Abstract:Total 12 maize inbreds were used as the explants,by using the isolated culture method,the effects of different plant growth regulators and genotypes on immature embryo regeneration were studied.The results showed that callus induction rate was 90.5% when the concentration of 2,4-D was 3 mg.L-1.There existed great differences among maize genotypes.Zheng58 showed the highest frequency of callus induction which was 96.3%.The minimum frequency of callus induction was observ-ed in the inbred line Dong46 which belonged to the local germplasm.Genotype was an important factor affecting the callus status,which would further affect the regeneration of callus and plantlets.BA 1.0 mg.L-1+IBA 0.5 mg.L-1 was more suitable for callus differentiation.

Abstract:The immature embryo of five maize lines (Zea mays.L) was used to study the influencing of genotypes, 2,4-D concentration and embryo-age on callus induction, influencing of 6-BA on callus differentiation ,and influencing of IBA on buds rooting. The results showed that, callis were obtained from Q31×Z31,Q31×Z3,B73,Z3 except Q31.In addition, there were dominant differences among the induction percentage of four genotypes on the same conditions.The concentration of 2,4-D in mediums and embryo-age had evident effects on the callus induction percentage of immature embryo. Adding 2.0mg/L 2,4-D in N6 medium and the embryo-age with 11d to 13d were optimal for improving the percentage of inducing callus. The 6-BA promoted callus differentiation and IBA promoted the buds rooting.

Abstract:After pollination 10—12 days,immature embryos from 19 maize inbreds(belonging to 5 maize heterotic groups)were used to determine the influence of genotypes and media on frequency of callus induction so as to study the culture characteristic of callus. Our research showed that callus could be induced from all the tested inbreds, but the quality of callus was quite different. And plant regeneration was obtained from 11 genotypes' callus;8 inbred lines,such as B73,Huang C,478,Zong 31,178,501 and so on,belonging to 3 elite inbreds groups of Reid,Luda Red cob and compound germplasm, were identified with higher performance both in induction of embryonic callus and plant regeneration. Furthermore,supplements of callus induction medium under N6 basic medium,such as concentration of 2,4-D,proline and casein hy-drolysate(CH) were used to study their effects on the induction frequency,embryogeny and quality of callus, the inhibition of embryo spontaneous shooting. In conclusion, N6 basic medium with the supplements of 4 mg/L 2,4-D,but without proline and CH,was investigated to be suitable for the callus induction of immature embryo from maize inbreds with the characteristics of little spontaneous shooting,and high regeneration ability.

Abstract:The young embryo of A188 inbred were cultured on N6 medium to induce callus. When 2,4-D concentration was 2mg/L, the frequency of callus induction reached to 100%. The high frequency (over 80%) was obtained on N6 medium plus 5mg/L 6-BA. The callus of A188 young embryo were highly sensitive to hygromycin. Callus growth could be inhibited completely by 10mg/L hygromycin. The type Ⅱcallus was used as target tissue for transformed with agrobacterium tumefaciens. After infection and coculture, transient expression assay of GUS suggested that 3.5% callus had blue blots.

Abstract:Callus was induced from immature embryos of 9 maize inbred lines on MB medium and N6 medium.The effects of different genotypes,different mediums,explants sizes and different concentrations of phytohormone on the callus induction of immature embryos were studied.Differences in callus growth state from different genotypes were also analyzed.Regeneration system was successfully established via the callus pathway.The results showed that genotypes have significant effects on callus induction.Both genotype and medium affected the frequency of embryogenic callus formation.The choice size was 1.5-2.0 mm young embryos as explants to be advantageous to obtain perfect callus.2,4-dichlorophenoxyacetic acid(2,4-D)was necessary in callus induction,and the optimal concentration for embryogenic callus formation was 2.0-2.5 mg/L.

Abstract:Objective:To induce callus from immature embryos of maize inbred lines Dan598 for establishment of regeneration system.Methods:The effects of embryo age,media type,concentration of 2,4-D on the callus induction were studied.Results:It was indicated that 16～18 days after post-pollination,2,4-D concentration at 2.0 mg/L are the best condition to induct embryogenic callus;set 5 kinds of culture medium such as N6,improved NB,NB,MS,MB,to observe the effects of callus induction,select the culture medium of induction with improved NB medium is better than other mediums.Differentiation medium suppled with 1 mg/L KT,0.5 mg/L 6-BA and 0.5 mg/L NAA can promote the shoot differentiation and root growth.Conclusion:An efficient plant regeneration system has been established for maize inbred lines Dan598,which provides basis for further genetic transformation.

Abstract:Several maize inbred lines were studied for the induction percentage of immature embryogeneic callus in this experiment.Different geneotypes and different induction mediums were evaluated.The results showed that there are significant differences among the different induction percentage of different maize inbred lines on the same conditions and the same maize inbred lines at different mediums.As for this experiment,N6A medium and 18-599 inbreed line have a better induction percentage of immature embryogeneic callus.This indicated there is an interaction on induction percentage of immature embryogeneic callus between genotype and medium.The content of 2,4-D in the induction mediums have an evident affect on the callus induction percentage of immature embryo.The accession of NAA to the induction mediums can not increase the induction percentage of embryogenic callus evidently.

Abstract:Inbred lines Huangzao4 and Mo17 were used to develop a population consisted of 239 Recombinant inbred lines(RILs).Based on the F_8 generation population,a SSR linkage map was constructed with 101 SSR loci,covering maize genome 1 422.7 cM with an average interval length of 15.6 cM.The 239 RILs and their parental lines were evaluated for tissue cultural traits,including calli initiation frequency,type Ⅱ calli initiation frequency,green buds initiation frequency and plant regeneration frequency.With the method of composite interval mapping,8 QTLs were identified and located on chromosome 2,3,5,6,8 and 9.Five QTLs of regeneration frequency were identified on chromosome 2,3,5,6 and 9,which can explained 6.29%,5.19%,7.88%,14.02% and 10.4% of the phenotypic variance,respectively.One QTL on chromosome 5,linked with markers umc1966 was identified with minor effects conferring type Ⅱ calli initiation frequency,which only explained 6.60% of the phenotypic variance.Two QTLs on chromosome 8,linked with markers bnlg2235 and bnlg240,were identified controlling calli initiation frequency and green buds initiation frequency,which explained 9.51% and 4.78% of the phenotypic variance,respectively.Two QTLs controlling type Ⅱ callus rate and plant regeneration rate were located in the neighboring region of chromsome 8,and these two traits were correlated between each other.However,further study is needed to make clear if the correlation is due to one pleiotropic gene or the linkage of two genes.

Abstract:Regenerable, embryogenic haploid cell suspensions were initiated and established from type II pollen calluses of two selected Chinese maize genotypes (No 592 Y and 592.A2 LY). The induction frequency of friable, embryogenic callus (type II) was highly dependent on three factors: genotype, medium, cold pretreatment, and on their interactions. Repeated callus and cell selection during the culture procedure led to stable haploid suspensions consisting of fine clusters each containing 20–50 cells. The selected cell lines were able to maintain their morphogenic ability during long-term subculture (2 years). Protoplasts were successfully isolated from subcultured, friable, embryogenic pollen calluses and cultured on N6BM and N6K media using a feeder layer, obtained from 2-day-old suspension culture. Healthy plants were regenerated from protoplast-derived calluses.
Agrobacterium tumefaciens-mediated Transformation of Maize Embryonic Calli

Abstract:Three types of Agrobacterium strains C58,LBA4404 and EHA105 containing standard binary vector pCAMBIA1301 with GUS as reporter gene were used to inoculate maize embryonic calli of two elite inbred lines Qi 319 and 18(red),respectively.The results indicated that the transformation efficiencies including efficiencies of GUS transient expression(F=24.92**)and regenerated resistant calli(F=19.43**)were significant different between six combinations of bacteria strains and maize inbred lines.EHA105-Qi 319 was the optimized combination for transformation,and average GUS transient expression efficiency was 55.5%,and some up to 71.1%.The efficiency of average regenerated resistant calli was 14.4%,and some up to 20%.PCR analysis was conducted with 22 regenerated resistant T0 generation plants and the efficiency of positive plants was 50%.Histochemical GUS assay was conducted with those resistant plants.The results indicated that intense blue staining was observed in the leaves of all PCR positive plants as expected,and no GUS expression was observed in the leaves of PCR negative and wild type maize plants.The results proved that foreign GUS gene was expressed stably in the T0 generation maize plants and also confirmed the consistency of PCR analysis with histochemical GUS assay.
Selection of Maize Inbred Lines with High Regeneration and Susceptibility to Agrobacterium Tumifacien

Abstract:Ten-maize inbred lines of maize (Zea mays L.) with high-induction rate and proliferation ability of embryonic calli were selected from 70-maize inbred lines by immature embryo culturing. Some of the embryonic calli were transferred onto regeneration medium to examine the ability of regeneration, some were transformed via Agrobacterium tumifaciens C58 carrying intron-β-glucuronidase (gus) gene, and GV3301 carrying the green fluorescent protein (gfp) gene to study the susceptibility of different genotypes in maize to A. tumifaciens. All embryonic calli initiated from 10-maize inbred lines were able to regenerate into plantlets, and the regeneration frequencies of inbred lines 6010, 6038, 6015, 6051, and 6060 were 61.11%, 31.94%, 45%, 33.33%, and 56.94%, respectively, which were higher than that of other lines. Analysis of variance indicated that the susceptibility of the various genotypes in maize to A. tumifacien C58 showed a significant difference among each other, and the inbred lines 6010, 6015, 6051, 6050, 6058, 6060, 6069, 6077 were susceptible to A. tumifacien C58, of which frequency of gus expression were over 70%. Expression of GFP was observed in six-inbred lines (6050, 6015, 6051, 6058, 6069, 6077). The inbred lines 6051, 6010, 6015, 6060, and 6050 had the high regeneration and the susceptibility to A. tumifaciens C58; and the inbred lines 6051, 6015, and 6060 had the high regeneration and the susceptibility to Agrobacterium tumifaciens GV3301.

Abstract:The regenerative capacity of the embryonic callus, a complex quantitative trait, is one of the main limiting factors for maize transformation. This trait was decomposed into five traits, namely, green callus rate (GCR), callus differentiating rate (CDR), callus plantlet number (CPN), callus rooting rate (CRR), and callus browning rate (CBR). To dissect the genetic foundation of maize transformation, in this study multi-locus genome-wide association studies (GWAS) for the five traits were performed in a population of 144 inbred lines genotyped with 43,427 SNPs. Using the phenotypic values in three environments and best linear unbiased prediction (BLUP) values, as a result, a total of 127, 56, 160, and 130 significant quantitative trait nucleotides (QTNs) were identified by mrMLM, FASTmrEMMA, ISIS EM-BLASSO, and pLARmEB, respectively. Of these QTNs, 63 QTNs were commonly detected, including 15 across multiple environments and 58 across multiple methods. Allele distribution analysis showed that the proportion of superior alleles for 36 QTNs was <50% in 31 elite inbred lines. Meanwhile, these superior alleles had obviously additive effect on the regenerative capacity. This indicates that the regenerative capacity-related traits can be improved by proper integration of the superior alleles using marker-assisted selection. Moreover, a total of 40 candidate genes were found based on these common QTNs. Some annotated genes were previously reported to relate with auxin transport, cell fate, seed germination, or embryo development, especially, GRMZM2G108933 (WOX2) was found to promote maize transgenic embryonic callus regeneration. These identified candidate genes will contribute to a further understanding of the genetic foundation of maize embryonic callus regeneration.

Abstract:The leaf base of maize was used as explants,inducing a lot of callus,and the influencing factors of induction and regeneration were investigated.The results showed that the frequency of callus induction existed significant differences among maize genotypes.B73 showed the highest frequency of callus induction which was 74.30%;the best responses were obtained from the leaf base of 7d old seedlings;when the concentration of 2,4-D was 2.2mg/L,it facilitates the formation of embryogenic callus;the level of 6-BA 1mg/L in differentiation medium resulted in the highest regeneration rate of Tie7922 and Nongda 178,and the level of 6-BA 2mg/L in differentiation medium resulted in the highest regeneration rate of B73 and Chang 7-2.Establishment of high efficient regeneration system of maize lay a foundation of further study on maize genetic transformation.

Abstract:The initiation,maintenance and differentiation of callus from seedling-derived leaves of maize(Zea mays L.) inbreds were studied and optimized.The results revealed that medium CIM1 was the best medium in the experiment.Callus obtained could achieve higher differentiation rates after modified treatment.All showed that,selecting appropriate induction medium and appropriately modified callus could improve callus regeneration rate,which benifits to the establishment of an efficient tissue-culture regeneration system for maize.

Abstract:We carried out a study of regeneration capacities of embryonic callus from maize immature embryo culture with 144 different inbred lines of natural groups from different countries,and found that the regeneration capacity was affected by three factors:environment,genotype and the interaction between the environment and genotype.We found that green embryonic callus rate (GCR),embryonic callus differentiating rate (CDR) and the plantlet number of embryonic callus regeneration (CPN) have significant positive correlations with each other,and they all have significant negative correlations with embryonic callus browning rate (CBR).Moreover,embryonic callus cloning index for the first subculture (CCI1) and embryonic callus cloning index for the second subculture (CCI2) have a significant positive correlation with each other,and CCI2 is positively correlated with green GCR,and is negatively correlated with CBR.Embryonic callus rooting rate (CRR) is positively correlated with GCR,CDR and CPN to some degree.Furthermore,we calculated Broad-Sense Heritability of each trait,and uncovered that the heritability index of CCI1,CCI2 and CRR was lower,and the heritability index of others was higher.In addition,by using the Ward method for two-way cluster analysis,we found eleven inbred lines with high regenerating abilities,and the rooting situation of regenerating plantlet was excellent by rooting culture,which could be used as the elite inbred lines of the maize transgenic receptor.

Abstract:bar gene was introduced into 3 maize inbred lines by Agrobacterium tumefacien mediated.The results showed that 2 mg·L-1 2,4-D was good to embryogenic callus.The suitable duration of infection was 20 min.The addition of AS(200 μmol·L-1) in infection and co-culture medium enhanced the instant expression frequency of immature embryo.The herbicide concentration should be lower than lethal concentration,8 mg·L-1 was better.Moderate selection fitted for transformation,the best screening mode was to use hygromycin in the concentration of 8,6 and 2 mg·L-1 in induction medium,buds elongation medium and rooting medium respectively.Seven plants showed positive reaction by PCR analysis and were confirmed to resist basta.

Abstract:In this report,four genotypes of maize(Zea mays L.)embryonic calli were transformed via Agrobacterium tumefaciens.The expression level of β-glucuronidase(Gus) gene was used as the evaluation index of influencing transformation efficiency.The results showed that there was dominant difference among different maize genotypes.The hybrid line Q31×Z31、Q31×Z3 and line B73 were more sensitive than line Z3.Improved transformation efficiencies were obtained when the embryonic calli were incubated with Agrobacterium suspension cells(OD600=0.4~0.6) for 10 min in the infection medium.Optimized co-culture condition was that infection was performed at 23℃ in dark for 3 days.The transgenic maize plants with basta-resistant were obtained via Agrobacterium-mediated transformation.

Abstract:Some influencing factors of induction and subculture by using maize inbred lines H99,Dan 988 and iron 7922’s immature embryo callus were investigated.The results showed that when the concentration of 2,4-D was 2mg/L,the highest rates of embryogenic-callus was obtained.Besides the optimal medium,embryo age and storing time at 4℃ were different among different genotypes.The addition of KT,Ca2 + and AgNO3 significantly stimulated embryogenic-callus of H99 and Dan 988,but the embryogenic-callus formation for iron 7922 was lower.The addi-tion of Ca2 + had little effect on embryogenic-callus of Dan 598.

Abstract:Genetic transformation has been an effective technology for improving the agronomic traits of maize. However, it is highly reliant on the use of embryonic callus (EC) and shows a serious genotype dependence. In this study, we performed genomic sequencing for 80 core maize germplasms and constructed a high-density genomic variation map using our newly developed pipeline (MQ2Gpipe). Based on the induction rate of EC (REC), these inbred lines were categorized into three subpopulations. The low-REC germplasms displayed more abundant genetic diversity than the high-REC germplasms. By integrating a genome-wide selective signature screen and region-based association analysis, we revealed 95.23 Mb of selective regions and 43 REC-associated variants. These variants had phenotypic variance explained values ranging between 21.46 and 49.46%. In total, 103 candidate genes were identified within the linkage disequilibrium regions of these REC-associated loci. These genes mainly participate in regulation of the cell cycle, regulation of cytokinesis, and other functions, among which MYB15 and EMB2745 were located within the previously reported QTL for EC induction. Numerous leaf area-associated variants with large effects were closely linked to several REC-related loci, implying a potential synergistic selection of REC and leaf size during modern maize breeding.
