators are challenging to meet the need for cost-effective, environmentally friendly, and flexible X-ray detection. Here, monodisperse nanocrystals (NCs) with small grain size and colloidal stability are obtained by adjusting the doping concentration of Zn2+ ions and controlling the morphology uniformity of Cs3Cu2I5 NCs. The photoluminescence quantum yield (PLQY) for the optimal doping concentration is as high as 92.8%, which is a 28.5% improvement compared to nondoped NCs. Density functional theory calculations reveal that the Zn2+ dopant inclines to occupy Cu sites and the I-rich condition suppresses the formation of I vacancy, enriching the excited electron density at the band-edge to enhance the self-trapped exciton emission. Moreover, high luminescence performance and flexible X-ray scintillator films are prepared using Zn2+-doped Cs3Cu2I5 NCs, with a spatial resolution of up to 15.7 lp mm(-1). This work provides an effective strategy for the development of environmentally friendly, low-cost, and efficient blue-emitting 0D all-inorganic metal halides, as well as shows their great potential for high-performance flexible lead-free and low-toxicity X-ray detector applications.

Abstract:Bulk HgS itself has proven to be a technologically important material; however, the poor stability and weak emission of HgS nanocrystals have greatly hindered their promising applications. Presently, a critical problem is the uncontrollable growth of HgS NCs and their intrinsic surface states which are susceptible to the local environment. Here, we address the issue by an ion-tuning approach to fabricating stable, highly fluorescent Cd:HgS/CdS NCs for the first time, which efficiently tuned the band-gap level of HgS NCs, pushing their intrinsic states far away from the surface, reducing the strong interaction of the environment with surface states and hence drastically boosting the exciton transition. As compared to bare HgS NCs, the obtained Cd:HgS/CdS NCs exhibited tunable luminescence peaks from 724 to 825 nm with an unprecedentedly high quantum yield up to 40% at room temperature and excellent thermal and photostability. Characterized by TEM, XRD, XPS, and AAS, the resultant Cd:HgS/CdS NCs possessed a zinc-blende structure and was composed of a homogeneous alloyed HgCdS structure coated with a thin-layer CdS shell. The formation mechanism of Cd:HgS/CdS NCs was proposed. These bright, stable HgS-based NCs presented promising applications as fluorescent inks for anticounterfeiting and as excellent light converters when coated onto a blue-light-emitting diode.

Abstract:Near-infrared emitting AgInS2/ZnS nanocrystals were synthesized by carefully controlling the growth conditions in a Ag/In/Zn/S solution with less zinc relative to the other precursors. The role of having a smaller amount of zinc (8 atom %) was systematically investigated in an effort to understand the mechanism of the largely red-shifted emission. The AgInS2/ZnS nanocrystals can be transferred to aqueous solutions while retaining the emission intensity. The near-infrared emission and solubility in aqueous solutions make AgInS2/ZnS nanocrystals excellent candidates for bioimaging and photocatalytic applications.

Abstract:High quality InAs/InP/ZnSe core/shell/shell quantum dots have been grown by a one-pot approach. This engineered quantum dots with unique near-infrared (NIR) fluorescence, possessing outstanding optical properties, and the biocompatibility desired for in vivo applications. The resulting quantum dots have significantly lower intrinsic toxicity compared to NIR emissive dots containing elements such as cadmium, mercury, or lead. Also, these newly developed ultrasmall non-Cd containing and NIR-emitting quantum dots showed significantly improved circulation half-life and minimal reticuloendothelial system (RES) uptake.

Abstract:AbstractNovel β‐NaGdF4/Na(Gd,Yb)F4:Er/NaYF4:Yb/NaNdF4:Yb core/shell 1/shell 2/shell 3 (C/S1/S2/S3) multi‐shell nanocrystals (NCs) have been synthesized and used as probes for in vivo imaging. They can be excited by near‐infrared (800 nm) radiation and emit short‐wavelength infrared (SWIR, 1525 nm) radiation. Excitation at 800 nm falls into the “biological transparency window”, which features low absorption by water and low heat generation and is considered to be the ideal excitation wavelength with the least impact on biological tissues. After coating with phospholipids, the water‐soluble NCs showed good biocompatibility and low toxicity. With efficient SWIR emission at 1525 nm, the probe is detectable in tissues at depths of up to 18 mm with a low detection threshold concentration (5 nM for the stomach of nude mice and 100 nM for the stomach of SD rats). These results highlight the potential of the probe for the in vivo monitoring of areas that are otherwise difficult to analyze.

Abstract:Zinc oxide quantum dots (ZnO QDs) have gained wide attention due to their wide excitation spectrum, large Stokes shift, adjustable photoluminescence spectrum, and excellent biocompatibility. However, low fluorescence intensity and poor stability restrict their further applications. In this work, zinc sulfide (ZnS) as surface modifier, ZnO/ZnS core-shell QDs with type-I core-shell structure and particle size of 5 nm were prepared. The ultraviolet fluorescence has been greatly enhanced. The maximum fluorescence intensity of ZnO/ZnS core-shell QDs increases by 5288.6% compared with that of bare ZnO QDs. After being stored for three weeks, the fluorescence performance can be well retained. Furthermore, the cytotoxicity tests confirm the excellent biocompatibility of ZnO/ZnS core-shell QDs, demonstrating they are good candidates for cell imaging.

Abstract:A simple, rapid and green aqueous approach to near-infrared (NIR)-emitting β-HgS nanocrystals (NCs) was demonstrated for the first time by using glutathione (GSH) as the stabilizer at room temperature. The resulting HgS NCs with zinc blend structure exhibited strong quantum size effect, and the emission peak could be tuned in a wide NIR region from ca. 775 to 1041 nm. As compared with early achievements, the emission intensity of GSH-stabilized HgS NCs enhanced, with the maximum quantum yield reaching ~2.8%. It was also found that the stability of the GSH-HgS NCs was improved noticeably, the PL peak red-shifting only 9 nm and 23 nm after stored at 4°C for 4 months and 25°C for 7 days, respectively. The better stability of the HgS NCs was elucidated by FT-IR due to the multiple coordination of GSH molecule to surface Hg of the NCs. The emission range of GSH-stabilized HgS NCs was located between the visible region (500-800 nm) and IR region (1000-1600 nm) of HgS NCs as reported previously, extending the emission region of HgS nanomaterial. Therefore, the continuous emission from visible to IR spectral ranges provided HgS material more potential applications.

Abstract:This work reports the study of optimization of the reaction parameters on the synthesis of high quality CuInS2 and AgInS2 nanocrystals for bioimaging applications. The concentration of reaction precursors (e. g. Ag, Cu, In and S) plays a key role in determining the emission profile of these ternary quantum dots (QDs). By carefully varying the precursor compositions, the emission of QD can be tuned from red to near infrared (NIR) region. Taking the advantages of NIR emission, which possesses minimal absorption in biological tissues, we have also prepared water-dispersible CuInS2/ZnS and AgInS2/ZnS nanocrystals and demonstrated the high biocompatibility for both deep tissue penetration and tumor targeting. The QDs were stabilized in Pluronic F127 block copolymer micelles, offering us optically and colloidally stable contrast agents for in vitro and in vivo imaging. Two-photon excitation of QD has also been demonstrated, accomplishing a NIR-to-NIR transaction. This study devotes the key steps in promoting the use of ternary QDs as low-toxic, photostable, and cadmium-free semiconductor nanocrystal formulation for multiple imaging applications.
Constructing NIR Silica–cyanine Hybrid Nanocomposite for Bioimaging in Vivo: a Breakthrough in Photo-Stability and Bright Fluorescence with Large Stokes Shift

Abstract:Optical near-infrared (NIR) nanomaterials provide a unique opportunity for applications in bioimaging and medical diagnosis. A kind of hydrophilic NIR fluorescent core-shell structured silica nanoparticle containing NIR cyanine chromophore, named as CyN-12@NHs, for in vivo bioimaging is developed through a facile one-pot strategy. The hydrophobic CyN-12 molecules can be successfully encapsulated into the core via the self-assembly of the amphiphilic block copolymer PS-b-PAA and subsequent shell cross-linking of silane. The as-prepared CyN-12@NHs exhibits typically spherical core-shell structure, which has a uniform size of 35 nm with a narrow size distribution, and excellent dispersity in aqueous solution. Moreover, NIR absorption (690 nm) and bright fluorescence (800 nm) of CyN-12@NHs with a large Stokes shift (110 nm) in aqueous system make it an amenable high quality bioimaging contrast agent. The core-shell nanostructure significantly enhances the chemical and photo-stability of CyN-12 via the encapsulation, which possesses a 50-times longer half-life period than free CyN-12, along with a better resistance to reactive oxygen species (ROS). Furthermore, in living cell imaging, CyN-12@NHs shows nearly no cytotoxicity and is able to outline the HepG2 cells. The in vivo imaging on a tumor-bearing mouse model indicates that CyN-12@NHs selectively accumulates in the liver after intravenous injection, and has a long retention in tumor after intra-tumor injection without decrease in fluorescence activity. Overall, the excellent photo-properties of CyN-12@NHs could meet the intricate requirements for tumor imaging, such as high sensitivity, sufficient tissue penetration, and high spatial resolution. The strategy of the silica-cyanine hybrid nanoparticles paves a desirable and efficient route to fabricate highly hydrophilic NIR fluorescent contrast agents for tumor imaging and therapy, especially with a breakthrough in photo-stability, bright fluorescence as well as large Stokes shift.

Abstract:Near-infrared (NIR) photostimulated luminescence (PSL) nanocrystals (NCs) exhibit deep penetrability and low autofluorescence, recently evoking extensive interest for their application in biosensing. Nevertheless, most of the NIR PSL NCs encounter substantial constraints in biomedicine applications, primarily stemming from the necessity of high-power diode laser stimulation or their larger particle size. Herein, we design a new class of NIR light-emitting diode (LED) PSL NCs based on SrS:Eu 2+ NCs, which were coated with SrS shell to improve their stability and photoluminescence (PL) intensity. The trap depth and density of the NCs were further tailored through thermal annealing and Dy 3+ co-doping, resulting in efficient NIR PSL with a duration time longer than 2 h by stimulation with an NIR LED in a broad region within 700–1400 nm. Benefiting from their excellent NIR PSL, we showcased the potential of SrS:Eu 2+ ,Dy 3+ @SrS NCs as sensitive nanoprobes for tumor-targeted imaging in zebrafish with a signal-to-noise ratio of 18.9. These findings demonstrate the significant promise of the proposed SrS:Eu 2+ ,Dy 3+ @SrS nanoprobes for in vivo bioimaging, which may pave the way for the development of effective NIR PSL nanoprobes for diverse bioapplications in the future.

Bandgap and Structure Engineering via Cation Exchange: From Binary Ag2S to Ternary AgInS2, Quaternary AgZnInS alloy and AgZnInS/ZnS Core/Shell Fluorescent Nanocrystals for Bioimaging

Abstract:Attention on semiconductor nanocrystals have been largely focused because of their unique optical and electrical properties, which can be applied as light absorber and luminophore. However, the band gap and structure engineering of nanomaterials is not so easy because of their finite size. Here we demonstrate an approach for preparing ternary AgInS2 (AIS), quaternary AgZnInS (AZIS), AgInS2/ZnS and AgZnInS/ZnS nanocompounds based on cation exchange. First, pristine Ag2S quantum dots (QDs) with different sizes were synthesized in one-pot, followed by the partial cation exchange between In(3+) and Ag(+). Changing the initial ratio of In(3+) to Ag(+), reaction time and temperature can control the components of the obtained AIS QDs. Under the optimized conditions, AIS QDs were obtained for the first time with a cation disordered cubic phase and high photoluminescence (PL) quantum yield (QY) up to 32% in aqueous solution, demonstrating the great potential of cation exchange in the synthesis for nanocrystals with excellent optical properties. Sequentially, Zn(2+) ions were incorporated in situ through a second exchange of Zn(2+) to Ag(+)/In(3+), leading to distinct results under different reaction temperature. Addition of Zn(2+) precursor at room temperature produced AIS/ZnS core/shell NCs with successively enhancement of QY, while subsequent heating could obtain AZIS homogeneous alloy QDs with a successively blue-shift of PL emission. This allow us to tune the PL emission of the products from 483 to 675 nm and fabricate the chemically stable QDs core/ZnS shell structure. Based on the above results, a mechanism about the cation exchange for the ternary nanocrystals of different structures was proposed that the balance between cation exchange and diffusion is the key factor of controlling the band gap and structure of the final products. Furthermore, photostability and in vitro experiment demonstrated quite low cytotoxicity and remarkably promising applications in the field of clinical diagnosis.

Abstract:Significance In vivo fluorescence imaging in near IR-IIb window (1,500–1,700 nm) can provide high spatial and temporal resolution and deep tissue penetration for fundamental research and potential translations. Herein, a bright fluorescent probe emitting at ∼1,600 nm based on lead sulfide (PbS)/CdS quantum dots was developed. The CdS shell helped to chemically passivate and retain the high fluorescence of the PbS core after phase transfer to aqueous solutions for biocompatibility. The 1,600-nm emitting probe allowed noninvasive, millimeter-deep fluorescence imaging at high speeds up to 60 frames per second with micrometer-scale spatial resolution in 2D wide-field and 3D confocal modes. The probes were nontoxic and largely excreted over 1 month, providing a tool for in vivo research of preclinical animal models.
Facile synthesis of water-soluble Zn-doped AgIn5S8/ZnS core/shell fluorescent nanocrystals and their biological application

Abstract:Here we demonstrate a novel and facile strategy of highly luminescent water-soluble Zn-doped AgIn5S8 (ZAIS) nanocrystals and ZAIS/ZnS core/shell structures, which were based on hydrothermal reaction between the acetate salts of the corresponding metals and sulfide precursor in the presence of l-cysteine at 110 °C in a Teflon-lined autoclave. The photoluminescent (PL) emission wavelength can be conveniently tuned from 560 to 650 nm by tailoring the stoichiometric ratio of [Ag]/[Zn]. The as prepared nanocrystals were characterized systematically and exhibit long PL lifetimes more than 100 ns. The influence of experimental conditions, including concentration of l-cysteine and reaction temperature, was investigated. In addition, we performed a coating procedure with the ZnS shell outside the ZAIS core and showed excellent PL quantum yields up to 35%. The in vitro experiment exhibited quite low cytotoxicity and marvelous biocompatibility, revealing their promising prospect in bioscience. Furthermore, the obtained ZAIS/ZnS nanocompounds (NCs) were covalently conjugated to alpha-fetoprotein antibodies and targeted fluorescent imaging for hepatocellular carcinoma cells was realized.

Abstract:In this paper, we report a facile method to synthesize high quality CdS: Eu nanocrystals (NCs) and CdS: Eu/ZnS NCs with strong photoluminescence (PL). The influence of various experimental variables including the concentration of Eu3+ ions, the reaction time and the reaction temperature were investigated systematically. In addition, the PL properties of CdS: Eu NCs exhibited pH sensitive. Under the acid condition, pH value of the CdS: Eu NCs solution played an important role in determining PL emission intensity. However, under the alkaline condition, the obtained CdS: Eu NCs exhibited a tunable PL emission wavelength (from 490 nm to 610 nm) when pH value was adjusted from pH 7 to 10. After coating with ZnS shell, the CdS: Eu/ZnS NCs showed enhanced PL intensity compare with one of the CdS: Eu NCs. The CdS: Eu NCs and CdS: Eu/ZnS NCs were characterized by X-ray diffraction (XRD) and high-resolution transmission electron microscopy (HRTEM). In addition, the biocompatibility of these NCs was measured by hemolytic test, which indicated that CdS: Eu/ZnS NCs were more biocompatible than CdS: Eu NCs at the same conditions. It can be expected that CdS: Eu/ZnS NCs are promising biolabeling materials.
Visible and Near-Infrared Dual-Emission Carbogenic Small Molecular Complex with High RNA Selectivity and Renal Clearance for Nucleolus and Tumor Imaging

Abstract:Fluorescence imaging requires bioselective, sensitive, nontoxic molecular probes to detect the precise location of lesions for fundamental research and clinical applications. Typical inorganic semiconductor nanomaterials with large sizes (>10 nm) can offer high-quality fluorescence imaging due to their fascinating optical properties but are limited to low selectivity as well as slow clearance pathway. We here report an N- and O-rich carbogenic small molecular complex (SMC, MW < 1000 Da) that exhibits high quantum yield (up to 80%), nucleic acid-binding enhanced excitation-dependent fluorescence (EDF), and a near-infrared (NIR) emission peaked at 850 nm with an ultralarge Stokes shift (∼500 nm). SMCs show strong rRNA affinity, and the resulting EDF enhancement allows multicolor visualization of nucleoli in cells for clear statistics. Furthermore, SMCs can be efficiently accumulated in tumor in vivo after injection into tumor-bearing mice. The NIR emission affords high signal/noise ratio imaging for delineating the true extent of tumor. Importantly, about 80% of injected SMCs can be rapidly excreted from the body in 24 h. No appreciable toxicological responses were observed up to 30 days by hematological, biochemical, and pathological examinations. SMCs have great potential as a promising nucleolus- and tumor-specific agent for medical diagnoses and biomedical research.

In particular, luminescence imaging in the tissue-transparent near-infrared (NIR, 700-1700 nm) region offers tremendous potential for visualizing biological architectures and pathophysiological events in living subjects with deep tissue penetration and high imaging contrast owing to the reduced light-tissue interactions of absorption, scattering, and autofluorescence. The distinctive quantum effects of nanocrystals have been harnessed to achieve exceptional photophysical properties, establishing them as a promising category of luminescent probes. In this comprehensive review, the interactions between light and biological tissues, as well as the advantages of NIR light for in vivo luminescence imaging, are initially elaborated. Subsequently, we focus on achieving deep tissue penetration and improved imaging contrast by optimizing the performance of nanocrystal fluorophores. The ingenious design strategies of NIR nanocrystal probes are discussed, along with their respective biomedical applications in versatile in vivo luminescence imaging modalities. Finally, thought-provoking reflections on the challenges and prospects for future clinical translation of nanocrystal-based in vivo luminescence imaging in the NIR region are wisely provided.
'Green'-Synthesized Near-Infrared PbS Quantum Dots with Silica-Peg Dual-Layer Coating: Ultrastable and Biocompatible Optical Probes for in Vivo Animal Imaging.

Abstract:In this paper, PbS semiconductor quantum dots (QDs) with near-infrared (NIR) photoluminescence were synthesized in oleic acid and paraffin liquid mixture by using an easily handled and 'green' approach. Surface functionalization of the QDs was accomplished with a silica and polyethylene glycol (PEG) phospholipid dual-layer coating and the excellent chemical stability of the nanoparticles is demonstrated. We then successfully applied the ultrastable PbS QDs to in vivo sentinel lymph node (SLN) mapping of mice. Histological analyses were also carried out to ensure that the intravenously injected nanoparticles did not produce any toxicity to the organism of mice. These experimental results suggested that our ultrastable NIR PbS QDs can serve as biocompatible and efficient probes for in vivo optical bioimaging and has great potentials for disease diagnosis and clinical therapies in the future.

Abstract:Biological applications of core/shell near-infrared quantum dots (QDs) have attracted broad interest due to their unique optical and chemical properties. Additionally, the use of multifunctional nanomaterials with near-infrared QDs and plasmonic functional nanoparticles are promising for applications in electronics, bioimaging, energy, and environmental-related studies. In this work, we experimentally demonstrate how to construct a multifunctional nanoparticle comprised of CdSe/ZnS QDs and gold nanorods (GNRs) where the GNRs were applied to enhance the photoluminescence (PL) of the CdSe/ZnS QDs. In particular, we have obtained the scattering PL spectrum of a single CdSe/ZnS QD and GNR@CdSe/ZnS nanoparticle and comparison results show that the CdSe/ZnS QDs have an apparent PL enhancement of four-times after binding with GNRs. In addition, in vitro experimental results show that the biostability of the GNR@CdSe/ZnS nanoparticles can be improved by using folic acid. A bioimaging study has also been performed where GNR@CdSe/ZnS nanoparticles were used as an optical process for MCF-7 breast cancer cells.
