Abstract:The hybrid inorganic/organic gel with immobilized papain enzyme in micro/nano size was prepared by absorption-flocculation co-immobilization method, in which polyacrylamide (PAM) was used as the flocculation agent, and characterized by scanning electron microscopy (SEM), energy dispersive spectrometer mapping (EDS), and size analyzer. Evolution of surface Zeta potential (ZP) of nano-gel containing enzyme with pH revealed that the main interaction between the enzyme and immobilization support was electrostatic attraction. The electrostatic field generated by the surface charges exhibited major impact on the enzyme loading and particle size distribution. At pH=7.0 and mass ratio of TiO2 to PA=7.5 : 1, the maximum amount of enzyme loading was achieved as 121.84 mg/g, corresponding to the efficiency of 91.38%. The size of nano-gel with immobilized enzyme was about 0.444 mu m and its distribution was very narrow in either low or high pH range. SEM/EDS results showed that immobilized papain was homogeneously dispersed in the porous hybrid gel. The stability test showed that the relative activity of immobilized papain still remained more than 50% even after five times cycle of reuse. It indicates that this novel method of immobilization could be propitious to the well dispersion, high stability and loading amount of papain.

Abstract:Papain was immobilized on the mesoporous molecular sieve SBA-16 by physical adsorption method.The effects of immobilization conditions on the relative activity of the papain were studied.At the same time,the pH stability of the free enzyme and immobilized enzyme was discussed at different pH values.Experimental results show that the optimum conditions for immobilization are as follows: concentration of the free enzyme,time,and pH are 30 mg/g,2.5 h and 7.0,respectively.Compared with the free enzyme,pH-stability of immobilized papain has also been improved.

Abstract:Freezing-vacuum adsorption was employed in the immobilization of enzyme in mesoporous material SBA-15.The results indicate that this method can guarantee a large amount of loading about 450 mg/g(support,) which is incomparable for other means.As long as vacuum degree is properly controlled,the(enzymatic) activity can be retained.This method provides a new solution for the immobilization of enzyme.

Abstract:Take the macroporous resin as carrier,the papain enzyme was immobilized using the glutaraldehyde as crosslinking agent,the influence of various carrier and preparation conditions on couple yield and relative activity of immobilized enzyme was studied,and the effect of temperature and pH on free and immobilized enzyme stability was also examined.As a result,the maximum relative activity of immobilized enzyme was obtained when enzyme amount was 40mg,immobilized temperature was 77℃,immobilized time was 4hand pH=6.5.The experiment result showed that the immobilized papain had better temperature and pH stability than free papain.

Abstract:Graphene oxide nanosheets was prepared, functionlized with 3-aminopropyltriethoxysilane and used as the carrier for immobilization of papain. A high immobilization yield and efficiency higher than 80% were achieved at pH 8.0 and 35 °C. The thermal stability of the immobilized papain was improved compared with the free enzyme. And the immobilized papain retained about 63% of its initial activity after 30 days storage at 4 °C, while free enzyme was only 41% at the same conditions. The immobilized enzyme displayed higher binding affinity to substrate because of the unique properties of GO. These results indicate that modified graphene oxide nanosheets has a good potential as carrier for immobilization of proteins, especially enzymes.

Abstract:Carboxyl-modified mesoporous materials have been demonstrated as efficient carriers for immobilizing enzymes. However, little attention has been paid to the effect of H2SO4 treatment, a key in the synthesis of carboxyl-modified carriers, on the immobilization. To examine the H2SO4 treatment’s influence, we contrasted the ability of immobilizing papain between SBA-15 and the one after H2SO4 treatment (H2SO4-treated SBA-15). Meanwhile, the contribution of the as-synthesized carboxyl to the immobilization was also rechecked by comparing carboxyl-modified SBA-15 (COOH-SBA-15) and H2SO4-treated SBA-15. In relation to the experiments, the H2SO4 treatment increases the immobilizing ability of SBA-15, especially when the immobilization performs near the pI of papain. Moreover, the as-synthesized carboxyl facilitates the immobilization by means of strengthening the hydrophobic dehydration at the interface, instead of merely through electrostatic forces. Additionally, the carboxyl makes a large contribution to inducing the favorable conformation change of papain, improving the specific activity, catalytic efficiency, and binding affinity of the immobilized enzyme when combined with the substrate.

Abstract:Papain was purified from spray-dried Carica papaya latex using aqueous two-phase system (ATPS). Then it was recovered from PEG phase by in situ immobilization or preparing cross-linked enzyme aggregates (CLEAs). The Plackett-Burman design and the central composite design (CCD) together with the response surface methodology (RSM) were used to optimize the APTS processes. The highly purified papain (96-100%) was achieved under the optimized conditions: 40% (w/w) 15 mg/ml enzyme solution, 14.33-17.65% (w/w) PEG 6000, 14.27-14.42% (w/w) NaH2PO4/K2HPO4 and pH 5.77-6.30 at 20°C. An in situ enzyme immobilization approach, carried out by directly dispersing aminated supports and chitosan beads into the PEG phase, was investigated to recover papain, in which a high immobilization yield (>90%) and activity recovery (>40%) was obtained. Moreover, CLEAs were successfully used in recovering papain from PEG phase with a hydrolytic activity hundreds times higher than the carrier-bound immobilized papain.

Abstract:Immobilized papain was prepared using microcrystalline cellulos as support which was activated by two-step method.The microcrystalline cellulose was first activated with p-toluenesulfonyl chloride, followed by coupling with ethyl diamine.After treated with glutaraldehyde, papain was immobilized to the support.Immobilized papain was obtained with activity 103 U/g support and high stability.

Abstract:Hydrogel composites based on pineapple peel carboxymethyl cellulose, polyvinyl alcohol and mesoporous silica SBA-15 were synthesized by an eco-friendly method of repeated freeze-thaw cycles for the application of papain immobilization. The experiment was optimized to obtain an efficient papain immobilization carrier. Simultaneously the immobilization conditions, including enzyme concentration, pH, crosslinker concentration and cross-linking time were optimized. The immobilized papain had maximum activity at low reaction temperature of 40°C and showed pH-sensitivity by exhibiting a rapid decrease of activity within a narrow range from pH 7.0 to pH 7.5. Compared with the free papain, the immobilized papain revealed enhanced pH, thermal and storage stability. After 2h incubation at 80°C, the immobilized papain retained 56% of its initial activity while the free papain only retained 16%. After 10days of storage, 79% of the initial activity was retained for the immobilized papain while only 27% for the free papain.

Abstract:Magnetic polystyrene(PS) microspheres were prepared by layer-by-layer self-assembly technique.The core-shell microspheres were constructed by alternately depositing negative and positive polyelectrolytes onto sulfate-stabilized PS latex particles.And then,papain was immobilized on them.The best circumstance of enzyme immobilization was determined:pH=6.0,1.0 h,30 ℃,mass ratio(papain∶microspheres) 37.98%.And the largest ratio of enzyme immobilization could reach 93.0%.The properties of the immobilized enzyme were also studied.The best application temperature and pH were 50 ℃ and 8.0,respectively.When used for 3 times or reserved for 1.5~2.0 h at 80 ℃,the activity of the immobilized papain would fall by nearly half.

Abstract:Lipase from Candida Ragusa (CRL) was successfully covalently immobilized on fibrous silica nanoparticles KCC-1, and the properties of immobilized enzyme were investigated. Mesoporous fibrous silica nanoparticles (MSFIPs) were synthesized with particles size 200 nm pore size 15-30 nm: followed by amino-functionalization. Scanning Electron Microscopy (SEM), Transmittance Electronic microscopy (TEM), Fourier Transform Infrared Spectroscopy (FT-IR) and N-2 adsorption were used for the characterization of nanoparticles. Further SiO2@NH2 nanoparticles were activated by glutaraldehyde as a bifunctional cross linker, and were used for lipase immobilization. The applied approach for support preparation, activation, and optimization of immobilization conditions, led to better resistance to temperature and pH inactivation in comparison to the free lipase, and hence widened the reaction pH and temperature regions, with the optimum pH and temperature of 7.5 and 40 degrees C, respectively. The immobilized Lipase Candida Ragusa (ICRL) maintained above 81% of the initial activity after 28 days and 80% activity after 8 repeated cycles. Thus ICRL showed improved storage stability reusability and 700 U/g of protein as immobilization efficiency. (C) 2016 Elsevier B.V. All rights reserved.

Abstract:Mesoporous materials, especially functionalized ones, have become a promising carrier for enzyme immobilization. We synthesized room temperature ionic liquid-decorated mesoporous SBA-15 (RTIL-SBA-15) for papain immobilization. The results of powder XRD, IR and N2 adsorption–desorption isotherms have confirmed that ionic liquid [Simim+][Cl−] was successfully grafted on the surface of SBA-15. As a consequence of the electrostatic attraction between the cation [Simim+] and the negatively charged papain, RTIL-SBA-15 had an advantage over SBA-15 when papain was immobilized at pH=9.00. The kinetic study showed that the interaction between papain and the carrier was stronger after ionic liquid modification. In the casein hydrolysis, the papain immobilized on RTIL-SBA-15 showed a higher specific activity than that on SBA-15, implying that the ionic liquid [Simim+][Cl−] was beneficial to improve the activity of the immobilized papain. The optimum pH of the immobilized papain was shifted to higher than that of free enzyme.

Abstract:A core/sheath nanofiber membrane with poly[bis(p-methylphenoxy)]phospha-zene (PMPPh) as the sheath and easily spinnable polyacrylonitrile (PAN) as the core was prepared via a coaxial electrospinning process. Field-emission scanning electron microscopy and transmission electron microscopy were used to characterize the morphology of the nanofiber membrane. It was found that the concentration of the PAN spinning solution and the ratio of the core/sheath solution flow rates played a decisive role in the coaxial electrospinning process. In addition, the stabilized core/sheath PMPPh nanofiber membrane was investigated as a support for enzyme immobilization because of its excellent biocompatibility, high surface/volume ratio, and large porosity. Lipase from Candida rugosa was immobilized on the nanofiber membrane by adsorption. The properties of the immobilized lipase on the polyphosphazene nanofiber membrane were studied and compared with those of a PAN nanofiber membrane. The results showed that the adsorption capacity (20.4 ± 2.7 mg/g) and activity retention (63.7%) of the immobilized lipase on the polyphosphazene nanofiber membrane were higher than those on the PAN membrane.

Abstract:Single enzyme nanoparticles of α-chymotrypsin (SEN-CT), in which each CT molecule is surrounded by a thin polymeric organic/inorganic network, stabilized the CT activity in a shaking condition as well as in a non-shaking condition. Since SEN-CT is soluble in a buffer solution and less than 10nm in size, SEN-CT could be further immobilized in nanoporous silica with an average pore size of 29nm. Free CT and SEN-CT were immobilized in nanoporous silica (NPS), and nanoporous silica that was first silanized with aminopropyltriethoxysilane (amino-NPS) to generate a positive surface charge. The SEN-CT adsorbed in amino-NPS was more stable than CT immobilized by either adsorption in NPS or covalent bonding to amino-NPS. In shaking conditions, nanoporous silica provided an additional stabilization by protecting SEN-CT from shear stresses. At 22°C with harsh shaking, free, NPS-adsorbed and NPS-covalently attached CT showed half lives of 1, 62, and 80h, respectively; whereas SEN-CT adsorbed in amino-NPS showed no activity loss within 12 days. The combination of SENs and nanoporous silica, which makes an active and stable immobilized enzyme system, represents a new structure for biocatalytic applications.

Abstract:Mesoporous materials have recently gained much attention owing to their large surface area, narrow pore size distribution, and superior pore structure. These materials have been demonstrated as excellent solid supports for immobilization of a variety of proteins and enzymes for their potential applications as biocatalysts in the chemical and pharmaceutical industries. However, the lack of efficient and reproducible methods for immobilization has limited the activity and recyclability of these biocatalysts. Furthermore, the biocatalysts are usually not robust owing to their rapid denaturation in bulk solvents. To solve these problems, we designed a novel hybrid material system, mesoporous silica immobilized with NiO nanoparticles (SBA-NiO), wherein enzyme immobilization is directed to specific sites on the pore surface of the material. This yielded the biocatalytic species with higher activity than free enzyme in solution. These biocatalytic species are recyclable with minimal loss of activity after several cycles, demonstrating an advantage over free enzymes.

Abstract:To investigate the effects of surface property of mesoporous supports on the lipase immobilization and the performance of immobilized lipase, the mesoporous molecular sieve SBA-15 is functionalized with three organic moieties, dimethyl (DM), diisopropyl (DIP), and diisobutyl (DIB), respectively, by post-synthesis grafting and one-pot synthesis methods. Porcine pancreas lipase (PPL) is immobilized oil SBA-15 supports through hydrogen bonding and hydrophobic interaction. The hydrophobic adsorption involves no active sites of PPL, and neither hyper-activation nor total inactivation occurs. The study on the intrinsic stability of PPL, including thermal stability, pH stability, and storage stability, indicates that the entrapment in mesoporous supports, and especially in organic-functionalized supports, makes PPL more resistant to temperature increment but more sensitive to pH change. The reusability investigation shows that the organic modification of mesoporous surface inhibits the enzyme leaching to some extent, resulting in a better operational stability.
