Charge-Reversal Polymers for Biodelivery

Abstract:Cationic polymer-based biodelivery has attracted more and more attention in recent years because of their advantages including facilitated cellular internalization, efficient endo/lysosomal escape, and nuclear targeting. However, direct applications of cationic polymers encounter numerous problems such as cell damage, nonspecific interactions with blood components, and rapid clearance from the bloodstream. Therefore, strategies to mask the amino groups in cationic polymers are widely under investigation. Among them, acid-labile amidized cationic polymers have emerged as an effective approach, making the polymers inert in the bloodstream but activated to recover such abilities as cellular adsorptive and endo/lysosome-lysing ability in the target sites by responding to the pH gradients in the tumoral interstitium or endo/lysosomes. Herein, we summarize and discuss the chemical mechanisms of the acid-labile amidized cationic polymers and their applications in biodelivery.
