Abstract:Nanoparticles are appealing drug delivery systems since they promise prolonged circulation time and predictable release behaviors. The current work reports a novel hyperbranched amphiphilic block copolymer synthesized using deactivation-enhanced atom transfer radical polymerization (DE-ATRP) for smart drug delivery. PEG2000-Br was applied as a macroinitiator to initiate acid-cleavable divinyl (ACD) monomers linked by acetal groups, formulating the hydrophilic (PEG) and hydrophobic (ACD) segments. The polymer appeared to self-assemble into micelles with diameters in the range of 70-100 nm, and was examined for the controlled release of doxorubicin (DOX). Results showed that DOX-loaded micelles (similar to 90 nm) could achieve drug loading as high as 8.2 wt%, with interesting pH-dependent release behaviors. Studies with flow cytometry (FCM) and confocal laser scanning microscopy (CLSM) showed that DOX-loaded micelles exhibited a high cellular uptake performance by HeLa cells, which indicated promising antitumor efficacy for such a drug delivery system. Additionally, such DOX-loaded micelles exhibited remarkable cytotoxicity against HeLa cells in a dose-and time-dependent manner due to the enhanced cell uptake behavior of micelles. These results indicated that the polymeric micelles might be used as a promising candidate for a pH-responsive drug delivery for cancer therapy.

Abstract:The synthesis of a new kind of amphiphilic poly(vinyl-co-ester)s via simple one-pot radical ring-opening polymerization (RROP) is reported. A series of biodegradable, biocompatible, and cross-linkable copolymers P(MDO-co-PEGMA-co-CMA) with different compositions were synthesized by copolymerization of 2-methylene-1,3-dioxepane (MDO), poly(ethylene glycol) methyl ether methacrylate (PEGMA) and 7-(2-methacryloyloxyethoxy)-4-methylcoumarin methacrylate (CMA). All the copolymers were well characterized by H-1 NMR, C-13 NMR, and gel permeation chromatography (GPC). The hydrolytic degradation and enzymatic degradation of the copolymers were investigated. The amphiphilic P(MDO-co-PEGMA-co-CMA) can self-assemble into micelles in aqueous solution. Polymeric micelles with different sizes were obtained through tuning the compositions of the copolymers, i.e., the hydrophilic/hydrophobic ratio. The size and morphology of the micelles were measured by dynamic light scattering (DLS) and transmission electron microscopy (TEM). Because of the coumarin side groups in the micellar core, the micelles can be cross-linked upon exposure to long-wavelength UV light (lambda = 365 nm). These micelles also showed excellent biocompatibility by the in vitro cytotoxicity experiments. Furthermore, the biodegradable micelles were applied for the delivery of an anticancer drug doxorubicin (DOX). The cell viability of DOX-loading micelles compared to free DOX was discussed. The effects of cross-linking and the copolymer composition on the drug release behavior were well studied. The results indicate that this new kind of amphiphilic poly(vinyl-co-ester) s could serve as promising nanocarriers for the delivery of anticancer drugs.
Synthesis of Branched Cyclic Copolymer for pH-responsive Drug Delivery System

Abstract:A branched cyclic amphiphilic block copolymer ( PEG-b-PACDs ) was synthesized via a deactivation-enhanced atom transfer radical polymerization ( DE-ATRP) with brominated polyethylene glycol (PEG2KBr) and di(2-acry-loyloxy ethoxy)-[4-methoxy-phenyl]methane (ACD) as initiator and hydrophobic monomer, respectively. And then its potential application for a drug delivery system was explored. The structure of copolymer was characterized by means of hydrogen spectrum nuclear magnetic resonance ( 1 HNMR ) and gel permeation chromatography ( GPC ) . The particle size and morphology of the micelles were measured by dynamic light scattering ( DLS ) and transmission scanning microscope ( TEM ) . The results showed that the resulting copolymer exhibited a branched cyclic structure. The blank micelles were spherical in shape with a particle size of 70 nm, while the particle size of doxorubicin ( DOX)-loaded micelles were about 90 nm. The encapsulation efficiencies of DOX-loaded micelles were 48. 7%. The DOX-loaded PEG-b-PACDs shows obvious pH-responsive drug release property under acidic condition.
Synthesis and pH-responsive self-assembly behavior of a fluorescent amphiphilic triblock copolymer mPEG-b-PCL-b-PDMAEMA-g-PC for the controlled intracellular delivery of doxorubicin

Abstract:In this work, well-defined pH-responsive methoxy poly(ethylene glycol)-block-poly(e-caprolactone)-block-poly[2-(dimethylamino)ethyl methacrylate]-g-7-propinyloxy coumarin triblock amphiphilic copolymers (mPEG-b-PCL-b-PDMAEMA-g-PC) were synthesized using a combination of atom transfer radical polymerization (ATRP), ring opening polymerization (ROP) and click chemistry. The chemical structures and compositions of these copolymers were characterized using Fourier transform infrared spectroscopy (FT-IR) and proton nuclear magnetic resonance (1H NMR). The molecular weights of the copolymers were obtained using 1H NMR spectroscopy and gel permeation chromatography (GPC) measurements. Subsequently, the polymers could self-assemble into micelles which were investigated using dynamic light scattering (DLS), transmission electron microscopy (TEM), and fluorescence spectroscopy. The pH-responsive self-assembly behavior of these triblock copolymers in water were investigated at different pH values of 5 and 7.4 for controlled doxorubicin release, the results indicated that the release rate of DOX could be effectively controlled by altering the pH, DOX was sealed in neutral surroundings and DOX release was triggered in acidic surroundings. CCK-8 assays and confocal laser scanning microscopy (CLSM) against HeLa cells indicated that the micelles had no associated cytotoxicity, possessed good biodegradability and biocompatibility, and identified the location of the DOX in HeLa cells. The DOX-loaded micelles possessed high cytotoxicity to HeLa cells and exhibited inhibition of the proliferation of HeLa cells. Moreover, these flexible micelles with an on–off switched drug release may offer a promising pattern to deliver a wide variety of hydrophobic payloads to tumor cells for cancer therapy.

Abstract:In this work, well-defined amphiphilic polyhedral oligomeric silsesquioxane (POSS) star-shaped inorganic/organic hybrid block copolymers with poly(epsilon-caprolactone)-poly(2-(dimethylamino)ethyl methacrylate)-co-poly(ethylene glycol) methacrylate (POSS-PCL-P(DMAEMA-co-PEGMA))(16) were synthesized with different PCL segments via thiol-ene click reaction, ring opening polymerization (ROP) and atom transfer radical polymerization (ATRP), which were confirmed by Fourier transform infrared spectroscopy (FT-IR), proton nuclear magnetic resonance (H-1 NMR), gel permeation chromatography (GPC), X-ray photoelectron spectroscopy (XPS) and thermogravimetric analysis (TGA). Subsequently, the polymers could self-assemble into micelles in aqueous solution, which were investigated by dynamic light scattering (DLS), ultraviolet-visible spectroscopy (UV-vis) and transmission electron microscopy (TEM). The pH-responsive self-assembly behavior of these triblock copolymers in water was investigated at different pH values of 5.0 and 7.4 for controlled doxorubicin release; the results indicated that the release rate of DOX could be effectively controlled by altering the pH and the release of drug loading efficiency (DLE) was up to 82% (w/w). Furthermore, CCK-8 assays and confocal laser scanning microscopy (CLSM) against HeLa cells indicated that the micelles had no associated cytotoxicity, possessed good biodegradability and biocompatibility, and identified the location of the DOX in HeLa cells. The DOX-loaded micelles could easily enter the cells and produce the desired pharmacological action, and minimize the side effects of free DOX. Moreover, these flexible micelles with on-off switched drug release may offer a promising method to deliver a wide variety of hydrophobic payloads to tumor cells for cancer therapy.

Abstract:Star-shaped polymeric materials provide very high efficiency toward various engineering and biomedical applications. Due to the absence of straightforward and versatile synthetic protocols, the synthesis of sequence-defined star-shaped (co)polymers has remained a major challenge. Here, a facile approach is developed that allows synthesis of a series of unprecedented discrete, multifunctional four-, six-, and eight-arm star-shaped complex macromolecular architectures based on a well-defined triple (thermo/pH/light)-stimuli-responsive poly(N-isopropylacrylamide)-block-poly(methacrylic acid)-umbelliferone (PNIPAM-b-PMAA)n-UMB diblock copolymer, based on temperature responsive PNIPAM segment, pH-responsive PMAA segment, and photoresponsive UMB end groups. Thus, developed star-shaped copolymers self-assemble in water to form spherical nanoaggregates of diameter 90 ± 20 nm, as measured by FESEM. The star-shaped copolymer's response to external stimuli has been assessed against changes in temperature, pH, and light irradiation. The star-shaped copolymer was employed as a nanocarrier for pH responsive release of an anticancer drug, doxorubicin. This study opens up new avenues for efficient star-shaped macromolecular architecture construction for engineering and biomedical applications.
A Convenient Scheme for Synthesizing Reduction‐sensitive Chitosan‐based Amphiphilic Copolymers for Drug Delivery

Abstract:A novel type of reduction-sensitive graft copolymers, chitosan-S-S-poly(e-caprolactone) (CS-S-S-PCL, here -S-S- means PCL was conjugated onto chitosan backbone through disulfide linkage), was synthesized through a convenient route using dithiodipropionic anhydride (DTDPA) as a disulfide donor. Reaction of hydroxy-terminated poly(e-caprolactone) (PCL) with DTDPA quantitatively yielded DTDPA functionalized PCL (PCL-S-S-COOH). The disulfide-containing polyester was regioselectively conjugated onto the hydroxy groups of chitosan under mild and homogeneous conditions, utilizing dodecyl sulfate-chitosan complexes (SCC) as an intermediate. The self-assembly and Doxorubicin (Dox) release behavior of the copolymers were investigated. Spherical micelles could be formed through self-assembly of CS-S-S-PCL in aqueous media. The reduction-sensitive behavior of CS-S-S-PCL micelles was investigated by using Dithiothreitol (DTT) as a reductive reagent. In the presence of 10 mM DTT, the micelles gradually lost their aggregation stability and were precipitated out after four days. ANTI-TUMOR DRUG DELIVERY OF A Ph-Sensitive POLY( ASPARTIC ACID)-CONTAINING BLOCK COPOLYMER

Abstract:A novel biodegradable ABC type triblock copolymer was synthesized by N-carboxyl anhydride ring-opening polymerization. Triblock copolymer mPEG-poly (asparthyl hydrazide) -PLAla (mPEG-PAHy-PLAla) was synthesized by hydrazine aminolysis. DOX was conjugated to the polymer through a pH-sensitive hydrazone bond. High drug-loaded polymeric micelles were obtained via the pi-pi interaction between conjugated and free DOX molecules. FTIR, dynamic light scattering, UV and TEM were used in the characterization and measurements. It was found that this polymer/drug complex could form nano-scaled core-shell-corona trilayer particles in aqueous solution. PLAla, PAHy (DOX) and PEG blocks served as the hydrophobic core, pH-sensitive shell and hydrophilic corona, respectively. The drug release experiments displayed a pH-dependent behavior that DOX release rate increased significantly as the environmental pH value dropped from physiologically 7. 4 to acidic values resulted from the cleavage of hydrazone bonds and the increasing of hydrophilicity of PAHy segements at lower pH values. This new platform represents a promising candidate in the formulation of targeted drug delivery systems to the low pH tumor tissues.

Abstract:A series of acid cleavable linkers based on tetrahydropyran/tetrahydrofuran (THP/THF) ether were synthesized, in which both of the two terminals are active group which is easy to react with other compounds. The structures for these linkers 1a and 1b were confirmed by H-1 NMR, C-13 NMR, HRMS, and IR. Model compound containing THP linkage was synthesized. Furthermore a pH-responsive amphiphilic diblock copolymer containing tetrahydrofuran (THF) linkage in which hydrophilic segment was poly(ethylene glycol) (PEG) and hydrophobic segment was hydrophobic polylactic acid (PLA) was synthesized and characterized by 1H NMR, IR and gel permeation chromatography (GPC). The size, stability and critical micelle concentration (CMC) of diblock copolymer micelle were also evaluated. The preliminary experimental results show that the acid sensitive amphiphilic diblock copolymer micelle is a potential alternative for future drug delivery.

Abstract:A series of amphiphilic terpolymers with miktoarm star and triblock architectures of poly(ethylene glycol) (PEG), poly(ε-caprolactone) (PCL) and poly(l-lactide acid) (PLLA) or poly(DL-lactide acid) (PDLLA) terpolymers were synthesized as carriers for drug delivery. The architecture, molecular weight and crystallization behavior of the terpolymers were characterized. Anticancer drug doxorubicin was encapsulated in the micelles to investigate their drug loading properties. The miktoarm star terpolymers exhibited stronger crystallization capability, smaller size and better stability than that of triblock polymeric micelle, owing to the lower CMC values of miktoarm star polymeric micelle. Furthermore, the drug-loaded miktoarm star polymeric micelles showed the cumulative DOX release account of the micelles with PDLLA blocks was 65.3% while the release account of the corresponding micelles containing PLLA blocks was 45.2%. The IC50 values of drug-loaded miktoarm star polymeric micelle were lower than triblock polymeric micelle. Meanwhile, Confocal laser scanning microscopy (CLSM) and Flow Cytometry results demonstrated that the miktoarm star micelles were more favorable for cellular internalization. The miktoarm star micelles with PDLLA blocks were promising carriers for anticancer drug delivery.
One-pot self-assembly directed fabrication of biocompatible core cross-linked polymeric micelles as a drug delivery system

Abstract:A series of pH-responsive diblock copolymers, methoxy poly(ethylene glycol)-poly(2-(diethylamino)ethyl methacrylate) (mPEG-PDEA), were synthesized via the atom transfer radical polymerization (ATRP) technique based on the poly(ethylene glycol) (PEG)-macroinitiator. The obtained diblock copolymers exhibit interesting micellization behavior in aqueous solution, and self-assemble into core-shell micelles by adjusting the solution pH at ambient temperature. In both cases, transmittance and transmission electron microscopy (TEM) reveal the presence of the near-monodispersed micelles. Novel core cross-linked (CCL) micelles with pH-responsive PDEA cores and biocompatible PEG coronas were fabricated with a one-pot self-assembly directed synthesis approach with N,N'-methylene bisacrylamide as the cross-linker in methanol at 60 degrees C. The reversible pH-dependent swelling-shrinking of the PDEA cores of the obtained CCL micelles was investigated in detail by DLS and TEM. As the structurally stable pH-induced CCL micelles have been exploited to develop a solvent-free protocol for drug loading, the release profile of model hydrophobic drugs, dipyridamole (DIP), ibuprofen and famotidine, initially loaded within the hydrophobic PDEA core via hydrogen bonding, can be controlled by the solution pH. Experiments on controlled release demonstrated that the pH-responsive CCL micelles have a potential application in fields such as drug delivery and smart release.

Abstract:A series of amphiphilic 4- and 6-armed star triblock copolymers based on poly( ɛ -caprolactone) (PCL), poly(2-(diethylamino)ethyl methacrylate) (PDEAEMA), and poly(poly(ethylene glycol) methyl ether methacrylate) (PPEGMA) were designed and synthesized by a combination of ring opening polymerization (ROP) and continuous activators regenerated by electron transfer atom transfer radical polymerization (ARGET ATRP). The continuous ARGET ATRP of DEAEMA and PEGMA was in situ monitored by react infrared spectroscopy (ReactIR) and showed good first-order kinetic characteristics. The molecular weights and chemical structures of the copolymers and their precursors were confirmed by gel permeation chromatography (GPC) and 1 H NMR. The critical micelle concentration (CMC) values of the star copolymers in aqueous solution were extremely low (2.2–4.0 mg/L), depending on the architecture of the copolymers. The pH-responsive self-assembly behavior of the star copolymers in aqueous solution was investigated by a combination of dynamic light scattering (DLS), UV-vis spectrometry and scanning electron microscopy (SEM). When the pH values decreased from 10 to 3, no obvious fluctuation of the visible light transmittance of the micelle solutions was observed for lower polymer concentrations of 0.1 and 1 mg/mL, while sharp increase occurred at higher concentration of 10 mg/mL. The hydrodynamic diameters ( D h ) of the micelle solutions appeared slight increase with the increase of concentration, and increased rapidly as the pH decreased from 10 to 4 followed by a slight decrease at pH 3. The effects of pH value on the zeta potentials exhibited almost the same tendency with the D h . This may due to the fact that the protonation of tertiary amine groups in DEAEMA can induce the swelling of micelles. The PCL and PDEAEMA contents and the topological structures (4- or 6-arm) showed significant influences on the pH-sensitivity of the micelles. Overall, the results demonstrated that the structures and pH-sensitivity of these amphiphilic copolymers could be well-controlled and their self-assembled micelles are promising carriers for delivery of anticancer hydrophobic drugs.
Supramolecular amphiphilic multiarm hyperbranched copolymer: Synthesis, self-assembly and drug delivery applications

Abstract:Novel supramolecular amphiphilic multiarm hyperbranched copolymers were successfully constructed through the molecular recognition of nucleobases. First, adenine-terminated H40-star-poly(epsilon-caprolactone)-adenine (H40-star-PCL-A) and uracil-terminated poly(ethylene glycol) (PEG-U) were successfully prepared. Due to the molecular recognition between A and U moieties, supramolecular multiarm hyperbranched copolymers were obtained by simply mixing the hydrophobic H40-star-PCL-A core and hydrophilic PEG-U shell. They not only had similar properties to conventional covalent-linked multiarm hyperbranched copolymers, but also possessed a dynamic and tunable nature. These supramolecular hyperbranched copolymers were found to self-assemble into pH-responsive micelles with low critical micelle concentration (CMC) because of non-covalent connection and hyperbranched architecture. The size of the self-assembled micelles could be easily tailored by changing the ratio of hydrophobic H40-star-PCL-A core and hydrophilic PEG-U arm. Moreover, encapsulation and controlled drug release were demonstrated with the chemotherapeutic drug doxorubicin (DOX). These supramolecular hyperbranched copolymer systems represent an evolution over conventional stimuli-responsive covalent-bonded hyperbranched copolymer systems and display a significant reduction in the viability of HeLa cells upon triggered release of DOX from the supramolecular micelles.
Acid-Cleavable Unimolecular Micelles from Amphiphilic Star Copolymers for Triggered Release of Anticancer Drugs

Abstract:In this contribution, amphiphilic star copolymers (H40-star-PCL-a-PEG) with an H40 hyperbranched polyester core and poly(ε-caprolactone)-a-poly(ethylene glycol) copolymer arms linked with acetal groups are synthesized using ring-opening polymerization and a copper (I)-catalyzed alkyne-azide cycloaddition click reaction. The acid-cleavable acetal groups between the hydrophilic and hydrophobic segments of the arms endow the amphiphilic star copolymers with pH responsiveness. In aqueous solution, unimolecular micelles can be formed with good stability and a unique acid degradability, as is desirable for anticancer drug carriers. For the model drug of doxorubicin, the in vitro release behavior, intracellular release, and inhibition of proliferation of HeLa cells show that the acid-cleavable unimolecular micelles with anticancer activity can be dissociated in an acidic environment and efficiently internalized by HeLa cells. Due to the acid-cleavable and biodegradable nature, unimolecular micelles from amphiphilic star copolymers are promising for applications in intracellular drug delivery for cancer chemotherapy.

Abstract:Amphiphilic triblock copolymer of poly(ethylene glycol)-block-poly(dimethylaminoethyl methacrylate)-block-poly(ε-caprolatone) (PEG-PDMA-PCL) was synthesized using a one-pot sequential oxyanionic polymerization of DMA and ε-CL, associated with a PEG-O−K+ macroinitiator. The pH-responsive micellization behavior of the copolymer was studied using dynamic light scattering (DLS), steady–state fluorescence and TEM techniques. The anti-cancer drug of doxorubicin (DOX) was chosen as a model drug to investigate the potential application of this triblock copolymer in drug controlled release. The results indicated the important roles of the PCL block for drug loading, the PDMA block for pH-responsive release, and PEG block for good bio-affinity. Cell cytotoxicity tests showed that the DOX-loaded PEG-PDMA-PCL micelles were pharmaceutically active to suppress the growth of SKOV-3 cells. This novel stimuli–responsive block copolymer is an attractive candidate as the “smart” pH-responsive carrier for intracellular delivery of hydrophobic drugs.
Acid and Reduction Dually Cleavable Amphiphilic Comb-Like Copolymer Micelles for Controlled Drug Delivery

Abstract:Disulfide- and acetal-functionalized comb-like copolymers (SACP) with poly(epsilon-caprolactone) (PCL) and poly(ethylene glycol) (PEG) side chains and a polymethacrylate backbone were synthesized and self-assembled for in vitro encapsulation and release of an anticancer drug, doxorubicin (DOX). Three step reactions including RAFT copolymerization of 2-hydroxyethyl methacrylate and poly(ethylene glycol) methyl ether methacrylate mediated by a disulfide-functionalized RAFT agent, hydroxyl-vinyloxy addition reaction to introduce acid cleavable PEG segments, and CL polymerization to grow PCL chains were used to generate the target dual-sensitive comb-like copolymers. Meanwhile, well-defined normal comb-like polymers (CP), acid-cleavable comb-like polymers (ACP) and disulfide-linked comb-like polymers (SCP) with PEG and PCL pendent chains were also prepared and acted as analogues of SACP. All the resultant comb-like copolymers had the desired chemical structures, well-controlled molecular weight and relatively low polydispersity (PDI = 1.09-1.16), evident from H-1 NMR and GPC-MALLS analyses. DLS and TEM measurements revealed that these copolymers were liable to self-assemble into micelles or large compound micelles in aqueous solution, and SACP aggregates could be dissociated and reaggregated in the presence of acidic PBS solution (pH 5.0) or 10 mM DL-dithiothreitol (DTT). Compared with CP aggregates with a lack of cleavable linkage, ACP, SCP and SACP aggregates upon acid and reduction stimuli could quickly release the encapsulated DOX, and the cumulative release from SACP aggregates could be roughly adjusted by addition of a single stimulus or dual stimuli. These results confirmed their great potential as novel nanocarriers and delivery systems.
Fabrication of Supramolecular Star-Shaped Amphiphilic Copolymers for ROS-triggered Drug Release.

Abstract:Star-shaped copolymers with branched structures can form unimolecular micelles with better stability than the micelles self-assembled from conventional linear copolymers. However, the synthesis of star-shaped copolymers with precisely controlled degree of branching (DB) suffers from complicated sequential polymerizations and multi-step purification procedures, as well as repeated optimizations of polymer compositions. The use of a supramolecular host-guest pair as the block junction would significantly simplify the preparation. Moreover, the star-shaped copolymer-based unimolecular micelle provides an elegant solution to the tradeoff between extracellular stability and intracellular high therapeutic efficacy if the association/dissociation of the supramolecular host-guest joint can be triggered by the biologically relevant stimuli. For this purpose, in this study, a panel of supramolecular star-shaped amphiphilic block copolymers with 9, 12, and 18 arms were designed and fabricated by host-guest complexations between the ring-opening polymerization (ROP)-synthesized star-shaped poly(ε-caprolactone) (PCL) with 3, 4, and 6 arms end-capped with ferrocene (Fc) (PCL-Fc) and the atom transfer radical polymerization (ATRP)-produced 3-arm poly(oligo ethylene glycol) methacrylates (POEGMA) with different degrees of polymerization (DPs) of 24, 30, 47 initiated by β-cyclodextrin (β-CD) (3Br-β-CD-POEGMA). The effect of DB and polymer composition on the self-assembled properties of the five star-shaped copolymers was investigated by dynamic light scattering (DLS), transmission electron microscopy (TEM), and fluorescence spectrometery. Interestingly, the micelles self-assembled from 12-arm star-shaped copolymers exhibited greater stability than the 9- and 18-arm formulations. The potential of the resulting supramolecular star-shaped amphiphilic copolymers as drug carriers was evaluated by an in vitro drug release study, which confirmed the ROS-triggered accelerated drug release from the doxorubicin (DOX)-loaded supramolecular star-shaped micelles due to the oxidation-induced dissociation of β-CD/Fc pair and the consequent loss of the colloidal stability of the star-shaped micelles. Studies of the delivery efficacy by an in vitro cytotoxicity study further indicated that higher DBs and longer hydrophilic arm compromised the therapeutic efficacy of the DOX-loaded supramolecular star-shaped micelles, resulting in significantly reduced cytotoxicity, as measured by increased IC50 value. Overall, our results revealed that the screening of hydrophilic block by DB and MW for an optimized star-shaped copolymer should balance the stability versus therapeutic efficacy tradeoff for a comprehensive consideration. Therefore, the 12-arm star-shaped copolymer with POEGMA30 is the best formulation tested.

Abstract:A novel pH-responsive and biodegradable triblock copolymer, poly(ethylene glycol)-poly(L-glutamic acid)-poly(L-Alanine acid) (PEG-PLGA-PLAA), was prepared by N-carboxyl anhydride ring-opening polymerization and acidic hydrolysis. It was found that this amphiphilic copolymer can self-assemble into micelles in aqueous solution. The PLAA, PLGA and mPEG segments serve as a hydrophobic core, a pH-responsive shell and a hydrophilic corona, respectively. The morphology and properties of the micelles such as pH sensitivity,zeta potential, secondary structure and mean diameter were investigated. Doxorubicin (DOX) was encapsulated into the micelles to explore the release profiles, which exhibited in vitro pH-dependence. In acidic environment,with the formation of a-helix secondary structure of poly(L-glutamic acid) segments, the PLGA segments in the triblock copolymers shrank and relocated in the micelles to trigger the release of drugs. As a result, these DOX-loaded polymeric micelles showed rapid release of DOX from the micelles in weakly acidic environments (pH = 5.5) but very slow release under physiological condition (pH = 7.4).
Biocompatible and Ph‐responsive Triblock Copolymer Mpeg‐b‐pcl‐b‐pdmaema: Synthesis, Self‐assembly, and Application

Abstract:A series of well-defined amphiphilic triblock copolymers [polyethylene glycol monomethyl ether]-block-poly(epsilon-caprolactone)-block-poly[2-(dimethylamino)ethyl methacrylate] (mPEG-b-PCL-b-PDMAEMA or abbreviated as mPEG-b-PCL-b-PDMA) were prepared by a combination of ring-opening polymerization and atom transfer radical polymerization. The chemical structures and compositions of these copolymers have been characterized by Fourier transform infrared spectroscopy, H-1 NMR, and thermogravimetric analysis. The molecular weights of the triblock copolymers were obtained by calculating from H-1 NMR spectra and gel permeation chromatography measurements. Subsequently, the self-assembly behavior of these copolymers was investigated by fluorescence probe method and transmission electron microscopy, which indicated that these amphiphilic triblock copolymers possess distinct pH-dependent critical aggregation concentrations and can self-assemble into micelles or vesicles in PBS buffer solution, depending on the length of PDMA in the copolymer. Agarose gel retardation assays demonstrated that these cationic nanoparticles can effectively condense plasmid DNA. Cell toxicity tests indicated that these triblock copolymers displayed lower cytotoxicity than that of branched polyethylenimine with molecular weight of 25 kDa. In addition, in vitro release of Naproxen from these nanoparticles in pH buffer solutions was conducted, demonstrating that higher PCL content would result in the higher drug loading content and lower release rate.
